상용 한약재의 부작용에 관한 연구 (A Study on the Side Effect of Crude Drugs)

  • As many crude drugs are used in the oriental medical field problems on the side effects of these drugs come to the front. To conduct delayed-type hypersensitivity we selected 29 kinds of drugs used frequently for therapeutic agents in oriental medical hospitals (Table I). The cell-mediated immune response was evaluated by measuring the foot pad swelling reaction and humoral immune response by measuring the antibody formation to these crude drugs. Mice were given these drugs intraperioneally for sensitization and challenged with same drug as used for sensitization respectively by intral dermal injection on the left and righ hind foot pad 4 days after senstization and then the foot pads were measured with the dial micrometer. The results were as follow; 1) Gentianae Scabrae Radix, Arecae Semen, Corydalis Tuber, and Paeoniae Radix were significant as delayed-type hypersensitivity inducers. 2) None of the crude drugs tested had effect on the induction of humoral immune response.

  • An experiment was carried out to measure the cellular immune response in the animals by sensitizing the animals with 2, 4-dinitrochlorobenzene(DNCB). The fowls could be sensitized with primary application of DNCB. The sensitizing and challenge dose was standardized. Heterophils were prominent in the early inflammatory response in the superficial and deep dermal regions with scattering of eosinophils. The histological response in the fowls was characteristic of a delayed hypersensitivity reaction.

염화아연의 세포성 면역독성에 미치는 황기 추출물의 효과 (Effects of Astragali Radix extract on the Cell Mediated Immunotoxicity of Zinc Chloride)

  • Effects of Astragali Radix extract (AG) on the cell mediated-and nonlpecific immunotoxic responses of zinc chloride (Zn) were studied usign ICR mice. Mice were divided into 4 groups (10 mice/group), and Zn was given to the mice 1 hr after i.p. injection with 0.5g/kg of AG by i.p. injection daily for 10 days at a dose of 25 mg/kg. Immune responses on the responses on the relative weight of thymus, delayed-type hypersensitivity to SRBC (DTH), phagocytic activity and circulating leukocytes were evaluated. Zn treatment decreased body weight gain, the relative weight of thymus, DTH and circulating leukocytes compared with those in controls. AG treatment increased DTH, phagocytic activity and circulating leukocytes compared with those in controls. Combination of AG and Zn increased DTH and circulating leukocytes compared with those in controls, but decreased body weight gain and the relative weight of thymus. These findings indicated that AG decreased immunotoxicity of Zn on the DTH and circulating leukocytes.

  • The immunopotentiating effect of ethanol extract, butanol fraction and petroleum ether extract of Panax ginseng on the immunotoxicity of benzo(a)pyrene were investigated in mice. A single administration of benzo(a)pyrene induced an apparent but relatively transient reduction in HY titer, Arthus reaction, delayed type hypersensitivity, rosette forming cell and natural killer cell activity Ethanol extract very significantly restored HY titer, Arthus reaction. RFC and natural killer cell activity. Butanol fraction have no effect. But petroleum ether extract very significantly restored humoral and cellular immune response and especially natural killer cell activity.

  • The effects of butylated hydroxyanisole and butylated hydroxytoluene on the immune status in normal male were evaluated. They exhibited significant decrease in the circulating leukocyte counts. Relative spleen and thymus weights were slightly decreased, but not stratistically significant. These were, however, significant liver hypertrophies in theier exposed mice. Splenic IgM PFCs per one million cells in 1/20 LD50 BHA and BHT exposed mice were significantly reduced IgM PFCs per spleen were similar tothose of control, except in 1/20 LD50 BHA exposed mice, where they were significantly suppressed. The precise nature of the inhibition is not clear. Direct cytotoxicity is not responsible for the depressed antibody response, even following relatively high doses of them, because the changes in spleen cellularity are not significant. Both substances, however, did not show any effects on the arthus reaction and delayed hypersensitivity reaction induced by heat aggreagted bovine serum albumin, and in vivo phagocytosis of colloidal carbon. In the light of the present results, in vivo antibody response as well as in vitro, may be sensitive to BHA of the present results, in vivo antibody response as well as in vitro, amy be sensitie to BHA and BHT. Further elucidation of the precise nature of antibody suppression in their exposed mice, is warranted.

베타-카로틴의 면역생물학적 연구 (Immunobiological Studies on Beta-Carotene)

  • Effects of beta-carotene on the immunobiological responses were studied in ICR mice. ICR male mice were divided into 8 groups (10 mice/group), and beta-carotene at doses of 4, 20 and 100 mg/kg were orally administered to ICR mice once daily for 28 consecutive days. Cyclophosphamide (CY) was injected intraperitoneally (i.p.) to ICR mice with a single dose of 5 mg/kg body weight at 2 days before secondary immunization. Mice were sensitized and challenged with sheep red blood cells (5-RBC). Immune responses were evaluated by humoral immunity, cellular immunity and non-specific immunity. The results of this study were summarized as follows: (1) Beta-carotene significantly increased the weight ratios of liver, spleen and thymus to body weight depending on dose, and significantly increased the increasing rate of body weight and the number of circulating leukocyte. (2) Beta-carotene dose-dependently increased hemagglutination titer, Arthus reaction and hemolytic plaque forming cell related to humoral immunity. (3) Beta-carotene significantly increased delayed-type hypersensitivity reaction and rosette forming cell related to cellular immunity. (4) Beta-carotene dose-dependently increased phagocytic activity, and significantly increased natural killer (NK) cell activity. (5) Beta-carotene dose-dependently inhibited reductions in humoral immunity, cellular immunity, NK cell activity and phagocytic activity by treatment with CY.

염화아연이 생쥐의 면역반응에 미치는 영향 (Effects of Zinc Chloride on the Immune Response in ICR Mice)

  • Effects of Zinc chloride on the immune responses were studied in ICR mice. ICR male mice were divided into 5 groups(10 mice/group) and Zinc chloride at doses of 0.3, 1.2, 4.8 and 19.2 mg/kg were orally administered to ICR male mice once a day for three weeks. Mice were sensitized and challenged with sheep red blood cells(S-RBC). The results of this study were summarized as follows; (1) Zinc chloride significantly increased the body weight rate, the weight ratios of spleen and thymus to body weight and the number of circulating leukocyte, but significantly decreased them at the high dose of it, and increased dose-dependently the weight ratio of liver to body weight. (2) Zinc chloride significantly increased hemagglutination titer, Arthus reaction and plaque forming cell related to humoral immunity, but significantly decreased them at the high dose of it. (3) Zinc chloride significantly increased delayed-type hypersensitivity reaction and rosette forming cell related to cellular immunity, but significantly decreased them at the high dose of it. (4) Zinc choride significantly enhanced phagocytic activity, but significantly decreased according to the increase of its dose. These results suggest that high dose of zinc chloride decreased humoral, cellular and non-specific immune responses.

가미보중익기탕이 알레르기반응에 미치는 영향에 대한 실험적 연구 (Experimental Study on the Anti-allergic Effects of Gamibojungikgi-tang)

  • This experimental research has been done to study the effects of Gmibojungikgi-tang(GBJIKT) on the anti-allergic reaction. We found the several important results from the research which has been performed by two experiments toward immediately type and delayed type in order to study the effects of GBJIKT on hypersensitivity response to mice. The results obtained from our research are as following: The survival rate of one group to which we injected only the compound 48/80 is almost 0% according to its density and timing test. In the other hand, the survival rates of the other group to which we injected both of the compound 48/80 and GBJIKT are 10%, 10%, 20%, 30%, 20%, and 40% according to 0.025, 0.05, 0.1, 0.25 0.5 and 1(mg/g) of compound 48/80. Time dependency test also shows the 0% survival rates in 5 and 10 minutes. GBJIKT revealed the significantly inhibitory effect on Compound 48/80 induced Mast cell degranulation. GBJIKT showed the significantly inhibitory effect in the delayed type hypersensitivity response to picry1 chloride GBJIKT showed the significantly inhibitory effect in the delayed type hypersensitivity response to sheep red blood cell. Our research provides the important evidence that GBJIKT is benificial to the prevention and treatment of allergy related diseases.

십전대보탕이 알레르기반응에 미치는 영향 (The Experimental Study on the Anti-allergic Effects of Sipjeondaebo-tang)

  • This experimental research has been done to study the effects of Sipjeondaebo-tang(SOT) on the anti-allergic reaction.
