띠미로 버섯 추출물의 항암효과

  • This study was carried out particutarly focusing on the antitumor effects of extracts obtained from mushroom, Daedalea dickinsii against Hela cell, Hep $G_{2}$, cell, L 929 cell and Sarcoma-180. Antitumor effect of hot water extracts obtained from Daedalea dickinsii against Hela cell showed at the highest level of 70% when adiministrated at the concentration of 1.5 mg/100 ml, however Hep $G_{2}$, cell inhibited 40% at the same concentration of hot water extracts. Antitumor effects of methanol extract of Daedalea dickinsii against Hela cell indicated at the highest level of 60% when 4 mg/ml was administerated. Antitumor effect of hot water extract of Daedalea dickinsii inhibited 51.03% against L929 cell. The solid tumor sarcoma-180 growth inhibition of methanol extract of Deadalea dickinsii inhibited 45.67% when administrated at the concentration of 60mg/kg, and the life prolongation was higher 30.88% than that control group.

Antioxidant Activities of Extracts from Fruiting Bodies of Mushrooms (버섯추출물의 항산화활성에 관한 연구)

  • Antioxidant activities of 80% ethanol extracts of 63 species of mushroom fruiting bodies were investigated. The ethanol extracts from Daedalea dickinsii, Armillariella mellea, and Fomitella fraxinea showed markedly inhibition on lipid peroxidation in rat liver microsome. The extracts from Daedaleopsis tricolor, Trametes suaveolens, Armillariella mellea, Trichaptium abietinum, Daedalea dickinsii, Fomitella fraxinea, Tylophilus neofelleus, Boletellus obscurecoccineus, and Xerocomus subtomentosus significantly inhibited the hepatic aldehyde oxidase activity, and the extracts from Daedaleopsis tricolor, Armillariella mellea, Daedalea dickinsii, and Fomitella fraxinea slightly stimulated the hepatic SOD activity. These results suggest that Daedalea dickinsii, Armillariella mellea, and Fomitella fraxinea contain the bioactive substances for natural antioxidant and may be useful for development of antioxidant from mushrooms.

  • Fruit bodies of the wood rotting basidiomycetes naturally grown in middle part of Korea were collected from August 1989 to May 1992 and identified to 24 genera and 31 species. Species diversity was differed little from areas, but index of species diversity was higher in Mt. Songni and Mt. Duckyu than that in other areas. The dominant species were Coriolus versicolor, Daedalea dickinsii, Daedaleopsis styracina, Lenzites betulina and Schizophyllum commune.

  • 31-Hydroxycarbowyacetylquercinic acid, a lanostenoid hydroxyl acid has been isolated from D. dickinsii by solbent extraction, silica gel column chromatography and recrystallization. The structure of this compound has been determined to be 31-hydroxycarboxyacetylquercinic acid by a combinationof spectral data and by HM-BC. This compound showed antimicrobial activities against human pathogenic fungi and bacteria.

  • In order to develope bioactive substances, dried fmit body of Daedalea dickinsii collected from Taeback mountain was extracted with ethanol, The compounds 1 and 2 were purified by solvent extraction, silica gel column chromatography and recrystallization from the ethanol extracts. Melting point, molecular weight and molar extinction coefficient of the compound 1 were estimated to be $167{\sim}168^{\circ}C$, 572 and 5,040 at 208 nm. The structure of this compound 1 has been elucidated to be 24S, 25S, $3{\alpha}-carboxyacetylquercinic$ acid using spectroscopic properties. This compound showed antibiotic activities against fungi, yeasts and bacteria and it also showed anticancer activity $IC_{50}=64.5\;{\mu}M$) against Korean stomach cancer cell. Melting point, molecular weight and molar extinction coefficient of the compound 2 were estimated to be $233{\sim}235^<\circ}$, 572 and 5,080 at 208 nm. The structure of this compound was elucidated to be 24S, 25R, $3{\alpha}-carboxyacetylquercinic$ acid. The compound 2 showed different antibiotic activities with the compound 1.

Screening for Antimutagenic Effects of the Wild Mushrooms in Korea (한국산 야생버섯류중 항변이원성 효과의 검색)

  • To detect naturally occuring antimutagenic substances from wild mushrooms in Korea, the screening for the antimutagenic compounds containing in ethanol and water extracts of 13 wild mushrooms toward benzo(a) pyrene (B(a)P) and $N-methyl-N'-nitro-N-nitrosoguanidine\;(MNNG)$ using the Ames assay system with Salmonella typhimurium TA98 and TA100 were studied. The ethanol extracts of Polyporus dispansus, Cantharellus infundibuliformis, Agaricus subrutilescens, Daedalea dickinsii, Panaeolus papilionaceus and Hydnum repandum showed significantly antimutagenic activity toward B(a)P. The water extracts of Hydnum repandum showed the strong antimutagenic activity toward B(a)P in S. typhimurium TA100, however the water extracts of the mushrooms did not show antimutagenic activity. Whereas 5 out of 13 samples exhibited antimutagenicity toward a direct mutagen of MNNG. The water extracts from mushrooms also not showed antimutagenic activity. The antimutagenic effect increased with increasing concentraion of the ethanol extracts from Polyporus dispansus.

Antioxidant activities and β-glucan contents of wild mushrooms in Korea (국내 야생버섯의 항산화 활성 및 베타글루칸 함량 분석)

  • The anti-oxidant activities and ${\beta}$-glucan contents of wild mushrooms collected from Songnisan, Soi-myeon, Joryeongsan, and Ganghwado, Korea, were investigated. Among the wild mushroom extracts, Pycnoporus sanguineus (OK1071), Piptoporus soloniensis (OK1090), and Daedalea dickinsii (OK1094) extracts showed the highest DPPH and $ABTS^+$ radical scavenging activities. The nitrite scavenging activity and total polyphenol content of the extract of P. sanguineus (OK1071) was 74.2% and 37.7 mg GAE/g, respectively, and these values were much higher than those of the other mushrooms analyzed in this study. The Pearson's correlation coefficients of the DPPH radical and nitrite scavenging activities with the total polyphenol contents of the wild mushroom extracts were r = 0.758 (p < .01) and r = 0.951 (p < .01), respectively. The ${\beta}$-glucan contents of Lenzites betulina (OK1040), Trametes versicolor (OK1044), and P. sanguineus (OK1071) were 47.8%, 43.9%, and 41.8%, respectively, which were higher than those of the other mushrooms. Therefore, the wild mushrooms analyzed were confirmed to have excellent antioxidant activities and high ${\beta}$-glucan contents. Accordingly, the fundamental data provided in this study can be used to isolate useful compounds from these wild mushrooms.
