Reactive oxygen species-dependent down-regulation of ubiquitin C-terminal hydrolase in Schizosaccharomyces pombe (Schizosaccharomyces pombe에서의 유비퀴틴 C-말단 가수분해효소의 활성산소종 의존성 하향조절)

  • The Schizosaccharomyces pombe $sdu1^+$ gene, belonging to the PPPDE superfamily of deubiquitinating enzyme (DUB) genes, was previously shown to encode a protein with ubiquitin C-terminal hydrolase (UCH) activity and to participate in the response against oxidative and nitrosative stresses. This work focused on the reactive oxygen species (ROS)-dependent regulation of the S. pombe $sdu1^+$ gene. UCH activities, encoded by the $sdu1^+$ gene, were attenuated in the S. pombe cells exposed to $H_2O_2$, superoxide radical-generating menadione (MD), and nitric oxide (NO)-generating sodium nitroprusside (SNP). Reduced glutathione (GSH) and its precursor N-acetylcysteine (NAC) were able to significantly enhance the UCH activities in the absence or presence of $H_2O_2$. However, the influences of both GSH and NAC on the ROS levels in the absence or presence of $H_2O_2$ were opposite to their effects on the UCH activities under the same conditions. The UCH activities in the Sdu1-overexpressing S. pombe cells were also diminished under exposure to $H_2O_2$, MD and SNP, but still remained to be higher than those in the vector control cells. In brief, it is proposed that the S. pombe $sdu1^+$ gene is regulated by ROS in a negative manner, the meaning of which largely remains elusive.

HIDE, a Testis Specific Deubiquitinating Enzyme, Interacts with HSP90 (고환 특이적으로 발현되는 탈유비퀴틴효소 HIDE와 HSP90의 상호작용)

  • 연구목적: 본 연구는 아직 그 기능이 파악되지 않은 탈유비퀴틴효소 중 하나인 HIDE에 대한 기본적인 생화학적 특징과 고환에서의 발현 양상을 파악하고 있다. 연구재료 및 방법: 인간의 HIDE 유전자를 클로닝하여 효소활성이 있는지 세포 외 실험을 통해 확인하였고, 아미노산 서열을 분석하여 진화상 보존된 부분을 찾아 그 기능을 파악한 다음 HSP90과의 상호작용을 공동면역침전반응으로 확인하였다. HIDE의 조직별 발현양상을 파악하기 위해서 인간과 쥐의 RNA 블롯과 쥐의 단백질 블롯을 이용하여 각각 노던 블롯팅과 웨스턴 블롯팅을 수행하여 고환에서 많이 발현된다는 것을 알았고 이 사실을 바탕으로 쥐의 고환을 절개하여 면역조직화학반응으로써 고환 내의 HIDE 단백질의 발현양상을 파악하였다. 결 과: HIDE는 세포 외에서 유비퀴틴 잔기를 제거하는 탈유비퀴틴 활성이 있으나 세포 내에서 전체적인 유비퀴틴 복합체를 줄여주는 효과는 없었다. HIDE는 HSP90이라는 분자 샤페론과 상호작용한다. HIDE의 전사체는 고환에서 가장 많이 발현되며 다른 조직에서도 소량 발현된다. HIDE의 단백질은 웨스턴 블롯상에서 고환에서만 확인되었다. 고환 내에서의 HIDE의 발현양상은 왕성한 감수분열을 하는 정모세포에서 높았으며 지지세포나 정조세포에는 발현되지 않았다. 결 론: HIDE는 분자 샤페론 HSP90과 상호작용하며 고환 내의 감수분열 중인 세포에서 많이 발현되는 것으로 보아 감수분열이나 정자형성에 관여하는 것으로 보인다.

The Process of Education in the Light of Xugua zhuan, the Sequence of the Hexagrams in I Ching (「서괘전」에 비추어 본 교육의 과정 -건(乾)·곤괘(坤卦)에서 태괘(泰卦)까지를 중심으로-)

  • The paper has focused on examining the sense of educational process in the light of Xugua zhuan[序卦傳], the Sequence of the Hexagrams as one among Ten Wings in I Ching, particularly on the 11 Iching-hexagrams from ch'ien[乾] to t'ai[泰]. The process involved in this part of the Book of Change can be recognized as both the micro and macro levels, which can eventually leads us to construct the circular system of thought[環相型]. This gives us a possibility to consider the values of education beyond the dualism such as intrinsic and extrinsic as well as traditional and progressive ones. Particularly some crucial defects from child-centered education have been critically examined so that the concept of education can be taken seriously in the light of the teacher-side. Thus this provides us a necessity to dub 'education' as '敎育', not as '兒育' in the Oriental context. As shown in the paper, the cosmic dual forces of I Ching also enables us to dissolve some conflicts arisen in the educational welfare system and situations, and then shows us why we should respect for the teachers' stances though we have to consider the interests of children to be educated. Thus we need to re-examine the content of the Book in the link with the Western systems of educational theory.

A Study on the Concept of 'Hyanghwa' of Chosun Dynasty (조선시대(朝鮮時代) '향화(向化)'개념에 대한 연구(硏究) - 『조선왕조실록(朝鮮王朝實錄)』을 중심(中心)으로 -)

  • The word, 'Naturalization(向化)' is an unfamiliar term for us; however, to us as citizens who live in a multicultural society, it is a word worthy of hearty appreciation even once. At the present Korean dictionary, it has been defined as the word related to those terms such as Enlightenment(敎化) which are used to explicate the governance through a virtue of moral excellence(德治主義) of Confucianism, and the word, 'Hyanghwa(向化)' itself has already included the meaning of Enlightenment(敎化). On many occasions, people who were dubbed the converts or the naturalized citizens(向化人) emigrated to Chosun because there were such substantial reasons as economic and political advantages, rather than living in a society for reasons to the governance through a virtue of moral excellence(德治主義). Therefore, to understand the converts or the Naturalized citizens(向化人) correctly, it shall be required to examine the cause which is the governance through a virtue of moral excellence together with the true realities which are economic and political advantages. The word, such as Hwangjoin(皇朝人) should be considered in order to understand the meaning of 'Naturalization(向化)'. Hwangjoin(皇朝人) were those displaced people who had been forced to leave the fallen Ming Dynasty, and they will be contrasted with the converts or Naturalized citizens(向化人) that were mainly composed of the Northern tribes or Japanese. Actually, Naturalized citizens(向化人) and Hwangjoin(皇朝人) had all emigrated to Chosun because of changes in international affairs, meanwhile, however, they were treated differently for the reason that they were 'Hwai'(華夷). If we dub Chosun 'Self'(我), Naturalized citizens(向化人) and Hwangjoin(皇朝人) are equally called as 'The Other persons'(他者). Can we say that Chosun had established the right relationship with Other persons(他者)? Judging from the facts that there was occurrence of convert or naturalization for the other persons, dubbed Naturalized citizens(向化人) and that there was another occurrence of emigrants, called Hwangjoin(皇朝人), it is evident that the Relationships between the two had not rightly established. Also, the names so called Naturalized citizens(向化人) and Hwangjoin(皇朝人) were not given according to the preference of the other person, but provided by Chosun one-sidedly. The reasons for occurrence of convert or naturalization for the other persons, dubbed Naturalized citizens(向化人) and for occurrence of other emigrants, called Hwangjoin(皇朝人) were ideology such as 'Hwai'(華夷). It is thought that the Relationships between 'Self'(我) and 'The Other Person'(他者) can be established only after excluding ideology.
