Measurement of the Anti-oxidative Properties of Extract from Medicinal Plants Using an On-line HPLC-DPPH Assay (HPLC와 DPPH radical 소거능 측정 방법의 결합에 의한 약용 식물 추출물의 항산화 활성 비교)

  • Natural anti-oxidative compounds have important disease prevention and food preservation properties, in addition to anti-bacterial, anti-inflammation, anti-cancer, and skin whitening effects. High-performance liquid chromatography (HPLC), with an ultra vilolet (UV) detector coupled to a reverse phase C18 column and an online measurement system for 1, 1-diphenyl-2-picryl hydrazyl (DPPH) radicals, was used to search for potent antioxidative compounds in crude extracts. The online HPLC-DPPH assay was then applied to confirm antioxidative compounds in water extracts from Radix of Pueraria lobata, Rhizoma of Zingiber officinale, Fructus of Chaenomeles sinensis, Cortex of Ulmus pumila, and Radix of Astragalus membranaceus. To determine the yields of the extracts, the Brix% of each extract solution was measured using a refractometer. When the relative DPPH radical scavenging ability values of the water extracts were compared with those of a positive control (ascorbic acid), the water extracts of P. lobata, C. sinensis, and U. pumila were 7.77%, 4.71%, and 4.19%, respectively. The results suggest that this method provides a useful assay for rapid measurement of DPPH radical scavenging abilities and conformation of antioxidative compounds in natural products. Moreover, it can reduce the time spent on the separation of active compounds from natural materials, such as medicinal plants, in addition to the use of reagents for separation.

Antioxidant Activities of Young and Mature Fruit in Three Asian Pear Cultivars (동양배 품종별 유과기와 수확기 과실의 항산화 활성)

  • This study was performed to compare antioxidant activities of 80% EtOH extracts from young and mature fruit of three pear cultivars ('Wonhwang', 'Whangkeumbae', and 'Chuwhangbae') classified by three parts (peel, core, and flesh) of mature fruit. The total phenolics compound and flavonoid contents of 80% EtOH extracts from young fruit were 1.5-2.5 times higher than the mature fruit with great cultivar difference. In particular, the contents of those compounds were highest in 'Chuwhangbae' pear. The total phenolics compound and flavonoid contents of 80% EtOH extracts from mature fruit were the most in the peel, core, and flesh respectively. DPPH radical scavenging and $ABTS^+$ radical scavenging of 80% EtOH extracts from the young fruit were higher than the mature fruit of 'Chuwhangbae' pear. DPPH radical scavenging and ABTS+ radical scavenging of 80% EtOH extracts from mature fruit were the highest in the peel, core, and flesh respectively.

Antioxidant Activities and Protective Effects of Hot Water Extract from Curcuma longa L. on Oxidative Stress-Induced C2C12 Myoblasts (강황 열수 추출물의 항산화 활성 및 C2C12 Myoblasts의 산화적 손상에 대한 보호 효과)

  • The aim of this study was to investigate the antioxidant activities and protective effects of hot water extract from Curcuma longa L. (CLW) on oxidative stress-induced C2C12 myoblasts. Antioxidant activities of CLW were evaluated based on 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activities. Protective effects of CLW on oxidative stress-induced C2C12 myoblasts were determined based on cytotoxicity, $H_2O_2$ protective activity, and intracellular reactive oxygen species (ROS) level. DPPH and ABTS radical scavenging activities represented by $SC_{50}$ were $188.5{\pm}3.0{\mu}g/mL$ and $92.0{\pm}0.9{\mu}g/mL$, respectively. Using C2C12 myoblasts, CLW treatment increased cell viability against oxidative stress-induced cell death. Further, CLW treatment reduced the intracellular ROS level in cells treated with $H_2O_2$. These results suggest that CLW might have the capability to protect oxidative stress-induced C2C12 myoblasts.

Effects of Fermentation on the Metabolic Activities of Pine Needle Juice (발효과정이 솔잎 착즙액의 항산화, alpha-Glucosidase 및 Angiotensin Converting Enzyme 저해 활성에 미치는 영향)

  • The objective of this study was to compare the content and metabolic activities between fresh pine needle juice (PNJ) and fermented pine needle juice (FPNJ). A variety of factors were measured, including total phenolic content (TPC), antioxidant activity [DPPH radical scavenging activity (RSA), total radical-trapping antioxidant potential (TRAP), oxygen radical absorbance capacity (ORAC), cellular antioxidant capacity (CAC)], anti-genotoxic activity, ${\alpha}$-glucosidase inhibitory activity, and angiotensin converting enzyme (ACE) inhibitory activity. The TPC was $17.3{\pm}0.2$ and $4.6{\pm}0.0$ mg GAE/g in PNJ and FPNJ, respectively. The DPPH RSA, TRAP, and ORAC values increased in a dose-dependent manner for both PNJ and FPNJ, with significantly higher activities in PNJ than FPNJ. The CAC against AAPH-induced oxidative stress in HepG2 cells was protected by both PNJ and FPNJ. Pretreatment with PNJ and FPNJ in human leukocytes produced significant reductions in $H_2O_2$-induced DNA damage at a concentration of $50{\mu}g/mL$. ${\alpha}$-Glucosidase inhibitory activity was significantly higher in FPNJ than PNJ. The ACE inhibitory activity was about 87.1% and 60.0% in 1:1 diluted PNJ and FPNJ, respectively. This study suggests that the fermentation of PNJ could enhance the regulation of blood glucose metabolism and both PNJ and FPNJ might be a new potential source of natural antioxidant, anti-diabetic, and anti-hypertensive agents applicable to food.

A Study on the Antimicrobial Activity and the Pharmacological Activities of matrial Isolated from Coptis Radix (황련(Coptis Radix)으로부터 분리된 물질의 항균효능 및 화장품 약리활성에 대한 연구)

  • This study evaluated antimicrobial efficacy and antioxidant effect of fraction isolated from Coptis Radix and confirmed its possibility as a cosmetic material. The extracts of isolated from Coptis Radix conducted an antimicrobial activity against Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Candida albicans by disc diffusion method and measure clear zone. As a result, it was confirmed that antimicrobial activity against S. aureus and candida. A was observed in all samples except Fr 1. The activity of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and The activity of 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) cation radical scavenging were determined by antioxidant assay according to the concentrations (50, 125, 250) ${\mu}g/mL$ of extracts of isolated from Coptis Radix. As a result, DPPH radical scavenging activity of Fr 1, 2, 3, 4 at $250{\mu}g/mL$ was 11.4%, 30.3%, 42.0% and 53.1%, respectively and $ABTS^+$ radical scavenging activity was 28.6%, 96.2%, 98.6% and 97.1% at the same concentration, respectively. Fr. 3 and 4 showed higher radical scavenging activity than the positive the control group BHT at the same concentration. In the WST assay results of measuring the cytotoxicity of Coptis Radix, except for Fr. 4, Fr. 1, 2 and 3 did not show toxicity. As a result, the fractions isolated from Coptis Radix can be regarded as a cosmetic material having antimicrobial activity and antioxidant ability.

Effect of Unripe Apple Peel Water Extracts on Tyrosinase Activity and Melanin Production in B16F10 Melanoma Cells (B16F10 melanoma 세포에서 미성숙 사과 과피 열수추출물의 tyrosinase 활성과 melanin 생 성에 미치는 영향)

  • This study was carried out to evaluate the possibility of unripe apple peel water extracts as cosmetic materials and to evaluate the biological activities of the antioxidant and whitening effects of the samples. The antioxidative properties of the samples were confirmed by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) cation radical scavenging ability. To evaluate the whitening effect of the samples, several analytical techniques were used, including toxicity evaluations of the samples by MTT assays. Measurements of the inhibition rates of cellular tyrosinase, melanin synthesis rates, and expression rates of whitening-related proteins and genes were confirmed using melanoma (B16F10 cell). At equivalent unripe apple peel water concentrations ($1,000{\mu}g/ml$), the DPPH radical scavenging and the ABTS cation radical scavenging activities were 77.3% and 93.1%, respectively. The whitening activity evaluation showed that tyrosinase activity and melanin synthesis were inhibited by 19.8% and 17.3%, respectively, at unripe apple peel water extract concentrations of $50{\mu}g/ml$. In B16F10 cells induced by ${\alpha}$-MSH, the expression of tyrosinase, TRP-1, and TRP-2 decreased. Also, the activity of the transcription factor MITF was inhibited. In real-time PCR experiments, the expression of related genes at the upstream signal level was also found to be progressively lowered as the concentration of unripe apple peel water extracts increased. From these results, it was confirmed that the unripe apple peel water extracts showed excellent whitening efficacy and could be used as safe, natural, raw cosmetic material in the future.

Antioxidant Activity of Branches of Fuyu and Nishimurawase Persimmon Trees (부유 및 서촌조생 감나무 가지의 항산화 활성)

  • The antioxidant activities of two persimmon (Diospyros kaki T.) branches were evaluated. After preparing acetone, 70% acetone, and methanol extracts from Fuyu and Nishimurawase persimmon branches, DPPH radical scavenging activity (RSA), ABTS RSA, reducing power (RP), and total phenol contents (TPC) were determined. The acetone extracts of Fuyu and Nishimurawase showed higher antioxidant properties than the other extracts. The highest TPC (22.48 mg/g), DPPH RSA ($IC_{50}=701.48\;{\mu}g/mL$), ABTS RSA ($IC_{50}=493.76\;{\mu}g/mL$), and RP ($IC_{50}=1218.57\;{\mu}g/mL$) were found in acetone extract of Fuyu, while the lowest antioxidant activities were in 70% acetone extract of Nishimurawase. The results suggest that sweet persimmon branches exhibit considerable antioxidant activity.

Anti-oxidative Activities for the Flavonoids of the Syzygium aqueum Burm.f. Alston Branches from Jeju Island (제주 자생 물사과 가지 유래 Flavonoid 화합물의 항산화 활성)

  • In this study, we investigated the anti-oxidative activities and cell protective effects of the constituents isolated from S. aqueum branches. DPPH and $ABTS^+$ radical scavenging activities were screened for the ethanol extract and solvent fractions, ethyl acetate (EtOAc) and butanol (BuOH) fractions showed potent activities. When HaCaT cells were treated with $H_2O_2$, the ethanol extract and EtOAc fractions ($20{\mu}g/mL$) protected the cells against oxidative damage. Two constituents were isolated from the EtOAc fraction of S. aqueum branches; pinocembrin (1), desmethoxymatteucinol (2). The chemical structures of the isolated compounds were elucidated based on the spectroscopic data including $^1H$ and $^{13}C$ NMR spectra, as well as comparison of the data to the literature values. Anti-oxidative activities and cell protective effects were studied for the isolated compounds. For the anti-oxidative activities, all of the compounds 1 and 2 showed DPPH and $ABTS^+$ radical scavenging activities. Also, from the cell protective effect test, the compounds 1 and 2 protected the cell against oxidative stress by $H_2O_2$. Based on these results, S. aqueum branches extract could be potentially applicable as anti-oxidant ingredients in cosmetic industries.

Antioxidant activities of soymilk added with green tea and rosemary extract (녹차와 로즈마리 추출물을 첨가한 두유의 항산화 활성)

  • The objective of this study was to compare antioxidant activities of green tea, rosemary, lemon and bamboo leaves extracts using three different extraction methods (80% ethanol sonication extraction, distilled water autoclave extraction, distilled water extraction at room temperature). As a result, green tea and rosemary extracts showed relatively high antioxidant activities compared with those of lemon and bamboo leaves. These green tea and rosemary extracts were subsequently added to soymilk and their mixtures were examined for antioxidant activities. Soymilk added with 10% green tea distilled water autoclave extracts and distilled water at room temperature extracts showed relatively high DPPH, $85.09{\pm}2.26%$, $84.38{\pm}1.97%$, and ABTS radical scavenging activities, $73.43{\pm}2.78%$, $81.34{\pm}4.78%$, respectively. On the other hand, soymilk added with 10% rosemary distilled water autoclave extracts showed remarkably higher DPPH radical scavenging activity while their ABTS radical scavenging ability was similar to that of non-added soymilk. In conclusion, our results suggest that the addition of green tea distilled water autoclave extracts, distilled water at room temperature extracts and rosemary distilled water autoclave extracts to soymilk could contribute to the development of added value soymilk products with increased antioxidant activities.

Cultural characteristics and antioxidant activity of wild-type collections of Hericium erinaceus (노루궁뎅이버섯 야생 수집균주의 배양적 특성과 항산화 활성)

  • This study aimed to characterize the mycelial morphologies of Hericium erinaceus isolates, including 18 wild-type collections and cultivatable species 'Noru 1' and 'Noru 2', in Korea. The morphological characteristics were used to classify the species based on aerial or branched mycelia and their brownish or whitish colors when grown on potato dextrose agar. Of the wild-type collections, the isolates KFRI 509, KFRI 1091, KFRI 1093, and KFRI 1623 showed faster mycelial growth than the cultivars 'Noru 1' and 'Noru 2'. Moreover, 60% ethanol extracts of the fruiting body of the mushroom showed the highest phenol content and DPPH and ABTS radical scavenging activity. The DPPH and ABTS radical scavenging activities of KFRI 507, KFRI 508, KFRI 842, and KFRI 1623 were 10-60% higher than those of 'Noru 1' and 'Noru 2', depending on the extract concentrations. Thus, results suggest that these wild-type collections could be useful for breeding genetic sources or processed food materials with high antioxidant activity.
