Comparative Evaluation of Silibinin Effects on Cell Cycling and Apoptosis in Human Breast Cancer MCF-7 and T47D Cell Lines

  • Silibinin is a natural polyphenol with high antioxidant and anticancer properties. In this study, its influence on two of the most commonly employed human breast cancer cell lines, MCF-7 and T47D, and one non-malignant MCF-10A cell line, were investigated and compared. Cell viability, the cell cycle distribution and apoptosis induction were analyzed by MTT and flow cytometry, respectively. The effect of silibinin on PTEN, Bcl-2, P21, and P27 mRNAs expression was also investigated by real-time RT-PCR. It was found that silibinin caused G1 cell cycle arrest in MCF-7 and MCF-10A cells but had no effect on the T47D cell cycle. Silibinin induced cytotoxic and apoptotic effects in T47D cells more than the MCF-7 cells and had no cytotoxic effect in MCF-10A cells under the same conditions. Silibinin upregulated PTEN in MCF-7 and caused slightly increased P21 mRNA expression in T47D cells and slightly increased PTEN and P21 expression in MCF-10A cells. Bcl-2 expression decreased in all of the examined cells under silibinin treatment. P27 mRNA expression upregulated in T47D and MCF-10A cells under silibinin treatment. PTEN mRNA in T47D and P21 and P27 mRNAsin MCF-7 were not affected by silibinin. These results suggest that silibinin has mostly different inhibitory effects in breast cancer cells and might be an effective anticancer agent for some cells linked to influence on cell cycle progression.

  • Power of loess ball on nitrogen and phosphorous removal in wastewater treatment were investigated. flow line A ( anaerobic${\rightarrow}$oxic${\rightarrow}$anoxic(organic source methanol)${\rightarrow}$p-absorption) showed the results of T-P 0.5, T-N 1.0, and COD 10ppm bellow, and flow line B ( oxic${\rightarrow}$anoxic, organic source: methanol${\rightarrow}$p-absorption) showed the results of T-P 0.3, T-N 5.0, and COD 15 ppm bellow. flow line C ( anaerobic${\rightarrow}$oxic${\rightarrow}$anoxic, organic source: wastewater ${\rightarrow}$ p-absorption) showed the results of T-P 0.6, T-N 10, and COD 15 ppm bellow, and flow line D ( oxic${\rightarrow}$anoxic, organic source: methanol${\rightarrow}$p-absorption) showed the results of T-P 1, T-N 8m, and COD 20 ppm bellow. So the results of these experiments showed the probability of loess ball in wastewater treatment.

(b) If $T^*$ is algebraically (p, k)-quasihyponormal, then a-Weyl's theorem holds for f(T) for every $f{\in}Hol({\sigma}T))$, where $Hol({\sigma}(T))$ is the space of all functions that analytic in an open neighborhoods of ${\sigma}(T)$ of T. (c) If $T^*$ is algebraically (p, k)-quasihyponormal, then generalized a-Weyl's theorem holds for f(T) for every $f{\in}Hol({\sigma}T))$. (d) If T is a (p, k)-quasihyponormal operator, then the spectral mapping theorem holds for semi-B-essential approximate point spectrum $\sigma_{SBF_+^-}(T)$, and for left Drazin spectrum ${\sigma}_{lD}(T)$ for every $f{\in}Hol({\sigma}T))$.

Effect of Vitamin E and Zinc Supplementation on Energy Metabolites, Lipid Peroxidation, and Milk Production in Peripartum Sahiwal Cows

  • The study was conducted to evaluate the effect of vitamin E and zinc supplementation on energy metabolites, lipid peroxidation, and milk production in peripartum Sahiwal cows. For this, thirty-two pregnant dry Sahiwal cows were selected at sixty days prepartum and divided into four groups viz control, $T_1$, $T_2$, and $T_3$ of eight each. Group $T_1$ were supplemented with zinc at 60 ppm/d/cow, group $T_2$ were supplemented with vitamin E at 1,000 IU/d/cow and group $T_3$ were supplemented with combination of vitamin E at 1,000 IU/d/cow and zinc at 60 ppm/d/cow during d 60 prepartum to d 90 postpartum. Blood samples were collected on d -60, -45, -30, -15, -7, -3, 0, 3, 7, 15, 30, 45, 60, 90, and 120 with respect to day of parturition and analysed for glucose, non esterified fatty acid, and thiobarbituric acid reactive substance. Body condition score was maintained significantly better (p<0.05) in $T_3$ than in the control, $T_1$ and $T_2$ groups. Overall glucose level was higher (p<0.05) in $T_3$ than control, $T_1$, and $T_2$ groups. Levels of nonesterified fatty acid, and thiobarbituric acid reactive substance were lower (p<0.05) in $T_3$ than control, $T_1$, and $T_2$ groups. Milk yield was higher (p<0.05) in $T_3$ than control, $T_1$, and $T_2$ groups. In conclusion, the present study indicated that the supplementation of vitamin E and zinc in peripartum Sahiwal cows enhanced milk production by reducing negative energy balance.

  • This study aims to examine the effects of watching 2D and 3D images on the blink rate. Regarding the image watch, their blink rate for 2D and 3D images was separately checked for 1 minute in the 1m distance, before the watch, after 15 minutes of watch, and after 30 minutes of watch. About the change of their blink rate in the 2D image watch, it tended to become higher than that before watching the image; however, there was no statistical significance (paired t-test, p=0.106, p=0.062). And in the 2D image watch, it tended to increase in comparison between after 15 minutes and after 30 minutes, but there was no statistical significance (paired t-test, p=0.623). Meanwhile, about the change of their blink rate in the 3D image watch, it tended to decrease statistically significantly both after 15 minutes and after 30 minutes when compared with that before watching the image (paired t-test, p=0.000, p=0.000). In the 3D image watch, it tended to increase in comparison between after 15 minutes and after 30 minutes; however, there was no statistical significance (paired t-test, p=0.867).

Clinical Analysis of 102 Cases of Open Heart Surgery (개심수술 102례 의 임상적 고찰)

  • A total of 102 patients who had an Open Heart Surgery from April 1976 to July 1981 were reviewed. 55 paeitnts were congenital heart disease and 47 patients were acquired heart disease. Among SS patients of congenital heart disease, 18 T 0 F, 18 V S D, 8 A S D, and each one case of l\ulcorner 0 R V, Truncus arteriosus, Ebstein anomaly, Single ventricle, P D A, P 5, A S D + P 5, E C D, V 5 D + P D A, A - P window, D C R V were noted respectively. In 47 patients of acquired heart disease and one Ebstein patient, 46 prosthetic values were implanted: 17 had M V R, 4 had A V R, 2 had M V R + A V R, and 4 had M V R + T V R and one T V R. The operative mortality was 8.S% in acquired heart disease and 17% in congenital heart disease. The follow up period was between 6 months and 6 years. There were 3 cases of late mortality in acquired heart disease and one case in congenital heart disease.

Effects of Growth Retardants on the Growth of Creeping Bentgrass (식물생장억제제가 Creeping Bentgrass의 생육에 미치는 영향)

  • This study was conducted to evaluate the effect of plant growth inhibitors on "Penncross" creeping bentgrass in the green of golf course for increasing the summer quality. Chlormequat chloride (C) 250$\times$, 500$\times$, 1000$\times$, daminozide (D) 50$\times$, 100$\times$, 200$\times$, paclobutralzol (P) 1000$\times$, 2000$\times$, 4000$\times$, trinexpac-ethyl (T) 1000$\times$, 2000$\times$, 4000$\times$, uniconazole (U) 25$\times$, 50$\times$, 100$\times$ with three concentrations, respectively, were applied to creeping bentgrass. The leaf length, leaf width, and internode length of stolon were inhibited by all plant growth retardants applied as compared to that of control in which D-50, T-2000, and trinexpac-ethyl were the most effective among treatments on the inhibition of leaf length and internode length of stolen, respectively. Leaf thickness increased more in the treatments of C-250, D-50. T-2000 and T-4000 than any other treatments. On the other hand, shoot density which was represented by number of leaves per unit area was found to be the highest in P-2000 and P-4000 treatment. In this case, all plant growth retardants with no significant differences showed increase of 15-20% in density as compared to control. Total dry weight increment was higher in P-2000 and P-4000 while leaf dry weight increment was higher in T-4000. and U-25 and 50 than other treatments. According to the visual evaluation for the purpose of turf quality measurement, it was found that paclo-butralzol, trinexpac-ethyl and uniconazole were not suitable for green maintenance and extension because of making leaves to yellow. In conclusion, it was postulated that C-250, D-50, and T-4000 had possibility to use practically in the green area of golf course.lf course.

  • Based on Computer graphics molecular modeling method, quinolone derivatives as DNA-gyrase inhibitors formed stable DNA-intercalation complex with deoxycytidilyl-3',5'-deoxy guanosine[d($C_{p}G)_{2}$] dinucleotide. When d($C_{p}G)_{2}$ and d($A_{p}T)_{2}$, were compared in order to find out which DNA could form more stable DNA-Drug complex based on interaction energy($\Delta$E) and DNA-Drug complex energy, d($C_{p}G)_{2}$ resulted in lower energy than d($A_{p}T)_{2}$.

The effects of berberine on ischemia-reperfusion injuries in an experimental model of ovarian torsion

  • Objective: Ovarian torsion is a gynecological disorder that causes ischemia-reperfusion injuries in the ovary. Our study investigated berberine's short- and long-term effects on ovarian ischemia-reperfusion injuries. Methods: This study included 28 Wistar albino female rats weighing 180 to 220 g, which were divided into four groups: sham (S), torsion/detorsion (T/D), torsion/ detorsion+single dose berberine (T/D+Bb), and torsion/detorsion+15 days berberine (T/D+15Bb). The torsion and detorsion model was applied in all non-sham groups. In the T/D+Bb group, a single dose of berberine was administered, while in the T/D+15Bb group, berberine was administered over a period of 15 days. After the rats were euthanized, their ovaries were excised. The left ovaries were used for histopathologic evaluation, which included ovarian injury scoring and follicle count, while the right ovaries were used for biochemical analyses (tissue transforming growth factor-β [TGF-β] and alpha-smooth muscle actin [α-SMA] levels). Results: The histopathologic evaluation scores for the ovaries were significantly lower in the T/D+B group (p<0.05) and the T/D+15B group (p<0.005) than in the T/D group. The follicle counts in the T/D group were lower than those in both the sham and treated groups (p<0.005). The TGF-β levels were significantly lower in the T/D+15B group (p<0.005), whereas the α-SMA levels did not show a significant difference. Conclusion: Both short- and long-term berberine use could potentially have therapeutic effects on ovarian torsion. Long-term berberine use exhibited anti-inflammatory effects by reducing TGF-β levels, thereby preventing ischemia-reperfusion injuries. Therefore, we suggest that long-term berberine use could be beneficial for ovarian torsion.

Recanalization Rate and Clinical Outcomes of Intravenous Tissue Plasminogen Activator Administration for Large Vessel Occlusion Stroke Patients

  • Objective : Stroke caused from large vessel occlusion (LVO) has emerged as the most common stroke subtype worldwide. Intravenous tissue plasminogen activator administration (IV-tPA) and additional intraarterial thrombectomy (IA-Tx) is regarded as standard treatment. In this study, the authors try to find the early recanalization rate of IV-tPA in LVO stroke patients. Methods : Total 300 patients undertook IA-Tx with confirmed anterior circulation LVO, were analyzed retrospectively. Brain computed tomography angiography (CTA) was the initial imaging study and acute stroke magnetic resonance angiography (MRA) followed after finished IV-tPA. Early recanalization rate was evaluated by acute stroke MRA within 2 hours after the IV-tPA. In 167 patients undertook IV-tPA only and 133 non-recanalized patients by IV-tPA, additional IA-Tx tried (IV-tPA + IA-Tx group). And 131 patients, non-recanalized by IV-tPA (IV-tPA group) additional IA-Tx recommend and tried according to the patient condition and compliance. Results : Early recanalization rate of LVO after IV-tPA was 12.0% (36/300). In recanalized patients, favorable outcome (modified Rankin Scale, 0-2) was 69.4% (25/36) while it was 32.1% (42/131, p<0.001) in non-recanalized patients. Among 133 patients, non-recanalized after intravenous recombinant tissue plasminogen activator and undertook additional IA-Tx, the clinical outcome was better than not undertaken additional IA-Tx (favorable outcome was 42.9% vs. 32.1%, p=0.046). Analysis according to the perfusion/diffusion (P/D)-mismatching or not, in patient with IV-tPA with IA-Tx (133 patients), favorable outcome was higher in P/D-mismatching patient (52/104; 50.0%) than P/D-matching patients (5/29; 17.2%; p=0.001). Which treatment tired, P/D-mismatching was favored in clinical outcome (iv-tPA only, p=0.008 and IV-tPA with IA-Tx, p=0.001). Conclusion : The P/D-mismatching influences on the recanalization and clinical outcomes of IV-tPA and IA-Tx. The authors would like to propose that we had better prepare IA-Tx when LVO is diagnosed on initial diagnostic imaging. Furthermore, if the patient shows P/D-mismatching on MRA after IV-tPA, additional IA-Tx improves treatment results and lessen the futile recanalization.
