Bioecological Characteristics of Coral Habitats around Munseom, Cheju Island, Korea - III. Seasonal and Diel Fluctuations of Gelatinous Zooplankton Biomass (제주도 문섬 산호서식지 주변의 생물생태학적 특성 - III. 젤라틴성 동물플랑크톤 생물량의 계절 및 일 변동)

  • The daily fluctuations of occurrence rate and standing stock of zooplankton, the relationship between dry and wet weights of gelatinous zooplankton, and the size distributions of body length and body weight of zooplankton were investigated in the sea around Cheju Island from September 1996 to August 1997. Mean ratios of wet to dry weights were 66.46 (raging from 47.05 to 84.64) in the gelatinous zooplankton and 10.89 (raging 9.21 to 14.85) in the non-gelatinous zooplankton which consisted of crustaceans such as copepods, decapods, mysids, and ostracods etc. Rarios of gelatinous zooplankton to non-gelatinous zooplankton (G/NG) in wet weight was 0.99 (0.28-2.30), indicating high occurrence rate of gelatinous in the study area. The seasonal and diel fluctuations of the gelatinous zooplankton were very large, and G/NG ratios greatly varied with seasons and dat/night cycles. The size distributions of body length of zooplankton (250-500 urn: 35.5% in maximum), and wet (0.1-0.2 mg: 34.4%) and dry weights (0.025-0.05 mg: 44.8%) showed much difference depending composition of the zooplankton groups. These results show that quantitative estimations of zooplankton produc- tivity only based on wet weight should be corrected to better understand and evaluate marine resources in Korean waters.

A Novel Complement Fixation Pathway Initiated by SIGN-R1 Interacting with C1q in Innate Immunity

  • Serum complement proteins comprise an important system that is responsible for several innate and adaptive immune defence mechanisms. There were three well described pathways known to lead to the generation of a C3 convertase, which catalyses the proteolysis of complement component C3, and leads to the formation of C3 opsonins (C3b, iC3b and C3d) that fix to bacteria. A pivotal step in the complement pathway is the assembly of a C3 convertase, which digests the C3 complement component to form microbial-binding C3 fragments recognized by leukocytes. The spleen clears microorganisms from the blood. Individuals lacking this organ are more susceptible to Streptococcus pneumoniae. Innate resistance to S. pneumoniae has previously been shown to involve complement components C3 and C4, however this resistance has only a partial requirement for mediators of these three pathways, such as immunoglobulin, factor B and mannose-binding lectin. Therefore it was likely that spleen and complement system provide resistance against blood-borne S. pneumoniae infection through unknown mechanism. To better understand the mechanisms involved, we studied Specific intracellular adhesion molecule-grabbing nonintegrin (SIGN)-R1. SIGN-R1, is a C-type lectin that is expressed at high levels by spleen marginal-zone macrophages and lymph-node macrophages. SIGN-R1 has previously been shown to be the main receptor for bacterial dextrans, as well as for the capsular pneumococcal polysaccharide (CPS) of S. pneumoniae. We examined the specific role of this receptor in the activation of complement. Using a monoclonal antibody that selectively downregulates SIGN-R1 expression in vivo, we show that in response to S. pneumoniae or CPS, SIGN-R1 mediates the immediate proteolysis of C3 and fixation of C3 opsonins to S. pneumoniae or to marginal-zone macrophages that had taken up CPS. These data indicate that SIGN-R1 is largely responsible for the rapid C3 convertase formation induced by S. pneumoniae in the spleen of mice. Also, we found that SIGN-R1 directly binds C1q and that C3 fixation by SIGN-R1 requires C1q and C4 but not factor B or immunoglobulin. Traditionally C3 convertase can be formed by the classical C1q- and immunoglobulin-dependent pathway, the alternative factor-B-dependent pathway and the soluble mannose-binding lectin pathway. Furthermore Conditional SIGN-R1 knockout mice developed deficits in C3 catabolism when given S. pneumoniae or its capsular polysaccharide intravenously. There were marked reductions in proteolysis of serum C3, deposition of C3 on organisms within SIGN-$R1^+$ spleen macrophages, and formation of C3 ligands. The transmembrane lectin SIGN-R1 therefore contributes to innate resistance by an unusual C3 activation pathway. We propose that in the SIGN-R1 mediated complement activation pathway, after binding to polysaccharide, SIGN-R1 captures C1q. SIGN-R1 can then, in association with several other complement proteins including C4, lead to the formation of a C3 convertase and fixation of C3. Therefore, this new pathway for C3 fixation by SIGN-R1, which is unusual as it is a classical C1q-dependent pathway that does not require immuno globulin, contributes to innate immune resistance to certain encapsulated microorganisms.

Determination of Volatile Organic Compounds emitted from Municipal Solid Waste Landfill Site by Thermal Desorption-Cryofocusing-GC/FID/FPD (열탈착-저온농축-GC/FID/FPD에 의한 도시 생활폐기물 매립장에서 방출되는 휘발성 유기화합물의 측정에 관한 연구)

  • In this study, the thermal desorption-cryofocusing-gas chromatographic(TD-C-GC) method was developed for determination of volatile organic compounds(VOCs) in ambient air and was applied at the municipal solid waste landfill sites. On-column cryofocusing was possible only with a 100 ml dewars bottle in TD-C-GC method with a stainless steel column. However, high operating pressure was needed for purging VOCs from the absorbent trap, which was able to solve by pressure programming with a electric pressure controller. By using both pressure and temperature programming brought increasing of resolution power in on-column cryofocusing method, but the high pressure caused a leakage of sample tube with repeated use. A loop cryofocusing devise was also developed and compared with the direct on-column method. In loop cryofocusing method, VOCs were concentrated on a 0.8mm i.d. loop which is located between the injector and separation column by using liquid nitrogen. In order to purge VOCs from the absorbent trap, only 0.4 psi of pressure was need in the loop cryofocusing method. Dual detection system was applied for the analysis of VOCs; a FID was used for hydrocarbons and a FPD was used for sulfur-containing compounds. Qualitative analysis was done by on-column cryofocusing GC-MS system. Among the large number of VOCs, toluene was the most abundant. Hydrogen sulfide, dimethyl sulfide, carbon disulfide, dimethyl disulfide and methyl propyl disulfide were detected at landfill site by FPD.

  • The metabolites of 1,2,4-trimethylbenzene (TMB) were synthesized and determined by capillary electrophoresis (CE). The optimum conditions of CE for the separation and determination of 3,4-, 2,4-, 2,5-dimethylbenzoic acid and 3,4-, 2,4-, 2,5-dimethylhippuric acid from the rat urine were as following: the fused silica capillary($75{\mu}m$ i.d. ${\times}$ 36 cm length, 29 cm to detector) was used and kept at $15^{\circ}C$. The applied voltage was 10㎸ and compounds were detected at UV 210 mnm and 254 nm. The running electrolyte was 0.1 M phosphate buffer (pH 7) containing 15 mM of ${\beta}-CD$ and 3% of 2-propanol. The relative amount of the metabolite of 1,2,4-TMB in the rat urine was 56.7% of 3,4-isomer, 30.5% of 2,4-isomer and 12.8% of 2,5-isomer. This method can be applied to the analysis of TMB-metabolites in human urine.

A Comparative Study on the Herbage Utilization of Turf Type Mixtures (하번초형 혼파초지의 목초이용성 비교에 관한 연구)

  • The objective of this experiment was to compare the dry matter intake, nutrients digestibility and nitrogen and energy utilization of herbages harvested from turf type mixtures {tall fescue(Rebell Jr.) 40%+ perennial ryegrass(Palmer II) 20%+ Kentucky bluegrass(Newport) 10%+ redtop(Banicuda) 10%+ red fescue(Salem) 10%+ creeping bentgrass(Crenshaw) 10%} and tall type mixtures {orchardgrass(Potomac) 40%+ tall fescue(Fawn) 20%+ Festuloium braunii(Paulita) 10%+ perennial ryegrass(Reveille) 10%+ timothy(Climax) 10%+ red clover(Kenblue) 10%+ alfalfa(Vernal) 10%} by Korean native goats. This experiment was conducted by total collection method in laboratory, 2001. The voluntary DM intake per body weight were slightly higher for turf type mixtures(28.9g) than that of tall type mixtures(26.2g), but there was no significant difference. The digestibility of cellular constituents was slightly higher for turf type mixtures than that of tall type mixtures(p<0.05), but digestibility of DM, NDF and ADF did not differ between two mixtures. The retained nitrogen percent(apparently biological value) was slightly higher for turf type mixtures(56.7%) than that of tall type mixtures(56.4%), but there was no significant difference between two mixtures. Apparently retained digestible energy was slightly higher for turf type mixtures(58.3%) than that of tall type mixtures(57.9%), but there was no significant difference between two mixtures. Based on the results, the herbage utilization by Korean native goats did not showed any significant difference between two mixtures, therefore there is a possibility that herbages of turf type mixtures can be used as a roughages by livestock.

Radioactivity Analysis of Soils Stored in KAERI for Regulatory Clearance (연구소 내 저장 중인 토양의 규제해제를 위한 방사능 분석)

  • In KAERI, about 3,100 drums containing soil have been stored. The soils were generated from the decommissioning process of Seoul office in 1988. Those soils occupy about $27\%$ of the capacity of the radioactive waste storage facility and make it difficult to maintain the storage facility. The major radioactive nuclides contained in the soils were expected to be Co-60 and Cs-137. As 16 years have passed, the radioactivity of those nuclides have decayed a lot. In this study, as a basis of regulatory clearance, radionuclides and radioactivity concentration of soils were analyzed. As a result, there are only Co-60 and Cs-137 in soils as ${\gamma}-emitters$. The total concentration of ${\gamma}-emitters$ in soil is analyzed as about $0.01\;{\sim}\;0.12$ Bq/g. As the soils are expected to be regulatory cleared in 2009, those concentrations will decay to be less than 0.1 Bq/g. This concentration can be meet the regulatory criteria suggested by IAEA. The regulatory clearance will be proceeded based on not only the assessment results of environmental influence but also related regulations.

  • From the 48 hour-cultured mycelial broth of Phellinus linteus, six compounds were isolated by means of ethyl acetate extraction, silica gel column chromatography and preparative thin layer chromatography, consecutively. Compound 1 was identified as a succinic acid by the comparison of its spectral data with authentic sample. Compounds 2 and 3 were identified as p-hydroxyphenyl acetic acid methyl ester and p-hydroxybenzaldehyde by spectroscopic studies, respectively. NMR and MS studies of compound 6 revealed that it was 2,5-dihydroxymethyl furan. Compound 4, which showed similar NMR absorptions and MS fragmentation pattern with those of compound 6 was identified as 2-hydroxymethyl-5-methoxymethylfuran. These structures were verified by the spectral data of the acetate derivatives of the compounds. Compound 5 was supposed to be a N-acetyltyramine from its $^1H-NMR$ and EI-MS data, and its structure was confirmed by a synthesis starting from tyramine.

  • The major volatiles from Allium species were found to be sulfide compounds and the ratio of sulfide to volatiles was 66.1% in garlic, 66.1% in scallion, 62.3% in green onion, 39.2% in onion, and 4.2% in chive. Trace of cyclooctasulfur was found to be present among the volatiles. The most oviposition of 17.2% occurred at diallyl sulfide and the least of 0.8% at acetylthiophene whereas the most oviposition of 43.3% occurred at ethyl alcohol if concentration was 100%. Among the organic solvents used for dilution, ethyl alcohol received the highest 52.5% of oviposition and ether the lowest of 5.9%. Furfuryl mercaptan which is also one of the volatiles, received 46.9% of oviposition. For oviposition site, D. antiqua preferred sulfides at near 1%, ethyl alcohol at high, and other volatiles at various concentrations. At 100% concentration, most volatiles except dimethyl disulfide and ethyl alcohol received less oviposition than the control which was watered sand with no volatiles added.

  • The morphological study on different types of cells of reproductive organ including spermatogenesis in the adult planaria was performed to observe their cytochemical and ultrastructural characteristics. 1. Spermatogenesis The circular luminated material appears immediately inside the nuclear envelope of early spermatid and is found also in the nucleus of sperm, but typical acrosomal structures cannot be observed. Approximately ten of small-sized mitochondria occur around the nucleus in the transitional phase from primary spermatocyte to secondary spermatocyte, but in sperm a long mitochondrion is closely associated with nucleus, parellel to long axis of it. The sperm has a relatively long head connected with two tails via hollow neck. 2. Reproductive organ The penis bulb and the bursa stalk were observed. (1) Penis bulb The cells constituted penis bulb are classified into six types on the basis of ultrastructure of the cells and cytochemistry of the cytoplasmic granules. 1) A-type cells: These cells exhibiting low electron density are mainly occupied by large nucleus. These cells possess two different types of granules: highly electron-dense round granules with an average size of $0.9{\mu}m$, and electron-dense granules exhibit PAS-positive reaction. 2) B-type cells contain PAS-positive granules with the size of about $0.4{\mu}m$. They are rich in free ribosomes and mitochondria. 3) C-type cells are found to be dark cells due to high electron-density. These cells are largely occupied by large nucleus. 4) D-type cells: These cells are seen as light cells which have poorly developed cell organelles. 5) E-type tells: These cells contain a large number of glycogen granules which occupy most of cell. 6) F-type cells: These arc parietal epidermal cells surrounding the genital antrum. These cells are characterized by their finger-like shapes and the presence of a number of electron-dense, irregularly-shaped structures inside cells. The relatively large electron-lucent granules can be also found. The F-type cells possess numerous microvilli on their free surfaces. (2) Bursa stalk The cells constituted bursa stalk are classified into 3 types on the basis of cell shapes and presences of electron-dense or electron-lucent granules. 7) G-type cells with a long cytoplasmic process. They have large nuclei and poorly developed cell organelles. 8) H-type cells: These cells are characterized by the presence of a long cytoplasmic process and relatively highly electron-dense cytoplasmic profile. They have poorly developed cell organelles. 9) I-type cells contain large electron-lucent granules which exhibit negative reactions with three kinds of cytochemical staining methods used in this experiment. The fine electron-dense structures can be found inside these granules.

Prediction of Chemical Composition and Fermentation Parameters in Forage Sorghum and Sudangrass Silage using Near Infrared Spectroscopy

  • This study was conducted to assess the potential of using NIRS to accurately determine the chemical composition and fermentation parameters in fresh coarse sorghum and sudangrass silage. Near Infrared Spectroscopy (NIRS) has been increasingly used as a rapid and accurate method to analyze the quality of cereals and dried animal forage. However, silage analysis by NIRS has a limitation in analyzing dried and ground samples in farm-scale applications because the fermentative products are lost during the drying process. Fresh coarse silage samples were scanned at 1 nm intervals over the wavelength range of 680~2500 nm, and the optical data were obtained as log 1/Reflectance (log 1/R). The spectral data were regressed, using partial least squares (PLS) multivariate analysis in conjunction with first and second order derivatization, with a scatter correction procedure (standard normal variate and detrend (SNV&D)) to reduce the effect of extraneous noise. The optimum calibrations were selected on the basis of minimizing the standard error of cross validation (SECV). The results of this study showed that NIRS predicted the chemical constituents with a high degree of accuracy (i.e. the correlation coefficient of cross validation ($R^2{_{cv}}$) ranged from 0.86~0.96), except for crude ash which had an $R^2{_{cv}}$ of 0.68. Comparison of the mathematical treatments for raw spectra showed that the second-order derivatization procedure produced the best result for all the treatments, except for neutral detergent fiber (NDF). The best mathematical treatment for moisture, acid detergent fiber (ADF), crude protein (CP) and pH was 2,16,16 respectively while the best mathematical treatment for crude ash, lactic acid and total acid was 2,8,8 respectively. The calibrations of fermentation products produced poorer calibrations (RPD < 2.5) with acetic and butyric acid. The pH, lactic acid and total acids were predicted with considerable accuracy at $R^2{_{cv}}$ 0.72~0.77. This study indicated that NIRS calibrations based on fresh coarse sorghum and sudangrass silage spectra have the capability of assessing the forage quality control
