Comparative Studies on Genetic Variabilities of Second Chromosomes in Sasang Natural and Experimental Populations of Drosophila melanogaster (자연과 실험집단내 Drosophila melanogaster 제 2 염색체의 유전적 변이성에 대한 비교 연구)

  • The genetic variabilities of second chromosomes concealed Sasang natural and experimental populations of Drosophila melanogaster have been analyzed. The experimental population was composed of D. melanognter which had the lethal-free second chromosome collected from Sasang natural population in 1982. The results were as follow; The mean frequencies of deleterious genes were estimated % be 33.33% in Sasang natural population and 31.72% in experimental population. The allelism rates in lethal genes isolated from the natural and experimental populations were calculated to be about 0.95% and 12.28%, respectively. The allelism rates between lethal genes isolated from the natural population and those of the experimental population were calculated to be about 0.01%. The mean values of elimination by frequencies of deleterious genes and allelism rates were 0.0011 in the natural population and 0.0124 in the experimental population. The frequencies of phenotypic sterility of males in the natural and experimental populations were estimated to be 1.49% and 1.36%, respectively. The frequencies of genotypic sterility of females and males were estimated to be 0.90% and 1.80% in the natural population, and that of males was 2.38% in the experimental population.

Drosophila melanogaster Is Susceptible to Vibrio cholerae Infection

  • Infection of Drosophila melanogaster adults with 6 Vibrio species revealed that V. cholerae was lethal (100% mortality) within 20 h as a result of systemic infection. Avirulent infection by V. vulnificus restricted the subsequent virulent infection by V. cholerae. The immediate transcription of antimicrobial peptides (AMPs), most notably Attacin A, was delayed in V. cholerae infection compared to V. vulnificus infection. Ectopic expression of Attacin A and Metchnikowin enhanced the survival of D. melanogaster upon V. cholerae infection. These results suggest that AMPs are important in the response to infections by Vibrio species and that the signaling pathways governing their expression may be targeted by V. cholerae virulence factors to elude the innate immunity of Drosophila.

  • In order to establish a biochemical genetic system in Drosophila populations in Korea, the $\\alpha$-glycerophosphate dehydrogenase alleles of eleven natural populations of D. melanogaster in Korea were examined by means of agarose gel electrophoresis. The results obtained are presented below: 1. $\\alpha$-Glycerophosphate dehydrogenase ($\\alpha$-GPDH) allele is scored for eleven natural populations of D. melanogaster in Korea, resulting that $\\alpha$-GPDH is found to be widely polymorphic for two electrophoretic variants. 2. The heterozygosity of $\\alpha$-GPDH is calculated as $40\\sim50%$. 3. The frequency of the FF genotype of $\\alpha$-GPDH is found to be roughly same as the SS genotype, but less than the FS genotype. 4. The F gene of $\\alpha$-GPDH is distributed almost frequently as the S gene.

  • 한국산 노랑초파리 군에 속하는 8종의 유전적 유연관계를 밝히고자 생식적 격리 그리고 수용성 단백질을 전기영동법으로 분석하였다. 생리적 격리 실험에서 교배전격리 실험결과를 Watanane와 Kawanishi model에 근거하여 보면 D. aurario complex 3종 중 D. triauror물가 원시종이며 남 auraria는 팍생종으로 나타났다. 교배후 격리 실험에서 나. melonogaster와 D. simuions의 교배시, D. melonogoster를 수컷으로 하였을 때는 불임의 수컷만이, 또 D. melonogaster를 암컷으로 곯였을 때는 불임의 암컷만이 출현하였다. 그리고 남 ouraria complex 3종간의 교배에서는 수정 능력이 있는 수컷과 암컷이 출현하였는데, 이는 아직 남 auraria Complex가 semispecies단계에 있음을 나타내는 것이라 할 수 있다. SDS-PAGE로 분석한 노랑초파리 군 8종의 band pattern을 densitometer로 scanning한 결과 남 susu상가 가장 특이하였으며, TDE에 의한 유전적 거리(Aquadro and Avise's)는 남 auror지와 봉 triauror지사이가 0.155로 가장 낮았고, D. melonogaster와 고. mfa 사이가 0.422로 가장 높았다. 본 연구의 결과를 UPGMA법 뜨로 분석하면, 한국산 노랑초파리 군 8종은 4개의 아군으로 나뉘어지며 이들은 2개의 다른 큰무리로 구분되었는데, D. suzu가기 아군, D. lutescens의 아군, D. melanogoster와 봉 simulons의 아군이 속한 큰무리와, D. mfa,0. ouroria, D. biauroria 그리고 남 triourario가 속한 다른 큰무리로 나눌 수 있다.

Proteome Analysis of Drosophila melanogaster Used 2-DE and MALDI- TOF-MS (이차원 전기영동과 펩타이드 지문 검색법을 이용한 초파리의 프로테옴 분석)

  • With the completely discovery of the Drosophila genome sequence, the next great challenge is to extract its biological information by systematic expression and to perform functional analysis of the gene. Here we reported a proteome analysis of D. melanogaster with two-dimensional electrophoresis (2-DE) and matrix-assisted laser desorption ionization time-of-flight mass spectrometer (MALDI-TOF-MS). The cell extracts of D. melanogaster, $200{\mu}g$ were resolved to more than 400 silver-stained spots by 2-DE. The most abundant protein spots were ranged from 4.0-7.5 of pI and from 15-90 kDa of molecular weight. The excised spots were destained and in-gel digested by trypsin. The masses of the resulting peptide mixtures were measured by MALDI-TOF-MS. Identified proteins were compared with measured peptide mass and a dynamic peptide searching database which is accessible via the internet. The results revealed that identified proteins were produced by 59 genes derived from 65 protein spots.

Construction of the Transgenic Drosophila melanogaster Expressing a Human Megsin Gene. (인간 신장질환 유발인자가 발현하는 형질전환 초파리 구축)

  • IgA nephropathy(IgAN) is considered to be a multifactorial disease with genetic and environmental factors contributing to its pathogenesis. The genes involved in susceptibility and progression of the disease have not yet been clearly elucidated. Megsin is an important candidate gene, predominantly expressed in glomerular mesangium and upregulated in IgAN. To understand biological function of megsin, in this work we have produced transgenic D. melanogaster fly over-expressing human megsin(actin-gal4>UAS-Megsin fly). Introduced human megsin was confirmed by RT-PCR and Western blotting, respectively. Its phenotype is melanin deficiency-abdomen and the megsin gene is stably transferred to the next generations.

A Study of the Effect of Monosodium Glutamate on the Development of Drosophila melanogaster (Monosodium Glutamate(MSG)가 초파리(Drosophila melanogaster))

  • Monosodium glutamate(MSG) is a widely used food additive. Some reports descried its positive effect and the others, negative effect on mouse, monkey, human or drosophilid flies. Because of the conflicting reports the present investigation was undertaken to study the effects of MSG on the development of Drosophila melanogaster. The two strains of D. melanogaster, Oregon-R and Sinchon-I were used and MSG as well as cane sugar (as the second control) media were prepared by adding MSG or cane sugar at various concentrations to the standard food media for the present study. Ten flies (Male 5, Female 5) were placed in each vial and the numbers of $F_1$ flies emerged from it were counted. The results are presented below: 1. The numbers of $F_1$ flies decrease as the concentrations of MSG increase, implying that MSG has an inhibitory effect in the development of D. melanogaster. 2. The effects of cane sugar show an enhancing effect rather than an inhibitory one. 3. The numbers of $F_1$ fies produced in the Sinchon-I strain are greater than in the Oregon-R. This may be due to the difference in the length of inbred period. 4. The Muller-5 test shows a negative result, suggesting that MSG may be not mutagenic.

Analysis of deleterious genes in natural and experimental populations of Drosophila melanogaster (Drosophila melanogaster 자연 및 실험집단내의 유해유전자 분석)

  • The genetic variabilities of second chromosomes of Drosophila melanogaster concealed in Busan natural and experimental populations have been analyzed by the Cy//Pm method. The experimental population was composed of D. melanogaster which had the lethal-free second chromosome, collected Sasang natural population in 1982. The frequencies of deleterious genes were estimated to be 14.3∼25.4% in Busan natural population and 26.5∼27.2% in experimental population. The allelism rates in lethal genes isolated from the natural and experimental populations were calculated to be about 0.76% and 9.76∼14.17%, respectively. The value of elimination by the frequencies of deleterious genes and allelism rate was 0.0106and the effective population size estimated to be about 430 flies at the 6570 days population.
