Isolation and Identification of Wild Yeasts from Soils of Fields in Daejeon Metropolitan City and Chungcheongnam-do, Korea (대전광역시와 충청남도 밭 토양으로부터 야생효모의 분리 및 동정)

  • This study focused on isolation and identification of wild yeasts from soils in fields near mountains and elucidation of its yeast distribution. Several kinds of yeasts were isolated from various soils of Daejeon metropolitan city and Chungcheongnam-do in Korea and identified by BLAST search of nucleotide sequences of internal transcribed spacer (ITS) region including 5.8S rRNA and D1/D2 region of 26S rDNA. Ninety-seven strains of 20 species from 61 soil samples were isolated, of which Cryptococcus podzolicus (11 strains), Debaryomyces hansenii (6 strains), and Trichosporon asahii (6 strains) were dominant species.

Wafer Level Packaging of RF-MEMS Devices with Vertical Feed-through (수직형 Feed-through 갖는 RF-MEMS 소자의 웨이퍼 레벨 패키징)

  • Wafer level packaging is gain mote momentum as a low cost, high performance solution for RF-MEMS devices. In this work, the flip-chip method was used for the wafer level packaging of RF-MEMS devices on the quartz substrate with low losses. For analyzing the EM (electromagnetic) characteristic of proposed packaging structure, we got the 3D structure simulation using FEM (finite element method). The electric field distribution of CPW and hole feed-through at 3 GHz were concentrated on the hole and the CPW. The reflection loss of the package was totally below 23 dB and the insertion loss that presents the signal transmission characteristic is above 0.06 dB. The 4-inch Pyrex glass was used as a package substrate and it was punched with air-blast with 250${\mu}{\textrm}{m}$ diameter holes. We made the vortical feed-throughs to reduce the electric path length and parasitic parameters. The vias were filled with plating gold. The package substrate was bonded with the silicon substrate with the B-stage epoxy. The loss of the overall package structure was tested with a network analyzer and was within 0.05 dB. This structure can be used for wafer level packaging of not only the RF-MEMS devices but also the MEMS devices.

Diversity of Culturable Soil Micro-fungi along Altitudinal Gradients of Eastern Himalayas

  • Very few studies have addressed the phylogenetic diversity of fungi from Northeast India under the Eastern Himalayan range. In the present study, an attempt has been made to study the phylogenetic diversity of culturable soil fungi along the altitudinal gradients of eastern Himalayas. Soil samples from 24 m above sea level to 2,000 m above sea level altitudes of North-East India were collected to investigate soil micro-fungal community structure and diversity. Molecular characterization of the isolates was done by PCR amplification of 18S rDNA using universal primers. Phylogenetic analysis using BLAST revealed variation in the distribution and richness of different fungal biodiversity over a wide range of altitudes. A total of 107 isolates were characterized belonging to the phyla Ascomycota and Zygomycota, corresponding to seven orders (Eurotiales, Hypocreales, Calosphaeriales, Capnodiales, Pleosporales, Mucorales, and Mortierellales) and Incertae sedis. The characterized isolates were analysed for richness, evenness and diversity indices. Fungal diversity had significant correlation with soil physico-chemical parameters and the altitude. Eurotiales and Hypocreales were most diverse and abundant group of fungi along the entire altitudinal stretch. Species of Penicillium (D=1.44) and Aspergillus (D=1.288) were found to have highest diversity index followed by Talaromyces (D=1.26) and Fusarium (D=1.26). Fungal distribution showed negative correlation with altitude and soil moisture content. Soil temperature, pH, humidity and ambient temperature showed positive correlation with fungal distribution.

Isolation and Identification of Wild Yeasts from Freshwaters and Soils of Nakdong and Yeongsan River, Korea, with Characterization of Two Unrecorded Yeasts (낙동강과 영산강 담수와 주변 토양으로부터 야생효모의 분리 및 동정)

  • Diverse wild yeast were isolated from freshwaters and soils of Nakdong and Yeongsan rivers in Korea and identified by the comparison of polymerase chain reaction-amplified nucleotide sequences of the internal transcribed spacer region (including the 5.8S rRNA) and D1/D2 regions of 26S rDNA, using BLAST. In total, 15 strains belonging to 9 species were isolated from 25 samples, out of which Aureobasidium pullulans and Cryptococcus bestiolae were dominant. Candida ghanaensis JSF0127 and Meira geulakonigii JSF0130 were identified as unrecorded yeasts, for which their mycological characteristics were investigated. These unrecorded yeasts formed ascospores and grew in yeast extract peptone dextrose medium containing 5% NaCl.

  • Sympathetic detonation (SD) of high explosives occurs when a detonating donor initiates neighboring acceptors. The present study focuses on the hydrodynamic simulation of one-on-one sympathetic detonation of 155 mm charge filled with PBXN-109. Both unbuffered and buffered SD configurations are performed while changing the distance between each charge, in order to investigate the detonation sensitivity to a donor initiation. The cause of a SD is by a shock impact for the unbuffered case at a close range, while at a distant range, blast fragment penetration is the primary cause. The buffers can reduce the incident sensitivity to a SD by reducing the strengths of shock wave and impinging fragments.

Analysis of Boundary Layer in Solid Rocket Nozzle and Numerical Analysis of Thermal Response of Carbon/Phenolic using Finite Difference Method (고체 로켓 노즐의 경계층 해석과 유한차분법을 이용한 탄소/페놀릭의 열반응 해석 연구)

  • The thermal response of carbon/phenolic used in a solid rocket nozzle liner was analyzed. In this paper, the numerical analysis of the thermal response of carbon/phenolic consists of (1) the integration equation of the boundary layer to obtain the convective heat transfer coefficient of the combustion gas on the rocket nozzle wall and (2) 1-D finite difference method for heat conduction of carbon/phenolic to calculate the ablation, char, and temperature. The calculated result was compared with the result of a blast-tube-type test motor. It is found that the calculated result shows good agreement with the thermal response of the test motor, except at the vicinity of the throat insert.

Flow-Chart for Influence Estimation of Underwater Blasting (수중발파의 영향평가를 위한 Flow-Chart)

  • In this study, ground vibration values and damping coefficient produced by underwater blasting were measured and analyzed. Equations of vibration, $V=K(SD)^{-0.536}$, were presented from quantitative experiment results. The K Values are classified with 1.507, 2.005 and 2.939 respectively at 50%, 90% and 95% reliability. Also, hydrospace noise in aquafarm and noise in atmosphere as well as ground vibrations were measured, and maximum values of these results were 86.8dB(A), 147.8dB(A), 0.244cm/s, respectively. Equations of hydrospace noise, $SL=293.2SD^{-0.164}$, was presented from quantitative experiment results. Also, the flow-chart for influence estimation and underwater blast design was presented from these results. The results of the study may be applied for the evaluation of the influence on aquafarm as a basic data before having main underwater blasting at construction sites.

Importance of Strain Improvement and Control of Fungal cells Morphology for Enhanced Production of Protein-bound Polysaccharides(β-D-glucan) in Suspended Cultures of Phellinus linteus Mycelia (Phellinus linteus의 균사체 액상배양에서 단백다당체(β-D-glucan)의 생산성 향상을 위한 균주 개량과 배양형태 조절의 중요성)

  • Strain improvement and morphology investigation in bioreactor cultures were undertaken in suspended cultures of Phellinus linteus mycelia for mass production of protein-bound polysaccharides(soluble ${\beta}$-D-glucan), a powerful immuno-stimulating agent. Phellineus sp. screened for this research was identified as Phellinus linteues through ITS rDNA sequencing method and blast search, demonstrating 99.7% similarity to other Phellinus linteus strains. Intensive strain improvement program was carried out by obtaining large amounts of protoplasts for the isolation of single cell colonies. Rapid and large screening of high-yielding producers was possible because large numbers of protoplasts ($1{\times}10^5{\sim}10^6\;protoplasts/ml$) formed using the banding filtration method with the cell wall-disrupting enzymes could be regenerated in relatively high regeneration frequency($10^{-2}{\sim}10^{-3}$) in the newly developed regeneration medium. It was demonstrated that the strains showing high performances in the protoplast regeneration and solid growth medium were able to produce 5.8~6.4%(w/w) of ${\beta}$-D-glucan and 13~15 g/L of biomass in stable manners in suspended shake-flask cultures of P. linteus mycelia. In addition, cell mass increase was observed to be the most important in order to enhance ${\beta}$-D-glucan productivity during the course of strain improvement program, since the amount of ${\beta}$-D-glucan extracted from the cell wall of P. linteus mycelia was almost constant on the unit biomass basis. Therefore we fully investigated the fungal cell morphology, generally known as one of the key factors affecting cell growth extent in the bioreactor cultures of mycelial fungal cells. It was found that, in order to obtain as high cell mass as possible in the final production bioreactor cultures, the producing cells should be proliferated in condensed filamentous forms in the growth cultures, and optimum amounts of these filamentous cells should be transferred as active inoculums to the production bioreactor. In this case, ideal morphologies consisting of compacted pellets less than 0.5mm in diameter were successfully induced in the production cultures, resulting in shorter period of lag phase, 1.5 fold higher specific cell growth rate and 3.3 fold increase in the final biomass production as compared to the parallel bioreactor cultures of different morphological forms. It was concluded that not only the high-yielding but also the good morphological characteristics led to the significantly higher biomass production and ${\beta}$-D-glucan productivity in the final production cultures.

Effects of Drying Method and Medicinal Herb Extract Addition on the Microstructure of Beef Jerky (건조방법과 한약재 추출물 첨가가 육포의 미세구조에 미치는 영향)

  • We investigated the effects of manufacturing method on the quality of beef jerky using electron micrography. Six types of beef jerky were prepared by the addition of sugar (A), licorice (B), one of three kinds of spice extract (clove: C, fennel fruit: D, and Chungyang green pepper extract: E), or a mixture of all spice extracts (F). Microstructural changes in beef jerky during preparation by drying, with respect to drying method and the nature of the added spice extract, were observed by scanning electron micrography (SEM) and transmission electron micrography (TEM). The latter technique showed that the microstructure of fresh meat showed actin and myosin in myofibril lines, and also mitochondria and inner membranes. Beef muscle structure was broken at many myofibril lines and decomposition of inner membrane material was evident after seasoning. SEM of air-blast dried beef jerky with added medicinal herb extracts showed both large spaces and regular myofibrils, whereas hot air-dried beef jerky had no spaces and the muscle myofibrils were still evident. After review of all available micrographs from SEM and TEM, we concluded that use of medicinal herb extracts could be helpful in preserving the muscle myofibril structure during drying, and the air-blast drying method is recommended to optimize the textural quality characteristics of beef jerky.

Utilization of Blast Furnace Slag Quenched with Water as a Source of Silicate Fertilizer -II. Effect of Particle Size Distribution of Quenched Slag on Rice Yields (급랭광재(急冷鑛滓)의 비료화(肥料化)에 관(關)한 연구(硏究) -II. 수도(水稻)에 대한 급랭광재(急冷鑛滓)의 입도별(粒度別) 비효(肥效))

  • The objective of this study was to attest the feasibility of the utilization of the blast furnace slag, quenched with water as a source of silicate fertilizer. A pot experiment was conducted to evaluate the effect of quenched slag and its particle size distribution on rice plant growth over the corresponding air-cooled slag of milled commercial silicate fertilizer. The yields of rice were slightly higher in quenched slag than in the commericial air-cooled slag, however, no significant statistical difference was observed. The silica content of rice plants through the growing period was high in quenched slag in any tested particle sizes. This indicated that the quenched slag might release silica fastly in soil. On the other hand, the available silica content retained in soil was high in air cooled slag, which implied that the silica of air-cooled slag was low acting. The results suggested that the quenched slag may be potentially utilized as a source of silicate fertilizer.
