Compound K, Ginseng Saponin Metabolite, Induces Apoptosis in Human Monocytic Leukemia cells

  • We report upon the cytotoxic activity of the ginseng saponin metabolite, Compound K (20-O-D-glucopyranosyl-20(S)-protopanaxadiol, IH90l) on various human leukemia cell lines. Compound K had most effect on U937, a human monocytic leukemia cell line, which on treatment showed; a exposure of phosphatidylserine from the inner cell membrane to the outer cell membrane, the formation of apoptotic bodies and DNA fragmentation, - characteristics of apoptosis. Compound K induced apoptosis by up-regulating Bax, disrupting the mitochondria membrane potential, and by activating caspase 9 and caspase 3. Therefore, we suggest that Compound K inhibit U937 cell growth by inducing apoptosis through the up-regulation of Bax and caspase activation.

  • Liver protective effects of ginsenosides as well as fractions of dammarane glycosides of Panax ginseng were studied using galactosamine (GalN)-induced cytotoxicity in primary cultured rat hepatocytes. Preventing effects on GalN-induced hepatotoxicity were found both microscopic observation and determination of GPT level with total dammarane glycosides fraction and $20(S)-ginsenoside-Rb_1$ as well as $20(S)-ginsenoside-Rg_1$ at the concentration of $50{\mu}g/ml$. The syntheses of both protein and RNA were significantly increased by the treatment of $50{\mu}g/ml$ of total dammarane glycoside fraction, $20(S)-ginsenoside-Rb_1$, -Rc, -Re and $-Rg_1$, respectively in both normal and GalN-induced cytotoxic hepatocytes.

Effect of Botulinum Toxin A on Proliferation and Apoptosis in the T47D Breast Cancer Cell Line

  • The present study was performed to assess the activity of the botulinum toxin A on breast cancer cells. The T47D cell line was exposed to diverse concentrations of the botulinum toxin A and cell viability and apoptosis were estimated using MTT and propidium iodine/annexin V methods, respectively. Botulinum toxin A exerted greater cytotoxic activity in T47D cells in comparison with MCF10A normal cells; this appeared to be via apoptotic processes caspase-3 and -7. In conclusion, botulinum toxin A induces caspase-3 and -7 dependent apoptotic processes in the T47D breast cancer cell line.

Roles of the Hsp90-Calcineurin Pathway in the Antifungal Activity of Honokiol

  • Honokiol, a bioactive compound isolated from the cone and bark of Magnolia officinalis, has been shown to have various activities including inhibition of the growth of Candida albicans. We investigated the roles of the Hsp90-calcineurin pathway in the antifungal activity of honokiol. The pharmacologic tool was employed to evaluate the effects of Hsp90 and calcineurin in the antifungal activity of honokiol. We also evaluated the protective effects of the calcineurin inhibitor cyclosporin A (CsA) on honokiol-induced mitochondrial dysfunction by the fluorescence staining method. The Hsp90 inhibitor potentiated the antifungal activity of honokiol. A C. albicans strain with the calcineurin gene deleted displayed enhanced sensitivity to honokiol. However, co-treatment with calcineurin inhibitor CsA attenuated the cytotoxic activity of honokiol due to the protective effect on mitochondria. Our results provide insight into the action mechanism of honokiol.

  • To know the in vitro and the in vivo cytotoicity of resin solution, resin solution was applied to cultured fibroblast and was injected into the mouse. The cytotoxic effect of resin solution was measured by MIT assay and in vivo cytotoxicity was examined after Hematoxylin and Eosin staining. The cell activity of resin solution in the concentration of 50% was significantly decreased compared to control group and 5 % group. In histopathologic study of resin solution, there were severe inflammatory cell infiltration, mild interstitial edema, trace hemorrhage, and moderate or severe muscle destruction in resin injected group. These results suggested that there might be some differences between the cell viability of fibroblast and in vivo cell cytotoxicity. Further study is needed to clarify the cytotoxicity by direct implanting of resin mass.

The Cytotoxic Action of New Ag-Porphyrin as a Potential Chemotherapeutic Agent

  • Earlier we have described new water-soluble Ag- and Zn-derivatives of tetrachloride meso-tetra (4-N-oxiethylpyridyl) porphyrin (TOEtPyP) as potential anticancer drugs. In this work the effect of one of these metal porphyrins, TOEtPyP Ag, on the cell population kinetics was studied in vitro using morphological and biochemical techniques. The results suggested that TOEtPyP Ag action consisted in the simultaneous suppression of the cell growth and activation of the cell death. About 40% of the cells were shown to die via apoptotic pathway. So, the porphyrin studied may be attributed to inducers of both necrotic and apoptotic processes. The results obtained support our previous assertion that TOEtPyP Ag may be considered as a potential chemotherapeutic agent.

  • The novel 1-(1-benzoylindoline-5-sulfonyl)-4-phenyl-4,5-dihydroimidazolones 2 shows highly potent and broad cytotoxicities. Their cytotoxicities against human lung carcinoma A549, human chronic myelogenous leukemia K562, and human ovarian adenocarcinoma SK-OV-3 are compatible with doxorubicin. Compound 2p (1-[(4-aminobenzoyl)indoline-5-sulfonyl])-4-phenyl-4,5-dihydroimidazolone) exhibits a cytotoxicity that is far more potent than doxorubicin and also exhibits highly effective antitumour activities against murine (3LL, Colon 26) and human xenograft (NCI-H23, SW620) tumor models.

  • The ethyl acetate fraction of Bupleurum longiradiatum was found to have an inhibitory effect on the tube-like formation of human umbilical venous endothelial (HUVE) cells. The active compounds, isolated from the fraction, were identified as acetylbupleurotoxin (P1) and bupleurotoxin (P2). The compounds P1 and P2 completely inhibited the tube-like formation of HUVE cells at 30 ${\mu}g$/ml, below the cytotoxic concentration. But, they did not exhibit antitumor activity on BDF1 mice bearing Lewis lung carcinoma cells despite their antiangiogenic activity.

Isolation of Isoguanosine from Croton tiglium and Its Antitumor Activity

  • This paper describes the isolation of isoguanosine from Croton tiglium L. and its cytotoxic effect anginst several tumor cell lines in culture and newly reports that isoguanosine has an antitumor activity against implanted S-180 ascitic turmor mice. Isoguanosine is effective at the dose of $24mg/kg/day{\times}5$, with T/C value of 168%. Isoguanosine inhibits the gorwth of S-180 and Ehrich solid tumor in mice at the optimal doses of $96\; mg/kg/day{\times}12$ and $48\;mg/kg/day{\times}12$, with 1-T/C values of 65% and 60% respecively.

Antimicrobial Activity of Some Schiff Bases Derived from Benzoin, Salicylaldehyde, Aminophenol and 2,4 Dinitrophenyl Hydrazine

  • The antibacterial and antifungal activities of three schiff bases were evaluated against some pathogenic bacteria and fungi. Parallel experiments were also carried out with standard drugs (Kanamycin for bacteria and Nystatin for fungi). Two compounds [N-(1-phenyl-2-hydroxy-2phenylethylidine)-2',4' dinitrophenyl hydrazine, abbreviated as PDH and N-(2-hydroxy benzylidine)-2'-hydroxy imine, abbreviated as HHP] showed significant antimicrobial activities. The rest one [N-(l-phenyl 2-hydroxy-2 phenyl ethylidine) 2'-hydroxy phenyl imine, abbreviated as PHP] showed moderate activity. All these three compounds were found to possess pronounced cytotoxic effect. These compounds can be considered as potent antimicrobial agents.
