Correlation between in vitro Flowering Frequency and the Structure of Cytokinins in Ginseng (Panax ginseng C.A. Meyer) (인삼 (Panax ginseng C.A. Meyer)의 기내 화아형성 빈도와 cytokinin 구조와의 관계)

  • To elucidate the relationship between in vitro flowering and the structure of cytokinins in ginseng (Panax ginseng C.A. Meyer), zygotic embryos, seedlings, and cotyledonary nodes were cultured on MS medium supplemented with 5 $\mu$M of various cytokinins (BA, kinetin, 2-iP, and zeatin) with or without GA$_3$ (5 $\mu$M). The frequency of in vitro flowering was the highest when explants were cultured on the medium containing BA regardless of the kinds of explants, followed by kinetin, 2-iP, and zeatin. Flowering frequency of cotyledonary node explants was significantly increased by the combined treatment of cytokinin and GA$_3$. Flowering frequency was highly correlated with the logP of cytokinins, indicating that the lipophilicity of each cytokinin may involved in the in vitro flowering of ginseng.

Proteomic Analysis of Cytokinin Induced Proteins in Arabidopsis (단백체를 이용한 애기장대 Cytokinin 유도 단백질의 분석)

  • Cytokinins are essential plant hormones that play crucial roles in various aspects of plant growth and development. To better understand the molecular mechanisms of cytokinin action, we identified cytokinin related proteins by a proteomic approach. Proteins extracted from control and trans-zeatin treated Arabidopsis seedlings were separated and analyzed by two dimensional gel analysis. Differentially expressed protein spots were identified with peptide mass fingerprinting based on matrix assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry and database searching, We obtained ten up-regulated and one down-regulated proteins upon t-zeatin treatment. The expression of the following proteins was induced; pollen allergen like protein, L-ascorbate peroxidase, tetrapyrrole methylase family protein, SGT1 protein homolog, disease resistance related protein, maternal embryogenesis control protein, paxneb related protein, gluthathione S-transferase and IAA amino acid hydrolase homolog.

Plant Regeneration and Effect of Auxin and Cytokinin on Adventitious Shoot Formation from Seedling Explant of Taraxacum platycarpum (민들레 [Taraxacum platycarpum]유식물 절편으로부터 부정아 발생에 미치는 auxin과 cytokinin의 영향 및 식물체 재생)

  • Taraxacum platycarpum has been used as a medicinal plant. We investigated optimal condition for efficient plant regeneration through adventitious shoot formation on medium with various kinds of growth regulators. Adventitious shoot formation was achieved when cytokinin was used alone. Shoot formation was higher on MS medium containing 2 mg/L BAP compared to that with 2 mg/L kinetin and 2 mg/L 2-ip. Among root, hypocotyl and cotyledon, roots were the best explant for the adventitious shoot induction. Adventitious shoot formation from roots declined markedly by the combination of both 0.1 mg/L NAA and 2 mg/L BAP, while shoot formation from cotyledons was stimulated by the same combination. Root formation from the regenerated shoots was achieved on 1/3MS medium containing 0.2 mg/L NAA. Regenerated plantlets was acclimatized and transplanted to the soil, showing 100% survival.

Improvement of Forage Crop Yield and Retardation of Leaf Senescence by Introduction of Gene for Cytokinin Synthetase into Plants (Cytokinin 합성효소의 도입에 따른 형질전환 식물체의 노화 지연 및 수량의 증가)

  • The bacterial isopentenyl transferase (ipt) gene involved in cytokinin biosynthesis was fused with 35S promoter of cauliflower mosaic virus (CaMV) and introduced into tobacco plants (Nicotiana tabacum L. cv. Samsun) via Agrobacterium-mediated transformation. As expected, ipt gene was constitutively expressed in all tissues of transgenic plants. Several primary transgenic plants were obtained that expressed different level of transcripts for ipt gene. Three of transgenic plants with different expression level of ipt gene were selected and selfed to obtain homozygous line for further analysis. A number of interesting phenotypic changes such as viviparous leaves, delayed senescence, larger axillary shoots, an abundance of tiny shoots at the apex and a release of lateral buds, were observed in transgenic plants. Chlorophyll content was 1.5- t.o 4-fold higher in transgenic plants as compared with non-transformed plants. These results indicate that the cytokinin synthesized in transgenic plants could improve forage crop yield by delay of leaf senescence and increase of leaf number.

Interaction of brassinosteroids and cytokinin in modulating light mediated signaling in Arabidopsis

  • Brassinosteroids (BRs) are a special class of plant steroid hormones that are essential for normal growth and development. Part of confusion is whether BRs are unique to plants, because they have overlapping physiological roles with other better-studied hormones and with physiological responses caused by light. In systems designed to assay for cytokinins, the effects of BRs vary. We measured hypocotyl length for testing the ability of brassinolide (BL) to rescue double mutant between det2 and the photoreceptor null mutant phytochrome B (phyB). PHYB involved in controlling hypocotyl elongation in increased concentration of BL whereas phyBdet2 double mutant just partially rescue to phyB in white and red light indicated the involvement of BRs in PHYB regulated cell elongation. BRs regulated hypocotyl growth was delayed by BAP, a cytokinin treatment but inhibitory effects of BAPs on hypocotyl growth was slightly recovered by BL. The result indicated that the mode of action of BR and cytokinin is independent or sequential in the downstream light-regulated response control on hypocotyl elongation and also light modulated the action of BR and cytokinin in some extent.

Novel Function of Cytokinin: A Signaling Molecule for Promotion of Antibiotic Production in Streptomycetes

  • Cytokinin has been known to act as a plant hormone to promote cell division and function in diverse processes in plant growth and development. Besides being produced in plants, it is also produced by various bacteria and fungi; however, its ecological significance is still unclear. In this report, we present an interesting finding that transzeatin riboside (tZR), a naturally occurring cytokinin compound, increased antibiotic production in many different streptomycetes, including Streptomyces coelicolor Ml3O, S. pristinaespiralis ATCC 25486, S. violaceoruber Tu22, S. anfibioticus ATCC l1891, and S. griseus IFO 13350. In vitro plate assays showed that the addition of 100 $\mu$M tZR increased the growth inhibition of Pseudomonas syringae pv. syringae, a plant pathogen, by S. griseus, a streptomycin producer. We suggest that cytokinin could act as a signaling molecule for antibiotic production in streptomycetes, a group of rhizosphere bacteria.

Studies on a PR4 Gene for Breeding Disease Resistant Forage Crops (내병성 목초 품종개량을 위한 PR4 유전자의 연구)

  • Cytokinins are essential plant hormones that play crucial roles in various aspects of plant growth and development. By using mRNA differential display, we isolated a cytokinine-inducible cDNA encoding pathogenesis-related (PR) 4 from Arabidopsis amp1 mutant. The full-length PR4 cDNA, designated AtPR4, contains an open reading frame of 212 amino acids with calculated molecular mass of 22,900 Da and isoelectric point (pI) of 7.89. Genomic DNA blotting showed that the Arabidopsis genome has one copy of AtPR4. AtPR4 mRNA was induced by cytokinin and NaCl, but decreased by SA or JA treatment. PR proteins are induced in response to pathogen attack. Thus the AtPR4 gene isolated in this study may be a useful candidate for genetic engineering of forage crops for increased tolerance against pathogen.

Effects of Plant Growth Regulators on the Regrowth of Perennial Ryegrass (Lolium perenne L.) (식물 생장 조절물질이 페레니얼 라이그라스의 재생에 미치는 영향)

  • Plant growth regulators were treated on the cut perennial ryegrass (Lolium perenne L. cv. Reveille) to investigate the effect on the regrowth after cutting. The growth showed better result when 0.1 or 0.5 mg/L cytokinin were treated. Among cytokinins, kinetin or 2iP gave the better effect on the growth than BAP. In 2.4-D as an auxin. cut plants grew best at the concentration of 0.1 mg/L. The initial regrowth was very vigorous when GA$_3$ was treated as a growth regulator, but the growth was retarded after 2 weeks later of cutting. Co-treatment of kinetin as a cytokinin and 2,4-D as an auxin showed synergistic effect on the regrowth of cut perennial ryegrass. Both plant growth regulators gave the same result at the same concentrations in the suspension culture of perennial ryegrass cells.
