  • Root-knot nematode damage was found on yam, Dioscorea bulbifera in Andong Korea. From the root-knots, female nematodes were isolated and subjected to DNA sequence analysis. Sequence of cytochrome oxidase subunit II (COII) was analyzed from the genomic DNA of the isolate. COII locus size and sequence of the nematode isolate were similar to those of Meloidogyne javanica or M. incognita. However, an analysis of HinfI restriction site, a species-specific character between these two species, showed that the isolate did not match to either M. javanica or M. incognita.

  • Cytochrome oxidase activities have been compared from crude sumibitochondrial preparations of rat skeletal muscle tissues. Fast red (type $II_A$) preparation has highest cytochrome oxidase activity, slow red (type I) next, and fast white (type $II_B$) the lowest. Differences of electrophoretic mobilities have been detected by heme staining. Migration of heme band is in the order of slow red>fast red>fast white from fastest to slowest. Results of immunoelectrophoresis have substantiated the above finding.

  • The topology of the enzyme has been investigated by biochemical studies including chemical labeling and cross linking. Thirteen subunits(polypeptides) of the cytochrome-c-oxidase have localistic characteristics of existing in the matrix side or cytoplasmic side in the mitochondria. In order to observe the distribution of the enzyme subunit on the mitochondria membrane, immunogold-labeling methods were employed. Antibody was obtained from the serum of immunized rabbit with enzyme subunit antigen which was obtained from cytochrome-c-oxidase of the beef heart muscle mitochondria. Beef heart muscle tissue as a tissue antigen was stained with immunized rabbit IgG and protein A gold complex. Electron microscopy has identified the existance of cytochrome-c-oxidase subunit $Mt_I,\;Mt_{II}\;and\;Mt_{III}$ on the membrane of cristae and outer chamber of mitochondria and the subunit $C_{IV}$ on the membrane of cristae and matrix of mitochondria. Particularly, the subunit $C_{IV}$ was also observed to exist in the sarcoplasm of muscle tissue.

Effect of Butylated Hydroxytoluene and 2-Acetylaminofluorene Administration and Microsomal Mixed Function Oxidase System in Young Rats fed different Fats (Butylated Hydroxytoluene첨가 식이 및 2-Acetylaminofluorene 투여가 식이지방을 달리한 쥐간의 Microsomal Mixed Function Oxidase계에 미치는 영향)

  • Sprague-Dawley male rats were fed the diet of p/s 4.0(soybean oil : I), p/s 0.08(Beef tallow : II) at the level of 15% fat until 8 weeks after weaning. I & II groups were divided into 4 sub-groups by diets with or without 0.3% butylated hydroxytoluene(BHT). 2-AAF was injected at the age of $5_{1/2}$, 6, $5_{1/2}$, 7 weeks. MFO system enzyme(cytochrome p-450, cytochrome p-450 reductase, cytochrome b5) activities and lipid peroxide were determined from isolated liver microsome. 2-AAF injected young rats had growth retardatiion. Lipid peroxide values were not influenced greatly by dietary fat, 2-AAF and BHT. Cytochrome p-450 contents were increased in I-BHT-AAF & II-AAF groups by 2-AAF and its contents were not affected by BHT. But cytochrome p-450 and cytochrome p-450 reductase were not increased in soybean oil diet ybean oil groups. Cytochrome b5 was not influenced by dietary fat, 2-AAF and BHT. Cytochrome p-450 and lipid peroxide, cytochrome p-450 reductase and cytochrome b5, which transfer to MFO system, appeared to have positive correlations(r=0.2474, r=0.2475, p<0.05) each other. This result suggests that MFO system metabolizing 2-AAF was influenced by dietary fats and BHT. 2-AAF induced growth retardation in young rats.

Relationship between Genetic Variants of Mitochondrial DNA and Growth Traits in Hanwoo Cattle

  • Genetic variants of Hanwoo mtDNA in the region of cytochrome oxidase subunit I, II and III complex were detected using restriction enzymes. PCR primers were designed based on the bovine mtDNA sequence, and 6 primer sets (Mt4, Mt5, Mt6, Mt7, Mt8 and Mt9) were used. A total of 20 restriction enzymes were used, and 6 restriction enzymes, which were Hinf I, Pvu II, Rsa I, Eco RI, Bgl II, and Msp I, showed genetic polymorphisms. Significant associations between genetic variants and weight traits were observed at WT15 (p<0.05) and WT18 (p<0.01) with Pvu II for Mt9, Bgl II for Mt6 and Rsa I for Mt8 segments in the region of cytochrome oxidase subunit complex. Significant associations were also observed at Mt9-Pvu II and Mt6-Bgl II segments for WT9 (p=0.01), WT12 (p=0.02), respectively. These results suggest that genetic variants of mtDNA in the region of cytochrome oxidase subunit complex may be candidate segments for improvement of animal growth as weight traits.

Effects of Mitochondrial DNA Polymorphism on Growth Traits of Hanwoo (mt DNA 다형이 한우 성장에 미치는 영향)

  • 한우의 mt DNA cytochrome oxidase subunit I, II, 및 III complex지역의 유전적 다형현상을 제한효소를 이용하여 검출하였다. PCR primer 6종에 대하여 20가지 제한효소를 처리하였으며, Pst I, Pvu II, Rsa I, Eco RI, Bgl II, and Msp I 제한효소를 사용하여 유전적 변이를 검출하였다. 검출된 변이체와 한우의 성장과의 관련성을 조사한 결과 cytochrome oxidase subunit III complex 지역의 유전염기서열을 근거로 제작한 primer Mt9 좌위에서 제한효소 PvuII를 이용한 절단형과 체중형질 인 WT15(P<0.05) 및 WT18(P<0.01)에서 고도의 유의성이 관찰되었다. 아울러 , Mt9-Pvu II(P=0.07), Mt6-Bgl II(P=0.05), and Mt8-Rsa I(P=0.05) 좌위 또한 WT9, WTl5, and WT15에서 각각 통계적 유의성이 관찰되었다. 따라서 본 결과는 cytochrome oxidase subunit III complex segments가 candidate gene으로서 기초적 유전정 보 제공은 물론 유전적 개량을 위해 사용될 수 있을 것으로 사료된다.

Cloning and Sequencing of the Mitochondrial Cytochrome c Oxidase Subunit II Gene from Rhabditidae Family Nematode (Rhabditidae과 선충의 CO II 유전자 클로닝 및 염기서열 분석)

  • Cytochrome c oxidase subunit II gene(CO II gene) is subunit of cytochrome oxidase, which is complex IV of mitochondria electron transport system. It has been frequently used in molecular phylogenetic studies because the speed of its DNA variation is faster than that of nucleus. It is especially useful in phylogenetic study of molecular biology in insects. In this study, we cloned and sequenced CO II gene of mitochondria DNA from Rhabditidae family nematode. Our results showed that this gene is comprised of 696 base pairs(bp). In the analysis of similarity of this gene with other known genes of 14 species of nematodes in Rhabditida order, we identified that this gene has high similarity with that of Caenorhabditis briggsae(86.0%) and C. elegans(85.6%) in Rhabditidae family. On the meanwhile, it has very low similarity with that of Angiostrongylus cantonensis(31.8%) in Angiostrongylidae family and Metastrongylus salmi(31.6%) in Metastrongylidae family. Based on the results of this study, we suggest that this nematode is closely related with that of Caenorhabditis genus in Rhabditidae family.

Cytochemical Study on the Cytochrome Oxidase Activity and Myocardial Adaptation to Treadmill Exercise in Rat (흰쥐에서 Treadmill 훈련에 따른 심근의 적응과 cytochrome oxidase 활성에 관한 세포화학적 연구)

  • The present study was to elucidate the cytochemical study on the cytochrome oxidase (CO) activity and myocardial adaptation to treadmill exercise in rat. The three month Sprague-Dowley male $(150{\pm}10g)$ were used in experimental animal. The experimental groups were divided into 2 groups: the normal sedentary group and the treadmill exercise group. On each 1st and 3rd day, 1st, 4th, 8th, and 12th experimental week four rats of each group were sacrificed for tests. The morphometrical measurements were used to evalute the change of heart weight, rate of myocardial fibers to capillaries, and cytochemical study of CO activities, using light and electron microscopy. The results were as follows: The heart weights were more increased in the treadmill exercise group than those of their sedentary group. The rate of myocardial fibers to capillaries were not changed in sedentary group, but those were significantly from 4th weeks in the treadmill exercise group. The CO activity was not changed in sedentary group, but increased in treadmill exercise group after 1 st week. I and III types of CO activity were increased In sedentary group, in contast to II and III types in treadmill exercise group on electron micrographic study. These results suggest that, the treadmill exercise-induced changes in CO activity and rate of myocardial fibers to capillaries appear to be related to exercise, and the adaptive response seems to occurs from 4th week of treadmill exercise.

Occurrence of Phytopythium vexans Causing Stem Rot on Anthurium andraeanum in Korea

  • In 2017, wilting symptom was observed on seedlings of Anthurium andraeranum in Youngin, Korea. Brownish lesions with water soaking were developed on the stems and roots of the infected plants. The stems and leaves wilted and finally died. One fungal isolate was obtained in pure culture. Morphological features and nucleotide sequences of internal transcribed spacer rDNA and cytochrome oxidase subunit II mt DNA were analyzed. The results of this study indicated that the fungus is identified as Phytopythium vexans. Pathogenicity tests showed the isolate was pathogenic to the seedlings of A. andraeanum. To our knowledge, this is the first report of P. vexans causing stem rot on A. andraeanum in Korea.
