Molecular Identification of Adenocephalus pacificus (Cestoda) from Three Human Cases in Lima Province, Peru

  • The Pacific tapeworm Adenocephalus pacifcus (syn. Diphyllobothrium pacificum) is a causative agent of diphyllobothriosis occurred in Pacific coast of South America, mainly in Peru. Source of infections are traditional meal from raw or undercooked marine fish such as "cebiche". We confirmed 3 new cases, one including scolex and the other two headless. A strobila 46 cm long without scolex was discharged from an 8-year-old boy before treatment. Specimens were confirmed morphologically by presence of tegumental protuberances on proglottids and small sized eggs. Partial sequence of cytochrome c oxidase subunit 1 gene was congruent with A. pacificus sequences.

Effects of dietary lipid sources on apoptotic and immune gene expression in head kidney of olive flounder (Paralichthys olivaceus)

  • It can be hypothesized that dietary fatty acids can modulate immune responses in fish by inducing apoptosis of immune cells since dietary polyunsaturated fatty acid (PUFA) increase apoptosis by oxygen radicals generated by peroxidation. Thus we examined the effects of deferent dietary lipid sources such as squid liver oil (FO), linseed oil (LO) and soybean oil (SO) on oxidation (Cytochrome C oxidase; COS), apoptosis (TNF-${\alpha}$ Scinderin like) and immune (IL-$1{\beta}$ and NKEF) gene expression in the main immune organ (head kidney) in olive flounder (Paralichthys olivaceus) by Q-PCR analysis after feeding diets containing each oil (5%) for 15 weeks. Linseed oil and soybean oil were chosen to compare n-3 or n-6 enriched vegetable oils, respectively. Consequently, COS, TNF-${\alpha}$ and Scinderin like gene expression was increased in SO group, indicating the induction of oxidation and apoptosis. Meanwhile, no significant difference was found in immune gene expression. In conclusion vegetable oils containing n-3 PUFA like linseed oil seems to be more suitable lipid source than soybean oil for replacement of fish oil in flounder since n-6 PUFA in SO leads to activation of apoptosis pathways within the cellular damage in head kidney.

First Record of the Interstitial Annelid Pharyngocirrus uchidai (Annelida: Saccocirridae) from Korea, Confirmed by Topotypic DNA Barcoding Data from Japan

  • The marine interstitial annelid Pharyngocirrus uchidai(Sasaki, 1981) has been only known from Japan. In this study, we report the occurrence of P. uchidai for the first time in four localities along the eastern coast of Korea: Bukmyeon, Gamchu, Gase, and Oeongchi. Species identification was confirmed by comparison of DNA barcoding sequences with morphological examination from the type locality, Oshoro, Japan. We generated a total of 25 sequences of a partial segment (580 bp) of the cytochrome c oxidase subunit I gene (COI), representing five specimens from each locality. Maximum intra-specific variation was 1.2% in terms of Kimura two-parameter (K2P) distance, observed between two individuals each from Gamchu (i.e., between two specimens from the single locality), Gamchu and Oeongchi, Gamchu and Oshoro, and Oeongchi and Oshoro. On the other hand, an identical haplotype was found in all the five localities, substantiating our species identification for the Korean populations. Inter-specific K2P distance between P. uchidai and an unidentified Saccocirrus sp. from Canada (based on public database entries) was 22.4-23.4%.

Occurrence and Molecular Identification of Microcotyle sebastis Isolated from Fish Farms of the Korean Rockfish, Sebastes schlegelii

  • Microcotyle sebastis is a gill monogenean ectoparasite that causes serious problems in the mariculture of the Korean rockfish, Sebastes schlegelii. In this study, we isolated the parasite from fish farms along the coasts of Tongyeong, South Korea in 2016, and characterized its infection, morphology and molecular phylogeny. The prevalence of M. sebastis infection during the study period ranged from 46.7% to 96.7%, and the mean intensity was 2.3 to 31.4 ind./fish, indicating that the fish was constantly exposed to parasitic infections throughout the year. Morphological observations under light and scanning electron microscopes of the M. sebastis isolates in this study showed the typical characteristics of the anterior prohaptor and posterior opisthaptor of monogenean parasites. In phylogenetic trees reconstructed using the nuclear 28S ribosomal RNA gene and the mitochondrial cytochrome c oxidase I gene (cox1), they consistently clustered together with their congeneric species, and showed the closest phylogenetic relationships to M. caudata and M. kasago in the cox1 tree.

Ocular Sparganosis: The First Report of Spirometra ranarum in Thailand

  • A 22-year-old Thai man from the Northeast region presented with acute eye swelling, itching, and discharge on his left eye. He was suspected of having gnathostomiasis and treated with albendazole and prednisolone for 3 weeks. Nine months later, he was treated with high-dose oral prednisolone for the preliminary and differential diagnoses with thyroid-associated orbitopathy and lymphoma. He had been administered prednisolone intermittently over a few years. Then he developed a painless movable mass at the left upper eyelid and recurrent pseudotumor oculi was suspected. The surgical removal of the mass was performed. A white pseudosegmented worm revealed a definite diagnosis of ocular sparganosis by a plerocercoid larva. Molecular diagnosis of the causative species was made based on the mitochondrial cytochrome c oxidase subunit 1 (cox1) gene. Proper technique of extraction and amplification of short fragments DNA from formalin-fixed paraffin-embedded tissue successfully identified parasite species. The result from the sequencing of the PCR-amplified cox1 fragments in this study showed 99.0% sequence homology to Spirometra ranarum. This is the first report of S. ranarum in Thailand.

DNA Barcoding of Benthic Ragworms of the Genus Nectoneanthes (Polychaeta: Nereididae) Collected in Korean Waters

  • To provide better taxonomic information of the genus Nectoneanthes, the two DNA barcode regions of mitochondrial cytochrome c oxidase subunit I (COI) and 16S ribosomal DNA (rDNA) sequences of Nectoneanthes oxypoda and N. uchiwa were determined. In addition, the respective sequences of four nereidid species closely related to Nectoneanthes were retrieved from GenBank for comparison and to estimate intra- and inter-specific genetic distances. The aligned sequence lengths of COI and 16S rDNA were 570 bp and 419 bp long, respectively. The mean intraspecific variation in both markers was less than 1% in all species except for that in COI of H. diadroma (1.87%). The mean interspecific variation between N. oxypoda and N. uchiwa was 12.02% regarding COI and 1.85% regarding 16S rDNA. In contrast, the mean interspecific variation between species of other genera was comparably higher(i.e., genus Perinereis: 20.5% in COI and 8.3% in 16S rDNA; genus Hediste: 13.18% in COI and 2.64% in 16S rDNA), compared with that between the two Nectoneanthes species. This result indicated that these Nectoneanthes species are genetically more closely related than other congeneric species of different genera. The DNA barcoding information on Nectoneanthes species generated in this study provides valuable insights for further biodiversity studies on nereidid species.

First record of Brachaluteres ulvarum (Tetraodontiformes: Monacanthidae) from Jeju-do Island, southern Korea

  • A single specimen (39.4 mm standard length) of Brachaluteres ulvarum, belonging to the family Monacanthidae has been recorded for the first time from Supseom, Jeju-do Island of southern Korea on 6 March 2021. This species was characterized by the presence of a small second dorsal spine present, the posterior end of pelvic without encasing scales, and numerous thin dark longitudinal lines on body. It was morphologically similar to the B. taylori, but distinguished by the ratio of the caudal peduncle to the head length (B. ulvarum, 2.5 to 3.7 times; B. taylori, 3.5 to 5.2 times). In addition, the former is well distinguished from the latter through mitochondrial cytochrome c oxidase subunit I (COI) sequences by genetic distances of range from 0.088 to 0.090. The new Korean name "Kko-ma-jwi-chi- sog" is proposed for the genus Brachaluteres, and "Kko-ma-jwi-chi" for the species B. ulvarum.

A New Record of Prionospio depauperata (Annelida: Polychaeta: Spionidae) with DNA Barcoding Data of Four Prionospio Species in South Korea

  • In this study, Prionospio depauperata Imajima, 1990 is newly reported in Korean fauna. Prionospio depauperata can be distinguished from other relatives by the four pairs of branchiae which are pinnate on chaetigers 2 and 5, and apinnate on chaetigers 3 and 4; caruncle extending to the end of chaetiger 2; and moderate dorsal crest present on chaetigers 7-13. The morphological diagnosis of P. depauperata are provided with the photographs of four Prionospio species. The mitochondrial cytochrome c oxidase subunit 1 (CO1), 16S ribosomal DNA (16S rDNA), and the nuclear 18S ribosomal DNA (18S rDNA) sequences of four Prionospio species from Korean waters, P. depauperata Imajima, 1990, P. japonica Okuda, 1935, P. krusadensis Fauvel, 1929, and P. membranacea Imajima, 1990, were determined for the first time. The inter-specific genetic distances among the congeners of four Prionospio species were 22.3-29.6% in CO1, 10.5-25.0% in 16S rDNA, and 0.3-3.6% in 18S rDNA.

Phylogeography of Gloiopeltis furcata sensu lato (Gigartinales, Rhodophyta) provides the evidence of glacial refugia in Korea and Japan

  • The present-day genetic structure of macroalgal species reflects both geographical history and oceanic circulation patterns as well as anthropogenic introduction across native ranges. To precisely understand the genetic diversity and how the factors shape the current population structure of Gloiopeltis furcata sensu lato, we determined the mitochondrial 5' end of cytochrome c oxidase subunit I (COI-5P) sequences for 677 individuals sampled from 67 sites spanning almost the entire distribution range in Korea and Japan. Results from the phylogenetic analysis and haplotype distribution revealed eleven distinct lineages within G. furcata s.l. along the Korea-Japan coastal areas and displayed divergent phylogeographic patterns among lineages. Despite the closely related lineages distributed in same habitats as high rocky intertidal zone, they display different phylogeographic patterns among lineages. The populations from the south of Korea-Japan harbored the highest genetic diversity and unique endemism in comparison with other populations in the distribution range. This could be the evidence of southern refugia for G. furcata s.l. in the Northwest (NW) Pacific and the recent migration from native to introduced region. The reason is that an exceptional distribution pattern was found high genetic diversity in Hakodate of Japan where is the northern location in the NW Pacific. Our results imply the contemporary influence on the distribution due to current circulation pattern and anthropogenic effects. These phylogeographic findings provide the important insight into cryptic species diversity and the detailed distribution pattern of Gloiopeltis in the NW Pacific.

봄철 제주 남부해역 난·자치어의 수직 분포 (Vertical Distribution of Icthyoplankton in the Southern Waters of Jeju Island During Spring)

  • The vertical distribution and abundance of icthyoplankton in the southern waters of Jeju Island during June 2020 were investigated. Fish eggs and larvae were identified using the mitochondrial DNA cytochrome c oxidase subunit I (mtDNA COI) and the 16S rRNA gene. During this period, fish eggs of 23 taxa belonging to 21 families and larvae of 27 taxa belonging to 25 families were collected. Fish eggs were located mostly from the surface to 30 m depth of the water column. Larvae were located from the surface to 80 m depth of the water column. Vertical distributions of fish eggs and larvae were influenced by oceanography conditions such as temperature, salinity, and thermocline depth. No discernible difference in mean thermocline depth was observed between day and night.
