HT-29 인체 대장암 세포에서 검정콩 된장의 in vitro 항암 효과 (Anticancer Effects of Black Soybean Doenjang in HT-29 Human Colon Cancer Cells)

  • 시료 된장의 pH, 아미노태, 암모니아태 수치는 각 군 간 특별한 차이는 보이지 않았다. 검정콩 된장이 가장 높은 폴리페놀 농도, DPPH를 이용한 항산화 효과를 보였다. 된장 추출물은 0.1~0.5 mg/mL 범위까지는 RAW 264.7 cells의 증식을 억제하지 않았으며, HT-29를 이용한 MTT에서 BD 군이 가장 높은 암세포 성장 억제율을 보였다. HT-29에서 pro-inflammatory cytokine인 $TNF-{\alpha}$, IL-6와 염증관련 인자 COX-2의 mRNA 발현은 시료 처리군에서 대조군에 비해 낮은 수치를 나타냈으며 BD군에서 가장 낮은 수치를 보였다(P<0.05). 세포 증식에 관여하는 p21, p53과 cyclin D1의 mRNA 발현은 p21과 p53가 BD군에서 발현이 증가하였고, cyclin D1은 BD군에서 낮아졌다. Apoptosis 관련 유전자인 Bcl-2는 BD군이 가장 낮은 발현을 보였다. 이상의 결과로 BD군은 CD, SD군에 비해 높은 폴리페놀 농도, 항산화 효과, 대장암세포에서 pro-inflammatory cytokines과 세포 증식에 관여하는 유전자 등을 조절한다. 이 결과는 아마도 서목태 된장의 높은 총 페놀화합물의 양과 안토시아닌의 함량으로 얻어진 결과로 생각된다.

A Bacterial Metabolite, Compound K, Induces Programmed Necrosis in MCF-7 Cells via GSK3β

  • Ginsenosides, the major active component of ginseng, are traditionally used to treat various diseases, including cancer, inflammation, and obesity. Among these, compound K (CK), an intestinal bacterial metabolite of the ginsenosides Rb1, Rb2, and Rc from Bacteroides JY-6, is reported to inhibit cancer cell growth by inducing cell-cycle arrest or cell death, including apoptosis and necrosis. However, the precise effect of CK on breast cancer cells remains unclear. MCF-7 cells were treated with CK ($0-70{\mu}M$) for 24 or 48 h. Cell proliferation and death were evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and flow cytometry assays, respectively. Changes in downstream signaling molecules involved in cell death, including glycogen synthase kinase $3\beta$ ($GSK3\beta$), $GSK3\beta$, $\beta$-catenin, and cyclin D1, were analyzed by western blot assay. To block $GSK3\beta$ signaling, MCF-7 cells were pretreated with $GSK3\beta$ inhibitors 1 h prior to CK treatment. Cell death and the expression of $\beta$-catenin and cyclin D1 were then examined. CK dose- and time-dependently inhibited MCF-7 cell proliferation. Interestingly, CK induced programmed necrosis, but not apoptosis, via the $GSK3\beta$ signaling pathway in MCF-7 cells. CK inhibited $GSK3\beta$ phosphorylation, thereby suppressing the expression of $\beta$-catenin and cyclin D1. Our results suggest that CK induces programmed necrosis in MCF-7 breast cancer cells via the $GSK3\beta$ signaling pathway.

백혈병세포주 Jurkat의 세포주기 억제에 미치는 합환피(Albizzia julibrissin) 물 추출물의 효과 (Effect of the Water Extract of Albizzia julibrissin on Cell Cycle Progression in the Human Leukemic Jurkat Cells)

  • Albizzia julibrissin belonging to the family Leguminosae has been used for the treatment of contusion, sore throat, amnesia, and insomnia in Oriental traditional medicine. The water extract of A. julibrissin induced apoptosis in Jurkat T-acute lymphoblastic leukemia (ALL) cells as measured by cell morphology. The capability of this herb medicine to induce apoptosis was associated with proteolytic cleavage of specific target protein such as beta-catenin protein suggesting the possible involvement of caspases. The purpose of the present study is also to investigate the effect of A. julibrissin on cell cycle progression. Our results showed that GI checkpoint related gene products (cyclin D1, cyclin dependent kinase 4, retinoblastoma, E2F1) were decreased in their protein levels in a dose-dependent manners after treatment of the extract. These results indicate that the increase of apoptotic cell death by A. julibrissin may be due to the inhibition of cell cycle progression in wild type p53-lacking Jurkat cells.

Tannic acid-induced apoptosis in FaDu hypopharyngeal squamous cell carcinoma

  • Tannic acid (TA) is a water-soluble polyphenol compound found in various herbal plants. We investigated the chemopreventive effects of TA on FaDu hypopharyngeal squamous carcinoma cells. In an 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, TA showed dose-dependent cytotoxicity with a half maximal inhibitory concentration (IC50) of 50 ?M. Cell cycle analysis and immunofluorescence imaging demonstrated that under low-dose ($25{\mu}M$) treatment, FaDu cells were arrested in G2/M phase, and as the dose of TA was increased, apoptosis was induced with the increase of cell population at sub-G1 phase. The expressions of various cyclins, including cyclin D1 and cyclin-dependent kinases (CDK-1 and CDK-2), were down-regulated at low doses of TA, whereas apoptotic effectors such as cleaved caspase 3, cleaved caspase 7, and poly (ADP-ribose) polymerase (PARP) were expressed in a dose-dependent manner in Western blotting. In addition, TA-induced apoptosis of FaDu cells might be mediated by the extracellular signal-regulated kinase (ERK)/mitogen-activated protein kinase pathway, with the upregulation of p-AKT/p-PKB (phosphorylated protein kinase B) and p-ERK. Overall, our data support the hypothesis that TA is a potential candidate agent for the treatment of hypopharyngeal cancer.

Anti-cancer Effect of Apigenin on Human Breast Carcinoma MDA-MB-231 through Cell Cycle Arrest and Apoptosis

  • Apigenin, a common natural product that is found in many plants and vegetables, has been reported to have many biological activities, including antioxidative, anti-inflammatory, and anticancer effects. The triple-negative breast carcinoma cell line MDA-MB-231 is known to be highly invasive and resistant to chemotherapy. In this study, we investigated the anticancer effect of apigenin on human MDA-MB-231 cells. First, the cytotoxicity of apigenin toward MDA-MB-231 cells was analyzed by MTT assay. Then, the cell cycle and apoptotic effects of apigenin were examined, and the molecular mechanism underlying its anticancer activity was explored. Apigenin inhibited the growth of the cells in a dose-dependent manner, correlating with the cell cycle arrest at the G2-M phase as well as an increase of early apoptosis. The cell-cycle inhibitory effect was highly associated with the increased expression of p21 and decreased expression of CDK6, cyclin D1, and cyclin B1. The induction of apoptosis by apigenin was associated with the upregulated expression of cleaved PARP and cleaved caspase-3, -7, and -9.

인체혈구암세포 U937의 D-Ala2-Leu5-enkephalin처리에 의한 세포 주기 억제 효과 (Cell Cycle Arrest by Treatment of D-Ala2-Leu5-enkephalin in Human Leukemia Cancer U937 Cell.)

  • 동면 개시인자로 알려진 DADLE는 여러 연구에 의해 in vivo와 in vitro 상에서 유사 동면 상태를 야기한다. 본 연구는 인체혈구암세포인 U937 세포주의 세포 사멸과 세포 주기 둥에 대한 DADLE의 영향을 살펴보았다. DADLE가 처리된 U937세포는 8${\sim}6$10 ${\mu}$M의 높은 농도에서 세포 증식이 감소하였으며, 0${\sim}6$ ${\mu}$M의 낮은 농도에서 영향이 없었다. DNA flow cytometer를 이용하여 세포 주기를 분석해본 결과 DADLE에 의한 세포 주기 억제가 관찰되었다. DADLE처리에 따른 세포 증식률 감소 및 세포 주기 억제효과를 전사 수준에서 조사한 결과 Bcl-XL, c-IAP-2의 발현 및 survivin의 발현 감소가 관찰되었으며, COX-2의 발현 역시 COX-1의 변화 없이 감소함을 확인하였다. 또한, cyclin E 와 cdk-2, -4 그리고 -6의 발현 역시 감소하는 것을 관찰하였다. Telomere 조절 관련 유전자의 경우도 c-myc과 TERT의 감소, 그리고 TEP-1가 증가하는 현상을 관찰하였다. 이상의 결과는 DADLE를 U937 암세포주에 처리했을 때 세포 주기의 억제를 통하여 life-time을 증가시킬 가능성을 시사하며 이에 관한 지속적인 연구가 필요할 것으로 사료된다.

Garcinol, an Acetyltransferase Inhibitor, Suppresses Proliferation of Breast Cancer Cell Line MCF-7 Promoted by 17β-Estradiol

  • The acetyltransferase inhibitor garcinol, a polyisoprenylated benzophenone, is extracted from the rind of the fruit of Garcinia indica, a plant found extensively in tropical regions. Anti-cancer activity has been suggested but there is no report on its action via inhibiting acetylation against cell proliferation, cell cycle progression, and apoptosis-inhibtion induced by estradiol ($E_2$) in human breast cancer MCF-7 cells. The main purposes of this study were to investigate the effects of the acetyltransferase inhibitor garcinol on cell proliferation, cell cycle progression and apoptosis inhibition in human breast cancer MCF-7 cells treated with estrogen, and to explore the significance of changes in acetylation levels in this process. We used a variety of techniques such as CCK-8 analysis of cell proliferation, FCM analysis of cell cycling and apoptosis, immunofluorescence analysis of NF-${\kappa}B$/p65 localization, and RT-PCR and Western blotting analysis of ac-H3, ac-H4, ac-p65, cyclin D1, Bcl-2 and Bcl-xl. We found that on treatment with garcinol in MCF-7 cells, $E_2$-induced proliferation was inhibited, cell cycle progression was arrested at G0/G1 phase, and the cell apoptosis rate was increased. Expression of ac-H3, ac-H4 and NF-${\kappa}B$/ac-p65 proteins in $E_2$-treated MCF-7 cells was increased, this being inhibited by garcinol but not ac-H4.The nuclear translocation of NF-${\kappa}B$/p65 in $E_2$-treated MCF-7 cells was also inhibited, along with cyclin D1, Bcl-2 and Bcl-xl in mRNA and protein expression levels. These results suggest that the effect of $E_2$ on promoting proliferation and inhibiting apoptosis is linked to hyperacetylation levels of histones and nonhistone NF-${\kappa}B$/p65 in MCF-7 cells. The acetyltransferase inhibitor garcinol plays an inhibitive role in MCF-7 cell proliferation promoted by $E_2$. Mechanisms are probably associated with decreasing ac-p65 protein expression level in the NF-${\kappa}B$ pathway, thus down-regulating the expression of cyclin D1, Bcl-2 and Bcl-xl.

Cyclin D1 Gene G870A Variants and Primary Brain Tumors

  • Alterations of cyclin D1, one of the main regulators of the cell cycle, are known to be involved in various cancers. The CCDN1 G870A polymorphism causes production of a truncated variant with a shorter half-life and thus thought to impact the regulatory effect of CCDN1. The aim of the present study was to contribute to existing results to help to determine the prognostic value of this specific gene variant and evaluate the role of CCDN1 G870A polymorphism in brain cancer susceptibility. A Turkish study group including 99 patients with primary brain tumors and 155 healthy controls were examined. Genotypes were determined by polymerase chain reaction-restriction fragment length polymorphism analysis. The CCDN1 genotype frequencies in meningioma, glioma and control cases were not significantly different (p>0.05). No significant association was detected according to clinical parameters or tumor characteristics; however, a higher frequency of AG genotype was recorded within patients with astrocytic or oligoastrocytic tumors. A significant association between AG genotype and gliobilastoma multiforme (GBM) was recorded within the patients with glial tumors (p value=0.048 OR: 1.87 CI% 1.010-3.463). According to tumor characteristics, no statistically significant difference was detected within astrocytic, oligoasltrocytic tumors and oligodentrioglias. However, patients with astrocytic astrocytic or oligoastrocytic tumors showed a higher frequency of AG genotype (50%) when compared to those with oligodendrioglial tumors (27.3%). Our results indicate a possible relation between GBM formation and CCDN1 genotype.

녹용(鹿茸)이 in vitro에서 자궁근종세포(子宮筋腫細胞)에 미치는 영향 (Effects of Cervi Pontotrichum Cornu on Human Uterine Leiomyoma Cell in vitro)

  • Purpose: This study was conducted to investigate the effects of Cervi Pontotrichum Cornu extract solution on the cell cycle regulation and apoptosis in human leiomyoma cell. Methods: The leiomyoma cell of patients was used in the study, and we administered the extract solution of Cervi Pontotrichum Cornu concentration at 1, $10mg/m{\ell}$ to the leiomyoma cell for 48 hours. We used flow cytometry and western blotting to confirm cell cycle and apoptosis. Results: In flow cytometry, G1 phase of the $1mg/m{\ell}$ group prolonged. But G1 phase of $10mg/m{\ell}$ group was shortened and S phase was increased. Cyclin D1 expression increased in higher concentration group. And Bax expression that regulates cell apoptosis increased in $1mg/m{\ell}$ and $10mg/m{\ell}$ group than control group. Bcl-2 expression decreased in 1, $10mg/m{\ell}$ groups than control group. VEGF expression rised in higher Cervi Pontotrichum Cornu concentration group. Conclusion: This study means that Cervi Pontotrichum Cornu could induce the apoptosis of leiomyoma cell by increasing Bax and decreasing Bcl-2 expression. But Cervi Pontotrichum Cornu could increase Cyclin D1 and VEGF expression, so more detailed studies would be needed.

마우스의 대뇌조직에서 방사선에 의한 아포토시스와 세포주기의 조절 (Regulation of Apoptosis and Cell Cycle in Irradiated Mouse Brain)

  • 목적 : 마우스 대뇌조직에 방사선이 조사되었을 경우 아포토시스와 세포주기의 조절작용에 어떤 영향을 미치는 지를 연구하고자 하였다. 대상 및 방법 : 8주간 성숙된 C57B1/6J 마우스의 전뇌에 코발트 방사선조사기로 25 Gy의 방사선을 단일 조사하였다. 방사선조사후 1, 2, 4, 8, 24시간 간격으로 마우스를 경추 탈구사시킨 후 뇌조직을 채취하였다. 채취한 뇌조직을 TUNEL 분석법에 의하며 아포토시스 유도 수준을 평가하였으며 Western blotting법을 이용하여 유전자 산물인 p53, Bcl-2, Bax 그리고 세포주기 조절인자인 cyclin Bl, Dl, E, cdk2, cdk4, $p34^{cdc2}$를 분석하였다. 세포주기의 변화는 유세포분석법에 의하여 분석되었다. 결과 : 아포토시스는 방사선조사후 8시간에서 최고치를 보였고 아포토시스 지수는 $24.0{\pm}0.25$ (p<0.05)였다. 세포주기에서 조절인자의 변화는 cyclin D1를 제외하고는 특이하지 않았다. 결론 : 마우스의 전뇌에 방사선을 조사한 결과 아포토시스는 대뇌의 상의하(subependyma)에서 주로 일어났으며 세포주기의 조절인자에는 영향을 미치지 않는 것으로 판명되었다.
