Purification and Characterization of a Thermostable Cellobiohydrolase from Fomitopsis pinicola

  • A screening for cellobiohydrolase (CBH) activity was performed and Fomitopsis pinicola KMJ812 was selected for further characterization as it produced a high level of CBH activity. An extracellular CBH was purified to homogeneity by sequential chromatography of F. pinicola culture supernatants. The molecular mass of the F. pinicola CBH was determined to be 64 kDa by SDS-PAGE and by size-exclusion chromatography, indicating that the enzyme is a monomer. The F. pinicola CBH showed a $t_{1/2}$ value of 42 h at $70^{\circ}C$ and catalytic efficiency of $15.8mM^{-1}s^{-1}(k_{cat}/K_m)$ for p-nitrophenyl-${\beta}$-D-cellobioside, one of the highest levels seen for CBH-producing microorganisms. Its internal amino acid sequences showed a significant homology with hydrolases from glycoside hydrolase family 7. Although CBHs have been purified and characterized from other sources, the F. pinicola CBH is distinguished from other CBHs by its high catalytic efficiency and thermostability.

  • The innovation city, which meets the best innovation condition to cooperate with the public institution and the industry-university-researcher closely and the good environment of housing, education, health and culture, was promoted to make the local city characteristic and independent. The plan to make the locally independent base have to consider the economical condition, the quality of life and the sustainable development. First of all The balanced city-planning is demanded to build friendly environmental and sustainable city. energy-efficient buildings shuld be designed to deal with the energy and environment problem. So we analyze the energy demand plan and the method to introduce the renewable energy system. As a result, the reduction ratio of the energy demand are greatly imbalanced between innovation cities. and only the Gwang-ju Jeon-nam innovation city is planed to apply the renewable energy to 5% of total energy demand.

Enhancing Factors of Electricity Generation in a Microbial Fuel Cell Using Geobacter sulfurreducens

  • In this study, we investigated various cultural and operational factors to enhance electricity generation in a microbial fuel cell (MFC) using Geobacter sulfurreducens. The pure culture of G. sulfurreducens was cultivated using various substrates including acetate, malate, succinate, and butyrate, with fumarate as an electron acceptor. Cell growth was observed only in acetate-fed medium, when the cell concentrations increased 4-fold for 3 days. A high acetate concentration suppressed electricity generation. As the acetate concentration was increased from 5 to 20 mM, the power density dropped from 16 to $13mW/m^2$, whereas the coulombic efficiency (CE) declined by about half. The immobilization of G. sulfurreducens on the anode considerably reduced the enrichment period from 15 to 7 days. Using argon gas to create an anaerobic condition in the anode chamber led to increased pH, and electricity generation subsequently dropped. When the plain carbon paper cathode was replaced by Pt-coated carbon paper (0.5 mg $Pt/cm^2$), the CE increased greatly from 39% to 83%.

  • Although somatic cell nuclear transfer (SCNT) is frequently employed to produce cloned animals in laboratories, this technique is expensive and inefficient. Therefore, the handmade cloning (HMC) technique has been suggested to simplify and advance the cloning process, however, HMC wastes many oocytes and leads to mitochondrial heteroplasmy. To solve these problems, we propose a modified handmade cloning (mHMC) technique that uses simple laboratory equipment, i.e., a Pasteur pipette and an alcohol lamp, applying it to porcine embryo cloning. To validate the application of mHMC to pig cloning, embryos produced through SCNT and mHMC are compared using multiple methods, such as enucleation efficiency, oxidative stress, embryo developmental competence, and gene expression. The results show no significant differences between techniques except in the enucleation efficiency. The 8-cell and 16-cell embryo developmental competence and Oct4 expression levels exhibit significant differences. However, the blastocyst rate is not significantly different between mHMC and SCNT. This study verifies that cloned embryos derived from the two techniques exhibit similar generation and developmental competence. Thus, we suggest that mHMC could replace SCNT for simpler and cheaper porcine cloning.

  • Laver aquaculture, which occupies a large proportion in the aquaculture industry in Korea, is still highly dependent on human labor. Therefore, it is necessary to study the development of an automatic system to improve the working environment and increase the efficiency of aquaculture production systems. The purpose of this study is to evaluate the economic feasibility of an improved system in a study for the loading-unloading and automatic weighing systems in laver aquaculture industry. Economic analysis of the developed unloading and automatic weighing system were implemented under various conditions to calculate more accurate benefits and costs. As a result of this study, the economic feasibility was found to be very high in the three models: net present value (NPV), benefit-cost ratio (B/C), internal rate of return (IRR). Moreover, the results of sensitivity analysis showed that the economical efficiency of the automatic loading, unloading, and weighing system in laver aquaculture was very high.

An Approach to Value Discourse on Translation of Korean Chinese written Classics (한국 한문고전 번역의 가치담론과 번역자상에 대한 시론적 접근)

  • This article deals with the reason for translating Korean Chinese written classics and the image of the person performing the translation. The scope of the research was restricted to South Korea and the translation value of translating the Korean classical texts from the 1960s to the 2018. In the 1960s and 1970s, the discourse of national culture and the Classical Sinology(漢學) discourse centered around the Minjokmunwhachujinhwe(民族文化推進會, National Culture Promotion Association). The discourse of the national culture was paired with the modernization, and the discourse of Classical Sinology(漢學) discourse was a certain antagonism to the discourse of modernization. The translator stereotype in this periods was close to a Classical Sinology(漢學) who could wright Korean letters. The discourse of the national culture led to the establishment of The Academy of Korean Studies by pairing with the discourse of the spiritual culture, and then changed into Korean study discourse in the 1980s. Since the mid 80s, the theory of translation has been introduced byo Kim Yong-ok. The translation of the Chosun dynasty annals, which started in the 70s, made the classical translation discourse in the classical translation field into the national project efficiency discourse. To the Early achievement of state-led gigantic project through group translation, they emphasized coherence and efficiency. On the contrary, the individuality of the translators and aspects of in-depth research have weakened. This discourse also influenced until the early 2000s. These large translation projects were produced by professional translator group. With the establishment of the Institute for the Translation of Korean Classics(Hankuk Kojon Bunyukwon) in 2007, he foundation for the stability of the classical translation business was established, and the classical translation discourse was shifted to the academic discourse centered on classical translation sudies. This discussion was expanded to the request of the establishment of an academic institution called the Classical Translation Graduate School, with a discussion on the academic identities of classical translation studies. The imagies of translators, paired with the academic discourse of this period, and that the classical translators must be classical scholars and translators, are begun to be requested. Thus, the classical translation value discourse changed with the passage of time, and the imagies of classical translators have been changed accordingly.

Anther Culture Efficiency affected by Growth Condition and Pre-treatment Methods in Barley (보리 생육환경 및 전처리 방법별 약배양 효율)

  • This experiment was carried out to improve the anther culture efficiency of barley (Hordeum vulgare L.). Callus induction rates from anther cultures of the five domestic naked barley and four unhulled varieties ranged from 0 to 5.6%, and plant regeneration rate to callus was 30.4% in the donor plants grown in a greenhouse during winter, among which the green plant regeneration rates ranged from 0 to 4.4%. Plant regeneration rate was 30.4% in the donor plants grown in a greenhouse during winter, whereas 21.3% in the normal field condition in spring. In addition, callus induction rates were 19.2% in plants grown in a normal field and 7.2% in drought-stressed condition, respectively. Being Considered the anther culture efficiency affected by the sampling time, the optimum sampling stage of anthers was 3~4 days before heading when the length between the 1st and 2nd auricles reaches 5 to 10 cm and at the uninucleate of pollen which the tip of the 2nd auricle aligns with the middle of panicle in the leaf sheath. Best callus induction rates came from the anthers stored at $4^{\circ}C$ for 3 weeks in a 10 to 15 cm diameter polyethylene bag with 5 to 10 panicles and Duwonchapssalbori and Saessalbori showed the higher induction rate of 4.8% and 1.7%, respectively.

Effect of Plant Growth Regulators, Media and Celling Agents on In Vitro Microtuber Production of Pinellia ternata Breit (1식물생장조절제, 배지와 고형지지물이 반하의 기내 소괴경 생산에 미치는 영향)

  • The study was carried out to establish in vitro microtuber production from leaf and petiole explant cultures of Pinellia ternata. Culture conditions were optimized by investigating the effect of plant growth regulators, different media, and gelling agents on the efficiency of microtuber indurtion. Among the different combinations of plant growth regulators tested, the combination of 0.1 mg/l 2,4-Dichlorophenoxyacetic (2,4-D) and 0.5 mg/l 6-benzylaminopurine (BAP) showed the highest yield fer microtuber production from leaf (3.9) and petiole (4.7) explant culture on MS medium far 6 weeks. SH(Schenk and Hildebrandt) medium was the most effective medium for microtuber induction and the half strength SH medium was better than SH medium for microtuber production from both leaf and petiole culture. Gelrite was better than agar in the formation of microtubers and 4% Gelrite showed the highest number of microtubers per explant frome leaf (5.9) and petiole (7.8) culture. Germination rate of microtubers after cold storaged for one months long was 86% from in vitro culture and 43% from autoclaved soil.

  • Recently, Daejeon Metropolitan City is planning a futuristic city with Daedeok Special District at the center of its transformation into a city of nature, science and culture along with the fourth industrial revolution. Therefore, this research will divide the 1.5 km distance from the in Doryong-dong, Daejeon, science park intersection to the (Former)Daedeok Science and Culture Center, and propose a space design concept that incorporates IT technologies that fit the characteristics. The center will analyze the space status and derive an experience-oriented space design concept for user-centered planning so that it can serve as a guideline that will provide the fundamental direction for the promotion of the Daedeok Science Culture. Through the field survey, we will look for ways to integrate IT technology with infrastructure that needs improvement and change the distance of Daedeok Science culture as a smart street. The results of the field survey were : (1) made by connecting the persistence of comfort and safety issues, (2) uniformity and efficiency issues, and (3) the experience of identity problems and the orientation of the smart city. To create user-centered distances, we attempted to avoid uniform zoning based solely on infrastructure and zoning focusing on user behavior.

Comparison of Biomass Production of Spirulina (Arthrospira) platensis in Outdoor Culture Conditions Using Different Media by Urea Addition (실외 배양 조건에서 요소를 첨가한 배지 성분에 따른 Spirulina (Arthrospira) platensis의 성장 비교)

  • One of the most important challenges facing the Spirulina mass cultivation industry is to find a way to reduce the high production costs involved in production. Although the most commercial medium (Zarrouk's medium) for Spirulina cultivation is too expensive to use, it contains higher amount of $NaHCO_3$ (16.80 g $L^{-1}$), trace metals and vitamin solutions. The purpose of this study was to increase the efficiency of Spirulina platensis biomass production by developing a low-cost culture medium at an isolated tropical island such as Chuuk State, Federated States of Micronesia (FSM). This study set out to formulate a lowcost medium for the culture of S. platensis, by substituting nutrients of Zarrouk's medium using fertilizer- grade urea and soil extract with a different concentration of carbon source under natural weather condition. In order to select a low-cost culture medium of S. platensis, 10 culture media were prepared with different concentrations of nitrogen (urea and $NaNO_3$) and $NaHCO_3$. The highest maximum specific growth rate (${\mu}max$) and mass production were 0.50 $day^{-1}$ and 1.05 g $L^{-1}$ in modified medium ($NaHCO_3$ 7.50 g $L^{-1}$, urea 2.00 g $L^{-1}$ without $NaNO_3$) among all the synthesized media. Protein (56.14%) and carbohydrate (16.21%) concentrations of the lyophilized standard samples were estimated with highest concentration of glutamic acid (14.93%). This study revealed that the use of a low concentration of urea and $NaHCO_3$ with soil extract was an affordable medium for natural mass cultivation in the FSM.
