Dynamics of lockstitch sewing process

  • During high speed sewing, the needle thread is exposed to dynamic loading, short strike loading, inertia forces, friction, rubbing, force of check spring, bending, pressure, friction, impact, shock and thermal influence. The dynamic thread loading/tension alters throughout the stitch formation cycle and along its passage through the machine. The greatest tensile force occurs at the moment of stitch stretching, when the take up lever pulls for required thread length through the tension regulator. These stresses act on the thread repeatedly and the thread passes 50-80 times through the fabric, the needle eye and the bobbin case mechanism, before getting incorporated into the seam, which result in upto 40% loss in tensile strength of the sewing thread. This damage in the sewing thread adversely affects its processing and functional performance. In this paper, the contribution of dynamic loading, passage through needle and fabric, and bobbin thread interaction in the loss in tensile properties has been studied. It is observed that the loss in tensile properties occurs mainly due to the bobbin thread interaction. Dynamic loading due to the action of take up lever also causes substantial loss in tenacity and breaking elongation of cotton threads.

Overexpression of Shinorhizobium meliloti Hemoprotein in Streptomyces lividans to Enhance Secondary Metabolite Production

  • It was found that Shinorhizobium meliloti hemoprotein (SM) was more effective than Vitreoscilla hemoglobin (Vhb) in promoting secondary metabolites production when overexpressed in Streptomyces lividans TK24. The transformant with sm (sm-transformant) produced 2.7-times and 3-times larger amounts of actinorhodin than the vhb-transformant in solid culture and flask culture, respectively. In both solid and flask cultures, a larger amount of undecylprodigiocin was produced by the sm-transformant. It is considered that the overexpression of SM especially has activated the pentose phosphate pathway through oxidative stress, as evidenced by an increased NADPH production observed, and that it has promoted secondary metabolites biosynthesis.

  • 지속가능한 한국문화축제 전략이 필요한 시기다. 한국문화축제는 2020년부터 전 세계적인 열풍을 이끌고 있는 K 콘텐츠를 중심으로 한식, 뷰티, 패션 등 다양한 한국문화를 종합적으로 소개하는 한류문화축제다. 문화체육관광부 신한류 진흥 핵심사업으로써 코로나 이후 실감 콘텐츠 무대를 적용한 첫 대면 콘서트 개최를 비롯하여 다양한 팬 참여 챌린지 진행 및 전시 등 월드 케이팝 콘서트와 K컬쳐 팬 페어를 바탕으로 했던 2021년 한국문화 축제는 이제 글로벌 한류 문화 축제로 도약해야 한다. 이를 위해서 드라마, 케이팝, K컬처 팬 페어, K밋업, K컬처 퍼레이드, 어워즈로 구성될 수 있다. 이러한 구분은 한류를 이끄는 프로슈머적 콘텐츠를 중심으로 고전적인 축제 프로그램을 보여주는데 이러한 철학이 동시대에 효율적으로 연계되기 위해서 학습된 유희성을 통해 잠재적 고객 확보를 위한 Business to Business(B2B), Businesss to Consumer(B2C) 테크테인먼트(Techtainment) 전략이 필요하다.

  • 인간의 모든 삶에 대한 디지털적 접근을 시도한 문화기술은 이제 IT 기술을 바탕으로 한 인간의 모든 생활에서 폭 넓게 적용되어 가고 있으며 앞으로 그 범위가 점차 확대되리라 기대된다. 문화기술은 어느 특정한 분야에서 연구되기보다는 모든 학문분야에서 학제적 연계를 통하여 연구, 발전되고 있다. 본 논문에서는 특히, 디지털기술에 바탕을 둔 문화콘텐츠의 개발 및 사용에 대한 효과적인 의사결정을 지원하기 위하여 데이터 마이닝 전략을 접목하고자 한다. 데이터 마이닝 분석기법올 적용하여 사용자로부터 만족될 수 있는 문화콘텐츠의 구현방안을 제안한다.

Secretory Production of Human Leptin in Bacillus subtilis

  • Human leptin is identified as a 16kDa (146 amino acids) protein secreted from adipocytes which influences body weight homeostasis. In order to produce active leptin, the human obese gene coding for leptin was expressed in Bacillus subtilis WB600 strain which is deficient in six extracellular proteases. The recombinant leptin was produced in a culture supernatant, and in a culture supernatant, it was contained as high as 48% for total proteins. After simple purification steps, which consisted of ammonium sulfate precipitation and anion-exchange column chromatography, 2.3 mg of leptin with a purity greater than 95% was obtained from the 0.51 culture with the recovery yield of 38.3%. The purified leptin showed the correct folding structure with one disulfied bond.

Assessment and Optimization of Xylanase Production Using Mono-Culture and Co-Cultures of Bacillus subtilis and Bacillus pumilus

  • Xylanase is an industrially relevant enzyme used for the production of xylobiose and xylose. Various methods are used to enhance the microbial yield of xylanase. In the present study, co-culturing of Bacillus subtilis and Bacillus pumilus were investigated using submerged fermentation for xylanase production, which was markedly increased when sal, sagwan, newspaper, wheat bran, and xylan were used as single carbon sources. Maximum xylanase production was reported after 5 days of incubation in optimized media at pH 7.0 and 37℃, resulting in 2.69 ± 0.25 µmol/min by coculture. The 1:1 ratio of sal and sagwan in optimized production media was shown to be suitable for xylanase synthesis in submerged fermentation (SMF). In comparison to mono-culture using B. pumilus and B. subtilis, co-culturing resulted in an overall 3.8-fold and 2.15-fold increase in xylanase production, respectively.

영지 액체 배양의 Wall Growth에 미치는 Polyacrylic Acid의 첨가 효과 (Effect of Polyacrylic Acid Addition on Wall Growth in Submerged Cultivation of Ganoderma lucidum)

  • This study was carried out to screen the effective polymeric additives preventing wall growth during mycelial submerged cultivation of Ganoderma. lucidum. Effects of additives on mycelial growth and exo-polysaccharide (EPS) production in flask culture and jar fermenter system under 3 different pH processes were investigated, and changes of mycelial morphology were also examined. From flask culture of G. lucidum with additives of different concentrations, 0.1%(w/v) polyacrylic acid was effective for EPS production. As the polyacrylic acid of 0.1%(w/v) was added in medium, wall growth of G. lucidum mycelium grown in jar fermenter system could be protected. The addition of 0.1%(w/v) polyacrylic acid to medium was also improved the mycelial growth and EPS production in the later of submerged culture G. lucidum and no changes of mycelial morphology were observed.

  • Aggregates of specific cells are often regarded as better from in artificial organs and mammalian cell bioreactors in terms of cell-specific functionality. In this study, the morphology and liver specific functions of freshly harvested primary rat hepatocytes, which were cultivated as spheroids and entrapped in a synthetic thermo-reversible extracellular matrix, were examined and compared to a control (hepatocytes in single cell form). A copolymer of N-isopropylacrylamide(98 mole % in feed) and acrylic acid (poly (NiPAAm-co-AAC)), a thermo- reversible copolymer gel ma- trix, was used to entrap hepatocytes either in spheroids or single cells. During a 7-day culture pe-riod, the spheroids maintained higher viability and produced albumin and urea at a relatively con-stant rate, while, the single cell culture showed a slight increase in cell numbers and a reduction in albumin secretion Hepatocytes cultrured as spheroids present a potentially useful three-dimensional cell culture system for application in bioartificial liver device.

Rapid Detection of Bacteria from Blood Culture by an Electronic Nose

  • The treatment of Patients with bacteraemia and septicemia requires accurate and rapid identification of the pathogen so that the physician can be guided regarding the selection of the proper antimicrobial therapy. The usual procedure is to withdraw an aliquot of the positive blood culture sample for gram staining and subculturing on the media for the growth and subsequent identification, and susceptibility determinations. It was noticed that during the process some microbiologists would sniff the effluent gases that are products of metabolism and in some cases guess the identity of the bacterium. That Prompted us to engage in systematic investigation of two gram positive and two gram negative bacteria using an electronic nose that had been proven successful in distinguishing the aroma of coffee beans from different sources. The investigation was successful in illustrating the efficacy of such a device in this clinical setting to distinguish Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus and Enterococcus faecalis. A representative set of patterns obtained with this apparatus is displayed as well. A representative set of patterns obtained with this apparatus is displayed as well. An Efficient Secretion of Type I Secretion Pathway-Dependent Lipase, TliA, in Escherichia coli: Effect of Relative Expression Levels and Timing of Passenger Protein and ABC Transporter

  • An ABC transporter apparatus of the Gram-negative bacterial type I secretion pathway can be used as a secretory protein expression system in Escherichia coli. Four types of coexpression systems for the Pseudomonas fluorescens lipase gene, tliA, and its cognate ABC transporter gene cluster, tliDEF, were constructed. When the relative expression levels were changed by adding different concentrations of IPTG, the secretion (16.9 U/ml of culture) of TliA in E. coli [pTliDEFA-223+pACYC184] was significantly higher than E. coli [pKK223-3+pTliDEFA-184] secreting the lowest level of TliA (5.2 U/ml of culture). Maximal accumulation of the lipase secreted occurred in the mid-exponential phase, implying that the efficient protein secretion via an ABC transporter was restricted only to actively growing cells. Finally, the secretion level of TliA in E. coli [pTliDEFA-223+pACYC184] was increased to 26.4 U/ml by inducing gene expression at the culture initiation time. These results indicate that a significant increase in the ABC transporter-dependent protein secretion can be achieved by simply controlling the relative expression levels between the ABC transporter and its passenger protein, even in the recombinant E. coli cells.
