Establishment of Analytical Method for Fenhexamid Residue in Korean Cabbage, Apple, Mandarin and Green Pepper (HPLC를 이용한 배추, 사과, 감귤, 고추 중 살균제 Fenhexamid의 정밀 분석법 확립)

  • This study was performed to develop a precise single residue analytical method of fungicide fenhexamid in representative crops for general residue analytical method which could be applied to most of crops. Korean cabbage, mandarin, apple and green pepper were selected, macerated, extracted with acetone, concentrated and partitioned with dichloromethane. Then the extracts were concentrated and cleaned-up through Florisil column with ethyl acetate/0.1% acetic acid in hexane [15:85, (v/v)] before concentration and analysis with HPLC. LOQ (Limit of Quantitation) of fenhexamid was 1 ng (S/N>10) and MQL (Method Quantitative Limit) was 0.01 mg/kg. Recoveries were measured at two fortification levels (10 MQL and 50 MQL) on crop samples and ranged from 85.2% to 94.8% (mean recoveries) and coefficients of variation were <10% regardless of sample type.

Elimination of Phenthoate Residues in the Washing and Cooking of Polished Rice (쌀의 취반 중 Phenthoate 농약 잔류분의 제거)

  • This study was undertaken in order to elucidate the elimination of phenthoate residues by washing and cooking processes of rice which if the most important food crop in Korea. When contaminated rice was washed with distilled water three times, the removal rate of total phenthoate was 51%. The removal rate in the successive washings was 37.3% (wash filtrate 7.8%, wash sediment 29.5%) in the first, 14.3% (wash filtrate 6.2%, wash sediment 8.1%) in the second and 8.9% (wash filtrate 5.8%, wash sediment 3.1%) in the third washings. More than half of the residue was removed by the first washing and most residues were found in the sediment rather than in the filtrate of the rice washings. The residue rate of phenthoate after cooking by an electric rice cooker was 41%, indicating that the removal rate after cooking was 59%, because phenthoate is thermally stable at the cooking temperature. In conclusion, phenthoate residues contaminated in rice grains are grcatly removed in the washing process and it is desirable to wash the grains before cooking in order to decrease the hazards from pesticide residues such as phenthoate. Reduction factor of phenthoate in rice cooking is proposed to be 0.4.

Analytical Methods of Fenpyroximate in Herbal Medicines (생약 중 Fenpyroximate의 분석법 연구)

  • Fenpyroximate is acaricide of pyrazole group. This acaricide have already been permitted for herb cultivation. This experiment was conducted to establish a determination method for fenpyroximate residue in herbal medicines using HPLC-PDA and HPLC-MS/MS. Fenpyroximate residue was extracted with acetone from samples of herbal which Liquorice Root (Glycyrrhiza uralensis) and Safflower Seed (Carthamus tinctorius Linne). The extract was diluted with saturated saline water and dichloromethane liquid-liquid partition (extraction) was followed to recover fenpyroximate from the aqueous phase. Amino propyl ($NH_2$) and florisil column chromatography was additionally employed for final clean up of the extract. The fenpyroximate was quantitated by HPLC-PDA and HPLC-MS/MS. The herbals were fortified with fenpyroximate at 2 or 3 levels per crop. Mean recovery ratio were ranged from 72.0 to 106.4%. The coefficients of variation were ranged from 0.2 to 4.4. Therefore, this analytical method was reproducible and sensitive enough to determine the residue of fenpyroximate in herbal medicines.

Effect of Hairy Vetch Green Manure on Nitrogen Enrichment in Soil and Corn Plant (토양 및 옥수수의 질소 집적에 미치는 헤어리벳치 녹비시용 효과)

  • Fresh hairy vetch (HV) as a green manure equivalent to $240kg\;N\;ha^{-1}$ were incorporated into soil at corn planting in 1997 and 1998 to clarify the effects on changes of nitrogen (N) content in soil and corn plant. The influences of HV for the N of soil and plant were compared with those of ammonium nitrate (AN) in terms of mineralization and microbial biomass. During early decomposition of HV residue, the content of $NO_3-N$ in HV plot was as much as 60-70% of that in AN plot in surface soil of 0-15 cm depth. In addition, soil microbial biomass N (SMBN) by HV residue was increased up to $10-20mg\;kg^{-1}$ more than that by AN. Some mineral N from HV seemed to be released slowly until late corn growth stage judging from high content of $NO_3-N$ in both corn stem at silking stage and soil at harvest. There were no difference of N accumulations in corn plant at silking stage between HV and AN plots in both 1997 and 1998. At harvesting stage, a total of plant N accumulation in HV plot in 1997 was 8% less than that in AN plot while in 1998 it was 19% more. It was concluded that fresh HV green manure equivalent to $240kg\;N\;ha^{-1}$ was good enough to substitute the same amount with chemical N fertilizer by slow releasing of mineral N from HV residue in soil.

The Allelopathic Effect of Alfalfa residues on Crops and Weeds (잡초와 작물에 대한 알팔파 잔유물의 Allelopathy 효과)

  • This experiment was conducted to test the effect on germination and seedling growth of major industrial crops and weed control potential using alfalfa plant. When dried alfalfa residues were mixed into vermiculite, germination, length of shoot and root of crops, such as Perilla frutescens, Sorghum nevosum, Platycodon grandiflorum and weeds, Digitaria saguinalis, Setaria viridis, Siegesbechia pubescens, Ammaranthus lividus, and Solanum nigrum, were significantly inhibited as the dried residue concentration increased. More than 10% concentration of the dired residue caused 80% germination and growth inhibition. The fresh alfalfa exudation also inhibited the germination and seedling growth of crop, barley, rye, alfalfa, and sesame, and weeds, Echinochloa crus-galli, Siegesbechia viridis, and Portulaca oleracea. The degree of inhibition showed the different response according to the fresh exudation concentration, types of crops and weeds. Generally, as the exudation concentration increased, the germination and seedling growth of crops and weeds inhibited. The exudation of dried residue also exhibited the strong inhibition effect on germination and seedling growth of crops, alfalfa, Platycodon grandiflorum, barley, sesame, rye and weeds, D. sanguinalis, S. pubescens, S. viridis, P. oleracea, E. crus-galli, At the 10% concentrations, S. pubescens, and P. oleracea were not germinated and showed only 15% germination in the S. viridis. From this study, would conclude that alfalfa plant contained water soluble phytotoxic substances which were inhibitory to weeds and crops. This results suggest that alfalfa had some possibility to control some weed species using toxic compounds like natural herbicide.

Establishment of Pre-Harvest Residue Limit (PHRL) of Fungicides Azoxystrobin and Difenoconazole on Prunus mume fruits (매실 중 살균제 azoxystrobin과 difenoconazole의 생산단계 잔류허용기준 설정)

  • This study was carried out to investigate the residual characteristics of fungicide azoxystrobin and difenoconazole in Prunus mume fruits, and establish pre-harvest residue limits (PHRL) based on dissipation and biological half-lives of fungicide residues. The fungicides were sprayed onto the crop at recommended dosage once and 3 times in 7 days interval, respectively. The samples were harvested at 0, 1, 2, 4, 6, 8, 10, 12 and 14 days after treatment. These residual pesticides were extracted with QuEChERS method, clean-up with $NH_2$ SPE cartridge, and residues were analyzed by HPLC/DAD and GLC/ECD, respectively. Method quantitative limits (MQL) of azoxystrobin were 0.03 mg $kg^{-1}$ and of difenoconazole were 0.006 mg $kg^{-1}$. Average recovery were $93.2{\pm}2.49%$, $85.5{\pm}1.97%$ for azoxystrobin at fortification levels at 0.3 and 1.5 mg $kg^{-1}$, and $100.8{\pm}6.74%$, $87.6{\pm}9.92%$ for difenoconazole at fortification levels at 0.06 and 0.3 mg $kg^{-1}$, respectively. The biological half-lives of azoxystrobin were 5.9 and 5.2 days at recommended dosage once and 3 times in 7 days interval, respectively. The biological half-lives of difenoconazole were 9.3 and 8.0 days at recommended dosage once and 3 times in 7 days interval, respectively. The PHRL of azoxystrobin and difenoconazole were recommended as 5.32 and 1.64 mg $kg^{-1}$ for 10 days before harvest, respectively.

Soil Quality Assessment for Environmentally Sound Agriculture in the Mountainous Soils; Analysis of Sediment Data and Suggestion of Best Management Practices (산지에서의 환경보전형 농업을 위한 토양의 질 평가;유사자료의 분석과 최적영농방법의 제안)

  • Eleven runoff $plots(3{\times}15\;m)$ were built on a sloping field of a high plateau in Kangwon Province, Korea. The plots were treated with different tillage, residue covers and fertilizers, corn and potato were cultivated, and sediment discharge was measured from the plots for 3 years. Agricultural management practices were monitored around the plots to develop adequate best management practices. The least sediment occurred from the plots with no-till and 100% residue cover $(corn,\;0.1{\sim}2.2\;t/ha/year)$ and with contour tillage and vinyl sheet cover plots $(potato,\;0.1{\sim}0.2\;t/ha/year)$. The largest sediment was measured from the plots with up-and-down till and no cover ($11{\sim}33$ and $16{\sim}31\;t/ha/year$ from corn and potato plots, respectively). The type of organic and commercial fertilizers seemed not to affect sediment discharge. Sediment discharge from contour plots were largely dependent on the collapse of ridges due to the flush of water stored in furrows. The sediment discharge from contour corn and potato plots with no residue cover was $10{\sim}27$ and $16{\sim}24\;t/ha/year$, respectively. No-till with residue cover, vinyl cover for tuber crops, construction of furrow dam, frost heave research, furrow and ridge tillage with furrow dam after tuber crop harvest, limited stalk harvest of row crops, use of winter cover crops, and other common best management practices were recommended to minimize the sediment discharge from a high plateau sloping land culture.

The residue property of fungicide boscalid and fluidioxonil at the same time harvest leafy-vegetables (일시수확 엽채류에서 살균제 Boscalid와 Fludioxonil의 잔류특성)

  • In order to use in the classification of minor crop for the mutual application of safe use guideline, it was investigated the residue property of fungicide boscalid and fludioxonil at the same time harvest leafy-vegetables, such as spinach, ulgaribaechu, vitaminchae and cheongkyungchae. After pesticides were applied 2 times with 1 week interval in that day of harvest, 2 days, 5 days and 7 days before harvest, vegetables were harvested, and the residue of pesticides was investigated. Base on the residue in that day of harvest, the deposit of spray solution in vegetables was calculated. The deposit of spray solution of boscalid was 253.9 mL/kg in spinach, 83.0 mL/kg in ulgaribaechu, 97.8 mL/kg in vitaminchae, and 88.3 mL/kg in cheongkyungchae, respectively. In case of fludioxonil, it was calculated 157.6 mL/kg in spinach, 67.6 mL/kg in ulgaribaechu, 64.8 mL/kg in vitaminchae, and 66.6 mL/kg in cheongkyungchae, respectively. When the amount of the deposit of both pesticides was compared in leafy-vegetables, it was the highest in the spinach. On the other hand, it was estimated the predicted dissipation curve of pesticides in leafy-vegetables during cultivation. The half-life of boscalid was 5.9 days in spinach, 7.4 days in ulgaribaechu, 4.6 days in vitaminchae, and 4.3 days in cheongkyungchae, respectively. Also, it was estimated half-life in fludioxonil, it was 3.0 days in spinach, 4.0 days in ulgaribaechu, 3.2 days in vitaminchae, and 3.5 days in cheongkyungchae, respectively. The half-life was the longest in the ulgaribaechu. When both pesticides were compared with the residue property, the deposit of spray solution and half-life of dissipation of boscalid were more than those of fludioxonil.

Comparision of the Residue Property of Insecticide Bifenthrin and Chlorfenapyr in Green Onion and Scallion under Greenhouse Condition (시설재배 쪽파와 부추에서 살충제 Bifenthrin과 Chlorfenapyr의 잔류특성 비교)

After pesticides were applied 2 times with 1 week interval in that day of harvest, 3 days, 7 days, 10 days and 14 days before harvest, vegetables were harvested, and the residue of pesticides was investigated. Base on the residue in that day of harvest, the deposit of spray solution in vegetables was calculated. The deposit of spray solution of bifenthrin was 123.0 mL/kg in a green onion, and 74 mL/kg in a scallion. In case of chlorfenapyr, it was calculated 126.5 mL/kg in a green onion, and 70.0 mL/kg in a scallion. When the amount of the deposit of both pesticides was compared a green onion with a scallion, it was higher in a green onion. On the other hand, it was estimated the predicted dissipation curve of pesticides in a green onion and a scallion during cultivation. The dissipation curve of bifenthrin was y = 1.0334 $e^{-0.0602x}$ ($R^2$= 0.8606) in a green onion, and y = 0.7693 $e^{-0.1823x}$ ($R^2$= 0.9756) in a scallion. In case of chlorfenapyr, it was y = 2.2603 $e^{-0.0519x}$ ($R^2$= 0.9043) in a green onion, and y = 1.2940 $e^{-0.1051x}$ ($R^2$ = 0.9782) in a scallion. The half-life of bifenthrin was 11.51 days in a green onion, and 3.80 days in a scallion, respectively. Also, it was estimated half-life in chlorfenapyr, it was 13.35 days in a green onion, and 6.59 days in a scallion, respectively. The half-life of both pesticides in a green onion was longer than in a scallion. When both vegetables were compared with the residue property, the deposit of spray solution and half-life of dissipation in a green onion were more than those in a scallion.

Evaluation of Neonicotinoid Pesticides' Residual Toxicity to Honeybees Following or Foliage Treatment (네오니코티노이드계 농약의 사용방법에 따른 꿀벌엽상잔류 독성 평가)

  • Neonicotinoid pesticides, widely used worldwide as potent insecticides, have been found to have detrimental effects on the environment and living organisms due to their persistent residues. This study aimed to investigate the neonicotinoid pesticides, imidacloprid, and clothianidin, focusing on their impact on honey bee toxicity and foliar residue levels. Alfalfa was selected as control crop while bell peppers, and cucumbers were chosen as representative application crops, respectively. The investigation involved comparing the toxicity and foliar residue levels resulting from soil and foliar treatments, with a focus on identifying potential shortcomings in conventional foliar residue toxicity testing methods. Imidacloprid and clothianidin were applied to crops or soil at recommended rates and through irrigation. The honey bee mortality rate (RT25) over time was determined, and pesticide residues on leaves were quantified using High-Performance Liquid Chromatography (HPLC). The results revealed that foliar treatment with imidacloprid on alfalfa resulted in an RT25 of less than 1 day, with residues ranging from 1.07 to 19.27 mg/kg. In contrast, application on bell peppers showed RT25 within 9 days, with residues ranging from 1.00 to 45.10 mg/kg. Clothianidin foliar treatment displayed RT25 within 10 days on alfalfa, with residues between 0.61 and 2.57 mg/kg. On bell peppers, RT25 was within 28 days, with residues ranging from 0.13 to 2.85 mg/kg. Soil treatment with imidacloprid and clothianidin in alfalfa exhibited minimal impact on honey bees and residues of 0.05 to 0.37 mg/kg. However, in applied crops, imidacloprid showed RT25 within 28 days and residues ranging from 4.47 to 130.43 mg/kg, while clothianidin exhibited RT25 within 35 days and residues between 5.96 and 42.32 mg/kg. In conclusion, when comparing honey bee toxicity and foliar residues among crops, application crops had a more significant impact on honey bee mortality and higher residue levels compared to control crops. Moreover, soil treatment for application crops resulted in higher RT25 and residue levels compared to foliar treatment. Therefore, to ensure pesticide safety and environmental sustainability, diverse research approaches considering different crops and application methods are necessary for the safety assessment of imidacloprid and clothianidin.
