  • Nanocrystalline CrN films were deposited on Si (100) substrates by means of asymmetric pulsed DC reactive magnetron sputtering. We investigated the growth behavior, corrosion resistance and mechanical properties of CrN films with a change in the duty cycle and pulse frequency. The grain size of the CrN films decreased from 25.4 nm to 11.2 nm upon a decrease in the duty cycle. The corrosion potentials for the CrN films by DC sputtering was approximately - 0.6 V, and it increased to - 0.3 V in the CrN films which underwent pulsed sputtering. The nanoindentation hardness of the CrN films also increased with a decrease in the duty cycle. This enhancement of the corrosion resistance and mechanical properties of pulsed sputtered CrN films could be attributed to the densification and surface smoothness of the microstructure of the films.

Membrane-Associated Hexavalent Chromium Reductase of Bacillus megaterium TKW3 with Induced Expression

  • Hexavalent chromium ($Cr^{6+}$) is a highly harmful pollutant, which can be detoxified and precipitated through reduction to $Cr^{3+}$. Bacillus megaterium TKW3 previously isolated from chromium-contaminated marine sediments was capable of reducing $Cr^{6+}$ in concomitance with metalloids ($Se^{4+}$, $Se^{6+}$, and $As^{5+}$). Notwithstanding approximately 50% inhibition, it was the first report of simultaneous bacterial reduction of $Cr^{6+}$ and $Se^{4+}$ (to elemental Se). No significant difference was observed among electron donors (glucose, maltose, and mannitol) on $Cr^{6+}$ reduction by B. megaterium TKW3. The reduction was constitutive and determined to be non-plasmid mediated. Peptide mass fingerprints (PMF) revealed a novel aerobic membrane-associated reductase with $Cr^{6+}$-induced expression and specific reductive activity (in nmol $Cr^{6+}$/mg protein/min) of 0.220 as compared with 0.087 of the soluble protein fraction. Respiratory inhibitor $NaN_3$ did not interfere with the reductase activity. Transmission electron microscopy with energy dispersive X-ray (TEM-EDX) analysis confirmed the aggregation of reduced chromium along the intracellular membrane region. Future identification of the N-terminal amino acid sequence of this reductase will facilitate purification and understanding of its enzymatic action.

  • 환경 친화형 전기자동차, 하이브리드 자동차, 전철 등에서는 고내압 및 소형으로 전력손실을 감소시킬 수 있는 파워 디바이스가 필수이다. 최근, 실리콘 카바이드(SiC, silcon carbide)는 기존 실리콘(Si)보다 스위칭 손실의 저감 및 고온환경에서의 동작 특성이 우수하여, 차세대 저 손실 전력반도체 재료로서 기대를 받고 있다. 용액 성장 법에서 고품질 SiC 결정을 만들 수 있다. 그러나 늦은 성장 속도 때문에 SiC의 양산을 어렵게 하고 있다. 현재까지 성장 속도 향상을 위한 Si용매에 Cr을 첨가하여 탄소 용해도를 높이는 방법이 사용되고 안정된 성장을 위한 Si-Cr용매에 Al를 첨가하는 등 다양한 금속을 첨가하는 방법이 이용되고 있다. 선행 연구에서는 다양한 용매인 탄소 용해도를 실측하고 특히 큰 탄소 용해도를 보인 것은 Co이었다. 본 연구에서는 $Si_{0.6}Cr_{0.4}$원료와 Co를 첨가한 $Si_{0.56}Cr_{0.4}Co_{0.04}$의 용매에 의한 SiC용액 성장을 실시하고 결정 성장 속도 및 표면 상태의 변화를 검토했다. on-axis 4H-SiC(000-1)을 사용한 Top-seeded solution growth(TSSG)법과 원자 비율로 $Si_{0.6}Cr_{0.4}$$Si_{0.56}Cr_{0.4}Co_{0.04}$의 용매를 이용하여 SiC 용액 성장을 실시했다. Ar가스에서 저항 가열로 내를 치환 후에 $1800^{\circ}C$까지 가열하고 종자화 후에 120분간 유지하고 결정 성장을 실시했다. 냉각 후에 성장의 표면에 남은 용매를 $HF+HNO_3$에서 제거했다. 광학 현미경을 이용하여 결정면과 두께를 관찰 측정했다. Co를 첨가한 $Si_{0.56}Cr_{0.4}Co_{0.04}$의 경우는 $Si_{0.6}Cr_{0.4}$의 경우보다 결정 성장 속도가 향상됐다. 또한 $Si_{0.6}Cr_{0.4}$보다 step-flow의 성장을 나타낸 결정의 표면이 전반적으로 관찰됐으며 안정된 결정성장을 나타냈다. 본 연구에서 실시한 연구 방법과 결과는 고품질 및 고속의 SiC 용액성장을 위한 매우 유용한 자료로 활용 될 수 있을 것으로 판단한다.

Study on the Protective Effect of Nelumbo nucifera GAERTN Extract on Cultured Cerebral Neuroglial Cells Damaged by Hexavalent Chromium (연꽃추출물이 6가 크롬으로 유도된 세포독성에 대한 보호효과에 관한 연구)

  • In order to investigate the cytotoxic effect of hexavalent chromium ($Cr0_3$) and the protective effect of Nelumbo nucifera GAERTN (NNG) extract, cultured cerebral neuroglial cells (C6 glioma cells) were treated with $4{\sim}55{\mu}M$ concentrations of $Cr0_3$ for 48 hours. Cell viability was measured by XTT assay. The superoxide dismutase (SOD)-like activity for the antioxidant effect was also examined on the extract of NNG stamen. In this study, $Cr0_3$ significantly decreased cell viability dose-dependently. The cytotoxicities of $XTT_{90}$ and $XTT_{50}$ determined with $10{\mu}M$ and $55{\mu}M$ of $Cr0_3$, respectively, showed that the $Cr0_3$ had highly toxic effect on cultured C6 glioma cells by the cytotoxic criteria. In the protective effect of NNG extract, the cell viability was significantly increased by the treatment of NNG extract, and NNG extract increased SOD-like activity. From these results, it is suggested that $Cr0_3$ showed highly toxic effect on cultured C6 glioma cell s and NNG extract was very effective in the protection of $Cr0_3$-mediated cytotoxicity by antioxidative effect in these cultures.

  • The adsorption and reduction characteristics of chromium(VI) by oak sawdust were studied. The optimum adsorption condition is obtained from the measurement of the distribution coefficient($K_d$) and adsorption capacity of Cr(VI) on the sawdust by changing pH of the solution. As a result, it was found that pH 2.0 was optimum because $K_d$ of Cr(VI) was maximum and reducing quantity was minimum. By the use of these characteristics of the oak sawdust, the removing of Cr(VI) in industrial waste water was examined.

  • This study investigates the acoustic property of aspiration realized in English voiceless stops of /Cr/ and /sCr/ clusters. VOT is measured from stops in these clusters produced by two groups; one from native speakers of English and the other from Korean native speakers. Aspiration of stops in different types of clusters is compared to various phonological factors such as location of stress, syllable type, and position in word. Pursuing the idea that phonetic realization is correlated with phonological representation, attempts are made to account for the gradient nature of aspiration of stops on the basis of syllable structure at the phonetic level, which may vary in the wake of resyllabification. Voiceless stops in /Cr/ and /sCr/ clusters are further compared to results obtained in the previous study on /sC/ cluster. Variations in aspiration are also characterized in terms of segmental precedence relation of stops in the clusters, namely, post-[s], pre-[r], or both.

Binding of the Hexavalent Chromium Ions in the Process of Cement Hydration (시멘트 수화에 따른 6가 크롬의 고정화 특성)

  • The hexavalent chromium (Cr(VI)) is well known as a hazardous ion, presumably inducing dermatic diseases and if serious cancer. The present study concerns the binding capacity of Cr(VI) ions in the cement powder and matrix for a quantitative technique of Cr(VI) ions in cement to influence human health. Both the water-soluble and acid-soluble Cr(VI) ions present in 3 types of ordinary Portland cement (OPC), pulverised fuel ash (PFA), ground granulated blast furnace slag (GGBS), and silica fume (SF) were measured using the spectrophotometer. As a result, it was found that the concentration of water-soluble Cr(VI) ion in cement ranged from 10.5 to 18.9mg/kg-cement, and in the additional materials a very low value of Cr(VI) ion was measured. Acid-soluble Cr(VI) ion was even higher than water-soluble Cr(VI) ion, ranging from 172.4 to 318.2mg/kg-cement. Nevertheless, the concentration of acid-soluble Cr(VI) ion is not proportional to addition of acid. It depends rather the variable pH of solvent involving cement paste. As enough cement hydration occurs, the binding capacity of Cr(VI) ion increases, inhibiting this ions from leaching out in the presence of hydration products such as ettringite or tri-calcium aluminate which bind Cr(VI) ion by ion-exchange.

Fate and Transport of Cr(VI) Contaminated Groundwater from the Industrial Area in Daejeon (대전 산업단지 지하수의 6가 크롬 오염 및 확산 평가)

  • The objective of this research was to characterize the fate and transport of Cr(VI) contaminated groundwater in the Daejeon industrial area. Five subsidiary monitoring wells were newly installed and two existing wells were utilized for the investigation and the reduction process of Cr(VI) contaminated groundwater of the Daejeon(Mun-pyeong) national groundwater monitoring station. The Cr(VI) concentrations at the shallow aquifer well of the station were in the range of 3.2-4.5 mg/L indicating continuous contamination. However, Cr was not detected at the deep bedrock well and the other monitoring wells except MPH-1 and 3. The Cr(VI) concentrations of MPH-1 and MPH-3 were below the drinking water guideline value (0.05 mg/L). Therefore, the plume of the Cr(VI) contaminated groundwater was predicted to be confined within the narrow boundary around the station. The soluble/exchangeable Cr(VI) concentrations were below the detection limit in all core and slime samples taken from the five newly installed wells. Although the exact source of contamination was not directly detected in the study area, the spatial Cr(VI) distribution in groundwater and characteristics of the core samples indicated that the source and the dispersion range were confined within the 100 m area from the monitoring station. The contamination might be induced from the unlined landfill of industrial wastes which was observed during the installation of an subsidiary monitoring well. For the evaluation of the natural attenuation of Cr(VI), available reduction capacities of Cr(VI) with an initial concentration of 5 mg/L were measured in soil and aquifer materials. Dark-gray clay layer samples have high capacities of Cr(VI) reduction ranging from 58 to 64%, which is obviously related to organic carbon contents of the samples. The analysis of reduction capacities implied that the soil and aquifer materials controlled the dispersion of Cr(VI) contamination in this area. However, some possibilities of dispersion by the preferential flow cannot be excluded due to the limited numbers of monitoring wells. We suggest the removal of Cr(VI) contaminated groundwater by periodical pumping, and the continuous groundwater quality monitoring for evaluation of the Cr(VI) dispersion should be followed in the study area.
