Currently, taxol is mainly extracted from the bark of yews; however, this method can not meet its increasing demand on the market because yews grow very slowly and are a rare and endangered species belonging to first-level conservation plants. Recently, increasing efforts have been made to develop alternative means of taxol production; microbe fermentation would be a very promising method to increase the production scale of taxol. To determine the activities of the taxol extracted from endophytic fungus N. sylviforme HDFS4-26 in inhibiting the growth and causing the apoptosis of cancer cells, on comparison with the taxol extracted from the bark of yew, we used cellular morphology, cell counting kit (CCK-8) assay, staining (HO33258/PI and Giemsa), DNA agarose gel electrophoresis and flow cytometry (FCM) analyses to determine the apoptosis status of breast cancer MCF-7 cells, cervical cancer HeLa cells and ovarian cancer HO8910 cells. Our results showed that the fungal taxol inhibited the growth of MCF-7, HeLa and HO8910 cells in a dose-and time-dependent manner. IC50 values of fungal taxol for HeLa, MCF-7 and HO8910 cells were $0.1-1.0{\mu}g/ml$, $0.001-0.01{\mu}g/ml$ and $0.01-0.1{\mu}g/ml$, respectively. The fungal taxol induced these tumor cells to undergo apoptosis with typical apoptotic characteristics, including morphological changes for chromatin condensation, chromatin crescent formation, nucleus fragmentation, apoptotic body formation and G2/M cell cycle arrest. The fungal taxol at the $0.01-1.0{\mu}g/ml$ had significant effects of inducing apoptosis between 24-48 h, which was the same as that of taxol extracted from yews. This study offers important information and a new resource for the production of an important anticancer drug by endofungus fermentation.
Background: Glioblastoma (GBM) is an immunosuppressive tumor whose median survival time is only 12-15 months, and patients with GBM have a uniformly poor prognosis. It is known that heredity contributes to formation of glioma, but there are few genetic studies concerning GBM. Materials and Methods: We genotyped six tagging SNPs (tSNP) in Han Chinese GBM and control patients. We used Microsoft Excel and SPSS 16.0 statistical package for statistical analysis and SNP Stats to test for associations between certain tSNPs and risk of GBM in five different models. ORs and 95%CIs were calculated for unconditional logistic-regression analysis with adjustment for age and gender. The SHEsis software platform was applied for analysis of linkage disequilibrium, haplotype construction, and genetic associations at polymorphism loci. Results: We found rs891835 in CCDC26 to be associated with GBM susceptibility at a level of p=0.009. The following genotypes of rs891835 were found to be associated with GBM risk in four different models of gene action: i) genotype GT (OR=2.26; 95%CI, 1.29-3.97; p=0.019) or GG (OR=1.33; 95%CI, 0.23-7.81; p=0.019) in the codominant model; ii) genotypes GT and GG (OR=2.18; 95%CI, 1.26-3.78; p=0.0061) in the dominant model; iii) GT (OR=2.24; 95%CI, 1.28-3.92; p=0.0053) in the overdominant model; iv) the allele G of rs891835 (OR=1.85; 95%CI, 1.14-3.00; p=0.015) in the additive model. In addition, "CG" and "CGGAG" were found by haplotype analysis to be associated with increased GBM risk. In contrast, genotype GG of CCDC26 rs6470745 was associated with decreased GBM risk (OR=0.34; 95%CI, 0.12-1.01; p=0.029) in the recessive model. Conclusions: Our results, combined with those from previous studies, suggest a potential genetic contribution of CCDC26 to GBM progression among Han Chinese.
The purpose of this study came to start to be helpful in that the teachers in field produced evaluation subject about the mathematics and applied by analyzing the performance capacity for the students about the subject developed through the application of subject about the teaching-learning model that can utilize the learners' learning experience and the development of items that can evaluate it by utilizing various and concrete operational materials as an object of the first year at the middle schools. The concrete purpose of realizing it is as follows. 1. This study is to develop the teaching-learning model centered on activities adequate to utilize various and concrete operational materials. 2. This study is to develop evaluation data for utilizing the concrete operational materials by making construction through the application of developed teaching-learning model as possible. 3. This study is to present standards initiative for reliable and fair marking about the evaluation data and to analyze the students' practice capacity by using it effectively. For accomplishing the purpose of this research, this study is the work-oriented teaching and learning model using learning data that can be easily found around the surroundings and 8 evaluation programs in order that the experience-oriented learning based on circumstances learning among the learning models of constructivism. Also, it is to examine the result after applying that on a basis of 25 students, consisted of only on class in a year, in the first year at the small size of agricultural middle schools. The result of this study from this research is as follows. First, as a result of frequency survey, of response about the developed evaluation data it showed positive response more than 80% of all the items. The atmosphere of self-directed learning was produced because the developed evaluation data could induce activity-based learning naturally by stimulating the students' curiosity and promoting interest. Second, this study executed t-test for the result of questionnaire about the mathematics propensity, and there were significant differences in 19 items among 25 items. It presented that the application of data about the teaching-learning and evaluation directly using concrete operational materials to the constructional learning by level might have desirable affect on the students' mathematical propensity. It came to be a motive that could increase value recognition and interest on the subject of mathematics by investigating a mathematical principle rule and confirming it through activity learning using the concrete operational materials.
With the recent increase in BIM orders in civil engineering, project owners are supplied with BIM-based products. Due to the lack of related guidelines, however, and the vague regulations on the creation and level of drawings as well as on the products list, many owners receive BIM-based products along with the existing 2D products. This can lead to task overlapping by designers or contractors and can cause the owners to have more data to maintain. If the existing 2D-based electronic supply system is changed to a BIM ordering system to reduce such a burden, it is necessary to examine the 2D products that can be extracted from or substituted for the BIM model so as to minimize the overlapping products. Therefore, in this study, BIM modeling of road construction projects whose enforcement plan by the Ministry of Land and Transportation has been completed was conducted, and based on the study results, the possibility of extracting 2D drawings from a total of 3,767 drawing items was determined. The results showed that 2,549 drawing items (67%) could be extracted as 2D drawings from the BIM model, and in particular, drawing items related to general connection work, earthwork, and amenities work could be most effectively substituted. The results of this study can be used as the base data for reducing the number of 2D drawings or substituting them in developing a BIM-based product supply system in the future.

This study has attempted to calculate the intensity of illumination for the optimal growth environment of indoor plants after analyzing both damage to plant species and growth conditions as impacted by light conditions for the Kyobo Life Insurance greenhouse. The optical intensity of illumination has been estimated after investigating the problems of growth conditions based on an analysis of illumination by light condition, dead tree replacement cycle(weeks) and rate of damage of plant species. According to the investigation of illumination, the lowest difference was observed between the shaded spot in the south(531lux) and the sunny spot(602lux) while the largest difference(nearly 500lux) was detected between the shaded spot in the central area(210lux) and the sunny spot(782lux). According to an analysis of dead trees from 1990 to 2004, in terms of dead tree replacement cycle, Viburnum awabuki was the highest(161weeks), followed by Phyllostachys spp.(84weeks), Camellia japonica and Ternstroemia japonica(40weeks). Regardless of plant species, damage rate of plant were lower in the shaded spot and higher in the sunny spot. According to correlation and regression analyses with the intensity of illumination as an independent variable and the damage rate of plant species as a dependent variable, the damage rate of plant species increased as the intensity of illumination decreased. A dramatic decline in the rate of damage was observed at $500{\sim}600lux$. At 700lux, it reached the lowest level.
A construction project of Incheon 2nd bridge, which is connected between the Incheon Song-Do New Town and the Incheon International Airport in Young-Jong-Do, has been proposed by the private capital in 1999. But the optimal width of the main span has not been decided in spite of the three investigations into the feasibility of ship's safe transit in this planned bridge. In this paper, we study the economic analysis on port operational efficiency according to traffic schemes, one-way or two-way of vessels over 10,000G/T, in the main span of this bridge. In this comparative result, total queueing time due to the one-way in the bridge main span is evaluated 20,362 hours in 2011 and 24,544 hours in 2020. Therefore the demurrage cost and the accumulation cost of freight are evaluated 19.7 billion won in 2011, and 23.3 billion won in 2020, then total accumulated costs during 33 years from 2008 until 2040 are evaluated about 768.9 billion won.
The Sejong Metropolitan Autonomous City is launched on July 1, 2012, and Phase 1 of the Multifunctional Administrative City Construction Project was completed in late 2015. Therefore, it is necessary through the results of the first phase of the project to check whether Sejong city can achieve the target population and number of households by 2030 and to use to determine the number and type of housing to be supplied next. Based on the presented results of the Phase 1 project period, this study estimate the population and number of households in 2030. For forecasting future population the population growth rate seen in the future of Sejong City's population forecast published by the National Statistical Office and the performance against plans Step 1 were used for forecasting future population. The results of analysis showed that the Multi-functional Administrative City is difficult to attract five hundred thousand people and two hundred thousand houses. In the analysis of households by type The Multi-functional Administrative City is The large proportion of 3-4 person households and high-income earners and Homeowners. But it increased the proportion of households with 1-2 people and rent house of the city grows in size and it is likely to change the level of income. Therefore, it is determined that there is a need to reflect these elements in next housing.
This study estimated the daily maximum snow depth using the Artificial Neural Network (ANN) model in Korean Peninsula. First, the optimal ANN model structure was determined through the trial-and-error approach. As a result, daily precipitation, daily mean temperature, and daily minimum temperature were chosen as the input data of the ANN. The number of hidden layer was set to 1 and the number of nodes in the hidden layer was set to 10. In case of using the observed value as the input data of the ANN model, the cross validation correlation coefficient was 0.87, which is higher than that of the case in which the daily maximum snow depth was spatially interpolated using the Ordinary Kriging method (0.40). In order to investigate the performance of the ANN model for estimating the daily maximum snow depth of the ungauged area, the input data of the ANN model was spatially interpolated using Ordinary Kriging. In this case, the correlation coefficient of 0.49 was obtained. The performance of the ANN model in mountainous areas above 200m above sea level was found to be somewhat lower than that in the rest of the study area. This result of this study implies that the ANN model can be used effectively for the accurate and immediate estimation of the maximum snow depth over the whole country.
In recent, the hydrological regime of the Mekong river is changing drastically due to climate change and haphazard watershed development including dam construction. Information of hydrologic feature like streamflow of the Mekong river are required for water disaster prevention and sustainable water resources development in the river sharing countries. In this study, runoff simulations at the Kratie station of the lower Mekong river are performed using SWAT (Soil and Water Assessment Tool), a physics-based hydrologic model, and LSTM (Long Short-Term Memory), a data-driven deep learning algorithm. The SWAT model was set up based on globally-available database (topography: HydroSHED, landuse: GLCF-MODIS, soil: FAO-Soil map, rainfall: APHRODITE, etc) and then simulated daily discharge from 2003 to 2007. The LSTM was built using deep learning open-source library TensorFlow and the deep-layer neural networks of the LSTM were trained based merely on daily water level data of 10 upper stations of the Kratie during two periods: 2000~2002 and 2008~2014. Then, LSTM simulated daily discharge for 2003~2007 as in SWAT model. The simulation results show that Nash-Sutcliffe Efficiency (NSE) of each model were calculated at 0.9(SWAT) and 0.99(LSTM), respectively. In order to simply simulate hydrological time series of ungauged large watersheds, data-driven model like the LSTM method is more applicable than the physics-based hydrological model having complexity due to various database pressure because it is able to memorize the preceding time series sequences and reflect them to prediction.
This study examined pollution level of sediment in Sookchun lake, and studied dredging validity by examining phosphorous release characteristics on surface polluted soil. Total phosphorous, the principal cause of algal blooms, exceeded dredging assessment standards regarding Daechung lake (1.5 mg/g) at all points. Also at all points, total nitrogen exceeded the dredging assessment standard regarding Paldang Lake (1.1 mg/g), but fell short of the standard regarding Daechung lake (3.0 mg/g). Dredging zone was suggested in this study is Chuso water body (WS-6~WS-12) in Sookchun lake. In relation to sediment pollution levels measured at different depths, LOI tended to decrease as it became deeper. The concentrations of T-N varied depending upon the depth as well as points, but no regular pattern was observed. The depth and site did not significantly influence T-P. From the results of phosphorous release tests, it was shown that total phosphorous release flux was calculated to be $7.2{\sim}15.4mg/m^2/d$ for anaerobic condition, $0.5{\sim}2.0mg/m^2/d$ for aerobic condition and $2.0{\sim}4.1mg/m^2/d$ for facultative condition. Release flux and T-P concentration of surface sediments had positive correlation ($R^2$ 0.7871). And The corelation between release flux and DO condition in reactor had strong negative correlation ($R^2$ 0.8824).
