Hydrolysis of Rice Bran Oil Using Immobilized Lipase in a Stirred-Batch Reactor

  • Candida cylindracea lipase was immobilized by adsorption on acid washed glass beads. It was observed that protein loading of the support depends on the size of the particle, with smaller particle containing higher amount of protein per unit weight. Initial reaction rate linearly varied up to enzyme concentration of 17.25 U/mL. Amount of free fatty acids produced was linearly proportional up to the enzyme loading of 1650 $\mu$g/g of bead. Achievement of chemical equilibrium took longer time in the case of less protein loading. Degree of hydrolysis was found to decrease in second and third consecutive batch operations on repeated use of immobilized lipase.

  • The potential energy surface (PES) for the loss of HCN or HNC from the pyrazine molecular ion was determined based on quantum chemical calculations using the G3//B3LYP method. Four possible dissociation pathways to form four $C_3H_3N^{+{{\bullet}}$ isomers were examined. A Rice-Ramsperger-Kassel-Marcus quasi-equilibrium theory model calculation was performed to predict the dissociation rate constant and the product branching ratio on the basis of the obtained PES. The resultant rate constant for the HCN loss agreed with the previous experimental result. The kinetic analysis predicted that the formation of $CH=CHN{\equiv}CH^{+{\bullet}}+HCN$ was predominant, which occurred by three consecutive steps, a C-C bond cleavage to form a linear intermediate, a rearrangement to form an H-bridged intermediate, and elimination of HCN.

  • The purpose of this research is for the development of a new type forging equipment H.C.G(Hyundai Continuous Grain-flow) by using two virtual build-up tools rigid viscoplastic FEM and downsized plasticine experiment. This forging equipment consists of consecutive horizontal and vertical pressure while the traditional forging method consists of only vertical pressure. Using this method high quality crankshafts can be forged as it can maintain a continuous grain flow. The factors considered in the development of equipment are die geometry for flawless deformed shape die reaction forces stress/strain distributions and continuous material flow. We carried out several numerical simulations and downsized plasticine experiments for the proper design of the forging equipment. The validity of those simulation results is confirmed by checking with the actual test results. Based on these simulation results the proper design of the H.C.G for ging equipment is enabled.

Influence of changes in cement fineness on lean mixture mortar quality (시멘트 분말도 변화가 빈배합 모르타르의 품질에 미치는 영향)

  • The fineness degree of Ordinary Portland Cement (OPC henceforth) usually used in Korea's construction sites, is designated as over 2,800㎠/g. But the higher the fineness, the surface area of hydration reaction on water increases as well, resulting in large early age strength and high-intensity; so the trend is to prefer a high degree of fineness. But from a pore-space filling perspective, fine-particled cement is not always beneficial to intensity. Therefore in this study artificial modifications were given to cement fineness to analyze the effect of various fineness changes on the liquidity, air quantity and intensity of lean mixture cement mortar. As a result, the greater the degree of fineness, the better the cement was, with fine particle+OPC having the most satisfactory results due to consecutive particle distribution.

Microarray Analysis of Gene Expression by Ginseng Water Extracts in a Mouse Adrenal Cortex after Immobilization Stress

  • To investigate the effects of repeated immobilization-stress challenge on the the hypothalamic-pituitary-adrenal axis, the genomic transcriptome in the adrenal cortex of immobilization-stressed mouse was analyzed by using a cDNA microarray. Mice were subjected to immobilization stress for 2 h per day for 5 consecutive d. With a 4.0-fold cutoff of arbitrary criteria, the expression levels of 168 out of 41,174 genes were significantly modulated in the adrenal cortex by stress when comparing the control and experimental groups. These genes were related to apoptosis, cell cycle, immune response, inflammatory responses, and signal transduction, and thus may be used as potential targets for the development of therapeutics for chronic stress or depression. Six significant genes among these were selected for real time polymerase chain reaction analysis to confirm the change of their expression levels. The gene for phospho 1 was also further investigated because its expression showed the greatest fold-change.

Effects of Quercetin on the Immune Responses in Mice (Quercetin이 마우스의 면역반응에 미치는 영향)

  • Effects of quercetin on the specific and non-specific immune responses were studied in vivo. Quercetin at a dose of 2.5, 5, 10, 20 and 40 mg/kg were orally administered to ICR male mice once daily for 28 consecutive days. Cyclophosphamide was injected intraperitoneally to ICR mice with a single dose of 5 mg/kg 2 days before secondary immunization. Mice were sensitized and challenged with sheep red blood cells (S-RBC). Immune responses were evaluated by humoral and cellular immune reponses and non-specific immune response. The results of this study were summarized as followings; 1. Quercetin significantly decreased the body weight, and introduced the atrophy of liver, spleen and thymus gland dose-dependently, but increased the numbers of white blood cell. 2. Querectin significantly depressed the hemagglutination titer, Arthus reaction and hemolytic plaque forming cell. 3. Quercetin significantly depressed the delayed type hypersensitivity and rosette forming cell. 4. Quercetin at a dose of 2.5, 5 and 40 mg/kg significantly depressed phagocytic activity. 5. Quercetin at a dose of 10 and 20 mg/kg significantly increased natural killer cell activity.

  • Carbonation of $Mg(OH)_2$ at 300 ${^{\circ}C}$ was studied by Infrared spectroscopy. Dehydroxylation and carbonation reactions were carried out in consecutive manner via 2-step procedure. Unidentate carbonates were produced only on defective surface of MgO in situ generated by dehydroxylation of $Mg(OH)_2$$Mg(OH)_2$${^{\circ}C}$ was proposed.

  • This research was aimed at developing new catalytic membrane reactors to be used for : i) partial oxidation of toluene (to benzaldehyde and benzoic acid) ii) oxidative dehydrogenation of propane iii) complete oxidation of propane and toluene. The reactor is particularly useful for the optimisation and the industrial development of heterogeneous catalytic processes, particularly for those processes where it is necessary to control the reactants stoichiometry in the reaction zone. This control limits consecutive reactions, thus obtaining high selectivity with industrially interesting conversions. This presentation will concentrate on the partial oxidation of toluene.

pH-Dependent Electrochemical Behavior of N-Monosubstituted-4,$4^{\prime}$-Bipyridinium Ions

  • The pH-dependent reduction behavior of N-monosubstituted-4,4'-bipyridinium ions ($RBPY^+: R=methyl(C_1)$; benzyl; n-octyl; n-dodecyl) has been investigated by electrochemical and spectroelectrochemical techniques. At acidic condition, $RBPY^+$ is protonated and the protonated species are reduced by two consecutive one-electron processes. The $2e^-$ reduced species undergoes a chemical reaction with $H^+$. The second-order rate constant $(k_H)$ of the homogeneous chemical process is $(3.7{\pm}0.3){\times}10^3M^{-1}s^{-1}$ for the two electron reduction product of $C_1BPY^+$. At high pH, the electrode reduction of $RBPY^+$ is one-step $2e^-$ transfer process with concomitant addition of $H^+$, which is confirmed by cyclic voltammetric study using a microdisk electrode.

Study on the Development of CVD Precursors II-Synthesis and Properties of New Lathanum β-diketonates

  • A new synthetic route for the lanthanum β-diketonate compounds via in-situ formed lanthanum alkyl complexes was developed in the process for the development of suitable MOCVD (metal-organic chemical vapor deposition) precursors of PLT, one of the promising material for the ferroelectric film. A series of lanthanum β-diketonate compounds were successfully synthesized by this method. This new method is found to have some merits; versatile method for almost every β-diketone, β-hydroxyketone, and β-hydroxyaldehyde, short reaction time, easy purification for high purity, moderate to high yield, and easy access to anhydrous compounds. In some cases, anhydrous oligomeric products fail to show the higher volatility. On the other hand, some lanthanum β-diketonates with aromatic groups such as La(1,3-biphenyl-l,3-propandione)3 are found to have favorable properties for a precursor of lanthanum oxide, one of major components of PLT, such as low melting point, and much higher decomposition temperature. A plausible pyrolysis mechanism is proposed by the TGA, where consecutive dissociation of R, CO, CH, C, and O fragments occurs.
