A composite microbial system (XDC-2) was used to pretreat and hydrolyze corn stalk to enhance anaerobic digestion. The results of pretreatment indicated that sCOD concentrations of hydrolysate were highest (8,233 mg/l) at the fifth day. XDC-2 efficiently degraded the corn stalk by nearly 45%, decreasing the cellulose content by 22.7% and the hemicellulose content by 74.1%. Total levels of volatile products peaked on the fifth day. The six major compounds present were ethanol (0.29 g/l), acetic acid (0.55 g/l), 1,2-ethanediol (0.49 g/l), propionic acid (0.15 g/l), butyric acid (0.22 g/l), and glycerine (2.48 g/l). The results of anaerobic digestion showed that corn stalks treated by XDC-2 produced 68.3% more total biogas and 87.9% more total methane than untreated controls. The technical digestion time for the treated corn stalks was 35.7% shorter than without treatment. The composite microbial system pretreatment could be a cost-effective and environmentally friendly microbial method for efficient biological conversion of corn stalk into bioenergy.
One chambered sediment microbial fuel cell (SMFC) was equipped with Fe, brass (Cu/Zn), Fe/Zn, Cu, Cu/carbon cloth and graphite felt anode. Graphite felt was used as common cathode. The SMFC was membrane-less and mediator-less as well. Order of anodic performance on the basis of power density was Fe/Zn ($6.90Wm^{-2}$) > Fe ($6.03Wm^{-2}$) > Cu/carbon cloth ($2.13Wm^{-2}$) > Cu ($1.13Wm^{-2}$) > brass ($Cu/Zn=0.24Wm^{-2}$) > graphite felt ($0.10Wm^{-2}$). Fe/Zn composite anode have twisted 6.73% more power than Fe alone, Cu/carbon cloth boosted power production by 65%, and brass (Cu/Zn) produced 65% less power than Cu alone. Graphite felt have shown the lowest electricity generation because of its poor galvanic potential. The estuarine sediment served as supplier of oxidants or electron producing microbial flora, which evoked electrons via a complicated direct microbial electron transfer mechanism or making biofilm, respectively. Oxidation reduction was kept to be stationary over time except at the very initial period (mostly for sediment positioning) at anodes. Based on these findings, cost effective and efficient anodic material can be suggested for better SMFC configurations and stimulate towards practical value and application.
To obtain an efficient natural lignocellulolytic complex enzyme, we screened an efficient lignocellulose-degrading composite microbial system (XDC-2) from composted agricultural and animal wastes amended soil following a long-term directed acclimation. Not only could the XDC-2 degrade natural lignocelluloses, but it could also secrete extracellular xylanase efficiently in liquid culture under static conditions at room temperature. The XDC-2 degraded rice straw by 60.3% after fermentation for 15 days. Hemicelluloses were decomposed effectively, whereas the extracellular xylanase activity was dominant with an activity of 8.357 U/ml on day 6 of the fermentation period. The extracellular crude enzyme noticeably hydrolyzed natural lignocelluloses. The optimum temperature and pH for the xylanase activity were $40^{\circ}C$ and 6.0. However, the xylanase was activated in a wide pH range of 3.0-10.0, and retained more than 80% of its activity at $25-35^{\circ}C$ and pH 5.0-8.0 after three days of incubation in liquid culture under static conditions. PCR-DGGE analysis of successive subcultures indicated that the XDC-2 was structurally stable over long-term restricted and directed cultivation. Analysis of the 168 rRNA gene clone library showed that the XDC-2 was mainly composed of mesophilic bacteria related to the genera Clostridium, Bacteroides, Alcaligenes, Pseudomonas, etc. Our results offer a new approach to exploring efficient lignocellulolytic enzymes by constructing a high-performance composite microbial system with synergistic complex enzymes.
Microbial fuel cells (MFCs) are a bioelectrochemical system where electrochemically active bacteria convert organic waste into electricity. Poly(vinyl alcohol) (PVA) and chitosan (CS) are polymers that have been studied as potential alternative ion exchange membranes to Nafion for many electrochemical systems. This study examined the optimal mixing ratio of PVA and chitosan CS in a PVA:CS composite membrane for MFC applications. PVA:CS composite membranes with 1:1, 2:1, and 3:1 ratios were synthesized and tested. The water uptake and ion exchange capacity, Fourier transform infrared spectra, and scanning electron microscopy images were analyzed to determine the physicochemical properties of PVA:CS membranes. The prepared membranes were applied to the ion exchange membrane of the MFC system, and their effects on the electrochemical performance were evaluated. These results showed that the composite membrane with a 3:1 (PVA:CS) ratio showed comparable performance to the commercialized Nafion membrane and produced more electricity than the other synthesized membranes. The PVA:CS membrane implemented MFCs produced a maximum power density of 0.026 mW cm-2 from organic waste with stable performance. Therefore, it can be applied to a cost-effective MFC system.
A cellulose-degrading composite microbial system containing a mixture of microbes was previously shown to demonstrate a high straw-degrading capacity. To estimate its potential utilization as an inoculant to accelerate straw biodegradation after returning straw to the field, two cellulose-degrading composite microbial systems named ADS3 and WSD5 were inoculated into wheat straw-amended soil in the laboratory. The microbial survival of the inoculant was confirmed by a denaturing gradient gel electrophoresis (DGGE) analysis, whereas the enhancement of straw degradation in soil was assessed by measuring the mineralization of the soil organic matter and the soil cellulase activity. The results indicated that most of the DGGE bands from ADS3 were detected after inoculation into straw-amended autoclaved soil, yet only certain bands from ADS3 and WSD5 were detected after inoculation into straw-amended non-autoclaved soil during five weeks of incubation; some bands were detected during the first two weeks after inoculation, and then disappeared in later stages. Organic matter mineralization was significantly higher in the soil inoculants ADS3 and WSD5 than in the uninoculated controls during the first week, yet the enhanced degradation did not persist during the subsequent incubation. Similar to the increase in soil organic matter, the cellulase activity also increased during the first week in the ADS3 and WSD5 treatments, yet decreased during the remainder of the incubation period. Thus, it was concluded that, although the survival and performance of the two inoculants did not persist in the soil, a significant enhancement of degradation was present during the early stage of incubation.
A microbial fuel cell (MFC) has been regarded as noble clean energy technology that can directly convert biomass to electricity. However, its low power density is a main limitation to be used as a new energy source. To overcome this limitation, we focused on the anode improvement in a mediator-type MFC using P. vulgaris as a biocatalyst. Fuel cell performance increased when the anode was coated with carbon black or polypyrrole. The best performance was observed when polypyrrole/carbon black (Ppy/CB) composite material was coated on a carbon paper electrode. Our obtained value of 452 mW $m^{-2}$ is the highest value among the reported ones for the similar system. The effects of amount of Ppy/CB, mediator concentration, and amount of P. vulgaris have also been examined.
To evaluate the potential utility of pretreatment of raw biomass with a complex microbial system, we investigated the degradation of rice straw by BMC-9, a lignocellulose decomposition strain obtained from a biogas slurry compost environment. The degradation characteristics and corresponding changes in the bacterial community were assessed. The results showed that rapid degradation occurred from day 0 to day 9, with a peak total biomass bacterium concentration of $3.3{\times}10^8$ copies/ml on day 1. The pH of the fermentation broth declined initially and then increased, and the mass of rice straw decreased steadily. The highest concentrations of volatile fatty acid contents (0.291 mg/l lactic acid, 0.31 mg/l formic acid, 1.93 mg/l acetic acid, and 0.73 mg/l propionic acid) as well as the highest xylanse activity (1.79 U/ml) and carboxymethyl cellulase activity (0.37 U/ml) occurred on day 9. The greatest diversity among the microbial community also occurred on day 9, with the presence of bacteria belonging to Clostridium sp., Bacillus sp., and Geobacillus sp. Together, our results indicate that BMC-9 has a strong ability to rapidly degrade the lignocelluloses of rice straw under relatively inexpensive conditions, and the optimum fermentation time is 9 days.
Microbial diversity in the soil is responsive to changes in soil composition. However, the impact of soil amendments on the diversity and structure of rare and abundant sub-communities in agricultural systems is poorly understood. We investigated the effects of different Chinese herb residue (CHR) soil amendments and cropping systems on bacterial rare and abundant sub-communities. Our results showed that the bacterial diversity and structure of these sub-communities in soil had a specific distribution under the application of different soil amendments. The CHR soil amendments with high nitrogen and organic matter additives significantly increased the relative abundance and stability of rare taxa, which increased the structural and functional redundancy of soil bacterial communities. Rare and abundant sub-communities also showed different preferences in terms of bacterial community composition, as the former was enriched with Bacteroidetes while the latter had more Alphaproteobacteria and Betaproteobacteria. All applications of soil amendments significantly improved soil quality of newly created farmlands in whole maize cropping system. Rare sub-communitiy genera Niastella and Ohtaekwangia were enriched during the maize cropping process, and Nitrososphaera was enriched under the application of simple amendment group soil. Thus, Chinese medicine residue soil amendments with appropriate additives could affect soil rare and abundant sub-communities and enhance physicochemical properties. These findings suggest that applying soil composite amendments based on CHR in the field could improve soil microbial diversity, microbial redundancy, and soil fertility for sustainable agriculture on the Loess Plateau.
Organic fouling and biofouling pose a significant challenge to the membrane filtration process. Photocatalysis-membrane hybrid system is a novel idea for reducing these membranes fouling however, when $TiO_2 photocatalyst nanoparticles are used in suspension, catalyst recovery is not only imposes an extra step on the process but also significantly contributes to increased membrane resistance and reduced permeate flux. In this study, $TiO_2$ photocatalyst has been immobilized by coating on the microfiltration (MF) membrane surface to minimize organic and microbial fouling. Nano-sized $TiO_2$ was first synthesized by a sol-gel method. The synthesized $TiO_2$ was coated on a Poly Vinyl Difluoride (PVDF) membrane (MF) surface using spray coating and dip coating techniques to obtain hybrid functional composite membrane. The characteristics of the synthesized photocatalyst and a functional composite membrane were studied using numerous instruments in terms of physical, chemical and electrical properties. In comparison to the clean PVDF membrane, the $TiO_2$ coated MF membrane was found more effective in removing methylene blue (20%) and E-coli (99%).
본 연구에서는 경제성 있는 생물학적 만니톨 고생산을 위해 선별균주의 만니톨 생산 최적화 배지조성을 RSM 방법을 이용하여 확립하고자 하였다. 먼저 김치로부터 분리된 10균주의 만니톨 생산량과 과당으로부터 만니톨 전환율 분석을 통하여 SRCM201425 균주를 선발하였으며, 선발균주는 16S rRNA 유전자 염기서열과 당 발효 분석을 통하여 Leuconostoc mesenteroides로 동정하였다. Plackett-Burman design (PBD)을 이용하여 만니톨 생산에 영향을 주는 배지 인자를 선별하기 위해 총 11개의 탄소원, 질소원, 무기원소의 영향을 조사하였으며, 통계학적 분석을 통하여 최종적으로 fructose와 sucrose, peptone을 선정하였다. 만니톨 생산을 위한 선별된 각 변수의 최적 농도를 결정하기 위한 방법으로 central composite design (CCD)과 반응표면 분석법을 이용하였으며, 최종적으로 CCD를 통해 만니톨 생산을 위한 배지 조성의 최적 농도는 fructose 38.68 g/l, sucrose 30 g/l, peptone 39.67 g/l으로 예측되었으며, 통계학적 분석을 통해 실험모델의 적합성을 확인하였다. 최종적으로 MRS 배지에서의 생산량의 약 20배의 만니톨을 생산할 수 있었으며, 100 g/l의 fructose가 포함된 MRS 배지 대비 97.46%의 만니톨을 생산하면서, 산업화시 기존 배지 대비 생산비용을 크게 절감할 수 있을 것으로 예상되었다. 본 연구를 통하여 만니톨 생산을 위한 배지 조성의 최적화를 확립하였으며, 만니톨을 생산하기 위한 방법으로 주로 사용되고 있는 고비용의 촉매환원 방법을 대체할 방법을 제시할 수 있는 기초자료로 활용될 수 있을 것으로 기대된다.
