A Formulation Study on the Combined Preparation of Nitroxynil and Levamisole for Subcutaneous Injection (Nitroxynil과 Levamisole의 피하 주사용 복합제제의 설계에 관한 연구)

  • In order to develop a broad-spectrum veterinary anthelmintic, a combined preparation of nitroxynil and levamisole was formulated for subcutaneous injection. The preformulation studies on solubilization, physicochemical stability and toxicity of combined preparation were performed. The combined preparation of nitroxynil-N-ethylglucamine and levamisole base could be solubilized up to 50.3%(w/v) of active ingredient concentration in propylene glycol/water system. Injectable solutions were most stable at $4^{\circ}C$. Local toxicities such as flare and edema were not shown when the usual dose of the combined preparation was injected subcutaneously to the rats.

Effects of the Combined-Preparation of Germinated Brown Rice, Cultured Mountain Ginseng and Longanae Arillus on Pentobarbital-induced Sleeping Time (발아현미, 배양산삼 및 용안육 혼합 제제가 Pentobarbital로 유도된 수면시간에 미치는 영향)

  • This experiment was performed to investigate whether the combined-preparation of water extracts of germinated brown rice (WGR), water extracts of cultured mountain ginseng (WCG) and 70% ethanol extracts of Longanae Arillus (ELA) has hypnotic effects and/or enhances pentobarbital-induced sleep behaviors through the GABAergic system. The combined-preparation of WGR and WCG reduced sleep latency and prolonged sleep time induced by pentobarbital. ELA also reduced sleep latency and prolonged sleep time induced by pentobarbital. However, WGR or WCG itself did not induce sleep. The combined-preparation of WGR, WCG and ELA strongly reduced sleep latency and prolonged sleep time via chloride influx into primary cultured cerebellar granule cells. In conclusion, the combined-preparation of WGR, WCG and ELA augments pentobarbital-induced sleep behaviors through the modification of GABAergic system.

Enhancing Effect of the Combined Preparation Containing Antacid and Aceglutamide Aluminium on Defensive Factors in Chronic Ulcer Model of the Rat (흰쥐의 만성궤양모델에서 Aceglutamide aluminium를 함유한 복합제산제의 점막방어인자 증강작용)

  • Antacid(AM, 600 mg/kg=aluminium hydroxide, magnesium hydroxide, and simethicone with a ratio of 1 : 1 : 0.1) and aceglutamide aluminium(AGA, 263 mg/kg)-Effect of the combined preparation containing on the gastric mucosal hexosamine, sialic acid, and aluminium contents adhered to the gastric wall of the rat was investigated. Severe ulcers were produced in rats by injecting of $30\;{\mu}l$ acetic acid(30%) into the subserosal layer of one position in the corpus. When given orally for 15 consecutive days, AM(1,200 mg/kg), AGA(525, 1,050 mg/kg), and the combined preparation significantly decreased the ulcer area. AGA(525, 1,050 mg/kg) and the combined preparation also increased the amount of hexosamine and sialic acid in the intact and ulcerated areas. On the other hand, the contents of hexosamine and sialic acid were not affected by AM (600, 1,200 mg/kg). The amount of aluminium adhered to the gastric wall of the rat was higher in the combined preparation when compared to the AM(600 mg/kg) and AGA(263 mg/kg). The aluminium contents adhered may play an important role protecting mucosa from aggresive action of gastric juice and potenting defensive factors through the increase of mucosa-forming components by AGA.

Effect of Several Combined Preparation of Crude Drugs on the Adverse Effects of Anticancer Agent-Mitomycin C (항암제 Mitomycin C의 부작용에 대한 수종 복합생약의 영향)

  • The studies were conducted to investigate the combined effects of several combined preparation of crude drugs and mitomycin C(MMC). The combined effects on the proliferation of HepG2, A549, KHOS-Np, A431 and HeLa cells were estimated by MTT colorimetric assays. Sa Kunja Tang(SKT), Boyang Hwanoh Tang(BHT) and Hyulbu Choogo Tang(HCT) inhibited the proliferation of A549 and HeLa cell. The inhibitory action of MMC was increased by the combined treatment of SKT and MMC, and Sa Mul Tang(SMT) and MMC, respectively. When the mice were treated by MMC, the number of leukocyte was decreased significantly at the 3rd day, but recovered at the 7th day. In the groups of MMC treated with SKT or HCT, the number of leukocyte was increased significantly that the group of MMC treated only at the 1st and 3rd day. The combined treatment of SKT, SMT, BHT, HCT and MMC retained the spleen weight of mice at the level of normal mice, but decreased the thymus weight of mice. The combined treatment of SKT, SMT, BHT, HCT and MMC increased the number of PFC significantly than the MMC treated group. The combined treatment of SKT, SMT, BHT, HCT and MMC increased the T cell proliferation significantly thant the MMC treated group.

Acute Toxicity of a Combined Preparation of the Standardized Extracts of Scutellaria baicalensis GEORGI and Salvia miltiorrhiza BUNGE (황금 및 단삼 표준화시료의 급성독성에 관한 연구)

  • Acute toxicity of a combined preparation of the standardized extracts Scutellaria baicalensis GEORGI and Salvia miltiorrhiza BUNGE in a ratio of 3:1 was examined in male and female ICR mice. Mice were treated with the test substance intragastrically at a dose of 0 mg/kg, 5 mg/kg, 50 mg/kg, 500 mg/kg or 2,000 mg/kg and observed for two weeks. No death or abnormal clinical sign was shown during the observation period. Also there were no difference in net body weight gain, organ weight, and gross pathological findings at the terminal sacrifice. The results suggested that acute oral toxicity of a combined preparation of the standardized extracts is very low at the conditions employed in this study.

Cross-Sectional Transmission Electron Microscopy Specimen Preparation Technique by Backside Ar Ion Milling

  • Backside Ar ion milling technique for the preparation of cross-sectional transmission electron microscopy (TEM) specimens, and backside-ion milling combined with focused ion beam (FIB) operation for electron holography were introduced in this paper. The backside Ar ion milling technique offers advantages in preparing cross-sectional specimens having thin, smooth and uniform surfaces with low surface damages. The back-side ion milling combined with the FIB technique could be used to observe the two-dimensional p-n junction profiles in semiconductors with the sample quality sufficient for an electron holography study. These techniques have useful applications for accurate TEM analysis of the microstructure of materials or electronic devices such as arrayed hole patterns, three-dimensional integrated circuits, and also relatively thick layers (> $1{\mu}m$).

COMPARISON OF MARGINAL LEAKAGE OF CLASS 5 GIC RESTORATIONS ACCORDING TO CAVITY DESIGNS (와동 형태에 따른 5급 글라스 아이오노머 시멘트 수복물의 변연누출 비교)

  • The purpose of this study was to assess the effects of cavity designs on the marginal leakage of class 5 glass ionomer restorations. The five cavity designs were as follows ; notch shape(A group), notch shape with groove(B group), combined lesion(C group), combined lesion with groove and deep chamfer margin(D group) and combined lesion with groove and shoulder(E group), and each design had 10 cavities. After the cavities were restored with GIC, they were immersed in 0.5% basic fuchsin solution for 6 hours. The specimens were washed thoroughly and sectioned longitudinally in a buccolingual direction through the center of the restorations. The degree of marginal leakage was measured as the extent of dye penetration under the stereoscope. The results of this study were as follows 1. The enamel margins of all groups showed lesser leakage than dentin/cementum margins(p<0.05). 2. The combined lesion(C group) showed more leakage than notch shape(A group), but there was no siginificant difference(p>0.05). 3. In the notch shape, there was no influence on the marginal leakage by the groove preparation. 4. In the combined lesion, marginal leakage was decreased by the groove preparation and marginal modification.

Oocysticidal effects of morestan combination preparations against oocysts of Eimeria tenella (Morestan 합제의 Eimeria tenella 오오시스트에 대한 살멸효과)

  • The tests on the oocysticidal effects of three formulations (75% o-dichlorobenzene, 7.5% cresol combined with morestan 0.5%, 1.0%, 1.5%, respectively) against immature oocysts of Eimeria tenella, were carried out. A o-dichlorobenzene preparation (72.5% o-dichlorobenzene, 7.5% cresol) on the market was selected and tested as the control. The tests were divided into two ways, the one was evaluated at different dilutions and contact times. The other was performed at different storage days. The test results are summarized as follows. Oocysticidal effects of 100% were revealed at 1 to 3 hours contact time of all morestan combined preparations. On the other hand, the perfect oocysticidal effect was revealed only at 24 hours exposure time of 100 times dilution among dilutes of control preparation. Combined rate of morestan was higher, oocysticidal effect was better. In the latter test, all morestan combined formulations had 100% of oocysticidal effect up to 5 days storage after dilution and 86.4% to 96.7% at two weeks storage after dilution, whereas the oocysticidal effects of the control preparation was gradually decreased by the storage time was passed and there was no effect at 2 weeks storage after dilution. With these results, morestan combined preparations have synergistic effect against oocysts of E tenella and can be used as the most effective oocysticidal disinfectant for broiler industry in future.

  • The effect of nonionic surfactant(polyoxyl 40 hydrogenated castor oil, PHCO), a common solubi-lizer, on the solubility of silymarin in the combined preparation containing ursoseoxycholic acid(UDCA) and silymarin was investigated in vivo using HPLC. The solubility of silybin, a major component of silymarin, was enhanced by increasing the amount of PHCO. The effect of PHCO on bioavailability was also evaluated in rats. The bioavailability was calculated by silybin content in bile juice that was excreted for 24 hr after oral administration. It was found that the bioavailability of silymarin containing PHCO was significantly increased compared to that of control. These results suggest that PHCO may improve the solubility and bioavailabilty of silymarin when it is combined with UDCA and silymarin.

  • This experiment was performed to investigate whether the combined-preparation of crude drugs (The $Well^{TM}$ Preparation, TW), has hypnotic effects and/or enhances pentobarbital-induced sleep behaviors. TW was mixed with water extracts of Ginseng Radix red, Germinated brown rice, cultured mountain ginseng, and 50% ethanol extracts of Longanae Arillus, Nelumbinis Folium and Chrysanthemi Flos. TW (100 mg/kg, p.o.) reduced sleep onset and prolonged sleep time induced by pentobarbital similar to muscimol (0.2 ${\mu}M$), a $GABA_A$ receptor agonist. Also, TW (2 ${\mu}g$/ml) and pentobarbital (2.5 ${\mu}M$) did not affect the chloride influx in primary cultured cerebellar granule cells, respectively, but the combined-treatment of TW (2 ${\mu}g$/ml) and pentobarbital (2.5 ${\mu}M$) increased the chloride influx onto the cells. In conclusion, TW augments pentobarbital-induced sleep behaviors; these effects may result from chloride channel activation.
