Conservation Treatment of the Seated Stone Bodhisattva Discovered in Pyeongchang and Restoration of the Statue Using 3D Digital Technologies (평창 발견 석조보살좌상의 보존처리와 3차원 디지털기술을 활용한 복원)

  • A stone seated bodhisattva (Sinsu5971) was discovered in Pyeongchang-gun, Gangwon-do in 1974 and was transferred to the Chuncheon National Museum upon its opening in 2002. The statue had damage to wide areas and was thus difficult to restore. This study utilized 3D scanning and 3D printing technologies to identify the overall form of the statue and the degree of damage, which allowed the restoration of lost portions that otherwise could not have been accurately restored to their original shape. Prior to the conservation treatment, the pigments used to decorate the surface were investigated using an optical microscope, and their main components were analyzed with a p-XRF (Potable X-ray Fluorescence Analyzer). The deteriorated lacquered surface was stabilized using animal glue and consolidated with stone strengthener (OH-100). The investigation found that the surface of the statue was made of zeolite that was lacquered and then gilded. As for pigments, white lead was used for the white color and red lead and cinnabar were used for red. The lost portions were redesigned by mirroring the remaining parts with 3D technologies. However, it was difficult to affix the 3D printing outputs to the statue without visible gaps since the damaged parts suffered flection. The portions of the outputs to be connected to the statue were thus modified and supplemented. It was also difficult to collect data on the properties of 3D printing materials due to the lack of previous in-depth study. These obstacles are subjects for further study.

  • Recently, intelligent packaging of foods has been increasingly developed in response to the growing interest of consumers in checking food quality. Indicators, an important element in intelligent packaging, change color to detect specific substances or indicate food quality changes. Gas indicators can be built into food packaging to detect volatile substances that are released when food quality changes. Ethylene gas is produced as climacteric fruits ripen. Climacteric fruit ripening results from a rapid increase in ethylene production and respiration. In the case of packaged fruits, the ethylene gas concentration in the headspace is closely related to the ripeness of each fruit variety. If an ethylene gas indicator that can be used in fruit packaging is available, the consumer will be able to eat the fruit at the optimal time. In this paper, the characteristics and pros and cons of the ethylene gas indicators developed so far were analyzed by reviewing various types of indicators such as metal reduction-based indicator, fluorescence-based indicator, pH indicator-based indicator, and liposome-based indicator.

Impact of Shading Net Color on Phytochemical Contents in Two Chili Pepper Hybrids Cultivated Under Greenhouse Conditions

  • The objective of this study was to understand the combined effect of shading treatment and harvest time on pungency, ascorbic acid content, market-quality pepper yield, and photosynthetic activity in a yellow 'Star Flame' and 'Fire Flame' F1 hybrid pepper cultivar (Capsicum annuum). Plants were cultivated in a greenhouse under white, red, and green shading nets and compared to those grown under unshaded control conditions. The ascorbic acid and pungency parameters were analyzed by high-performance liquid chromatographic methods. Ascorbic acid, a compound of high nutritive value in pepper, was highest in fruits gathered during the first summer harvest in both hybrids. The ascorbic acid content was found to be further increased in fruits from plants grown under white net shading in both 'Star Flame' and 'Fire Flame'. Total capsaicinoid contents in 'Star Flame' were influenced by the interaction of shading treatment and harvest time (p = 0.004), with the lowest amounts ($241.2-251.5mg{\cdot}kg^{-1}$ fresh weight) resulting from white and red shading treatments at the early autumn harvest. In 'Fire Flame', both shading treatment (p = 0.009) and harvest time (p < 0.001) affected total capsaicinoid contents. We observed significantly higher total capsaicinoid contents in control fruits compared to that resulting from red and green shading treatments at the second harvest, and that resulting from red shading treatment at the third harvest. Analysis of photosynthetic activity revealed a significant and negative correlation between Fv/Fm values and total capsaicinoid contents in both hybrids (p < 0.001). Based on the obtained results, the harvest of peppers during summer should be promoted to produce fruits with high pungency.

A study on mitigation of membrane fouling by ozonation/coagulation in ultrafiltration (오존산화/응집 혼성공정에 의한 UF 분리막의 막오염 저감에 관한 연구)

  • Microfiltration (MF) and Ultrafiltration (UF) membrane processes capable of producing highly purified water have been extensively applied as a pretreatment process in the wastewater reuse field with the improvement of membrane properties and resistance, development of operating protocols, and improvement of technologies of backwashing and physicochemical cleaning, and improvement of scale and antifoulants. However, despite of the development of membrane production and process technologies, fouling still remains unresolved. This study confirmed that foulants such as polysaccharides, proteins and humic substances existed in final treated effluent (secondary effluent) by fluorescence excitation emission matrix (FEEM) and fourier transform infrared spectroscopy (FTIR) analysis. In addition, when constructing ozone oxidation and coagulation processes as a hybrid process, the removal efficiency was 5.8%, 6.9%, 5.9%, and 28.2% higher than that of the single process using coagulation in turbidity, color, dissolved organic carbon (DOC), and UV254, respectively. The reversible and irreversible resistances in applying the hybrid process consisting of ozone oxidation and coagulation processes were lower than those in applying ozone oxidation and coagulation processes separately in UF membrane process. Therefore, it is considered possible to apply ozonation/coagulation as a pretreatment process for stable wastewater reuse by and then contributing to the reduction of fouling when calculating the optimal conditions for ozone oxidation and coagulation and then to applying them to membrane processes.

FISH Karyotype and GISH Meiotic Pairing Analyses of a Stable Intergeneric Hybrid xBrassicoraphanus Line BB#5

  • xBrassicoraphanus line BB#5, a new synthetic intergeneric hybrid between Brassica rapa L. ssp. pekinensis and Raphanus sativus L. var. rafiphera induced by N-methyl-N-nitroso-urethane mutagenesis in microspore culture, shows high seed fertility and morphological uniformity. Dual-color fluorescence in situ hybridization (FISH) using 5S and 45S rDNA probes and genomic in situ hybridization (GISH) using B. rapa genomic DNA probe were carried out to analyze the chromosome composition and the meiosis pairing pattern compared to its parental lines. The somatic chromosome complement is 2n = 38, which consists of 17 metacentric and two submetacentric chromosomes with lengths of 2.18 to $5.01{\mu}m$. FISH karyotype analysis showed five and eight pairs of 5S and 45S rDNA loci. GISH meiosis pairing analysis showed that 19 complete bivalents were most frequent and accounted for 42% of the 100 pollen mother cells examined. Based on chromosome number, size, morphology, rDNA distribution, and meiosis pairing pattern, both parental genomes of B. rapa and R. sativus appear to exist in xBrassicoraphanus line BB#5, demonstrating its genome integrity. Such stable chromosome constitutions and meiotic pairing patterns in somatic and meiotic cells are very rare in natural and synthetic intergeneric hybrids. Chromosomal studies and genetic and phenotypic changes in allopolyploids a re discussed. Cytogenetic Profile of De Novo B lineage Acute Lymphoblastic Leukemia: Determination of Frequency, Distribution Pattern and Identification of Rare and Novel Chromosomal Aberrations in Indian Patients

  • Background: Chromosomal aberrations identified in acute lymphoblastic leukemia (ALL) have an important role in disease diagnosis, prognosis and management. Information on karyotype and associated clinical parameters are essential to physicians for planning cancer control interventions in different geographical regions. Materials and Methods: In this study, we present the overall frequency and distribution patterns of chromosomal aberrations in both children and adult de novo B lineage ALL Indian patients using conventional cytogenetics, interphase FISH and multiplex RT-PCR. Results: Among the 215 subjects, cytogenetic results were achieved in 172 (80%) patients; normal karyotype represented 37.2% and abnormal 62.8% with a distribution as follows: 15.3% hypodiploidy; 10.3% hyperdiploidy; 15.8% t(9;22); 9.8% t(1;19); 3.7% t(12;21); 2.8% t(4;11); 2.8% complex karyotypes. Apart from these, we observed several novel, rare and common chromosomal rearrangements. Also, FISH studies using LSI extra-signal dual-color probes revealed additional structural or numerical changes. Conclusions: These results demonstrate cytogenetic heterogeneity of ALL and confirm that the incidence of chromosomal abnormalities varies considerably. To the best of our knowledge, this is one of the largest reported series of cytogenetic investigations in Indian B-lineage ALL cases. In addition, ongoing cytogenetic studies are warranted in larger groups of B-lineage ALL cases to identify newly acquired chromosomal abnormalities that may contribute to disease diagnosis and management.

Dissolved organic matter characteristics and bacteriological changes during phosphorus removal using ladle furnace slag

  • A sidestream contains the filtrate or concentrate from the belt filter press, filter backwash and supernatant from sludge digesters. The sidestream flow, which heads back into the sewage treatment train, is about 1-3% less than the influent flow. However, the sidestream can increase the nutrient load since it contains high concentrations of phosphorus and nitrogen. In this study, the removal of PO4-P with organic matter characteristics and bacteriological changes during the sidestream treatment via ladle furnace (LF) slag was investigated. The sidestream used in this study consisted of 11-14% PO4-P and 3.2-3.6% soluble chemical oxygen demand in influent loading rates. LF slag, which had a relatively high $Ca^{2+}$ release compared to other slags, was used to remove $PO_4-P$ from the sidestream. The phosphate removal rates increased as the slag particle size decreased 19.1% (2.0-4.0 mm, 25.2% (1.0-2.0 mm) and 79.9% (0.5-1.0 mm). The removal rates of dissolved organic carbon, soluble chemical oxygen demand, color and aromatic organic matter ($UV_{254}$) were 17.6, 41.7, 90.2 and 77.3%, respectively. Fluorescence excitation-emission matrices and liquid chromatography-organic carbon detection demonstrated that the sidestream treatment via LF slag was effective in the removal of biopolymers. However, the removal of dissolved organic matter was not significant during the treatment. The intact bacterial biomass decreased from $1.64{\times}10^8cells/mL$ to $1.05{\times}10^8cells/mL$. The use of LF slag was effective for the removal of phosphate and the removal efficiency of phosphate was greater than 80% for up to 100 bed volumes.

Synthesis and Characterization of Red Organic Fluorescent of Perylene Bisimide Derivatives (Perylene Bisimide 유도체의 적색 유기 형광체 합성 및 특성 연구)

  • The white light of a hybrid LED is obtained by using red and green organic fluorescent layers made of polymethylmethacrylate (PMMA) films, which function as color down-conversion layers of blue light-emitting diodes. In this research, we studied the fluorescence properties of a red organic fluorophore, employing perylene bisimide derivatives applicable to hybrid LEDs. The solubility, thermal stability, and luminous efficiency are important characteristics of organic fluorophores for use in hybrid LEDs. The perylene fluorescent compounds (1A and 1B) were prepared by the reaction of 4-bromophenol and 4-iodophenol with N,N'-bis(4-bromo-2,6-diisopropylphenyl)-1, 6,7,12-tetrachloroperylene-3,4,9,10-tetracarboxyl diimide (1) in the presence of dimethyl formaldehyde (DMF) at $70^{\circ}C$. The synthesized derivatives were characterized by using $^1H-NMR$, FT-IR, UV/Vis absorption and PL spectra, and TGA analysis. Compounds 1A and 1B showed absorption and emission at 570 nm and 604 nm in the UV/Vis spectrum. We also documented favorable solubility and thermal stability characteristics of the perylene fluorophores in our work. Perylene fluorophore 1, with the 4-bromophenol substituent 1A, exhibited particularly good thermal stability and solubility in organic solvents.

Functionality Changes of Rapeseed Protein upon Proteolysis (유채단백질의 Proteolysis에 의한 기능성 변화)

  • purified rapeseed(Brassica napus var. Youngsan) protein was hydrolyzed by pronase. The hyrolysate protein was investigated for the some physicochemical and functional properties. UV and intrinsic fluorescence spectra of the hydrolysate showed the maximum absorption at 274nm and 360nm respectively. Intensity of yellow color decreased in the process of hydrolysis and the surface hydrophobicity decreased up to fourfold. The main bands of hydrolysate by sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE) were observed at 14,000 to 12,000 dalton molecular weight. Solubilities of hydrolyzed protein increased by 10~15% compared to those of unhydrolyzed protein at acidic pH. In the hydrolysate, while absorption of both water and oil, foam expansion and emulsion stability were increased, absolute viscosity, heat coagulation, calcium coagulation, foam stability and emulsion activity were decreased.

Characterization and bacterial anti-adherent effect on modified PMMA denture acrylic resin containing platinum nanoparticles

  • PURPOSE. This study characterized the synthesis of a modified PMMA (Polymethyl methacrylate) denture acrylic loading platinum nanoparticles (PtN) and assessed its bacterial inhibitory efficacy to produce novel antimicrobial denture base material. MATERIALS AND METHODS. Polymerized PMMA denture acrylic disc ($20mm{\times}2mm$) specimens containing 0 (control), 10, 50, 100 and 200 mg/L of PtN were fabricated respectively. The obtained platinum-PMMA nanocomposite (PtNC) was characterized by TEM (transmission electron microscopy), SEM/EDX (scanning electron microscope/energy dispersive X-ray spectroscopy), thermogravimetric and atomic absorption spectrophotometer analysis. In antimicrobial assay, specimens were placed on the cell culture plate, and $100{\mu}L$ of microbial suspensions of S. mutans (Streptococcus mutans) and S. sobrinus (Streptococcus sobrinus) were inoculated then incubated at $37^{\circ}C$ for 24 hours. The bacterial attachment was tested by FACS (fluorescence-activated cell sorting) analysis after staining with fluorescent probe. RESULTS. PtN were successfully loaded and uniformly immobilized into PMMA denture acrylic with a proper thermal stability and similar surface morphology as compared to control. PtNC expressed significant bacterial anti-adherent effect rather than bactericidal effect above 50 mg/L PtN loaded when compared to pristine PMMA (P=.01) with no or extremely small amounts of Pt ion eluted. CONCLUSION. This is the first report on the synthesis and its antibacterial activity of Pt-PMMA nanocomposite. PMMA denture acrylic loading PtN could be a possible intrinsic antimicrobial denture material with proper mechanical characteristics, meeting those specified for denture bases. For clinical application, future studies including biocompatibility, color stability and warranting the long-term effect were still required.
