Use of Antibody Displayed Phage for the Detection of Dextran Using a Dipstick Assay and Transmission Electron Micrograph

  • An antibody displayed phage collection (SBAE-2R), screened from a human synthetic phage antibody library (Fab 21ox), was used for the determination of dextran. The dextran-binding affinity was determined by serologically specific transmission electron microscopy (TEM) and a paper dipstick assay. The phage collection was distributed over the dextrancoated grids with 39$\pm$25 phages/$\mu$m$^2$ on the grids. Phages were not seen on dextran-coated grids exposed to the Fab 2lox phage library. The phage collection (SBAE-2R) produced 54$\pm$3 color normalized intensity (N.I.) from 125 ppm to 1,000 ppm of dextran and 5$\pm$1 (N.I.) for 63 ppm of dextran in a paper dipstick assay. This research extends the analytical options for dextran analysis by antibody displayed phage with a minimum of equipment usage.

Effective Control of fine Particles Using an Electrostatic Coagulation Between Particle and Water Droplet (입자와 액적간의 정전기적 응집을 통한 미세입자의 효율적인 제어)

  • A charged droplet scrubber was introduced to remove visible smokes generated in many industrial facilities. Lab-scale and field tests were conducted in this study. The system consists of a corona discharger to effectively charge the fine particles, fellowed by an electrostatic chamber to promote coagulation between charged fine particles and oppositely charged droplets and a demister to remove resultant particles. Overall collection efficiency, 98.4% was obtained from a lab-scale test, when a high voltage was applied to an ionizer and a charged droplet scrubber. Field tests also show the high collection efficiencies, 93.5% with one stage and 99.4% with two stage system. This system can be used to increase the collection efficiency of the conventional air pollution control devices to satisfy the national emission standard.

Characteristics of Current Collection Signals during Test Run of High-speed Train (주행 중 발생하는 고속전철 집전계 신호의 특성)

  • The dynamic characteristics of the current collection process of the high-speed railway are investigated through signals acquired during a test run. The signals are obtained from accelerometers, load cells, and strain gauges attached to various positions of the pantograph, and they are processed in time-and frequency-domains to obtain the dynamic characteristics. The main natural frequency of the pantograph is found to be 8.5Hz. There also are components at low frequencies varying linearly with the train speed. The contact frequency components above 20Hz is attenuated as they pass through the secondary suspension. The main frequency component of the load cell signal is found to be related with the rolling motion of the panhead generated by the stagger in the catenary.

  • This study examines the Jukchoui and Jukchosang from the late 19th century archived at Wonkwang University Museum focusing on the order and techniques of sewing used to make them to arrive at a better understanding of the sewing methods in late Chosen period and of 'Po'(Robe), one of the most symbolic outfits in Eastern Asia. Since there is no document on the items in Wonkwang collection, I reproduced and studies them based on the 19th-century sample. The sewing methods used for these items included fine broad-stitching, saddle-stitching, blind-stitching, and hemming, and there was no lining for the robe. The items in Wonkwang collection seem to reflect the trend of the late Chosun period when various systems including that of clothing were simplified: Jukchoui and Jukchosang in the period had same design for all levels, but colors and lengths were varied to mark the status of the wearer.

  • There have been concerns about possible pathogenicity and antimicrobial resistance in Enterococcus, which constitute more than 50% of probiotics in the worldwide market. In this study, Enterococcus in sixteen products manufactured by ten different companies was tested for the presence of six virulence genes and two vancomycin resistance genes. Results in this study showed the safety of Enterococcus on the Korean market and the importance of screening vanA, vanE, agg, cylA, esp, and gelE. Pulse-field gel electrophoresis showed that the sixteen isolates tested in this study are originated from three strains.

  • A check list which includes the items that are to be considered in the process of the statistical data collection and analysis by non-scientific organizations is proposed. Based on the suggested check list, the output resulting from the statistical survey conducted by private organizations, banks, organs of expression and enterprises in Gwangju and Chonnam are examined about the validity of data collection and statistical analysis.

A Study on the Elasticity Disuniformity for Catenary using by Beam Model (빔 모델을 이용한 전차선 불균일율에 관한 연구)

  • A catenary system should be designed to be an uniform elasticity over a span in order to maintain the lowest possible loss of contact between a pantograph and a contact wire. A elasticity disuniformity of a catenary can be regarded as a important design factor used for predicting the current collection performance for a catenary. There are a couple of formulas to calculate elasticity disuniformity of a catenary according to the literature survey, The effectiveness of these formulas is reviewed by performing catenary elasticity and loss of contact analysis for 5 different configurations of catenary systems using a beam element based FEM program, KRRI developed program, and the loss of contact by GASENDO, RTRI developed program, respective]y. The results reveals that these formulas are not suitable to predict the current collection performance for a catenary. Therefore, a new formula based on the standard deviation of the elasticity over a span is proposed in this study. The analysis results show that the new formula for an elasticity disuniformity of a catenary is very effective in predicting the current collection performance for a catenary.

Factors Affecting the Motility and Fertility of Frozen-thawed Stallion Semen (말 동결융해 정자의 생존성 및 수정능에 영향을 미치는 인자)

  • The techniques for the collection, cooling and freezing of semen and artificial insemination of horses are not fully understood in Korea. We investigated the percentages of total motile (TM) and progressively motile (PM) sperms after the collection, cooling and freezing of stallion semen. The average volume of semen was 167 ml in Thoroughbred and 68 ml in Arab. The average numbers of spermatozoa in Thoroughbred and Arab were $104\times10^6/ml$ and $86\times10^6/ml$ respectively. The average percentages of TM and PM were 82.3% and 88.6% in Thoroughbred, and 61.4% and 82.6% in Arab, respectively. The average percentage of TM at 4 hr after cooling at $5^{\circ}C$ was significantly lower than that at 0 hr ($30.0\pm4.1%\;vs.\;78.0\pm2.5%,\;p<0.05$), but the percentage of PM was similar between 66.5 and 73.2% at 0, 1, and 4hr. The average percentage of frozen-thawed Thoroughbred semen frozen in MFR5 extender was 56.2%, which was significantly higher than that of the semen frozen in LE extender (average 32.9%, p<0.05). The percentage of TM in Arab was similar for semen frozen in MFR5 extender and LE extender (18.2% and 21.2%, respectively), but the percentage of PM was significantly higher in sperm frozen in MFR5 extender than in sperm frozen in LE extender (69.0% vs. 36.4%, p<0.05). Four mares were artificially inseminated by Thoroughbred frozen-thawed semen and one of them fertilized at 11 day after artificial insemination. In this study, the collection, cooling and freezing of equine semen were possible under domestic conditions.

  • Collection Efficiency of a tritium bubbler being used at domestic nuclear power plants was studied. Equilibrium is attained while tritium vapour in air passes through the bubbler. When a lot of gas sample passes through the bubbler, the tritium concentration of water is raised and volume of water is decreased or increased. These phenomena give effect to the collection efficiency. Estimation of the tritium concentration of the water agreed with our experiments. If relative humidity is higher than 0.5, the efficiency becomes lower than 1 and if relative humidity is lower than 0.5, the efficiency becomes higher than 1, provided collected tritium is evaluated with original volume of the water. Though the relative humidity is riot 0.5, the efficiency can be maintained near 1 by making $\frac{saturated\; water\; vapour(cm^3_w)\times volume \; of \; gas \; sample(cm^3)}{volume\; of\; water(cm^3)}$ small

Factors Affecting Superovulatory Responses in Dairy Donor Cows (공란유우의 과배란반응에 영향하는 요인에 관한 연구)

  • This study was carried out in order to find out better ways for superovuiation and egg collection by checking some factors affecting on donor cows such as iactating and dried, age, season of treatment and sequence of treatment. The results were summarized as follows:1. Number of corpus luteum and collected eggs of eactating and dried doner were 9.8 vs 8.0 and 9.6 vs 7.9,respectively. However, the rate of transferable embryos of lactating doner were higher than that of dried, 82.5% vs 48.1%. 2. The average number of corpus luteum and collected eggs of lactating donors under 7 years of age (6.7 vs lactating 5.3) were slightly lower than those of over 8 years of age (11.1 vs 9.2). But the rate of transferable embryoswas better in under 7 years old donors than over 8 years (81.1% vs 49.3%). were 6.0, 4.8, 1.5, 1.8 and 75%, and those in the summer were 2.9, 3.8, 2.2 and 46.7%, respectively. 3. The average number of corpus luteum was the highest in winter (10.5) and the rate of egg collection was the best in autumn (94.7%). The rate of the transferable embryo was the highest in winter (64.3%). 4. The average number of corpus luteum was the lowest (5.5) in the animals treated six or more superovulating treatments. The rate of egg collection was the best in the third treatment group (90.2%), but it was getting worse after fifth treatment, The rate of transferable embryos was the highest in the second treatment group (94.1%), and it was decreased thereafter.
