해양생물 유래 피부건강 기능성의 소재를 발굴할 목적으로 굴 단백질을 Protamex와 Neutrase로 가수분해 한 후 생성된 굴 가수분해물 펩타이드의 주름 개선 효과를 조사하였다. 굴 가수분해물은 약 7.9%의 유리아미노산을 포함하며 urea, taurine, alanine, glycine 등의 아미노산의 함량이 높았다. 굴 가수분해물은 collagenase 저해 활성을 가지고 있었고, 피부세포인 CCD986sk에 대한 독성은 없었다. 분획물의 수율은 1,000 Da 미만이 40%였으며, 5,000 Da 미만이 60.4%를 차지하였다. 1,000~3,000 Da인 분획물이 항산화 활성, procollagen의 생성능 및 MMP-1 효소 저해 활성이 가장 높았다. 굴 가수분해물과 5,000 Da 미만의 분획물은 주름 개선을 위한 피부건강 기능성 소재로서 화장품 등의 제품에 응용 가능함을 확인하였다.
Hoichunyonggyuksan(HCYGS) and Yongseksan(YSS) are important prescriptions that have been used in oriental medicine for stomatitis and wound healing. The study was done to evaluate the inhibitory effects of cytotoxicity, formation of superoxide on the macrophage and neutrophil, prostaglandins($PGE_2$), interleukins($IL-1{\Beta}$), collagenase activity and synthesis of collagen and DNA. The results were obtained as fo1lows: 1. HCYGS and YSS were not showed the proliferation difference of human fibroblast and monocyte in all concentrations to be experimented and in result, it was concluded that they have no cytotoxicity. 2. YSS inhibited the formation of superoxide to $48\% at the concentration of $0.001\%$ in the mouse monocyte. 3. HCYGS inhibited the formation of superoxide to $13\%$ at the concentration of $0.001\%$ as compared with control in the human monocyte. 4. HCYGS and YSS inhibited the formation of superoxide to $25\%\;and\;35\%$ respectively at the concentration of $0.0001\%\;and\;HCYGS\;showed\;38\%$ inhihitory rate on the formation of superoxide at concentration of $0.001\%$ in the human neutrophil. 5. The concentration of inhibiting the production of prostaglandins($PGE_2$) to $11\%$ in the human monocyte stimulated with E. coli were $0.01\%$ of HCYGS. 6. The concentration of inhibiting the production of interleukins($IL-l{\Beta}$) to $43\%\;and\;39\%$ in the human monocyte stimulated with E. coli were $0.01\%$ of HCYGS and YSS. 7. HCYGS and YSS didn't influence on collagen synthesis and total protein in fibroblasts. 8. HCYGS and YSS inhibited the collagenase activity to $22\%\;and\;19\%\;at\;0.1\%,\;45\%\;and\;56\%\;at\;0.2\%,\;57\%\;and\;52\%$ at $0.5\%$ respectively, but YSS exerted the inhibitory effect on collagenase activity to $27\%$ at the lower concentration of $0.01\%$. 9. HCYGS and YSS didn't show the faster recovary than control group hut exerted the consistant effect on the anti-inflammations and the formation of granulation tissue.
Objective : This study was performed to assess the effects of Aloe and Violae herba extracts on skin disease and skin beauty. Methods : Anti-oxidant effects were measured by the scavenging for DPPH radical, xanthine oxidase activity. Anti-inflammatory effects were examined by relations in NO synthesis, IL-$1{\beta}$, IL-6, TNF-$\alpha$, NF-kB, COX-2, MAP kinase. The skin wrinkle formation effects were measured by collagenase and elastase activities. The whitening effects were examined by tyrosinase activities, melanin synthesis in MNT-1 cell. Results : 1. In an anti-oxidant test, Aloe and Violae herba extracts showed high radical scavenging activity. 2. In an anti-inflammatory test, Aloe and Violae herba extracts strongly inhibited the lipopolysaccharide(LPS)-induced nitric oxide(NO) release from the RAW 246.7 macrophage cells. Aloe and Violae herba extracts also inhibited the LPS-induced IL-$1{\beta}$ and COX-2 expressions. The inhibitory effects of Aloe and Violae herba extracts on macrophage activation were via the inhibition of NF-kB, evidenced by transient transfection assay. Furthermore, Aloe and Violae herba extracts weakly inhibited the activation of Jun-N-terminal kinase(JNK) but they did not have any effects on p38 MAP kinase in RAW 264.7 cells. 3. In the skin wrinkle formation assay, Aloe extract strongly inhibited collagenase and elastase, whose activity are tightly related with the wrinkle formation. 4. In the skin whitening assay, Aloe and Viloae herba extracts weakly inhibited tyrosinase activity, however, it was not statistically significant. Besides they did not have any effects on melanin synthesis, indicating that they could not be applicable for skin whitening. Conclusion : This study show that Aloe and Violae herba extracts may play a significant role in skin disease and skin beauty.
Marine resources have various biological activities and their constituents are more novel than those of land organisms. Several biologically active constituents have been found in marine organisms. Recently, many studies have reported that marine animals (MAs) can be used as functional ingredients in functional foods or nutraceutical due to their health benefits. However, no studies have extensively investigated the cosmeceutical activities of MAs extracts. Here, 70% ethanol extracts of 38 MAs were investigated for their activities of whitening and anti-aging properties for use as materials in novel cosmeceuticals. Anti-aging activities were determined by skin aging-related enzyme activities (anti-collagenase, anti-elastase, anti-hyaluronidase) and whitening activities (anti-tyrosinase, anti-3,4-dihydroxyl-L-phenylalanine [DOPA] oxidation) evaluated by colorimetric method. Among the 38 MAs, we found that Urechis unicinctus and Petrosia corticata extracts showed the strongest inhibitory effects against tyrosinase and DOPA oxidation, respectively. Our results additionally showed that Protankyra bidentata extract might provide a major source of anti-hyaluronidase and anti-elastase; meanwhile, anti-collagenase effects were similar in most MAs. Overall, these results suggest that extracts of marine animals have potential as a tyrosinase, collagenase, elastase, and hyaluronidase inhibitors. Taken together, MA resources could be considered as a novel cosmeceutical agent to be applied in cosmetic industry.
Objectives : This study was designed to investigate the collagen metabolism and tyrosinase activity of Polygonati Rhizoma extracts (PR). It's effects are to tonify spleen qi and augment the spleen yin. It enrichs the yin and moisten the lung. Methods : The effect of PR on type I procollagen production and collagenase (matrix metalloproteinase-1, henceforth referred as MMP-1) activity in human normal fibroblasts Hs68 after ultraviolet B (UVB, 312 nm) irradiation was measured by ELISA method. The tyrosinase activity after treatment of PR was measured. Results : There were no cytotoxicity at concentrations of 10, 30, $100{\mu}g/ml$. The reduced type I procollagen production was recovered by PR in UVB damaged Hs68 cells at a concentration of $100{\mu}g/ml$ ($16.2{\pm}0.0$ ng/ml) from control group ($13.9{\pm}0.5$ ng/ml). However there was no statistical significance. PR reduced The increased MMP-1 activity after UVB damage at concentrations of $10{\mu}g/ml$, $30{\mu}g/ml$, and $100{\mu}g/ml$ in a dose dependent manner ($42.2{\pm}20.5%$, $44.8{\pm}8.5%$, and $22.0{\pm}5.8%$). PR $100{\mu}g/ml$ treatment showed the statistical significace (p < 0.05). PR significantly reduced the tyrosinase activity at a concentration of 10 mg/ml ($32.0{\pm}12.8%$, p < 0.05). However, the L-DOPA oxidation was not changed. Conclusion : PR showed the anti-wrinkle effects and whitening effects in vitro. Although more researches are needed to validate the efficacy, these results suggest that PR may have potential as an anti-aging ingredient in cosmetic herb markets.
Objective : Herbal medicinal mixture (JMB) are consisted of Caryophylli Flos, Aucklandiae Radix, and Angelicae Dahuricae Radix. Each of these herbal medicines has studied on anti-aging effect in vitro. So this study was conducted to investigate efficacy and potency of JMB extract on dermal anti-aging and whitening. Methods : The JMB was extracted at room temperature by 80% ethanol. Collagenase and elastase inhibition activity in JMB ethanol extract were determined at 10, 50, 100, 500, 1000 mg/ml concentrations by colorimetric method. The toxic range of JMB ethanol extract was evaluated using MTT assay. Also, The inhibitory effect of JMB ethanol extract on tyrosinase activity and melanin contents in mouse melanoma cell line (B16F10 cell) was identified at 50, 100, 200 ㎍/㎖ levels by spectrometric assay. In each analysis, EGCG (epigallocatechin gallate) and Kojic acid were used as positive controls, respectively. Results : The elastase and collagenase inhibitory activity of JMB ethanol extract increased dose dependently. Also, The MTT assay showed that JMB, up to 400 ㎍/㎖ concentration, exhibited no toxic effect to the B16F10 cell. And following the JMB ethanol extract treat, cellular melanin contents and tyrosinase activity were dose-dependently decreased compared to those of control. Conclusion : These results suggest that JMB ethanol extract has effects to inhibitory activity on dermal wrinkle enzyme and melanogenesis. Therefore, JMB has applicable benefits for development of materials or products to have whitening and anti-aging functions on skin.
NUC1 (Nutraceutical compound 1) is an ethanol extract composed of a formulation based on medicinal herbs traditionally used for the treatment of arthritis in Korea and China. This study investigated the therapeutic effects of NUC1 on osteoarthritis (OA). The protective effect of NUC1 on OA was tested in a rabbit model of collagenase-induced arthritis (CIA) for 4 weeks. Results were compared among four groups (n = 9 per group): the normal group (untreated), the CIA group (vehicle control), the NUC1 group (CIA rabbits treated with 200 mg/kg NUC1), and the JOINS group (positive control, CIA rabbits treated with 200 mg/kg JOINS tablet). NUC1 significantly inhibited NO production (p < 0.05 at 125 ㎍/ml, p < 0.01 at 250 ㎍/ml, and p < 0.001 at 500 ㎍/ml) and iNOS expression in macrophages, in a concentration-dependent manner. NUC1 also inhibited the release and protein expression of MMP-1, 3, and 13, in TNF-α-induced chondrosarcoma cells in a concentration-dependent manner. In vivo, the MMP-1 and MMP-3 levels in synovial fluids were significantly (p < 0.05) lower in NUC1 group (77.50 ± 20.56 and 22.50 ± 7.39 pg/ml, respectively) than in the CIA group (148.33 ± 68.58 and 77.50 ± 20.46 pg/ml, respectively). Also, in histopathological, NUC1 ameliorated articular cartilage damage in OA by increasing the abundance of chondrocytes and proteoglycan in the articular cartilage. Thus, NUC1 showed promise as a potential therapeutic agent, and it can be generalized to a broader study population in different OA animal models.
그린볼 사과 적과의 phenolic compound를 50-200 ㎍/mL 농도로 처리하여 DPPH radical 소거능을 측정한 결과 200 ㎍/mL 농도의 물과 ethanol 추출물은 94.69과 92.24%의 DPPH radical 소거능을 나타내었다. ABTS radical 소거능은 200 ㎍/mL 농도의 물과 ethanol 추출물에서 각각 100.30과 99.16%의 ABTS radical 소거능을 나타내었다. PF는 200 ㎍/mL 농도의 물과 ethanol 추출물에서 각각 1.76, 1.76 PF를 나타내었다. TBARs는 phenolic compound 농도가 200 ㎍/mL에서 물과 ethanol 추출물 각각 101.46, 99.64%의 항산화능을 나타내었다. 염증에 관련이 있는 HAase의 저해 효과를 측정한 결과 150 ㎍/mL 농도의 물과 ethanol 추출물은 각각 33.28, 32.14%의 저해효과를 나타내었다. 미용식품활성을 나타내는 elastase 저해활성을 측정해 본 결과 phenolic compound 농도가 200 ㎍/mL에서 물과 ethanol 추출물 각각 47.33, 40.92%의 저해 효과를 나타내었다. 피부의 주름과 탄력에 관련이 있는 collagenase 저해 효과를 측정한 결과 200 ㎍/mL 농도의 물과 ethanol 추출물은 각각 46.19, 65.58%를 나타내었다. 따라서 그린볼 사과 적과는 항산화, 항염증, 주름 개선 등 다양한 생리활성을 나타내는 기능성 소재로 개발이 가능할 것으로 판단되었다.
본 연구는 노령목의 산림의 경제적 가치를 증진하고자 밤나무 잎에 함유된 물질의 기능성화장품 원료로의 기능을 규명하여 화장품 산업에 적용하기 위하여 천연소재로써의 이용 가능성을 살펴보았다. 항산화능 측정을 위한 실험으로 DPPH radical 소거능과 ABTS cation radical 소거능을 실시하였으며, 양성 대조군으로는 항산화 작용이 있는 것으로 알려진 BHA를 이용하여 밤잎 추출물의 항산화 효과를 비교하였다. 그 결과 아세톤 추출물의 경우 시료 농도에 따라 DPPH radical 및 ABTS radical 소거 증가 정도가 컸으며, 열수 추출물은 10 ug/ml의 농도에서 50% 이상의 소거활성을 나타내었다. 이러한 결과로 미루어 볼 때 밤잎 추출물은 항산화 작용이 높고 피부에서 흔히 일어나는 활성산소에 대한 노화 방지에 도움이 될 것으로 사료된다. 주름개선 효과 측정으로 elastase 저해활성을 측정한 결과 아세톤과 열수 추출물에서는 그 효과가 없었으나 에탄올 추출물 100 ug/ml에서 64.6%의 우수한 elastase 저해활성을 나타내었으며, collagenase 저해활성을 측정한 결과 3가지 추출물은 100 ug/ml에서 열수 추출물 18.0%, 아세톤 추출물 14.3%, 에탄올 추출물 5.3%의 순으로 collagenase 저해활성능을 나타내었으며, 다른 천연물질을 이용한 연구결과와 비교 시 밤잎 추출물은 주름개선에 우수함을 확인 할 수 있었다. 주름개선 원료로의 이용가치가 있을 것으로 보인다. 이상의 연구 결과로 보아 밤잎은 항산화 및 항노화 효소 활성을 갖는 천연소재로서 주름개선 소재로 활용 될 수 있을 것으로 사료되며, 임삼 효능 평가 등의 계속적인 연구를 통해 기능성 화장품 소재로 개발된 가능성이 매우 높다고 사료된다.
본 연구에서는 7가지 한방원료(음양곽, 복분자, 오미자, 구기자 , 건지황 l 사상자 , 토사자)의 항산화 효과, collagen 합성 촉진 효과, collagenase 활성 저해 효과를 비교 평가함으로써 주름개선 기능성 화장품 원료로서의 사용 가능성을 검토하였다. 항산화 실험인 DPPH 소거 효과, 리놀산 자동산화 억제 효과, superoxide radical 소거 효과, hydroxyl radical 소거 효과에서는 음양곽, 복분자, 오미자, 구기자가 대체적으로 뛰어난 활성을 보여 항노화 화장품 소재로서의 가능성을 보여주었으며, 주름개선 실험인 collagen 합성 촉진 효과, collagenase 저해 효과에서는 건지황, 사상자, 토사자가 다른 네 가지 물질에 비해 뛰어난 효과를 나타내어 주름개선 화장품 소재로서의 가능성을 보여주었다. 이러한 결과는 7가지 한방원료가 항산화 및 주름개선 효능을 나타내므로 기능성 화장품 소재로 활용될 수 있음을 보여 주었다.
