Isolation and Characterization of an immunomodulatory Protein from Bovine Colostrum

  • A colostral protein that augments natural killer (NK) cell activity was isolated from bovine colostrum. This protein, designated matemal immunity enhancing factor (MIEF), increased NK cell-mediated cytotxicity against human tumor targets wheb added to cultures of resting peripheral blood lymphocytes. The NK cell stimulatory activity of the MIEF was demonstrated at the concentrations as low as 0.1-0.01 $\mu$g/ml. Purified MIEF showed an apparent molecular weight of 22,000 in SDS-polyacrylamide gel electrophoresis. The unusual biochemical characteristics of the MIEF distinguish it from other cytokines. The MIEF was soluble at a cold tgemperature, and precipitated by raising the temperature. This themal precipitability was reversible, and dependent on the concentration, pH and ionic strength. Maximal precipitation was observed at neutral pH, and higher ionic strength.

Lack of Sunlight Exposure Influence on Primary Glioblastoma Survival

  • Background: The prognosis of primary glioblastoma (GBM) is poor. Approximately 2/3 of primary brain tumor diagnoses are GBM, of which 95% are primary lesions. In this study, we aimed to evaluate whether more sunlight exposure has an effect on survival of patients with primary GBM. Materials and Methods: A total of 111 patients with primary GBM were enrolled from Kayseri in inner Anatolia which has a cold climate (n: 40) and Mersin in Mediterranean region with a warm climate and more sunlight exposure (n: 71). The patients with primary GBM were divided into two groups as Kayseri and Mersin and compared for progression free survival (PFS) and overall survival (OS).Results: The PFS values were 7.0 and 4.7 months for Kayseri and Mersin groups, respectively (p=0.10) and the repsective OS values were 13.3 and 9.4 months (p=0.13). We did not found any significant difference regarding age, sex, comorbidity, smoking, surgery, resurgery, adjuvant chemoradiotherapy and palliative chemotherapy between the groups. Conclusions: We found that more sunlight exposure had no impact on prognosis of patients with primary GBM, adding inconsistency to the literature about the relationship between sunlight and GBM.

  • This report describes the studies as follows through the research into the data of Fructus Forsythiae main blended prescriptions from Dongeuybogam. Through investigation on prescriptions from Dongeuybogam with Fructus Forsythiae as an ingredient, the following conclusions were reached. 1. All kinds of infection in a boil and the malignant tumor recorded the largest number of clinical frequency of the prescriptions in therapeutic use when Fructus Forsythiae was taken as a principle medicine. 2. The prescriptions are compounded with Fructus Forsythiae as a principle medicine can apply to eye disease, nose and ear disease, throat disease, trauma, the malignant tumor, skin disease, dental disease and heat pathogenic disease. 3. Various pathogenic factors such as wind-heat, heat, fire, wind, qi, poison, cold are put to practical use. 4. The dosage of Fructus Forsythiae is 2bun(about 0.75 gram) to 3jeon(about 11.25 gram), however 1joen(about 3.75 gram) has been taken the most for clinical application. 5. With examination the formulae of presciptions containing Fructus Forsythiae, I found the basic prescriptions as well as the medicines are in conjunction with Fructus Forsythiae and the theoretic grounds of formula in combining Fructus Forsythiae as a group of medicine for prescription through this research.

『황제내경(黃帝內經)』의 비(痺)와 적(積)의 관계에 대한 고찰(考察) (A Study on the Relation of Tingling Disease and Accumulation in 『Huangdineijing』)

  • Objectives : In this study, the relationship between Tingling disease(痺證) and Accumulation (積) was examined focusing on the occurrence of Tingling disease and its development into tangible lesion. Methods : Based on related contents in the "Huangdineijing", the process of creation and development of Tingling and its transformation to Accumulation was mainly examined. Results : While Tingling disease is usually caused by the three Qis of Wind-Cold-Dampness, due to its Yin nature there is high tendency of Qi and blood to be blocked and Blood and Fluid-Humor agglomerating into Accumulation. Symptoms of dysaesthesia are merely expressions manifesting in this process. Development into colic accumulation [疝瘕], Gu(蠱), or convulsion[瘛] after Tingling has traveled to the five zang is also related to its transformation into Accumulation. In the case of Tingling disease of the five zang in the "Huangdineijing", it is not a diagnostic category for treatment but actual lesions in the five zang six fu. In other words, in the beginning stages of Tingling disease, some sort of solidification that causes abnormal senses among other symptoms happens, and this solidification starts taking up space within the flesh. If it is not eliminated and persists, it enters into the inner organs and develops Tingling disease of the zangfu. Conclusions : Understanding Tingling disease(痺證) as a presymptom to developing Accumulation, rather than abnormality of sense will enable people to have higher chances in treating tumor.

지방육종형성 동물모델에서 $^{123}I$-15-(p-iodophenyl)-3-R, S-methylpentadecanoic acid (BMIPP)의 생체분포와 생체영상 (In Vivo Image and Biodistribution of $^{123}I$-15-(p-iodophenyl)-3-R, S-methylpentadecanoic acid (BMIPP) in Liposarcoma Bearing Nude Mice)

  • 목적: $^{123}I$ 표지된 지방산 유도체는 심근에너지대사를 평가하는데 사용되어 왔다. 본 연구는 $^{123}I$-BMIPP를 지방육종 진단물질로서의 유용성을 평가하기 위하여, 지방육종 세포주와 신경교종 세포주간의 시험관내의 섭취율을 비교하고 지방육종 형성 nude mice 동물모델에서의 생체분포와 생체영상을 확인하였다. 대상 및 방법: Cold-BMIPP에 $^{123}I$의 표지는 $Cu^{2+}$를 촉매로 이용하여 반응 후 Sep-pak으로 분리정제하여 TLC를 이용하여 방사화학적 순도를 확인하였다. 시험관내 섭취율 시험은 $^{123}I$-BMIPP를 지방육종 세포주인 SW872와 신경교종 세포주인 9L에 각각 시간대별로 5분, 15분, 30분, 60분, 120분, 180분씩 처리한 후 섭취율을 비교하였다. 정상 백서에서의 생체분포를 평가하기 위하여 18시간 금식시킨 군(n=6/군)과 식이를 한 군으로하여 $^{123}I$-BMIPP를 꼬리정맥에 주사하고 30분, 2시간, 24시간후의 각 장기별 방사능을 감마카운터로 계측하였다. 지방육종 세포주인 SW872를 왼쪽 대퇴부에 피하접종한후 종양이 형성된 누드마우스(n=5/군)를 18시간 금식시키고 $^{123}I$-BMIPP를 주사한 후 30분, 2시간, 24시간후의 생체분포와 2시간, 24시간의 생체영상을 평가하였다. 결과: $^{123}I$-BMIPP의 표지수율은 95 % 이상이었으며, 방사화학적 순도는 99 % 이상이었다. 지방육종 세포주인 SW872가 신경교종 세포주인 9L보다 180분에서 1.5배 정도의 섭취율의 증가를 나타내었다. 정상 백서에서의 생체분포를 확인한 결과, 24시간에서 금식을 한 경우가 식이를 한 경우보다 방사성표지 지방산의 제거율이 지연됨을 알 수가 있었으며 주요 배출장기는 위장관으로 확인되었다. 지방육종이 형성된 누드마우스에서 각 시간대별로 Tumor/Blood 비율은 0.94, 0.75, 1.38이었고, Tumor/Muscle의 비율은 0.66, 1.53, 1.11을 나타내었다. 24시간의 감마카메라영상에서 지방육종으로의 국소적인 집적이 확인되었다. 결론: 이상의 결과를 종합해볼 때 지방육종에서 $^{123}I$-BMIPP를 지방육종 영상물질로서의 사용 가능성을 확인할 수 있었다.

파래 추출물의 항응고 활성과 항암 활성에 관한 연구 (The Anticoagulant and Anticancer Activities of Enteromorpha intestinalis Extracts)

  • The study was performed to investigate the biological activity of Enteromorpha intestinalis. In order to examine its blood anti-coagulant effects, Enteromorpha intestinalis was extracted with cold water, methanol, hot water, HCl and NaOH. In general, the alkali extract of Enteromorpha intestinalis was approximately 17 times stronger than the control. The anti-cancer effects of select extracts(methanol, hot water, 0.1 N NaOH, 1 N NaOH) were determined in human melanoma cells(Bl6/F10), fibrosarcoma cells(HTl080) and breast cancer cells(MCF7) by MTT assay. With the treatment of 250 ${\mu}g/m{\ell}$ of methanol extracts. HT1080, B16/F10 and MCF7 cell viabilities significantly decreased to 8.06%, 3.62% and 10.10%, respectively. Thus these results strongly support the possibie use of Enteromorpha intestinalis as a functional materials.

모유수유 중 인삼의 적응증과 용량에 관한 기존한약서 고찰 (Textual Research : Indication and Dosage of Panax Ginseng during Lactation)

  • Purpose: To provide basic informations for guidelines of Ginseng usage during lactation. Method: Based on a regulation of scope and preparation of herbal prescriptions by Ministry of health and welfare of Korea, we selected the formulae for postpartum care and disease in 7 Korean Medical Classics. And we searched the number of formulae including Ginseng, dosage and indications of Ginseng in formulae in those books. Results: The range of Ginseng dosage during lactation from medical classics is 1.5-18.75g/day for medical purposes. Indications of Ginseng are lethargy, excessive bleeding, asthma, fever, cold, pain, dizziness, mental disorder, spasm, digestive problem, constipation, diarrhea, urinary incontinence, edema, breast engorgement, lack of breastmilk, breast ulcer, etc. But Ginseng had not been used in the case of stroke, epistaxis, beginning of mastitis, tumor of lower abdomen. Conclusions: Ginseng should be recommended less than 1.5g/d as a nutritional supplement. If breastfeeding woman wants to take Ginseng as a medication or as a tonic, she should consult with a official specialist about proper prescription and dosage instead of risky random medication.

Cytotoxicity, Apoptosis Induction and Anti-Metastatic Potential of Oroxylum indicum in Human Breast Cancer Cells

  • Despite clinical advances in anticancer therapy, there is still a need for novel anticancer metabolites, with higher efficacy and lesser side effects. Oroxylum indicum (L.) Vent. is a small tree of the Bignoniaceae family which is well known for its food and medicinal properties. In present study, the chemopreventive properties of O. indicum hot and cold non-polar extracts (petroleum ether and chloroform) were investigated with MDA-MB-231 (cancer cells) and WRL-68 (non-tumor cells) by XTT assay. All the extracts, and particularly the petroleum ether hot extract (PHO), exhibited significantly (P<0.05) higher cytotoxicity in MDA-MB-231 when compared to WRL-68 cells. PHO was then tested for apoptosis induction in estrogen receptor (ER)-negative (MDA-MB-231) and ER-positive (MCF-7) breast cancer cells by cellular DNA fragmentation ELISA, where it proved more efficient in the MDA-MB-231 cells. Further, when PHO was tested for anti-metastatic potential in a cell migration inhibition assay, it exhibited beneficial effects. Thus non-polar extracts of O. indicum (especially PHO) can effectively target ER-negative breast cancer cells to induce apoptosis, without harming normal cells by cancer-specific cytotoxicity. Hence, it could be considered as an extract with candidate precursors to possibly harness or alleviate ER-negative breast cancer progression even in advanced stages of malignancy.

A New Rat Model of Cisplatin-induced Neuropathic Pain

  • Background: Chemotherapy-induced peripheral neuropathy is a major side effect of anti-cancer drugs, and our knowledge of its mechanisms is lacking. Several models for chemotherapy-induced neuropathy have been introduced. However, the outcomes of these models differ significantly among laboratories. Our object was to create a model of chemotherapy-induced neuropathy in rats with cancer. Methods: Female Sprague-Dawley rats were used. Mammary rat metastasis tumor (MRMT-1) cells were implanted subcutaneously in rats. Chemotherapy-induced peripheral neuropathy was induced by injection of cisplatin once a day for four days. The responses to mechanical and thermal stimuli were examined using von Frey filaments, acetone, and radiant heat. Results: Cisplatin (2 mg/kg/day) produced mechanical allodynia, while it did not induce cold allodynia or thermal hyperalgesia. This dose of cisplatin could work successfully against cancer. Body weight loss was not observed in cisplatin-treated rats, nor were other abnormal behaviors noted in the same rats. Conclusions: Repeated injection of intraperitoneal cisplatin induced peripheral neuropathic pain in rats. Thus, this type of rat model has broad applicability in studies related to searching for the mechanism of cisplatin-induced mechanical allodynia and agents for the treatment of neuropathic pain.

Caffeic acid phenethyl ester protects against photothrombotic cortical ischemic injury in mice

  • In this study, we aimed to investigate the neuroprotective effects of caffeic acid phenethyl ester (CAPE), an active component of propolis purified from honeybee hives, on photothrombotic cortical ischemic injury in mice. Permanent focal ischemia was achieved in the medial frontal and somatosensory cortices of anesthetized male C57BL/6 mice by irradiation of the skull with cold light laser in combination with systemic administration of rose bengal. The animals were treated with CAPE (0.5-5 mg/kg, i.p.) twice 1 and 6 h after ischemic insult. CAPE significantly reduced the infarct size as well as the expression of tumor necrosis $factor-{\alpha}$, hypoxiainducible $factor-1{\alpha}$ monocyte chemoattractant protein-1, $interleukin-1{\alpha}$, and indoleamine 2,3-dioxygenase in the cerebral cortex ipsilateral to the photothrombosis. Moreover, it induced an increase in heme oxygenase-1 immunoreactivity and interleukin-10 expression. These results suggest that CAPE exerts a remarkable neuroprotective effect on ischemic brain injury via its anti-inflammatory properties, thereby providing a benefit to the therapy of cerebral infarction.
