Effects of herbal acupuncture(Atratylodes japonica, Coix lachrymajobi, Ephedra sinica, Atratylodes japonica mixed with Coix lachrymajobi and Ephedra sinica mixed with Green tea) at Pungnyung(ST40) and Umnungchon(SP9) in obese Rats induced by high fat diet (고지방식이(高脂肪食餌)로 유발(誘發)된 비만백서(肥滿白鼠)모델에서 창출(蒼朮), 의이인(薏苡仁), 마황(麻黃), 창출합의이인(蒼朮合薏苡仁) 마황합록차(麻黃合綠茶) 약침(藥鍼)이 혈청지질(血淸脂質) 및 간기능(肝機能)에 미치는 영향(影響))

  • Objective : This research was performed to investigate the effect of herbal acupuncture(Atratylodes japonica, Coix lachrymajobi, Ephedra sinica, Atratylodes japonica mixed with Coix lachrymajobi and Ephedra sinica mixed with Green tea) at Pungnyung(ST40) and Umnungchon(SP9) on weight gain, food intake, food efficiency, serum of lipid concentrations, liver function and HDL to total cholesterol ratio of rats fed high fat diet for 5weeks. Method : Experimental groups were divided into normal group(Normal), high fat diet group(Control), high fat diet and Atractylodes japonica-herbal acupuncture group(AJ), high fat diet and Coix lachrymajobi-herbal acupuncture group(CL), high fat diet and Ephedra sinica-herbaI acupuncture group(ES), high fat diet and Atractylodes japonica+Coix lachrymajobi-herbal acupuncture group(AJ+CL), Ephedra sinica+Green tea-herbal acupuncture group(ES+GT). Herbal acupuncture was bilaterally treated at the level of 132.5mg/kg body weight per 2day. Results : Body weight and food efficiency were decreased in AJ, ES, AJ+CL, ES+GT. The level of serum total cholesterol, triglyceride and free fatty acid were increased in AJ, ES, ES+GT. That of serum HDL-cholesterol was increased in AJ. The change of food intake, the level of serum phospholipid and ALP were not significant. The HDL to Total cholesterol ratio was increased in AJ and ES. Conclusion : Atractylodes japonica-herbal acupuncture in ST40 SP9 is effective on Body weight, food efficiency ratio, the level of serum lipid, protection of liver function and prevention cardiovascular risk by obesity induced by high fat diet. Herbal acupuncture mixed Ephedra sinica with Green tea can control the body weight, food efficiency ratio and the level of serum lipid.

The level of serum triglyceride, free fatty acid were increased in AJ. The level of serum AST was decreased in AJ, AJ+CL. The level of serum ALT was decreased in AJ, CL, AJ+CL. The level of serum ALP was decreased in AJ+CL. The HDL to Total cholesterol ratio was increased in AJ. Conclusions : Atractylodes japonica-herbal acupuncture in ST40 SP9 can control the body weight, feed intake and feed efficiency ratio, HDL-cholesterol and herbal acupuncture combined Atractylodes japonica with Coix lachrymajobi var. mayeun Stapf is effective on Body weight, feed intake, feed efficiency ratio, protection of liver function and prevention cardiovascular risk by obesity.

Optimal Extraction Condition and Characterization of Antidementia Acetylcholinesterase Inhibitor from Job's Tears (Coix lachrymajobi L.) (율무로부터 항치매성 Acetylcholinesterase 저해물질의 최적추출 조건 및 특성)

  • For the development of a new antidementia functional food or alternative drug using agricultural products, Job's tears (Coix lachrymajobi L.), which shows high acetylcholinesterase (AChE) inhibitory activity (55.1%) was selected and the extraction conditions of AChE inhibitor were optimized. AChE inhibitor of Job's tears was maximally extracted when it was treated with 60% methanol at $40^{\circ}C$ for 6 h. The AChE inhibitor of the methanol extracts was partially purified by systematic solvent extraction, thin layer chromatography, silica gel chromatography and reverse-phase HPLC and the partial purified AChE inhibitor with inhibitory activity ($IC_{50}$) of $0.608\;{\mu}g$ was obtained. The partial purified AChE inhibitor was soluble in methanol and hexane, and insoluble in water. Its maximum absorption spectra was 230 nm and also it was stable in the range of $30^{\circ}C$ and $70^{\circ}C$ and pH 4.0-8.0 for 1 h.

Identification of Cultivate Sites for Job's-tears (Coix lachrymajobi var. mayuen) using Capillary Electrophoresis (Capillary electrophoresis를 이용한 율무의 원산지 판별)

  • Optimal extraction, separation, and capillary rinsing conditions for capillary electrophoresis (CE) were established to identify the cultivation site (domestic vs. foreign) of Job's-tears (Coix lachrymajobi var. mayuen) using 240 samples (domestic sample n = 121, foreign sample n = 119). Job's-tears was extracted with 30% ethanol and separated on a $50-{\mu}m-I.D.$ untreated fused-silica capillary. Optimal analytic conditions were: temperature, $45^{\circ}C$; voltage, 15 kV; detector rise time, 0.1 sec; and pressure injection, 20 sec. Separation of peak investigated using 0.1 M phosphate buffer (pH 2.5) containing 0.05% hydroxypropylmethylcellulose (P buffer) revealed the optimal separation buffer was P buffer containing 26 mM hexane sulfonic acid with 30% methanol. Under the optimal conditions established for CE, the average correct identification percentage of domestic or foreign Job's-tears was 82%.

  • The objective of this study was to establish the stable cultivating method of adlay in paddy fields. The effect of soil moisture on the early of the transplanted adlay seeding was evaluated in 1996. the suitable transplanting period and the relationship between water management and leaf blight disease were tested in 1997. The critical transplanting date was June 30. rooting of transplanted adlay plants was retarded when soil moisture was saturated. However, sufficient soil moisture since one month after transplanting was required for the control of leaf blight disease and high yield of adlay.

  • In order to determine if the mRNA and protein expression levels of 3T3-L1 adipocytes are influenced by oriental medicines, adipocytes were treated with $100\;{\mu}g/ml$ of G-Re and aqueous extract of a Coix lachrymajobi var. mayuen (AEC) every other day for 12 days, respectively. The tumor necrosis factor alpha ($TNF-{\alpha}$). mRNA and protein expressions were suppressed markedly in treated mature adipocytes. Those of lipoprotein lipase (LPL) levels were found to increase gradually in preadipocytes differentiating into mature adipocytes. Those were higher than that of the untreated mature adipocytes. The treated adipocytes showed reduction of leptin expression levels, while in untreated mature adipocytes cell, those of levels were significantly higher after the conversion of preadipocytes into mature adipocytes. The resistin levels in the treated adipocytes were significantly decreased comparing to that of the untreated mature adipocytes. In conclusion, the expression levels of LPL, $TNF-{\alpha}$, leptin and resistin mRNA and proteins are shown to be regulated by G-Re and AEC, making them potential candidates for controlling fat mass related obesity.

Physicochemical Properties of Yullmoo (Coix lachryma-jobi var. mayuen stapf.) and Yeomjoo (Coix lachryma-jobi L.) Starches (율무와 염주 전분의 이화학적 특성)

  • The physicochemical properties of starch isolated from Yullmoo (Coix lachrymajobi var. mayuen stapf.) and Yeomjoo (Coix lachryma-jobi L.) were investigated. The average diameters of starch granules of Yullmoo and Yeomjoo were 12.0 microns, both of all, and the shape of these starch granules were hexagon, octagon and round. X-ray diffraction patterns of two samples were A-types and amylose contents of Yullmoo and Yeomjoo starch were 0% and 23%, respectively, iodine affinities of these were 0.08% and 4.2%, respectively, blue values and alkali numbers of these were 0.13 and 0.43, 2.4 and 7.2, and raising power of these were 280 and 20, respectively. Yullmoo starch had higher swelling power than Yeomjoo starch. The increase in optical transmittance of 0.1% suspensions of Yullmoo and Yeomjoo starches occurred at $60^{\circ}C$ and continued up to $75^{\circ}C$ for Yullmoo, $85^{\circ}C$ for Yeomjoo. Amylogram data on 5% of Yullmoo and Yeomjoo starch suspensions showed that gelatinization temperatures and maximum peak heights of Yullmoo and Yeomjoo were $68.5^{\circ}C\;and\;78^{\circ}C$, 920 and 310 B.U., respectiyely. Intrinsic viscosities of Yullmoo and Yeomjoo starches were 1.49 and 1.77, respectively, and interaction coefficients of the two starches were 0.57 and 0.56, respectively The extent of retrogradation determined at $2^{\circ}C$ showed that retrogradation occurred slowly with Yullmoo starch paste tut little with Yullmoo.

A study of dietetic therapy on the edema (부종(浮腫)의 식요방법(食療方法)에 관한 연구)

  • Objectives : The purpose of this research was to examine dietary therapy on the edema based on the literary findings, for clinical treatment and prevention of the edema. Methods : This Research was examined on histories, compositions, applications, and effects of dietetic treatment about 100 cases of dietary therapy of the edema from Chinese literatures. Results : 1. Various vegetables and animals including herbs, grains, vegetables, fruits, food and drink were used for the dietary therapy. 2. Methods of the preparation for use as therapeutics were decoction, pulvis, gruel, cake, tea, paste, gelatin and etc. 3. Frequently used materials were Cyprinus carpio Linnaeus, Cucumis sativus L. Coix lachrymajobi var. mayuen, Zea mays, Maydis Stigma, Citrullus vulgaris, Zingiber officinale, Cinnamomum cassia Blume, Allium scorodorpasum var. viviparum Regel, Camellia sinensis, Armeniacae amarum Semen, Phyllostachys bambusoides, Luffa cylindrica, Dioscorea batatas Decne, Panax ginseng, Brassica oleracea and Raphanus sativus. Conclusion : Though dietary therapy for the edema is not based on clinical or experimental data, but through experience. It is mostly based on Yin-Yang and five elements, visceral manifestation, channels and their collateral channels and Oriental herbal medicine theories. If we use them properly according to oriental medicine method, it will be effective on treating and preventing the edema.
