Natural Dyeing of Silks Dyed Singly with Caesalpinia sappan, Dactylopius coccus, Combination-Dyed with Caesalpinia sappan and Phellodendron Amurense Rupr., and with Dactylopius coccus and Phellodendron Amurense Rupr. (소목 및 코치닐로 단독 염색한 견직물, 그리고 소목과 황벽 및 코치닐과 황벽으로 복합 염색한 견직물의 천연염색)

  • We studied the dyeability and functionality of silks dyed singly with Caesalpinia sappan, Dactylopius coccus, combination-dyed with Caesalpinia sappan and Phellodendron Amurense Rupr., and with Dactylopius coccus and Phellodendron Amurense Rupr.. The color of silk dyed singly with Caesalpinia sappan was Red with Hue of 2.2 R, the color of silk dyed singly with Dactylopius coccus was Reddish Purple with Hue of 3.0 RP, The color of silk combination-dyed with Caesalpinia sappan and Phellodendron Amurense Rupr. was 2.7YR being in the Orange family, and the color of silk combination-dyed with Dactylopius coccus and Phellodendron Amurense Rupr. was Red with Hue of 4.3R. In the case of antimicrobial activity, except for silk dyed only with Dactylopius coccus, the other three types of dyed silk showed excellent antimicrobial activity against Staphylococcus aureus achieving higher than 91.1% of bacteria reduction rate. Against Klebsiella pneumoniae, silk combination-dyed with Dactylopius coccus and Phellodendron Amurense Rupr., and with Caesalpinia sappan and Phellodendron Amurense Rupr. presented very excellent antimicrobial property achieving bacteria reduction rate of 99.9%.

  • Since Kanda's paper on Korean scale insects in 1942, no work on scale insects has been done in Korea. The author has collected 8 species of scale insects in 5 green houses in Seoul and Suwon, of which 4 species, i.e., Chionaspis kentiae, Chrysomphalus aonidum, Planococcus kraunhiae and Aspidiotus hederae are new to Korea. Dominant species were Planococcus citri and Coccus hesperidum and common species to 4 green houses were Planococcus citri, Coccus hesperidum and Saissitia coffeae.

A study on effective disinfection methods of medical ultrasound probe resident floras (의료용 초음파 프로브 상제균의 효과적인 소독법에 관한 연구)

  • In the probe of a medical ultrasound device, three parts were selected randomly by the examiner and the bacteria in the probe were detected by the blood examiners. In addition, the degree of death of the pathogenic bacteria after each disinfection of the detected pathogens, disinfecting ethanol, and disinfecting tissue of the detected pathogens were analyzed quantitatively. The following was detected: S Aureus (32.3 %), Bacillus spp. (26.5 %), Micrococcus spp. (21.5 %), and CNS (20 %). With the conventional probe, S. aureus (26.2 %), a playback curve (24.2 %), and a micron (19.5 %), Micrococcus spp. (15.5 %), and CNS (14.6 %) were observed. In the fan probe, S. aureus (24.7 %), Enterococcus (24.7 %), Enterococcus (17.7 %), and CNS (13.8 %) were detected. The disinfection of the three pathogens detected revealed sterilization of most of the pathogens, and most cases contained at least 91.3 % of the total sterilizing effect (P>0.05). In addition, for the disinfection of Propolis extract and disinfecting tissue, the disinfection effect was lower than that of disinfecting ethanol, but the difference was not statistically significant (P>0.05). The results revealed bacteria on most of the ultrasound probes. Antiseptic disinfection of surgical instruments using an extract of propolis works with results similar to those of ethanol. A blood test along with disinfection can help prevent infection if an ultrasound probe is applied to food.

  • Microbial communities were analyzed in an anaerobic/aerobic sequencing batch reactor (SBR) fed with glucose as a sole carbon source. Scanning electron microscopy (SEM) showed that tetrad or cuboidal packet bacteria dominated the microbial sludge. Quinone, slot hybridization, and 165 rRNA gene sequencing analyses showed that the Proteobacteria beta subclass and the Actinobacteria group were the main microbial species in the SBR sludge. However, according to transmission electron microscopy (TEM), the packet bacteria did not contain polyphosphate granules or glycogen inclusions, but only separate coccus-shaped bacteria contained these, suggesting that coccus-shaped bacteria accumulated polyphosphate directly and the packet bacteria played other role in the enhanced biological phosphorus removal (EBPR). Based on previous reports, the Actinobacteria group and the Proteobacteria beta subclass were very likely responsible for acid formation and polyphosphate accumulation, respectively, and their cooperation achieved the EBPR in the SBR operation which was supplied with glucose.

  • Serratia marcescens characterized by the ability to produce red pigments inhabits various ecological niches. A strain Serratia marcescens PYU was isolated from abalone (Haliotis discus) collected at the West Sea in Korea. The isolated strain was gram-negative, motile, rods like coccus, oxidase-negative, and catalase-positive; and formed red pigment. S. marcescens PYU was grown in the presence of 0~10% (w/v) NaCl, at pH 4~9, and at $10{\sim}40^{\circ}C$. The strain PYU produced red pigment, and the extracted pigment showed antibacterial activity against Streptococcus iniae and Lactococcus garviae which has been known as an important fish pathogens. Further studies are underway to elucidate the direct relationship between the red pigment and antibacterial activity.

Isolation of Bacteria from Freshwater Plant Roots (담수식물 뿌리로부터 세균의 분리)

  • The isolation and identification of bacteria from freshwater plant root system were performed. Twenty four different strains were identified by microbiological identification system. Gram negative bacteria were isolated three times more than Gram positive ones. The ratio of rod and coccus was 11:1. The similarity above 0.5 was 37.5% from the tested samples in identification. Among these isolated bacteria, three dominant strains Pseudomonas cepacia JH10, Xanthomonas maltophilia JH12, Aeromonas salmonicida JH13 were selected to investigate biochemical properties and optimal growth conditions. These strains reached maximum growth after 24-36 hr of incubation in peptone broth, and their optimal temperature was 28$^{\circ}C$ and that of pH was between 7 and 8.

  • Antibacterial effects of the water-soluble extract of Ulmus pumila L. on Staphylococcus aureus and Escherichia coli were studied by cylinder plate dilution method. The cells of S. aureus and E. coli treated with the extract were comparatively examined by scanning and transmission electron microscopies for their morphological features. The normal cells of S. aureus and E. coli showed thypical coccus and rod shapes, respectively, but the cells of S. aureus treated with the extract showed rough surface structures with many granular protrusions and were destroyed to form ghost cells by liberating their cytoplasmic components. E. coli cells treated with the extract were destoryed without enlarging in size and producing granules on their surfaces.

  • A survey of pest occurrence and status of farmer's pest management was conducted at 45 cymbidium farms in 10 major cultivation areas in Korea. The pest species collected from the cymbidium farms were identified as follows: Tetranychus urticae Koch, Frankliniella intonsa Trybom, Pinnaspis aspidistrae Signoret, Incilaria confusa Cockarel, Halyomorpha brevis Walker, Myzus persicae S$\ddot{u}$lzer, and Aphis gossypii Glover, Coccus hesperidum Linnaeus, Thrips flavus Schrank, and Thrips tabaci Lindeman. The two-spotted spider mite, T. urticae, was the key pest in cymbidium production, occurring on 45 farms, followed by scales (20 farms), slugs (6), thrips (8), aphids (5), and stinkbug (1). PCR-RFLP of the rDNA ITS2 region revealed that two thrips species, Thrips flavus Schrank and Thrips tabaci Lindeman, occur on cymbidium farms. Therefore, it is necessary for the cymbidium farmers to establish an integrated pest management system to meet quarantine standards.
