Characterization and phylogenetic analysis of halophilic bacteria isolated from rhizosphere soils of coastal plants in Dokdo islands (독도 해안식물로부터 분리된 호염성 세균들의 특성 및 계통학적 분석)

  • To study the halobacterial diversity at the rhizospheric soil of coastal plant native to Dokdo islands, several host plant were selected and its rhizospheric soil was sampled. Soil sample was diluted serially and pure isolation was done by sub-culture using marine agar media. 26 halophilic strains cultivable at the marine medium containig concentration of 9.0% sodium chloride were selected among total 161 isolates. Their partial 16S rRNA gene sequences extracted from genomic DNA were analyzed and partially identified. Furthermore, to identify their genetic relationship, phylogenetic tree was deduced. Total 26 strains were belongs to Firmicutes (30.8%), Gamma proteobacteria (53.8%), Bacteroidetes (7.7%), Alpha proteobacteria (7.7%), and Actinobacteria (7.7%). These results showed the specific difference from previous researches which has been reported the microbial flora of soil or sea water around the Dokdo islands. Furthermore, 4 among 26 halophilic strains grew at above 12.0% NaCl concentrated marine broth, and 2 strains Idiomarina abyssalis LM4H23 and Halomonas huangheensis AS4H13 grew at 15.0% concentration. These halophilic strains thought to overcoming the severe stress like high salt concentration or variation derived from Dokdo-specific climate and might have unknown, specific relationship with their host coastal plant native to Dokdo islands.

The Effects of Field Trip Learning Program on Plant Inquiry in Coastal Dune using Artificial Intelligence on the Affective Domain of Gifted Elementary Science Studentt (인공지능을 활용한 해안사구 식물 탐구 프로그램이 초등 과학영재의 정의적 영역에 미치는 영향)

  • In the application and composition of learning content, the field trip learning of scientific inquiry could provide a positive effect. Also, it can arouse an experience of various inquiry activities through open thinking. In addition, it could take a positive effect by providing the diversity and specificity of wildlife experience for the living organism. The biology inquiry program of the field trip is a necessary process to acquire ecological experiences in the learning context. However, there is some problem to solve before the performance of field trip learning as professional knowledge of the outdoors inquiry. Therefore, this study developed a field trip inquiry program for the plant in a coastal dune using artificial intelligence to assist professional knowledge. The researcher carried out literature reviews and analysis related to studies and programs to investigate learning steps, content, and strategy. Also, this study investigated the effects of the program on the affective domain of gifted elementary science students. According to the results of this study, the program can provide a positive effect on motivation, task commitment, and attitude level. Consequently, the field trip learning program for plant in the coastal dune using artificial intelligence developed in this study can arouse a positive effect on the affective domain. Therefore, additional study is necessary related to inquiry programs of the field trip for various students and sites.

The Prediction of Littoral Transport in the Vicinity of Coastal Structures (해안구조물 주위의 표사이동예측)

  • This study describes a procedure of movable bed model tests and discusses the similitude with a view to state of the art. The prediction of littoral transport in the vicinity of coastal structure of nuclear power #9, 10 has been attempted. From the result of the present movable model study, it was found that the offshore breakwater is more excellent than the groyne and the rip-current barrier for the prevention against in take siltations and for the stability of investigated coastal structure of nuclear power plant.

A Three-Dimensional Numerical Model of Circulation and Heat Transport in Coastal Region (연안 해수유동 및 온배수 확산에 관한 3차원 수치모형)

  • This paper is concerned with the development of a three-dimensional numerical model for coastal circulation and heat transport with improved prediction ability. The model uses fully nonlinear, time-dependent three-dimensional, $\sigma$-transformed equations of motion and equation of heat transport The model was verified with experimental data for wind-driven current in a one-dimensional channel and thermal jets flowing into stagnant waters and applied for unsteady flow induced by tide and thermal jets in coastal waters around Kori nuclear power plant. The model results were in good agreements with experimental data sets for wind-driven current and thermal jet, and field observed data sets in coastal waters. This study has shown that the $\kappa$-$\varepsilon$ turbulence model is applicable to various coastal conditions without any modification of turbulence constants.

  • Geotextile tube method is the latest application process to construct a variety of civil structures such as river and coastal structures by using geotextile which is a high polymer synthetic fiber. In this paper, laboratory tests and field tests were conducted in order to identify the behavior, stability and application possibility of geotextile tube which prevents the erosion of coastal sand. As a result of large-scale direct shear test, which is one of laboratory tests, the increase in friction angle was shown as the relative density increased, and friction angle of sand/geotextile was larger than that of sand/sand. As a result of field test, the behavior and stability during construction and after construction were identified through measurement, and the effect of preventing erosion was confirmed.

Ecological Division of Habitats by Analysis of Vegetation Structure and Soil Environment -A Case Study on the Vegetation in the Kimpo Landfills and Its Periphery Region- (식생구조와 토양환경 분석을 통한 서식처의 생태학적 구분 -김포매립지와 그 근린 지역의 식생을 사례로 -)

  • Division of ecoregions having respective functions was attempted through quantitative and qualitative analysis on vegetation diversity, and heterogeneity and on soil environment of the study sites. Field research was carried out in a square of 81 ㎢ around Andongpo (126°38'E, 37°30'N), Kimpo-gun, Kyonggi provice. Conventional methods applied are as follows: classical syntaxonomy by the Zurich-Montpellier School, interpolation method to determine the degree of diversity, heterogeneity and distribution pattern of vegetation, and correlation analysis between soil properties and plant communities. 41 plant communities were identified and composed of 6 forests, 4 mantle and 31 herb communities including 6 saltmarsh plant communities. In a mesh, number of plant communities was highly correlated to the number of species. The highest number of plant community and species was 25 communities·km-2·mesh-1 and 381 species· km-2·mesh-1 ,and the highest value of vegetation heterogeneity was 28.1 species· community-1·mesh-1. Their lowest numbers were 4 communities·km-2·mesh-1. and 28 species·km-2·mesh-1. and 7 species·community-1·mesh-1, respectively. Contour map on vegetation diversity and heterogeneity enabled us to establish two regions; coastal and inland vegetation. Isoline 〔150〕,〔10〕and〔10〕and〔15〕on the species diversity, the community diversity and the vegetation heterogeneity, respectively, were regarded as ecolines in the study area. Cl- content was recognized as the most important factor from correlation analysis between soil properties. Ordination of sites indicated that the study area be divided into two edaphic types: inland and coastal habitats. It was considered that the extent of desalinization in soil played a major role in determining the species composition in the reclamed area. By matching edaphic division of habitats with division of vegetation structures, designation of ecoregion was endorsed. The approach of current study was suggested as an effective tool to implement an assessment of the vegetation dynamics by the disparity of natural environment and anthropogenic interferences.

Isolation, Characterization, and Use for Plant Growth Promotion Under Salt Stress, of ACC Deaminase-Producing Halotolerant Bacteria Derived from Coastal Soil

  • In total, 140 halotolerant bacterial strains were isolated from both the soil of barren fields and the rhizosphere of six naturally growing halophytic plants in the vicinity of the Yellow Sea, near the city of Incheon in the Republic of Korea. All of these strains were characterized for multiple plant growth promoting traits, such as the production of indole acetic acid (IAA), nitrogen fixation, phosphorus (P) and zinc (Zn) solubilization, thiosulfate ($S_2O_3$) oxidation, the production of ammonia ($NH_3$), and the production of extracellular hydrolytic enzymes such as protease, chitinase, pectinase, cellulase, and lipase under in vitro conditions. From the original 140 strains tested, on the basis of the latter tests for plant growth promotional activity, 36 were selected for further examination. These 36 halotolerant bacterial strains were then tested for 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase activity. Twenty-five of these were found to be positive, and to be exhibiting significantly varying levels of activity. 16S rRNA gene sequencing analyses of the 36 halotolerant strains showed that they belong to 10 different bacterial genera: Bacillus, Brevibacterium, Planococcus, Zhihengliuella, Halomonas, Exiguobacterium, Oceanimonas, Corynebacterium, Arthrobacter, and Micrococcus. Inoculation of the 14 halotolerant bacterial strains to ameliorate salt stress (150 mM NaCl) in canola plants produced an increase in root length of between 5.2% and 47.8%, and dry weight of between 16.2% and 43%, in comparison with the uninoculated positive controls. In particular, three of the bacteria, Brevibacterium epidermidis RS15, Micrococcus yunnanensis RS222, and Bacillus aryabhattai RS341, all showed more than 40% increase in root elongation and dry weight when compared with uninoculated salt-stressed canola seedlings. These results indicate that certain halotolerant bacteria, isolated from coastal soils, have a real potential to enhance plant growth under saline stress, through the reduction of ethylene production via ACC deaminase activity.

  • Numerical models have been widely used to understand the structure of coastal currents and the transport mechanisms in regard to the fate of pollutants. This study focuses on the development of a three-dimensional model of coastal circulation and mass transport. The model was used to calculate coastal currents and temperature distributions of the thermal plume discharged from a power plant. The model results were compared with field-observed data. They showed the relatively good agreements with the data. The model can be used to estimate the currents and its mass transport in coastal waters.

An Investigation of the Medicinal Plant Flora of Baek-Ah Island (백아도 약용식물 분포조사)

  • The author has investigated the plant flora of Baek-Ah Island during the period from August 4 to August 10 1970, and found that the flora consist of the plants of coastal, temperature and warm zones. The present report lists 81 families, 190 genera and 235 species of plants, of which 73 families and 170 species are medicinal plants.
