Gene Expression Profiling of Early Renal Toxicity Induced by Gentamicin in Mice

  • To elucidate the molecular mechanisms associated with early renal injury induced by gentamicin, the most commonly used antibiotics worldwide in the treatment of Gram-negative bacterial infections. We have identified genes differentially expressed at different duration of gentamicin administration. C57BL/6 female mice were treated daily with gentamicin (20 mg/kg, 100 mg/kg, and 200mg/kg) for 7 days and then sacrificed at day 1, 3, and 7 after administration. Standard blood biochemistry and histopathological observation indicative of nephrotoxicity were made. Total RNA was extracted from the kidney for microarray analysis using Affymetrix $GeneChip^{\circledR}$. Five hundred and seventy eight genes were identified as being either up-or down-regulated over 2-fold changes during early renal injury (p<0.05) and were analyzed by hierarchical clustering. The results showed that the genes involved in early immune responses were differentially regulated during early renal injury. Principal component analysis (PCA) confirmed sample separation according to the degree of renal toxicity. In addition, we identified two potential biomarkers that may predict early renal toxicity. This data may contribute to elucidate of the genetic events during early renal injury and to discover the potential biomarkers for nephrotoxicity induced by gentamicin.

Anti-multi drug resistant pathogen activity of siderochelin A, produced by a novel Amycolatopsis sp. KCTC 29142 (Amycolatopsis sp. KCTC 29142로부터 유래된 siderochelin A의 다제 내성 균주에 대한 항균활성)

  • A novel Amycolatopsis strain KCTC 29142 was isolated and characterized based on the polyphasic taxonomic analysis including morphological observation, phylogenetic analysis, physiological and chemotaxonomic characteristics. The ethyl acetate extract of strain KCTC 29142 culture broth showed strong antibacterial activity and the active compound was identified as siderochelin A, a ferrous-ion chelating compound. In this study, siderochelin A showed good activity against multi-drug resistant pathogens, including Acinetobacter baumanii, methicillin-resistant Staphylococcus aureus (MRSA), vancomycin-resistant Staphylococcus aureus (VRSA), and Escherichia coli (E. coli). The minimum inhibitory activity against clinical isolates was also determined.

Single Oral Dose Toxicity Study of WK-38 in Rats (랫트에서 WK-38의 단회경구투여 독성에 관한 연구)

  • Acute toxicity of WK-38, a herbal preparation for the atherosclerosis, was examined using male and female Sprague-Dawley rats. WK-38 is composed of Rhei Rhizoma, Magonoliae Cortx, Moutan Cortex Radicis. Rats were treated with the WK-38 intragastrically at 0 mg/kg, 5 mg/kg, 50 mg/kg, 500 mg/kg or 2,000 mg/kg and observed for two weeks. No mortality was observed at the doses used. Abnormal clinical signs such as eye bleeding, nasal bleeding and hyperemia had been shown temporary after administration. All rats were appeared to be healthy and normal during the 2 week observation. Also there was no difference in net body weight gain, gross pathological findings, and urine analysis among the groups rats treated with different doses of the WK-38.

A Clinical Analysis of Peripheral Nerve Sheath Tumors in the Head and Neck Region (두경부 말초신경초종의 임상적 고찰)

  • Background and Objectives:Peripheral nerve sheath tumors(PNSTs) are uncommon neoplasm in the head and neck region. The treatment of PNSTs is surgical removal, but excision cause neurologic complications. This study was performed to evaluate the proper diagnosis and treatment of PNSTs with our experiences. Subjects and Method:During the period from October 1994 to July 2007, 58 patients were diagonised with PNSTs in head and neck region. We reviewed medical records and imaging study retrospectively. Result:95%(55/58) of the PNSTs in head and neck were benign;5%(3/58) were malignant peripheralnerve sheath tumors(MPNSTs). 63%(37/58) were neurilemoma, 20%(17/20) were neurofibroma, 2%(1/58) was perineuroma. 55 patients underwent surgery. 80%(45/55) of cases were treated with excision. Enucleation was performed in 7(14%) patients. All cases of benign neurogenic tumors showed no recurrence. Among 3 of MPNSTs 2 patients were Von Recklinghausen’s disease and expired with regional recurrence and lung metastasis. Conclusion:The benign PNSTs can be treated with enucleation if possible and observation can be another choice to minimize neurologic sequele.

The Effects of Bangkibokryeong-tang on Papain-induced OsteoArthritis C57BL/10 Mouse (방기복령탕(防己茯苓湯)이 papain으로 유도된 생쥐의 골관절염에 미치는 영향)

  • Objectives : This study was carried out to know the anti-osteroarthritic effects of Bangkibokryeong-tang(Fanjifuling-tang(BBT)) on the papain-induced osteoarthritis C57BL/10 mouse. Methods : Osteoarthritis was induced by injection of papain(6 ${\mu}l$) into knee joint of mouse. Osteoarthritic mice were divided into 4 groups(normal, control, joins(R), BBT). The injection did not fit the normal group. A week later, after the injection of papain, control group was taken normal saline 200 ${\mu}l$, positive control group was taken joins(R)(100 mg/kg), treated group was taken extract of Bangkibokryeong-tang(Fanjifuling-tang(BBT))(400 mg/kg). After then, we examined hepatotoxicity, nephrotoxicity, inflammation cytokines, expression of inflammation factor mRNA, hemotology, histology through the micro CT-arthrography, and etc. Results : 1. Hepatotoxicity and nephrotoxicity have not expressed. 2. The levels of IL-$1{\beta}$, TNF-${\alpha}$, IL-6, MCP-1, Thromboxane B2, Leukotriene B4, Prostaglandin E2 in serum were significantly decreased. 3. In hematology, the levels of neutrophils and monocytes were significantly decreased. 4. The expression of inflammation factor mRNA like TNF-${\alpha}$ and IL-6, COX-2, iNOS-II were significantly inhibited. 5. In micro CT-arthrography, cartilage volume was less decreased. 6. The degree of osteoarthritis induced damage of joint of BBT group is low in histopathologic observation(hematoxylin&eosin(H&E), Safranin-O). Conclusions : According to this study, BBT has effect of anti-osteoarthritis. Further clinical research for the cartilage protective effect is necessary.

GUIDANCE OF ROOT FORMATION BY FORCED ERUPTION FOR INVERTED MAXILLARY CENTRAL INCISOR (역위 매복된 상악 중절치의 교정적 처치를 통한 치근 형성유도)

  • It is a relatively common clinical experience to see an unerupted maxillary central incisor. This phenomenon is apparent at the dental age of almost eight years and over. Among the possible cause for failure of eruption, ectopic development of the tooth germ is mentioned. This is not fully understood but trauma or periapical imflammation of primary predecessors is accepted. The case with no history of trauma may be impacted by the periapical imflammation of primary predecessors. For bringing into the tooth eruption and the continued normal root developement by the Hertwig's epithelial root sheath, there are early considered of surgical invention and orthodontic traction with removable appliance. We reported successful treatment for inverted maxillary central incisor with proper eruption and normal root developement by forced eruption using removable appliance. But further observation will be required to evaluate the final root developement state and amount of keratinized attachment gingiva.

Prognostic Factors of Necrotizing Enterocolitis (신생아 괴사성 장염의 예후 인자에 관한 연구)

  • Necrotizing enterocolitis (NEC) is the most common gastrointestinal emergency in the neonatal population. The aim of this study is to evaluate surgical indication and prognostic factors of NEC. Clinical data of seventy patients, diagnosed as NEC between January 2000 & January 2007, were reviewed retrospectively. Fifty-seven patients had medical treatment and 7 of them died. Thirteen patients who presented with pneumoperitoneum on plain abdominal film or were refractory to medical treatment received surgical treatment, and 5 of them died. All the expired 12 patients weighed less than 2500 g. Twenty out of seventy patients showed thrombocytopenia, and 11 patients of them died. The finding of pneumoperitoneum and thrombocytopenia could be the most important surgical indication. Prematurity, low birth weight and thrombocytopenia were related to a bad prognosis. NEC patients who presents with these findings must be considered for close observation and intensive care.

The Early Childhood Caring Experience of North Korean Refugee Mothers (북한이탈 어머니의 영유아 자녀 양육 경험)

  • Purpose: This study was done to develop a theory on the early childhood caring experience of North Korean refugee mothers and how such caring affects how they deal with the socio-psychological problems they face as North Korean refugees. Methods: Data were gathered by indepth interviews, participant observation, and medical records, and were analyzed using the Grounded Theory methods of Strauss and Corbin (1998). Results: From open coding 62 concepts, 23 subcategories and 11 categories were derived, and the North Korean refugee mothers' caring experience was revealed to be 'hopeful upbringing'. A central theme common to the participants was 'coexistence of expectations and worries'. Conclusion: The results of this study provide theoretical grounds to understand North Korean refugee mothers' child care experiences and offer personalized nursing and a deeper understanding of their needs by looking at their experience. Findings may also be useful to help nurses who care for North Korean refugee mother-child dyads in the community and in clinical settings to gain insight on this special needs group, and facilitate the development of interventions based on better understanding of the mothers' experiences.

  • Objective : The purpose of this study is to investigite the more effective oriental medical treatment in pediatric diseases and its clinical applicability and chief complaints at oriental pediatrics in an area of busan and kyeungnam. Method : 884 new patients of theirs ages between 0 term 20 years who were treated at the pediatrics in Dong-Eui Oriental Medicine Busan Hospital from 1. 2000 to 29. Feb. 2001 Result : 1. The number of male cases was 514 (58%) and female cases 370 (42%) an so male cases 1.38 more than female. In age distribution, 0-1year: 242 patients(27.4%), 2-6years: 455 patients(51.4%), 7-20years: 187 patients(21.2%). 2. The Chief Complaint according to systemical division was Resparatory diseases (37.4%) for common cold, chronic cought, take a easy cold etc and the Digestive diseases(23.6%) for anorexia, diarrhea, abdominal pain etc and Nervous also Mental(11.8%) diseases for sezure, tic, etc and Allergic diseases(6.2%) for atopic dermititis, allergic rhinitis, etc and Physique diseases(5.4%) for short stature, leg pain, etc and Genaral symptoms(4.5%) for sweat profusely, headache, etc and Urinary diseases(3.3%) for bed wetting, frequently voiding, etc Discussion : 1. The Chief Complaint cannot be found a regional specificity 2. The oriental medicine treatment was still congnated as a way to improve the weakness by patients, also was widely noised about preventive medicine. In particular, Respiratory and Disestive diseases are dependent on the oriental medicine. 3. The new diseases and the new treatment method should be investigated as field of oriental medical treatment.

Mouse Embryo Culture used in Quality Control of Water for Human in Vitro Fertilization : The One-cell Stage Versus the Two-cell Stage Model (수질에 대한 1-세포기 및 2-세포기 생쥐배아를 이용한 생물학적 정도관리에 관한 연구)

  • This study was carried out investigate the effect of water quality and the kind of media on the in vitro development of 1-cell and stage mouse embryos. $F_1$ hybrid mice were superovulated and timely mated. 1-cell stage and 2-cell stage mouse embryos were recruited and taken into Ham's F-10 or m-KRB media which was made of two of two kinds of water having different quality, highly purified water and tap water. 2-cell stage embryos grew up well in vitro to blastocyst or hatching blastocyst regardless of the composition of culture media, but 1-cell stage mouse embryo didn't develop well to blastocyst or hatching blastocyst in simple media like m-KRB. These results meant in vitro devleopment of 1-cell stage mouse embryo neded complex media like Ham's F-10 which contained abundant protein components. In case of quality control for water, in vitro fertilization program. observation of in vitro development of 2-cell mouse embryos up to blastocyst or hatching blastocyst media such as m-KRB would be efficatious in detecting the difference of water quality.
