허혈/재관류 환경하에서 청폐사간탕의 염증 관련 반응 억제 효과 (Inhibitory Effects of Chungpesagan-Tang on Ischemia/Reperfusion-Induced Inflammatory Responses In vitro)

  • Chungpesagan-Tang (CST), a Korean traditional prescription composed of oriental medical herbs, has been used successfully to improve human health and regimen. This study was designed to examine the inhibitory effects of CST against in vitro ischemia/reperfusion-induced inflammatory response. We have characterized the production of prostaglandin $E_2$ and arachidonic acid during ischemia/reperfusion in the human neuroblastoma SK-N-MC and human monocytic macrophage U937 cells and the inhibitory effect of CST on these inflammation-related substance formation has been found out in this study. This result suggested that CST used in this experiment reinforced antiinflammatory potentials and protected cells against ischemia/reperfusion-induced inflammatory resopnse.

저산소/재관류로부터 청폐사간탕의 PC12 세포 보호 효과 (Protective Effect of Metabolized Chungpesagan-tang on Hypoxia/Reperfusion Induced-PC12 Cell Damage)

  • This research was performed to investigate the protective effect of Chungpesagan-tang (CST) from hypoxia/reperfusion induced-PC12 cell damage. To elucidate the mechanism of the protective effect of CST, cell viability, changes in activities of superoxide dismutase, glutathione peroxidase, catalase, caspase 3 and the production of malondialdehyde were observed after treating PC12 cells with CST which was metabolized by rat liver homogenate. Pretreatment of CST with liver homogenate appeared to increase its protective effect against hypoxia/reperfusion insult. The result showed that CST exhibited the highest protective effect against hypoxia/reperfusion at the dose of $1\;{\mu}g/ml$ in PC12 cells, probably by recovering the redox enzyme activities and MDA to control level.

  • This research was performed to investigate the protective effect of Chungpesagan-tang (CPS) against ischemic damage in PC12 cells. To elucidate the mechanism of the protective effect of CPS on ischemic insult, cell viability and changes in activities of Superoxide dismutase, Glutathione Peroxidase, Catalase, Caspase 3 and the production of Malondialdehyde were observed after treating PC12 cells with CPS which was metabolized by rat liver homogenate. Pretreatment of CPS with liver homogenate increased its protective effect against ischemic insult by reducing the harmful effect of CPS itself. (omitted)

Hexane Soluble Fraction of Chungpesagan-tang Exhibits Protective Effect against Hypoxia/Reoxygenation-Induced N2a Cell Damage

  • Chungpesagan-tang (CST) has been traditionally used in Korea as a therapeutic for cerebral ischemia. To understand the protective mechanism of CST on hypoxia/reoxygenation insults in N2a cells, the cell viability was determined with the treatment of water solution and several solvent fractions of CST. The highest cell viability occurred when the cells were treated with the hexane soluble fraction of CST. Hypoxia/reoxygenation insults were shown to decrease the glutathione peroxidase (GPx) activity and the level of glutathione (GSH) and increase the superoxide dismutase (SOD) activity. However, treatment with hexane soluble fraction of CST ranging from 0.1 ${\mu}g$/ml to 10 ${\mu}g$/ml recovered the activities of GPx and SOD and maintained the levels of MDA and GSH at control levels. While hypoxia/reoxygenation insults induced the activation of ERK in N2a cells, treatment with the hexane soluble fraction of CST inhibited the activation of ERK in a concentration dependent manner. In this study, we were able to demonstrate that the bioactive compounds of CST can be effectively transferred into the hexane soluble fraction, and more importantly that CST exhibits protective effects against hypoxia/reoxygenation insults most likely by recovering redox enzyme activities.

청폐사간탕(淸肺瀉肝湯)이 중대뇌동맥 폐쇄 후 재관류에 미치는 영향 (The Effects of Chungpesagan-Tang Extracts on Reversible Forebrain Ischemia in Sprague-Dawley Rats)

  • Objectives : The purpose of this investigation is to evaluate the effect of Chungpesagan-Tang Extracts on reversible forebrain ischemia in Sprague-Dawley rats. Methods : the volume of cerebral infarction and edema, the pathohistological change of neurons, the number of survived neurons, neurotransmitters through immunohistochemical methods, proteins connected with neurotransmitters through immunohistochemical methods and the pathohistological change of neurons through electro-microscopy were investigated. From these reseach data, the protection of neurons and the activity of brain cells were examined. Results : 1. The infaction volume of the control group was 23.9%, and that of the sample group was 16%. 2. The brain edema volume of the control group increased by 17% compared to the normal group and that of the sample group increased by 10%. 3. The light microscopy revealed that the neurons in the ischemia-induced area and CA1 area of hippocampus were most heavily damaged and that the sample group was less damaged compared with the control group. Most pyramidal neurons died in 7 days when brain ischemia was induced. 4. The number of survived pyramidal neurons in the CA1 area of the hippocampus were studied. The normal group had 93 neurons/mm, survived the control group(after 3 days) had 21/mm, the control group(after 7 days) had 3/mm and the sample group 33/mm. 5. The immunohistochemical methods revealed that: (1) In the control group, the sensitivity of GABA, NOS, DBH were increased, and those of Synapsin, eEF-$1{\alpha}$ decreased. NOS and DBH had positive reactions in the control group, but negative in the normal group. (2) In thd sample group, the sensitivity of GABA, NOS, DBH were attenuated, and those of NPY, Synapsin, CaMKII, eEF-$1{\alpha}$ increased when compared to the control group. 6. The electro-microscopy revealed that most neurons died by necrosis and some neurons died by apoptosis. Several imflammation cells appeared in the injured area of neurons. The number of neurons in the sample group that died by ischemia decreased. But, the number that died by apoptosis did not significantly change. Conclusions : The data shows that the effect of Chungpesagan-Tang Extracts on reversible forebrain ischemia in Sprague-Dawley rats is significant.

사상방을 이용한 20대 여성 안면홍조 환자 치험 2례 (Two Case Reports of Facial Flushing of Women in Their Twenties Treated by Sasang Constitutional Medicine)

  • Objectives: The purpose of this study is to report the effect of Sasang constitutional medicine and acupuncture on facial flushing of women in their twenties. Methods: Two patients who had facial flushing were treated with Sasang constitutional medicine and acupuncture. In the first case, the patient diagnosed with Taeeumin was treated by Chungpesagan-tang and Sa-Am Liver-sedating Acupuncture. In the second case, the patient diagnosed with Soeumin was treated by Palmulgunja-tang, Sa-Am Spleen-tonifying Acupuncture and Seungyangsungi-tang which is not belong to Sasang constitutional medicine. The progress of symptoms was evaluated by NRS, frequency of facial flushing, duration time of facial flushing, amount of sweating accompanied by facial flushing. Results: After treatment, the patients' symptoms of facial flushing were improved. Conclusions: These case studies show that Sasang constitutional medicine and acupuncture are effective for the patient in their twenties who had facial flushing.

봉와직염(蜂窩織炎)의 한방(韓方) 임상치험(臨床治驗) 1례(例) (A Case Report on Cellulitis Treated with Therapeutic Intervention of Oriental Medicine)

  • A patient diagnosed as cellulitis by bone scan was taken the therapeutic intervention of Oriental medicine for 35 days. Cellulitis is characterized by acute purulence inflammation deeply diffused throughout epidermal connective tissue to subcutaneous adipose tissue. This patient presented a finding of soft tissue radioactive isotope uptake increase on bone scan of both foot and ankle. Cellulitis belongs to the category of ong(癰) in Oriental medicine. During therapeutic intervention, 3 herbal prescriptions were applied. Yeonkyopedok-san was applied for high fever and severe chilling on acute stage. After the application of Yeonkyopedok-san and Sunbangwhalmyong-eum, clinical symptoms improved with changes of CRP($112{\rightarro}12),\;WBC(13{\rightarro}8.8),\;ESR(65{\rightarro}46$). After the application of Chungpesagan-tang, clinical symptoms improved with changes of $CRP(12{\rightarro}3),\;WBC(8.8{\rightarro}5),\;ESR(46{\rightarro}13$).

  • A Case Report of Communication disorder associated with Post-stroke Objectives : The Objective Is the Report of Improvement in Communication Disorder Associated with Post-Stroke. Methods : Acupuncture And Medicinal Treatments Were Performed from 17th March 2006 until 8th April 2006. The Medicines Bangpungtongsung-san and Chungpesagan-tang Were Used to Improve Patient Bowel Movement. The Sa-Am Acupuncture Method Was Used to Improve Patient Communication Disorders. Every 5 Days Articulation Accuracy, Vowel Accuracy, Alternation and Speed of Reading Sentences Were Evaluated. Results : After 23 Days of Treatment There was Improvement of Articulation Accuracy, Alternation and Speed of Reading Sentences.

허혈/재관류 세포 손상에서 청폐사간탕의 보호 효과 (Protective effects of Chungpesagan-tang against ischemia/reperfusion induced cell injury)

  • 세포열을 이용한 허혈/재관류 환경에서 청폐사간탕의 세포보호능을 측정하였다. 청폐사간탕 추출물은 허혈/재관류 환경하에서 발생하는 세포 독성으로부터 대표적 수용성 항산화제인 ascorbic acid보다 높은 세포 보호 활성을 나타내었으며, 산화적 손상의 지표로서 사용되는 지질과산화물(TBARS)를 측정한 결과에서도 ASA와 유사한 활성을 나타내었다. 또한, 허혈/재관류 환경하에서 활성이 증가하여 세포에 산화적 손상을 일으키는 활성 산소종을 유발하는 것으로 알려진 xanthine oxidase 활성 측정에서는 청폐사간탕이 ASA보도 높은 xanthine oxidase 활성 억제능을 보였으며, xanthine oxidase 효소 표품을 이용한 활성 억제능 측정에서도 ASA보다 뛰어난 결과를 보였다. 따라서, 청폐사간탕은 허혈/재관류 환경하에서 세포 보호능이 있는 것으로 추측이되며, 이러한 보호능은 항산화 활성과 더불어서 xanthine oxidase 활성 억제능이 공동 작용의 결과로 사료된다.
