  • This study was performed to investigate the effect of chitosan and NOCC from different sources and of different molecular weights on cadmium toxicity. Sprague-Dawley rats were blocked into 26 groups according to body weight, and were raised for 4 weeks. Cadmium chloride was given at the level of 0 or 400 ppm in diet. Various forms of chitosan and NOCC were given at the level of 0 or 4%(w/w) of diet. Cd toxicity was alleviated by various chitosan and NOCC supplements. However, the alleviating effects were different with fiber source(crab and shrimp), type(chitosan and NOCC), and molecular weight (low, medium, and high). Molecular weight had no significant effect. Compared with shrimp-source-fiber-fed groups, crab-source-fiber-fed groups showed lower Cd concentrations in blood and kidney, lower kidney metallothionein concentration, and lower liver and kidney MT-Cd contents. Compared with NOCC -fed groups, chitosan-fed groups showed lower intestine and liver Cd concentrations, lower liver and kidney MT concentrations and MT-Cd content, and higher fecal Cd excretion. Among cadmium-exposed group, low molecular weight shrimp chitosan group showed low Cd concentrations of liver and kidney, high fecal Cd excretions, and the lowest intestine and liver MT concentrations and liver MT-Cd content. In summary, a crab source fiber was more effective than shrimp source fiber, chitosan was more effective than NOCC, and the most effective one in alleviating Cd toxicity was low molecular weight shrimp chitosan.

다이옥신계 TCDD (2, 3, 7, 8-tetrachlorodibenzo-p-dioxin)에 노출된 흰쥐의 지질대사 및 간 독성물질대사에 관한 키토산의 예방효과 (Preventive Effects of Chitosan on the Disorders of Hepatic Functions and Lipid Metabolism in Rats Treated with 2, 3, 7, 8-tetrachlorodibenzo-p-dioxin (TCDD))

  • This study was conducted to fine out the preventive effects of chitosan and chitosan oligomer on the disorders of hepatic functions and lipid metabolism induced by 2, 3, 7, 8-tetrachlorodibenzo-p-dioxin (TCDD) using adult male rats (SD) for four weeks. Rats were fed chitosan ($4\%$) or chitosan oligomer ($4\%$) diets respectively before 3weeks of TCDD treatment (50 ug/kg BW) by intraperitoneal injection and then continually supplied these diets for one week until being sacrificed. The elevation of serum total and LDL cholesterol levels induced by TCDD treatment was significantly reduced in the rats fed chitosan diets. The increment of liver triglyceride levels caused by TCDD treatment was tended to suppress in all rats fed chitosan and chitosan oligomer diets. Fecal total lipid and cholesterol excretion were high levels in the rats fed chitosan diets. The hepatic cytosolic catalase activities significantly decreased by TCDD treatment appeared recovering trend by chitosan diets. In hepatic microsomal cytochrome p-450, NADPH cytochrome p-450 reductase, ethoxycoumarin-o-deethylase (ECOD) and benzphetamin N-demethylase (BPND) chitosan than chitosan oligomer diets apparently decreased the increasing levels by TCDD treatment. In histochemical observation the fat droplets and apoptosis of hepatocytes by TCDD treatment were markedly alleviated by chitosan and chitosan oligomer diets. These results indicate that chitosan, more than chitosan oligomer can exert preventive effects on some disorders of hepatic functions and lipids accumulation by TCDD.

Various chitosan and NOCC sources were given at the level of 0%(w/w) of diet. Total serum cholesterol and serum triglyceride concentrations were little affected by chitosan and NOCC supplements. However, HDL-cholesterol concentration and HDL : total cholesterol ratio were increased , and liver lipid, cholesterol , and triglyceride concentration were decreased by chitosan and NOCC supplements. The cholesterol and lipid lowering activity depends on fiber sources(crab and shrimp); type (chitosan and NOCC); and molecular weight(low , medium , and high). Among cadmium-free groups, chitosan-fed groups showed greater activity than NOCC -fed groups in lowering cholesterol and lipid levels, and greater fecal excretion of lipids and bile acids. Crab chitosans were more effective in cholesterol and lipid lowering activity than shrimp chitosans. The group fed high molecular weight crab chitosan showed the highest fecal excretion of lipids and bile acids. This indicated that high molecular weight crab chitosan was most effective in interfering with cholesterol and lipid absorption.

The Growth, Innate Immunity and Protection against H2O2-Induced Oxidative Damage of a Chitosan-Coated Diet in the Olive Flounder Paralichthys olivaceus

  • We demonstrate enhanced growth, innate immunity and protection against hydrogen peroxide ($H_2O_2$)-induced protein oxidation and cellular DNA damage in olive flounder Paralichthys olivaceus fed a chitosan-coated moist pallet (MP) diet. A chitosan-based biopolymer coated MP as the experimental diet and a non-coated MP (control) was fed to olive flounder fish. Growth, including the average weight gain (g/fish), weight gain (%) and feed intake (g) of the fish group fed a chitosan-coated MP diet increased significantly. The survival rate was reported as 100% throughout the experimental period. Immunological parameters indicated higher mucus lysozyme activity and significantly higher fish skin mucus total protein content was observed in fish fed the chitosan-coated MP diet compared to the control. A blood plasma analysis revealed attenuation of cellular DNA and protein oxidative damage caused by $H_2O_2$-induced oxidative stress in the fish fed the chitosan-coated MP diet compared to the control group. Moreover, blood serum biochemical analysis revealed health-promoting effects, including significantly higher hemoglobin and total cholesterol levels in the fish fed the chitosan-coated MP diet compared to the control group. In conclusion, growth, innate immunity and protection against oxidative stresses were improved by feeding of the chitosan-coated MP diet to olive flounder reared in aquaculture.

  • The effects of chitin and chitosan on the contents of plasma and liver lipids and the fecal excretion of neutral and acidic steroids were investigated in rats fed with atherogenic diet. Dietary chitin did not affect plasma total cholesterol, LDL cholesterol and HDL cholesterol, while chitosan diet de creased total cholesterol and LDL cholesterol. Plasma content of triglyceride was reduced signif icantly by chitin diet but not by chitosan. Increases in fecal weight were observed in rats fed with either chitin or chitosan, and the increase was greater with chitosan than with chitin. Dietary chitosan resulted in an increase of fecal excretion of neutral steroids without any changes in the excretion of acidic sterols. Putting the results together, dietary chitosan reduces plasma cholesterol probably due to the reduction of cholesterol absorption. The decrease in plasma triglyceride level by dietary chitin requires further research to understand the mechinism.

키토산 두부가 고지질 식이를 급여한 흰쥐의 혈청 지질대사에 미치는 영향 (Effect of Chitosan-Soybean Curd on Serum Lipid Metabolism in Rats Fed High-Fat Diet)

  • 키토산을 응고제로 사용하여 상업 적 조건에서 제조한 두부와 일반 두부 및 키토산 분말을 고지질 급여 흰쥐 에6주간 급여한 후 혈청 지질대사에 미치는 영향을 조사하였다. 그 결과, 흰쥐의 식이 섭취량은 정상 식이군(N)에 비해 모두낮았으며, 키토산 두부 첨가군(CSC)과 키토산 분말 첨가군(CP)의 식이섭취량이 특히 낮았다. 체중 증가량은 고지질 식이군(HF)에 비해 CSC군, 일반 두부 첨가군(SC) 그리고 CP군들은 모두 유의적으로 낮았으며, 특히 CP군이 가장 낮은 체중증가를 나타내었다. 혈청중의 총 콜레스테롤 농도는 전 실험군간에 유의적인 차이는 없었으나, HDL-콜레스테롤 함량에서는 CSC, SC, CP군 모두 HF군에 비하여 높게 나타났다 따라서 총 콜레스테롤중 HDL-콜레스테롤이 차지하는 비율은 CSC, SC, CP군이 HF군에 비하여 높고 동맥경화지수는 반대로 낮게 나타났다. LDL-콜레스테롤 농도는 HF군에 비해 CSC, SC, CP 실험 군에서 유의적으로 낮게 나타났다. 총 지질 함량에서는 HF군에 비하여 SC와 CP군에서 유의적으로 감소하였다. 고지질 식이에 의해 증가한 중성지질 및 감소한 인지질 함량은 CSC, SC, CP군에서 유의적으로 회복되었다. 본 실험 에서는 HF군에 비해 CP군 뿐 아니라 CSC, SC군에서 HDL-콜레스테롤 및 인지질 농도의 증가, 중성지질 및 LDL-콜레스테롤 농도의 감소가 관찰됨으로써 상업용 키토산 두부가 고지질 관련 성인병 예방 및 치료에 효과가 있을 것으로 사료되었다. 그러나 현 실험조건하에서는 일반두부와 키토산 두부간에 효능에서 유의적인 차이를 발견하긴 어려웠다.

  • 키토산 첨가 혼합사료를 섭취한 양돈 근육 조직과 일반 사료를 섭취한 조직에서의 품질을 비교하기 위한 방법으로 생체광자를 이용한 광전자증폭관(Photomultiplier Tube, PMT) 기기를 사용하여 측정한 결과 서로 다른 결과를 얻었다. 키토산 첨가제를 사용한 돼지 등심조직에서의 육질 비교를 PMT와 현미경을 사용하여 측정한 결과 키토산 첨가 혼합 사료를 먹인 돼지 등심 조직의 생체광자 양을 측정한 결과 서로 상이한 수치를 얻을 수 있었다. 이것으로 조직의 품질을 비교 측정이 가능했다. 또한 이조직의 형태를 해부현미경으로 관찰한 결과 키토산 첨가 혼합사료 근육 조직의 육질이 더 섬세하고 촘촘하여 지방질이 적고 맛이 좋다는 것을 알 수 있었다. 따라서 PMT에 의한 조직 육질 측정은 생화학적 상태변화를 측정하는 새로운 분석법의 하나가 될 것으로 기대되어진다.

랫드에 있어서 키토산이 납중독의 대사에 미치는 영향 (Effects of Dietary Supplementation of Chitosan on the Metabolism of Lead in Rats)

  • This study was carried out to investigate the effect of chitosan on lead metabolism in SD rats. Ten male rats were divided into a experimental group and a control group. Each experimental and control rats were administered by water contaminated with 100 mg/l of lead for 4 weeks, respectively. Experimental group received diets supplemented artificially with 5% of chitosan for 4 weeks. Body weight change, food and water consumption, fecal and urinary excretion, and fecal and urinary lead excretion were measured. There were no significant differences in body weight gain, food and water consumption, and fecal and urinary excretion between the two groups. However, fecal lead excretion of rats fed the diet containing 5% of chitosan were higher than the control group. Whereas urinary lead excretion of rats fed the diet containing 5% of chitosan were lower than the control group. The results suggested that the oral administration of chitosan prevents the gastrointestinal lead absorption in rats.

납에 노출된 흰쥐의 혈액과 조직의 납 함량 및 병변에 대한 키토산의 섭취효과 (Effects of Chitosan on the Lead Level and Histological Changes in Rats Exposed to Various Levels of Lead)

  • Chitosan, which is a biopolymer, composed of glucosamine units linked by $\beta$-1, 4 glycoside bonds, is rich in shells of crustacean such as crabs and shrimps. Consumption of chitosan has been rapidly increased as a functional food. We examined effects of chitosan on the damages caused by lead (Pb) exposure in rats. Male Sprague-Dawley rats were divided into 8 groups (n = 64), then fed diets containing 3% cellulose (control) or 3% chitosan, each with 4 different lead doses (0 mg/d, 20 mg/d, 50 mg/d, and 100 mg/d) for 4 wks. Lead doses were given 3 times per week by oral administration. Blood lead levels in rats increased depending on the administered doses of lead. Rats fed chitosan diets showed lower blood lead concentration than did their respective controls. Effect of chitosan on the blood lead was more beneficial in rats exposed to lower lead (20 mg/d) than in rats exposed to higher lead (50 mg/d and 100 mg/d). Histological changes in erythrocytes and liver were also examined. Chitosan tended to reduce numbers of basophilic stippling erythrocytes and improve the histological liver changes in rats given various lead doses. The preventive effects of chitosan on liver damages were stronger in rats with higher lead than those with lower lead. These results indicate that chitosan has beneficial effects on both blood toxicological responses and histological damages of erythrocytes and liver induced by the administration of various lead doses.

Effect of Chitosan on Nitric Oxide Content and Inducible Nitric Oxide Synthase Activity in Serum and Expression of Inducible Nitric Oxide Synthase mRNA in Small Intestine of Broiler Chickens

  • The present study was conducted to determine the effects of chitosan on nitric oxide (NO) content and inducible nitric oxide synthase (iNOS) activity in serum, and relative expression of iNOS mRNA in the duodenum, jejunum, and ileum of broiler chickens. A total of 240 one-day-old Arbor Acre mixed-sex broiler chickens were randomly allotted to six dietary treatments with five replicates in each treatment and eight chickens in each replicate. The broiler chickens in the six treatments were fed the basal diet supplemented with 0 (control), 0.05, 0.2, 0.5, 1.0 or 2.0 g/kg chitosan. The trial lasted for 42 days. The results showed that dietary chitosan enhanced NO content and iNOS activity in serum as well as iNOS mRNA expression in the duodenum and ileum of broiler chickens in a quadratic dose-dependent manner (p<0.05), and improved jejunum iNOS mRNA expression in a quadratic dose-dependent manner (p<0.10) with increasing addition of chitosan. Chicks fed a diet containing 0.5-1.0 g/kg chitosan had higher NO content and iNOS activity in serum as well as small-intestinal iNOS mRNA expression compared with birds given the control diet, but positive effects of chitosan tended to be suppressed when addition of chitosan in the diet was increased to 2.0 g/kg. These results implied that there was a threshold level of chitosan inclusion beyond which progressive reductions in serum NO content and small intestinal iNOS expression occured, and the regulation of chitosan on immune functions in chickens is probably associated with activated expression of iNOS and NO secretion.
