Effect of Chitosan on Expression of Osteogenic Genes during the Healing of Rat Extraction Socket

  • Purpose: This study was performed to evaluate the effect of chitosan combined with absorbable gelatin compressed sponge on the expression of osteoblastic differentiation marker genes during the healing of rat extraction socket. Materials and Methods: Twenty-four male Wistar rats were used. In control group, the extraction socket was closed with suture. In chitosan group, the socket was filled with chitosan combined with Gelfoam (Pharmacia & Upjohn Co.) and closed with suture. In each group, the animals were sacrificed at 3 days, 1 week, 2 weeks, and 4 weeks postoperatively. The expression of osteoblastic differentiation marker genes, including BSP, OCN, Runx2, and Col1 were quantified by real-time polymerase chain reaction. Result: Compared to control group, the mRNA level of BSP in chitosan group increased significantly at 2 weeks after extraction and the level of OCN decreased significantly at 3 days and 4 weeks after extraction (P<0.05). The mRNA levels of OCN, Runx2, and Col1 in chitosan group increased slightly at 2 weeks after extraction, but there was no statistical difference between groups. Conclusion: The results indicate that chitosan has some effects on the expression of osteogenic genes during the healing of extraction sockets.

Effects of Foliar Application of Chitosan and Wood Extraction on Rooting and Tuber Formation of Plug Seedlings in Potatoes (Chitosan과 목초액 엽면살포에 의한 감자 플러그 삽목표의 발근 및 괴경형성효율)

  • To do mass multiplication of plug seedlings in potatoes, apical stem cuttings originated from virus-free microtubers were cut to one-two internodes and transplanted into the plug-tray. After 10days, we applied Chitosan and Wood Extraction on rooting and tuber formation of plug seedlings. To improve field adaptability of plug seedlings, rooted cuttings with a height of 20cm after 20days of cutting were transplanted ito the fields, We applied 500~2000ppm Chitosan on growth characteristics and tuber formation of that. The above and underground growths, such as plant height and number of leaves were significantly more vigorous after treatment with 500~1,000ppm Chitosan and 2,000ppm Wood Extraction, the spray treatment was carried out five times at intervals of four days after ten days of transplanting. T-N, K, P, Mg and Na, were higher as the concentrations of chitosan and Wood Extraction were higher. The growth and tuber yield in plug seedlings planting plot and seed potatoes planting plot were effectively highter as foliar application of Chitosan(500~2,000ppm) was done after planting the plot. T-N content in leaves and tuber was higher as the concentration of Chitosan was high. A similar tendency was shown in K, P and Mg. In the small tuber(under 30g), the number of tubers and tuber yield were relatively increased in the seed potatoes planting plot, but the large tubers(over 80g) yield was higher in the plug seedlings planting plot, and in order to increase tuber yield in plug seedlings it was necessary to add plant density to the field.

  • By introducing chitosan treatment prior to dyeing, examination was given to the effect of chitosan coated on the surface of fabrics on the color change of dyed fabric based on th e change of ${\Delta}E$, a*, and b* values. At the same time, the dyeing mechanism of Caesalpinia sappan dyestuff was predicted by the investigation of the change of air-permeability ac cording to the chitosan treatment. The change of elution was investigated by the examination of the elution of metallic ions employed as mordants after soaking dyed fabrics in the solution of alkaline perspiration ba sed upon the fact that chitosan carries excellent absorption ability toward metallic ions.

  • The Louisiana crawfish industry comprises the largest concentration of crustacean aquaculture in the United States. Processing plants throughout the culture region annually generate as much as 80 million pounds of peeling waste during recovery of the 15% (by weight) edible tail meat. A commercial oil extraction process for recovery of carotenoid astaxanthin from crawfish waste has been developed. Crawfish pigment in its various forms finds applications as a source of red intensifying agents for use in aquaculture and poultry industries. Crawfish shell, separated in the initial pigment extraction step, is an excellent source of chitin. Applicable physicochemical procedures for isolation of chitin from crawfish shell and its conversion to chitosan have been developed. Crawfish chitosan has been demonstrated to be both an effective coagulant and ligand-exchange column material , respectively, for recovery of valuable organic compounds from seafood processing wastewater.

Physicochemical properties of chitosans produced from shrimp shell under the different conditions and their influences on the properties of Kakdugi during storage (제조조건이 다른 새우껍질 chitosan의 물리.화학적 성질 및 깍두기의 보존성에 미치는 영향)

  • This study was undertaken to investigate the physicochemical properties of chitosans produced from chitins which were prepared from shrimp shell under the 4 different conditions(HCl concentration : 1 N, 4 N and reaction temperature $0^{\circ}C,\;20^{\circ}C$). The effects of chitosan on the properties of Kakdugi during storage also were examined. Viscosity of chitosan was markedly different$(313.1{\sim}98.8\;cps)$ depending on the production conditon. Chitosan solution showed pseudoplastic property. The infrared(IR) spectrums of chitosans prepared under the conditions of this study were very simillar, and the degree of deacetylation of chitosan was relatively high$(92{\sim}96%)$ independent of the extraction conditions of chitin. As the storage period was extended, markedly lower pH and higher titratable acidity were resulted in all the groups. Throughout the storage period, pH and titratable acidity of kakdugies with chitosan were higher and lower, respectively than those of control. Viscosity of kakdugi juice was significantly different among the groups and as the storage period increased, the juice viscosity decreased. At the eighth day of storage, the juice of control group was more viscous than those of kakdugies containing chitosan. Throughout the storage period the numbers of total microorganisms and of microorgarnisms of Leuconostoc genus in control kakdugi tended to be higher and lower, respectively than those in kakdugies containing chitosan. The growth of Lac. plantarum was slightly lower in kakdugi containing chitosan C produced at low HCl concentration and high temperature compared to the others.

Current status and prospects of chitosan for industrial applications (키토산 관련 산업의 현황과 전망)

  • For the development of the chitosan industry in Korea, the catch of red snow crabs caught on the east coast is rapidly decreasing. Therefore, it is urgent to develop raw materials that can replace the red snow crab as the top priority to solve the supply and demand problems, as well as wastewater treatment costs account for a large proportion of the cost of chitosan. In order to solve the problems, continuous research on biological extraction methods such as enzymatic extraction and microbial fermentation will increase production efficiency and lower unit cost as well as chemical extraction methods. Further efficient manufacturing method can be established. Establishing of novel techniques is indispensable for production of high-purity chitosan and the ability to regulate and separate the molecular weight, as well as joint research with industry, academia and research institute for the research and development of high-functional chitosan derivatives.

Characteristics of Glycyrrhiza uralensis extract-loaded chitosan nanocapsules and their antioxidant activity (감초 추출물 함유 키토산 나노캡슐의 특성 및 항산화 활성)

  • To improve the oxidative stability of Glycyrrhiza uralensis extract (GU), GU extraction conditions were optimized for maximal antioxidant activity, and GU-loaded nanocapsules were prepared by chitosan ionic gelation. The optimized ethanol concentration and extraction time were 83.0% and 32.6 min, respectively, using response surface methodology. The particle size of the GU-loaded nanocapsules ranged from 280 to 370 nm. A GU extract of 0.8 mg/mL and chitosan concentration of 2.0 mg/mL were selected as the optimal conditions for entrapment and loading efficiency. Both free GU and GU-loaded chitosan nanocapsules exhibited concentration-dependent antioxidant activity. However, the antioxidant protection factor of GU was effectively maintained when it was entrapped within the chitosan nanocapsules. In conclusion, chitosan nanoencapsulation is a potentially valuable technique for improving the oxidative stability of GU.

Optimum Extraction Condition of Peach Liqueur Containing Chitosan (키토산을 첨가한 복숭아 리큐르의 침출조건 최적화)

  • This study was performed for obtaining optimal extraction conditions to produce peach liqueur containing chitosan. Alcohol content of unsliced fruits was the largest recording 30% after extracting for 8 weeks and those of fruits sliced into four pieces were high recording around 28% after extracting for $6{\sim}8$ weeks. Sugar content was $11{\sim}13^{\circ}Brix$, total acidity was around 0.2% and pH was about 4.8. While soluble solids content was around 2.8% in unsliced fruits with a little change of the content, those of fruits sliced into four pieces were ranged from 2.8% to 3.1% and showing a tendency that longer extraction time increased the content. Brownness of unsliced fruits and fruits sliced into four pieces were $0.33{\sim}0.54$ and $0.56{\sim}0.73$ respectively. Total phenol content showed a similar tendency with brownness and the total phenol content of sliced fruits was higher than that of unsliced fruits by only around 0.2 mg%. When peach liqueur was made by extracting unsliced fruits massively by following conditions set above, methanol content grew with longer extraction time but the difference was not significant and the content was detected to be extremely small recording about 50 ppb. Among organic acids, oxalic, malic and citric acids were found, and fructose, glucose, sucrose and maltose were detected as free sugars. In conclusion, extracting unsliced fruits for 8 weeks as extraction conditions showed an excellent quality in overall for peach liqueur containing chitosan.

  • African marigold is a common plant easily available in many flower beds. It has been reported as a practical and prospective resource of dyes since the dyes can be extracted from their bodies as well as petals. In this research, cotton and ramie fabrics which are made from natural cellulose were dyed using the extract of the African Marigold which is a variety of marigold. Dyeing tests were carried out under different pH of the dye solution and mordants. Dyeability was evaluated by examining and measuring surface color, K/S value, and the changes in the maximum absorption wavelength. The probability of improving dyeability was investigated by pre-mordanting with pre-treated chitosan. For the dyeing with marigold extract, the color tone did not differ by pre-mordanting and non-mordanting. Reaction with post-mordanting was excellent, which was colored in various yellow series. The best dyeability was achieved in dye solution of pH 6.5-7.0 which is not conditioned. The largest K/S value and color difference were obtained in tin mordanting. The dye uptake was greatly increased in chitosan pre-treated mordanting compared with the post-mordanting without chitosan pre-treatment. Due to its high heat resistance, African Marigold extract is easy for dye extraction and dyeing, and its dyeability is excellent for natural cellulose fibers. Also, colorfastness was proved to be practically usable.

  • The purpose of this study is to evaluate the bioresorbability of Calcium Polyphosphate added with $Na_2O$ and chitosan. Though calcium phosphate ceramics meet some of the needs for bone replacement, they have some limitation of unresorbability and fibrous encapsulation without direct bone apposition during bone remodelling. To solve these problem, we developed a new ceramic, calcium polyphosphate(CPP), and report the biologic response to CPP in extraction sites of beagle dog. Porous CPP granules were prepared by condensation of anhydrous $Ca(H_2PO_4)_2$ to form non-crystalline $Ca(PO_3)_2$. CPP granules added with $Na_2O$ and chitosan were implanted in extraction sockets and histologic observation were performed at 12 weeks later. Histologic observation at 12 weeks revealed that CPP matrix were mingled with and directly apposed to new bone without any intervention of fibrous connective tissue. CPP granules added with chitosan were well adatped without any adverse tissue reaction and resorbed slowly and spontaneously. CPP granules added with $Na_2O$ and chitosan show multinucleated giant cells and osteoblast-like cells around grafted material and newly formed bone. This result revealed that CPP, regardless of its additive component, had a high affinity for bone and had been resorbed slowly. From this results, it was suggested that CPP is promising ceramic as a bone substitute and addition of $Na_2O$ and chitosan help biodegradation. In further study , it will be determined which concentration of $Na_2O$ help biodegradation and the other additive components increase the degradation rate.
