Study on the Antimicrobial Activities of Natural Products

  • The antimicrobial activities of extracts from some of natural products and bacteriocins produced from lactic acid bacteria such as L. mesenteroides, L. citreum and P. pentosaceus have been investigated with using disc diffusion assay, challenge method and minimum inhibitory concentration method. The bacteriocin substances produced from L. mesenteroides, L. citreum and P, pentosaceus have shown strong antimicrobial activities against E. coli, S. aureus, P. aerugionosa, and B. subtillis while the extracts from phenonip and sandal wood have shown strong antimicrobial activities only against C. albicans. The bacteriocins produced from L. mesenteroides, L. citreum and P. pentosaceus could be a promising preservatives for cosmetics, pharmaceutical products and foods in future.

Angular-based Measurement for Quantitative assay of Albumin in three-dimensional Paper-based analytical Device (회전각도를 이용한 알부민 농도 측정용 3차원 종이 칩)

  • This study presents an angular-based measurement on three-dimensional paper-based analytical devices (3D-PADs) for quantitative detection of albumin without using an image analyzer. We demonstrate a simple quantitative and straightforward approach based on the angle of the discolored area as detection criteria. 3D-PADs are rapidly fabricated by the wax-printing and laminating process. The 3D-PADs are treated with citrate buffer and tetrabromophenol blue to react with albumin in a sample solution. Dropping sample solution into sample pad in the 3D-PAD, fluid flows toward the assay zone laterally and vertically by capillary action. We find that the change of angle of the discolored area correctly reflects the concentration of albumin and is reliable determinant for the measurement of the albumin concentration. It is the first demonstration of angular-based detection as a simple, inexpensive, and equipment-free approach for point-of-care diagnosis.

  • Scutellaria baicalensis Georgi (SBG) is traditionally used medicinal herb that has anti-oxidant, anticancer and anti-inflammatory effects. In this study, we investigated whether the extracts of SBG have the inhibitory activity in the osteoclast differentiation by using mouse monocytes RAW264.7 cells and primary mouse bone marrow-derived macrophages (BMMs). Methanol extract (ME) from SBG was successively fractionated into methylene chloride (MF), ethylacetate (EF) and n-butanol fraction (BF). The activity assay for tartrateresistant acid phosphatase (TRAP) and Western blot analysis were employed to evaluate the osteoclasts differentiation and the activation of mitogen-activated protein (MAP) kinases, respectively. ME, MF, EF and BF significantly and dose-dependently inhibited osteoclast differentiation without the decrease of cell viability at the concentrations used in this study. In addition, ME significantly inhibited the activation of c-jun-N-terminal kinase (JNK). In conclusion, this study firstly demonstrated that ME of SBG has the potential to inhibit the osteoclast differentiation through the suppression of JNK activation partially.

The control effect of some fungicides against cucumber sclerotinia rot and the sensitivity of sclerotinia isolates to fungicides (오이 균핵병에 대한 몇 가지 살균제의 방제 효과와 살균제에 대한 균핵병균의 감수성 정도 조사)

  • As Sclerotinia sclerotiorum causing cucumber sclerotinia rot was the fastest in the mycelial growth at $25^{\circ}C$, its pathogenicity was strong at the same temperature among several temperatures. All the isolates of Sclerotinia sclerotiorum showed a strong pathogenicity against cucumber fruits, which was confirmed by a disk assay and a wound assay. A wound assay was superior to a disk assay to develop the assay system for assessing the fungicidal activity of several fungicides against Sclerotinia sclerotiorum. In a disk assay, it was very difficult to assess the fungicidal activity, because the pathogenicity of isolates used in the experiment was very strong. At 500 and $3.0{\mu}g/mL$, the activity of dichloflouanid and the mixture of carbendazim and diethofencarb against cucumber sclerotinia rot was 14.3 and 42.3%, respectively, by using a disk assay. However, at same concentration two fungicides showed the high controlling activity as 100 and 92.5%, through a wound assay in a laboratory. Also, the activity of two fungicides was good against cucumber sclerotinia rot in the greenhouse where cucumber plants were cultivated in the field, showing the control value as 91.1 and 82.9% at 100 and $825{\mu}g/mL$, respectively. All the isolates of Sclerotinia sclerotiorum from cucumber fruits sampled in the polyvinyl house were subjected to monitoring for the resistance to 7 fungicides. The $EC_{50}$ value of 7 fungicides was as follows: fenhexamid; $0.13{\mu}g/mL$, procymidon and iprodione; 0.18 and $0.24{\mu}g/mL$, carbendazim and the mixture of carbendazim and diethofencarb; 0.13과 $0.05{\mu}g/mL$, iminoctadine and dichlofluanid; 1.94 and $8.95{\mu}g/mL$. Ultimately it was not found that resistant isolates of Sclerotinia sclerotiorum were appeared in the field.

  • A series of new $N^7$-tetrazolo[5,1-f ]-1,2,4-triazin-8-(7H)-one derivatives 4-16 were designed and synthesized from 3 with different reagents. The newly prepared compounds were characterized by spectral data and screened for their antimicrobial activities against various bacteria and fungi strains.

  • The adhesion of hydrocarbon degrading bacteria(HDB) differing in surface hydrophobicity was investigated. Cell wall hydrophobicity was modified chemically and physiologically. Modified adhesion deficient mutant of HDB was selected in a soil column assay Physiologically and chemical modification increased cell surface hydrophobicity. Cell surface charcteristis including BATH and zeta potential were measured. Physiological modification using ampicillin was not stable, but chemical modification was stabel. Hydrocarbon degrading potential was measured for modified and unmodifed HDB.
