Developmental Characteristics of Eggs and Yolk Sac Larvae of Korean Striped Bitterling, Acheilognathus yamatsutae (Cyprinidae), Spawning in Mussels

  • This study investigated the characteristics of the eggs and yolk sac larvae of Korean striped bitterling, Acheilognathus yamatsutae, spawned and grown In mussels. The number of eggs in the ovary was small ($358{\pm}108$ SD). The eggs were oval and large, and the formation of the perivitelline space was narrow. The eggs were hatched at only 41 hours after fertilization but the hatched larvae were underdeveloped. The development of yolk projection and minute tubercles on the skin surface was notable, along with the vividly moving tail in the hatched larvae. The yolk projection and minute tubercles were disappeared upon enhancement of the motor ability of the larvae was enhanced. The formation of eyes and body pigments of the larvae was relatively delayed in comparison with that of other cyprinid larvae. After completely consuming the yolks the larvae escaped from the mussel for free swimming and exogenous feeding.

  • Six new serovarieties of B. thuringiensis carrying specific H-antigen have minor differences in biochemical characteristics and morphological characteristics of crystals, which are commonly resistant against four antibiotics. The B. thuringiensis serovar. coreanensis is nontoxic to silkworm larvae, but it is moderately toxic against the Culex pipiens larvae. The B. thuringiensis serovar. konkukian and leesis are nontoxic against mosquitos larvae, but are toxic against silkworm larvae. The B. thuringiensis serovar. seoulensis, sooncheon, and yosoo are highly toxic to B. mori larvae and moderately toxic to C. pipiens larvae. The six serovarieties harbor different plasmid DNA patterns. A 102-kDa protein is a major crystal protein in the four serovarieties and a 86-kDa protein is in one serovariety.

Ontogenetic variation in Chironomus flaviplumus (Diptera, Chironomidae) larvae

  • Chironomus is a large genus of Diptera comprising about 400 species and occurs worldwide except for Antarctica. The strong morphological cross-taxon similarity of chironomid larvae renders identification at the species level difficult. Here, we analyzed the morphology of larvae of Chironomus flaviplumus, an easily cultured species employed as a bioindicator in polluted environments, to determine identifying morphological characteristics at the first through fourth instar. Observed differences appearing at each instar include the presence or absence of setae on the body and tubules on the 10th and 11th body segment, the number of seta interna in the mandible, and the presence or absence of ring organs in the antennae. Some specific morphological characteristics did not change after hatching. Our findings provide a reliable method for identifying C. flaviplumus larvae.

환경정화곤충의 활용이 한우분에 미치는 영향 (Effects of Using Environmental Purification Insects on Hanwoo Manure)

  • This study was conducted to evaluate the effects of using Hermetia illucens larvae as environmental purification insects on ammonia and Hanwoo manure for 15 days in the Hanwoo farms. H.illucens larvae were allocated into four treatment groups and three replicates using a randomized block design. The treatments were as follows: control, normal 1000 g of Hanwoo manure, T1, 1000 g of Hanwoo manure + 100 g of 2nd instar H.illucens L. larvae, T2, 1000 g of Hanwoo manure + 100 g of 3rd instar H.illucens L. larvae, and T3, 1000 g of Hanwoo manure + 100 g of 4th instar H.illucens L. larvae. After 0 days, ammonia fluxes did not occur in the 2nd, 3rd, and 4th instar H.illucens larvae treatments (p<0.05). Among the characteristics of Hanwoo manure, the pH and nitrogen content increased for each larvae instar of H.illucens, whereas ADF and NDF content decreased (p<0.05). In particular, nitrogen contents were found to be highest in the 3rd instar larvae of H.illucens owing to their greater ability to decompose Hanwoo manure than larvae at other growth stages.

Morphological Differences between Larvae of the Oriental Fruit Moth (Grapholita molesta Busck) and the Peach Fruit Moth (Carposina sasakii Matsumura) in Korea

  • The oriental fruit moth (Grapholita molesta Busck) and the peach fruit moth (Carposina sasakii Matsumura) are the most severe insect pests affecting apple orchards in Korea. To prevent an outbreak of these two species and to control these agricultural insect pests, it is important to identify them accurately. However, it is hard to classify them when they were in the larval stage since they tunnel into the apple fruit. In this study, surface structures of the two species of larvae were observed using stereo microscope and scanning electron microscope. Distinct differences between the two species of larvae were found. The prothorax spiracles of oriental fruit moth larvae were approximately twice as large as those of peach fruit moth larvae. The arrangements of subventral setae, located around the proleg, were different between oriental fruit moth and peach fruit moth larvae. Furthermore, subdorsal setae of oriental fruit moth were located next to the spiracle on the 8th abdominal segment, while that of peach fruit moth was located above the spiracle. The identification of the two species of larvae observed in this study was confirmed using polymerase chain reaction-restriction fragment length polymorphism method. Surface structural differences are intrinsic characteristics for each species of larvae and can easily be identified using stereo microscope. These specificities will be helpful where a large number of field-collected larvae need to be identified routinely in pest control research.

한강 수계 내 서식하는 깔따구류 유충의 속 수준에서의 분류 형질 (Taxonomic Characteristics of Chironomids Larvae from the Hangang River at the Genus Level.)

  • 깔따구류 유충(Chironomidae larvae)은 저서성대형무척추동물로 수질 및 환경오염 생물 모니터링에 중요한 지표생물이다. 본 연구에서는 2022년 7월에서 9월 동안 한강의 다양한 수계에서 서식하는 깔따구류 유충을 채집하여 형태적 동정 및 유전자 계통 분석을 실시하였다. 20개 지점에서 총 3아과 18속 32종의 깔따구류 유충이 출현하였으며, 반지깔따구속에 속하는 1개 종이 6개 지점에서 우점하였다. 깔따구류 유충을 분류하는 데 이용하는 몸통, 두부, 하순기절, 촉각 등을 관찰하여 특징을 Pictorial key로 제시하였고, 유전자 계통분석 결과 3개 아과가 명확하게 구분되었으며, 속 수준에서도 구분이 되었다. 이러한 결과들은 깔따구류 유충 동정에 도움이 될 것이고, 수질 조사 및 관리에 기초적인 자료로 활용할 것으로 기대된다.

흰점박이꽃무지 산란 특성 조사(2) - 월동(동면) 유충을 중심으로 - (Investigation of Oviposition Characteristics of Protaetia brevitarsis seulensis (2) - Winter Larvae -)

  • The present study was investigated the oviposition characteristics of Protaetia brevitarsis seulensis with a focus on winter larvae to improve the utilization of insect resources. The average oviposition tendency and average cumulative number of eggs laid during a 7-week period were measured in adults emerging from larvae that had hibernated for 4 months in five farmhouses. The study covered five zones: Gimje, Jangsu, Wanju, Iksan and Yeoju. The average oviposition tendency over a 7-week period showed increased egg laying between 2 to 3 weeks and 5 to 6 weeks. Overall, it was found that the average cumulative number of eggs laid during the 7-week period was initially low and increased proportionally with time. Our results suggest that using adults that have emerged from hibernated larvae is effective in increasing the oviposition rate and utilization of the insect.

  • Tenebrio molitor is a major pest of stored grain, although its larvae have potential for use as food. However, little is known about the characteristics of individual larval instars of this species, and the number of instars remains disputed. Therefore, we assessed T. molitor for the average number of instars and its characteristics at different larval stages. The focus of this study was to establish a foundation for further studies on the characteristics of each larval instar. All of the T. molitor larvae showed incubation periods of 7 to 8 d and a period of 3 to 4 d for the $1^{st}$ instar. Beyond the $1^{st}$ instar, there were relatively large variations in the number of days in each instar period. Before emergence, most of the larvae had typically gone through 15 to 17 instars. The highest rate of pupae formation, 28.32%, was observed in the $17^{th}$ instars. The body length gradually increased with each successive instar, reaching its maximum at the 17th instar. Beyond the $17^{th}$ instar, however, the body length decreased. The larvae were white in the first instar, and gradually turned brown after the $2^{nd}$ instar.

신돌연변이 불면잠 nm-f의 유전형질 (Phenotypic Characteristics of New Mutant, Non-molting f(nm-f) of Bombyx mori)

  • Studies were carried out to investigate the phenotypic characteristics of the non-molting mutant (nm-f) which was mapped on the 2nd linkage group. The results obtained were as follows : The nm-f mutant was distinguishable in the 3rd day of hatching. About 80 percentage of the non-molting mutant larvae died at the first instar within 10 days of hatching. The remaining larvae survived to the 2nd ad the 3rd instar but did not live to the final instar. There was no difference in non-molting nutant manifestation between hibernating and artificial hatching eggs. As a result of hemolymph protein analysis, the protein content on nm-f mutant was less than the normal larvae's. Therefore, we conclude that the characterization of nm-f is similar to the already known strains of non-molting mutant and the shortage of hemolymph protein is closely related to the non-molting characteristic in nm-f.

Molecular Analysis of Anisakis Type I Larvae in Marine Fish from Three Different Sea Areas in Korea

  • Anisakiasis, a human infection of Anisakis L3 larvae, is one of the common foodborne parasitic diseases in Korea. Studies on the identification of anisakid larvae have been performed in the country, but most of them have been focused on morphological identification of the larvae. In this study, we analyzed the molecular characteristics of 174 Anisakis type I larvae collected from 10 species of fish caught in 3 different sea areas in Korea. PCR-RFLP and sequence analyses of rDNA ITS and mtDNA cox1 revealed that the larvae showed interesting distribution patterns depending on fish species and geographical locations. Anisakis pegreffii was predominant in fish from the Yellow Sea and the South Sea. Meanwhile, both A. pegreffii and A. simplex sensu stricto (A. simplex s.str.) larvae were identified in fish from the East Sea, depending on fish species infected. These results suggested that A. pegreffii was primarily distributed in a diverse species of fish in 3 sea areas around Korea, but A. simplex s.str. was dominantly identified in Oncorhynchus spp. in the East Sea.
