Filtration Rates of Juvenile Purple Clam, Saxidomus purpuratus (Sowerby) Feeding on Red Tide Dinoflagellates

  • The purpose of this study is to compare the clearance rate (CR) and intake rate (IR) of juvenile purple clam, Saxidomus purpuratus when feeding on different unialgal diet of red tide dinoflagellates (RTDs), and to know what is the most important cell characteristic of RTDs to cause the differences in feeding parameters. Experiments were performed to measure the CR and IR of juvenile S. purpuratus as a function of algal concentration when food was either the standard food, Isochrysis galbana or one of 9 RTDs. Patterns of CR with increasing algal concentration were similar among different RTDs. The highest $C_{max}$ was observed when S. purpuratus was feeding on A. affine, while the lowest on C. polykrikoides. The patterns of IR with increasing algal concentration were also similar among different RTDs. However, there were great differences in the maximum value of IR ($I_{max}$) among different RTDs. The highest $I_{max}$ was observed when S. purpuratus was feeding on A. carterae, while the lowest on G. catenatum. Some RTDs similar in size showed different $C_{max}$. Other RTDs different in size showed similar $I_{max}$. Life form of each RTD affected significantly the $I_{max}$, which was higher for single-celled RTDs than chain-forming RTDs. There were no significant differences in feeding parameters between toxic and nontoxic RTDs. Moreover, a toxic dinoflagellate, A. carterae recorded the highest $I_{max}$ among RTDs. The most important characteristic of RTD as a factor affecting the feeding rate of S. purpuratus was life form, not size or toxicity of RTD species.

  • The advancement of industry have increased domestic energy demands and energy facilities such as storage facility, compressed gas pipe, station, and tank lorry. Also, concern about environment have diversified energy source to clean energy such as LNG. In these major energy facilities, major accident can happen to result in fire, explosion, toxic release and etc. In addition, it may cause chain accidents to the adjacent energy facilities. In this research, safety assessment was performed through the consequence analysis of LPG liquefied petroleum gas) station, gasoline station and LNG(liquiefied natural gas) station. The obtained result will be helpful to make a safety guideline of the LPG/LNG station built adjacent to the gasoline station.

Virulence Factor Profiles of Escherichia coli O157:H7 Bacteriophage Isolates from Sewage and Livestock Stools (하수와 가축분변에서 분리된 대장균 O157:H7 박테리오파지의 병원성인자 프로파일)

  • The aim of study was to investigate the virulence profile of Escherichia coli O157:H7 bacteriophages isolated from sewage and livestock stools. Among 23 E. coli O157:H7 bacteriophages, 14 strains were isolated from sewage and 9 were from animal stools collected from 10 livestock farms in Korea. For each bacteriophage DNA sample, the presence of stx1, stx2, eae, aafII, ial, elt, estI, estII, astA, afa, and cnf was examined by polymerase chain reaction. The detection rate of eae, stx2, estI, astA, and ial was 100%, 69.6%, 13.0%, 13.0%, 8.7%, respectively. While all E. coli O157:H7 bacteriophages isolated from stools carried eae+stx2, stx2+eae, eae+astA, eae, stx2+eae+estI, eae+estI, stx2+eae+ial, and eae+ial were observed in bacteriophages isolated from sewage. As several plasmid-carrying virulence factors (estI, astA, and ial) were found in E. coli O157:H7 bacteriophages obtained from sewage and stools, the microbial safety of bacteriophages should be investigated in further study.

Application of a Kiwifruit (Actinidia chinensis) to Improve the Textural Quality on Beef Bulgogi Treated with Hydrostatic Pressure

  • In order to reduce the increased hardness of beef bulgogi due to hydrostatic pressure (HP), kiwifruit (Actinidia chinensis) was applied. To understand the changes of shear force in beef bulgogi with kiwifruit induced by HP, changes in chemical properties of myofibril (Mf) with 10% kiwifruit induced by HP were investigated. From the SDS-PAGE patterns of Mf with 10% kiwifruit, there was an observed increase in the degradation of myosin heavy chain (MHC) by HP (300-500 MPa) to that by 0.1 MPa. This result indicates that HP may enhance enzyme action from a kiwifruit for the degradation of MHC, and the similar phenomenon occurred in the beef bulgogi with kiwifruit induced by HP. The shear force of beef bulgogi without a kiwifruit induced by 400 and 500 MPa significantly increased compared to that by 0.1 MPa (p<0.05). However, in the beef bulgogi with 10% or 20% kiwifruit, the shear force induced by 400 or 500 MPa was similar or slightly lower than that by 0.1 MPa. Consequently, adding kiwifruit to bulgogi could reduce the hardness of HP-induced beef bulgogi due to the enzyme action in the kiwifruit accelerated by HP.

A Lung Granuloma Case Possibly Associated with a Working Environment: A Case Report

  • Lung granulomas are uncommon in Thailand. The disease typically develops from an occupational environment and is mostly caused by infection. Herein is a case report of a female patient, aged 48, working as a nurse in an Accident and Emergency Department at a hospital. Eighteen years prior to admission the patient was diagnosed with myasthenia gravis and pulmonary tuberculosis. The chest X-ray and CT scans showed a solitary pulmonary nodule in the lower left lung. The patient received an open thoracotomy with a left lobectomy. Granulomatous and nonseptate hyphae were found in the pathology diagnosis. The patient was thus diagnosed as having a lung granuloma. The galactomannan antigen test was positive. The solitary pulmonary nodule-found from the use of a Polymerase Chain Reaction (PCR) test-was an Aspergillus spp. The fungus culture was collected from air samples. The air samples were collected by the impaction technique using a microbial air sampler. Three types of Aspergillus spp. were found as well as Penicillium spp. and Monilia sitophila. The Aspergillus spp. was a match for the patient's disease. The patient was diagnosed as having a lung granuloma possibly Aspergillus nodule which was caused by airborne Aspergillus spp. from the occupational environment.

Development of a ladder-shape melting temperature isothermal amplification (LMTIA) assay for detection of African swine fever virus (ASFV)

  • Background: Due to the unavailability of an effective vaccine or antiviral drug against the African swine fever virus (ASFV), rapid diagnosis methods are needed to prevent highly contagious African swine fever. Objectives: The objective of this study was to establish the ladder-shape melting temperature isothermal amplification (LMTIA) assay for the detection of ASFV. Methods: LMTIA primers were designed with the p72 gene of ASFV as the target, and plasmid pUC57 was used to clone the gene. The LMTIA reaction system was optimized with the plasmid as the positive control, and the performance of the LMTIA assay was compared with that of the commercial real-time polymerase chain reaction (PCR) kit in terms of sensitivity and detection rate using 200 serum samples. Results: Our results showed that the LMTIA assay could detect the 104 dilution of DNA extracted from the positive reference serum sample, which was the same as that of the commercial real-time PCR kit. The coincidence rate between the two assays was 100%. Conclusions: The LMTIA assay had high sensitivity, good detection, and simple operation. Thus, it is suitable for facilitating preliminary and cost-effective surveillance for the prevention and control of ASFV.

  • Purpose: Based on the preceding studies in supply chain management, factors were analyzed to verify the effect of risk assessment and risk management factors of the business continuity management system (BCMS) on management performance. The purpose of this study is to establish a systematic risk management plan by deriving the risk factors of BCMS and evaluating unpredictable risks, and at the same time, contributing to a company's competitive advantage without interruption of work. Method: The structural relationship between risk assessment, risk management and management performance of BCMS was derived. To this end, a questionnaire survey was conducted of 124 managers and managers in Korean companies. Frequency analysis, validity analysis, reliability analysis, correlation analysis, and simple regression analysis were performed. Result: First, risk assessment had a positive (+) effect on risk management. Second, risk management had a positive (+) effect on management performance. Finally, risk assessment had a positive (+) effect on management performance. Conclusion: BCMS's risk assessment and risk management capabilities should be managed through financial performance, and risk management activities should be managed through non-financial performance.

Detection of foot-and-mouth disease virus and coxsakievirus in the soil and leachate of modeled carcass burial site (시험 가축 매몰지 토양 및 침출수 내에서의 구제역 바이러스 검출)

  • Foot and mouth disease (FMD) is highly infectious disease of cloven-hoofed animals, particularly cattle, sheep, pigs and goats. Last outbreak reported in November, 2010 induced the enormous social and economical impacts. Culling of infected animals, movement control, and vaccination are the major control measures of FMD. The aim of this study was to detection foot-and-mouth disease virus (FMDV) in the soil and leachate from modeling burial for pig carcass as measured by real-time reverse transcriptase polymerase chain reaction (RT-PCR). FMDV and Coxsakievirus B1 (CVB1) were detected in soil by week 16 and Coxsakievirus B1 (CVB1) by weeks 12, respectively. FMDV and CVB1 also detected by weeks 8 in the leachate. Results from this study provides an evidence that FMDV could be inactivated for safe of pig carcasses infected with FMDV within 4 month in the carcass burial site.

  • Nuclear power plants have a lower cost of power generation, and they are more eco-friendly than other power generation plants. Also, we need to prepare nuclear plant accidents because of their severe damage. In the event of a safety accident, such as a radiation leak, by applying a wireless sensor network to a nuclear power plant, many sensor nodes can be used to monitor radiation and transmit information to an external base station to appropriately respond to the accident. However, applying a wireless sensor network to nuclear power plants requires routing protocols that consider the sensor network size and bypass obstacles such as plant buildings. In general, the hierarchical-based routing protocols are efficient in energy consumption. In this study, we look into the problems that may occur if hierarchical-based routing protocols are applied to nuclear power plants and propose improved routing protocols to solve these problems. Simulation results show that the proposed routing protocol is more effective in energy consumption than the existing LEACH protocol.

  • Globalization of production, changing technology in ocean shipping sector, shifting bargaining power from port service provider to port users, changing distribution patterns of the containerization and new implementation for the environment and safety concerns introduced new ways of administration in port sector. In these days, port industries cannot survive alone. Port operation and management has to be proactive to cooperate and integrate with other port operators or other industries. In the 21st century, cooperation in maritime sector is more common than that in the past with several different reasons in a mixture of forms of cooperation. Cooperation among port authorities, port operators, shipping line, inland transport service providers and/or related organizations themselves becomes to exist to overcome the challenges and development of the global transport chain. This paper investigates the concept of port cooperation and the case analysis of the cooperation in the port industry.
