Skin ageing is associated with compromised performance of its fundamental barrier functions, with undesirable changes in appearance. Since this may introduce a detrimental impact on the quality of life, significant effort to discover effective ingredients against ageing is being invested. Recently, collagen hydrolysates containing tripeptides such as GlyPro-Hyp (GPH) have been developed with anticipation of improved effects compared to that of existing collagen hydrolysate-products. To evaluate the cutaneous hydration effect of collagen tripeptides (CTP), meaningful biomarkers in human dermal fibroblasts (HDF) and NC/Nga Tnd mice were analyzed in this study. Increased levels of ceramide kinase, hyaluronic acid, collagen 1A, and hyaluronan synthase-2 (HAS2), and decreased levels of hyaluronidase-1 (HYAL1) and CD44 in HDF cells were demonstrated. Furthermore, significant reduction of transepidermal water loss (TEWL), scratching behavior, HYAL1, $TNF-{\alpha}$ and IL-6 and increased water content and HAS2 were verified by in vivo tests. These results strongly suggest the potential of CTP as a skin hydration agent.
Ceramide, cholesterol and free fatty acids are the major intercellular lipids, maintaining the integrity of the skin barrier. However, the roles of these lipids in canine skin barrier function are little known. The aim of this study was to evaluate the repairing effects of 2% ceramide (CER), 2% cholesterol (CHO), 2% linoleic acid (LIN) and 2% intercellular lipid mixture (ILM) on damaged canine skin barrier by 1.25% sodium lauryl sulphate (SLS). Transepidermal water loss (TEWL), skin hydration, skin pH and skin thickness were assessed. Histological profiles and transmission electron microscopic (TEM) profiles were assessed on day 12. SLS effectively induced the canine skin barrier damage. TEWL was significantly decreased by topical application of CER and ILM in SLS and vehicle-treated skin on day 8 and 12, respectively (p < 0.05, p < 0.0 I). By end of the experiment all lipids significantly decreased the TEWL as compared with SLS and vehicle control, but CER and ILM more significantly decreased the TEWL than UN and CHO, respectively (p < 0.01). Skin hydration was significantly increased by CER and ILM during experimental periods (p < 0.01). Skin pH was significantly decreased by CER, LIN and ILM. In histological profiles, the thickness of the stratum corneum (SC) was significantly increased by the SC lipids as compared with vehicle and SLS (p < 0.01). Especially, CER and ILM showed more prominent improvement of barrier recovery. In TEM of the SC, SLS induced exfoliations of corneodesmosomes in the SC, and CER and ILM effectively protected exfoliations of corneodesmosomes on SLS-damaged canine skin. These results indicated that topical application of CER and ILM dramatically improved damaged-skin barrier function by SLS. Also, it was considered that the use of CER or ILM was recommended for the management of skin barrier dysfunction by irritant and inflammatory skin disorders such as atopic dermatitis.

This study was conducted to discover substances that regulate skin surface acidification using human epidermal keratinocyte cell lines, and to investigate their effects on the moisturizing ability and skin barrier function of the stratum corneum. Prunella vulgaris (P. vulgaris) is an herb widely distributed in Northwest Africa and North America that has been studied for its anti-apoptotic, antioxidant, and anti-inflammatory effects. However, research on the regulation of NHE1 expression and the restoration of skin barrier function has not been conducted. Analysis of P. vulgaris revealed the presence of rosmarinic acid and caffeic acid as active ingredients, which were tested for toxicity in human epidermal keratinocyte cell lines (HaCaT), and showed no toxic effects were observed at high concentarion (100 ㎍/mL or 100 µM). It is known that sodium-hydrogen ion exchange pumps (NHE1) decrease in expression in aging skin to maintain the acidic pH of the stratum corneum, and it is hypothesized that this decrease plays an important role in the impaired restoration of skin barrier function in aging skin. P. vulgaris extract and caffeic acid increased the expression of NHE1 in keratinocytes, increased the expression of natural moisturizing factor (NMF) precursor filaggrin and ceramide synthesis enzyme serine palmitoyl transferase (SPT). In addition, P. vulgaris and caffeic acid decreased the extracellular pH of keratinocytes, indicating a direct effect on skin pH regulation. Taken together, these results suggest that P. vulgaris and caffeic acid can regulate skin pH through NHE1 modulation, and may help to restore skin barrier function by increasing NMF and ceramide synthesis. These results show the possibility that honeysuckle and caffeic acid can have a positive effect on skin health, and can be the basis for the development of new skin protection products using them.
Fumonisins are a family of mycotoxins produced by the fungus Fusarium moniliforme which is a common contaminant in corn. Fumonisins are potent inhibitors of sphingosine and sphinganine N-acyltransferase (ceramide synthase), key enzymes in sphingolipid metabolism. The purpose of this study was to provide the evidence that the elevated levels of free sphingoid bases (primarily sphinganine) and depletion of complex sphingolipids were closely related to the inhibition of cell growth in LLC-$PK_1$ cells exposed to fumonisin $B_1$$(\leq 35 {\mu}M)$. Concentrations of fumonisin $B_1$ between 10 and $35 {\mu}M$ were known to inhibit cell growth without cytotoxicity in $LLC-PK_1$ cells (Yoo et al. Toxicol. Appl. Pharmacol. 114, 9-15, 1992). Cells exposed to 35$\mu M$ fumonisin B$_1$ for 48 and 72 hr developed a fibroblast-like (elongated and spindle-shaped) appearance and were less confluent than normal cells. At between 24 and 48 hr after exposure to fumonisin $B_1$ cells were beginning to show the inhibition of cell growth and at 72 hr the number of viable cells in fumonisin-treated cultures was about 50% of concurrent control cultures. During the 24 hr lag period preceding inhibition of cell growth, the free sphinganine levels in cells exposed to $35 {\mu}M$ fumonisin $B_1$ were highly elevated (approximately 230 fold higher than normal cells). The elevated levels of free sphinganine were $435\pm14$$pmoles/{10^6}$ cells at 48 hr and approximately TEX>$333\pm11$$pmoles/{10^6}$ cells in cells exposed to $35{\mu}M$ fumonisin$B_1$ at 72 hr, while the levels of free sphinganine in normal cells were less than 2$pmoles/{10^6}$ cells. Under the same condition, depletion of intracellular complex sphingolipids as a consequence of fumonisin inhibition of de novo sphingolipid biosynthesis and turnover pathway was appeared. Content of free sphingold bases in dividing cells was more elevated than in confluent cells at 24-48 hr after cells were exposed to $20{\mu}M$ fumonisin $B_1$. The dividing cells were showing the inhibition of cell growth at 48-72 hr and $20{\mu}M$ fumonisin $B_1$. The results of this study support the hypothesis that the inhibition of cell growth is very well related to the disruption of sphingolipid metabolism in $LLC-PK_1$ cells.

Gromwell (LE, Lithospermum erythrorhizon), a perennial herbal plant, has been used for the treatment of various problems associated with atopic dermatitis of the skin, such as water loss, epidermal hyperproliferation, and severe inflammation. Previously, it was shown that oral supplementation with a 70% ethanol extract of gromwell prevented the development of atopic dermatitis in NC/Nga mice. In this study, in order to identify the fraction that mediates gromwell's efficacy, the dietary effects of water and ethyl acetate fractions from the ethanol extract of gromwell were assessed in the development of dermatitis using NC/Nga mice. Dietary supplementation of the hot water fraction significantly reduced scores for epidermal hyperproliferation in parallel with a marked increase of ceramides. Supplementation of the gromwell hot water fraction also decreased scratching behavior, which was accompanied by a decrease in plasma levels of IgE. These results showed that the hot water fraction of the gromwell ethanolic extract prevented the development of atopic dermatitis by increasing ceramides in NC/Nga mice.
Physiological lipid mixtures comprised of cholesterol, ceramide and free fatty acid better maintain epidermal homeostasis and have been recently used for dermatoses induced by skin barrier damage, for example for atopic dermatitis and xerotic skin. Itching and dry atopic dermatitis of the skin may be related to altered skin barrier function. In a previous study, the use of multi-lamellar emulsion (MLE), which is a lipid mixtures containing cholesterol, pseudoceramide and free fatty acid, has been shown to accelerate the recovery of the epidermal permeability barrier. In this study, we assessed the efficacy of MLE compared with a currently used anti-itch moisturizer (AIM), the active ingredients of which are menthol and camphor, on barrier recovery after barrier disruption. To clarify the effect of MLE and AIM after acute barrier perturbation, we measured the relation between transepidermal water loss (TEWL) and the barrier recovery rate at 3, 6, 24, and 48 hours after tape stripping hairless mice and then observed changes in the stratum corneum (SC), including the intercellular lipid structure and secretion of lamellar bodies, by electron microscopy. MLE treated skin recover skin barrier function more rapidly, and AIM treated skin delayed barrier repair. Morphological changes in the epidermis, of MLE treated skin revealed well-conserved lipid multi-lamellar structures at 24 h after tape stripping, whereas AIM treated skin showed altered lamellar bilayers within the SC interstices at 48 h. In addition, MLE treated skin showed an increase in the number of LBs and in their secretions and a decrease in the number of SC layers versus AIM treated skin. These results suggest that MLE may accelerate the production of an epidermal permeability barrier in hairless mice by increasing the number and secretion of LB and improve the dryness and itch associated with an altered epidermal permeability barrier.

The barrier structure of the skin's epidermis is a key structure to prevent the loss of water inside the body and the invasion of foreign substances, and is composed of keratinocytes and intercellular lipids. At this time, the intercellular lipids of the skin barrier has the strongest structure when packed in an orthorhombic structure. However, it is damaged by various external causes and changes to a hexagonal structure. This change in physical structure can be analyzed non-invasively by analyzing the signal of the CH2-CH2 scissoring band of lipids using FT-IR. In this study, SDS was treated on porcine skin to construct a skin barrier damage model, and the degree of change in packing structure was quantified by analyzing FT-IR signals. We then judged whether the barrier of the damage model was recovered according to the treatment of the cosmetic formulation. From these results, an indirect method of measuring the water evaporation of the skin barrier to date can be supplemented. In addition, physical changes in the structure of the skin barrier can be utilized in a direct and efficient manner to identify the function and verify the formulation of various materials.
Fumonisins are specific inhibitors of ceramide synthase in sphingolipid metabolism. An alteration in sphingolipid metabolism as a result of fumonisin exposure is related to cell death (Yoo et al., 1992). The objective of this study was to investigate whether elevated free sphinganine levels are related to the sensitivity of cultured cells to fumonisin exposure. Fumonisin $B_1$ elevated the intracellular free sphinganine concentraions in both LLC-$PK_1$ and Chinese hamster ovary (CHO) cells. However, CHO cells are resistant to fumonisin cytotoxicity at 50${u}m$, while LLC-$PK_1$ cells are sensitive at concentrations greater than 357M. The intracellular concentration of free sphinganine in LLC-$PK_1$ cells treated at 50${u}m$ fumonisin $B_1$ for 72 h was approximately 1450 pmol/mg protein relative to the 37 pmol observed in the control culture. Under the same conditions, the population of apoptotic cells in the 50${u}m$ fumonisin $B_1$-treated culture was approximately 37% of the total compared to 12% in the control. The caspase III-like activity after 72 h in the 50${\mu}$M fumonisin $B_1$-exposed culture Increased to approximately 50 $pmol/mg$ protein/hr compared to 6 $pmol/mg$ protein/hr in the control. L-cycloserine, a serine palmitoyltransferase inhibitory reduced the fumonisin $B_1$-stimulated caspase III-like activity down to the control level. Under the same culture conditions, the intracellular concentration of free sphinganine after-cycloserine plus fumonisin $B_1$ treatment was 140 pmol/mg protein compared to 1450 $pmol/mg$ protein in fumonisin $B_1$ alone. The intracellular concentration of free sphinganine in CHO cells treated with 50${u}m$ fumonisin $B_1$ for 72 h was al)proximately 460 pmol/mg protein, indicating that the mass amount of elevated free sphinganine in the CHO cells was about 32% of that in LLC-$PK_1$ cells. Adding exogenous sphinganine to the CHO cells along with 50${u}m$ fumonisin $B_1$ treatment for 72 h caused both necrosis and apoptosis. In conclusion, the elevated endogenous sphinganine acts as a contributing factor to the fumonisin-induced cell death.

The structure and physiological function of human skin continuously weaken due to growing older. The reasons of aging from external conditions are long term exposure to sun, wind, heat, cigarette smoke, and etc. This also palmitoyl oligopeptide or ceramide oligopeptide are known asc ingredient stimulating collagens and have the effect of reproducing the upper level of skin. Acetyl hexapeptide is an ingredient that makes the skin and muscle suppler and reduces wrinkles. It is a major high function beauty ingredient that substitutes botox. After dividing 7%, 14%, and 20% Acetyl hexapeptide experimental groups as groups A, B, and C the control group and experiment groups' change of wrinkles, hair follicles, moisture content, and dead skin cells was analyzed. According to the results, Acetyl hexapeptide seems to affect wrinkles, hair follicles and moisture content contrasting to the control group. Experimental groups and control group showed similar change in dead skin cells. In contrast to the control group satisfaction of examines was affected in wrinkles, hair follicles and moisture but removing dead skin cells had similar result in experimental groups and control group.
