Immunomodulatory Effects of Aqueous-extracted Zingiberis rhizoma on Methorexate Induced Immune Suppression in Mouse Spleen Cell (건강 열수추출액이 Methotrexate에 의해 유도된 마우스 면역억제 조절에 미치는 영향)

  • Zingiberis rhizoma(ZB) has been used to treat a various condition and disease in traditional oriental medicine. The present study was conducted to evaluate the immunomodulatory effect of aqueous-extracted ZB(ZBE) on methotrexate (MTX)-induced immune suppression in mouse spleen cells. In spleen cell proliferation assay, ZBE enhanced mitogenic activity in mouse spleen cells. In RT-PCR, ZBE induced IL-2, IFNr and IL-6 cytokine gene expression in mouse spleen cells. In spite of MTX treatment, IL-2, IFNr and IL-6 gene expressions sustained in MTX treated spleen cells. CD45R/B220, pan B marker was slightly increased in ZBE treated mouse spleen cells. IL-6, B cell tropical cytokine, production was induced by ZBE-treated mouse spleen cells and IL-6 production was sustained on MTX-ZBE co-cultured cells. ZBE administration enhanced suNival of S-180 bearing mouse. These data indicate that ZBE has a protective effect of immune suppression caused by MTX, and ZBE may be enhance cellular and humoral function by regulate cytokine gene expression as well as the mitogenic effect on spleen cells.

Immunosuppressive effects of a Korean isolate of reticuloendotheliosis virus (국내 분리 세망내피증 바이러스의 면역억제능)

  • Humoral and cellular immune responses are depressed in chickens infected with reticuloendotheliosis virus(REV). The extent of depression is influenced by the age of infection and strain of virus. This study was conducted for investigation of immunosuppressive effects of a Korean isolate of REV. Chickens infected with REV-HI, a Korean isolate, at 1 day old were severely suppressed in the vaccinal immunity against Newcastle disease, infectious bronchitis and infectious bursal disease. But these immunosuppressive effects were not observed in chickens infected with the virus at 2 weeks of age, or contact infected by growing in-contact with inoculated chickens from one day old. The clinical signs following infectious laryngotracheitis(ILT) vaccination in chickens infected with REV-HI at 1 day old were more severe than those of uninfected chickens, and some of REV-infected chickens(21.4%) were died after the vaccination. Mortality following virulent ILT virus infection was increased in REV-HI infected chickens. Effects of REV infection at one day old to susceptibilities to subsequent Chicken anemia agent (CAA) infection were also studied. Chickens were infected with REV-HI at 1 day old and subsequently inoculated CAA at 1, 7, 14 and 28 days old, respectively. Mortalities of the chickens infected with REV-HI and subsequent CAA infection were 100, 100, 40 and 0%, respectively, whereas 23, 8, 0 and 0% of chickens infected with only CAA were died, respectively. These above all results suggest that a Korean isolate of REV may be highly immunosuppressive.

Enhanced expression of the structural protein of porcine reproductive and respiratory syndrome virus (PRRSV) by SUMO fusion

  • The major structural proteins of porcine reproductive and respiratory syndrome virus (PRRSV) are derived from ORFs 4, 5, and 6. They have been considered very important to arouse the humoral and cellular immune responses against PRRSV infection and proposed to be the excellent candidate proteins in the design of PRRS bioengineering vaccine. However, the PRRSV structural proteins are produced in low levels in the infected cells because it forms insoluble protein and possesses several transmembrane regions. To overcome this problem, we fused the ORF4, ORF5, and ORF6 with SUMO (small ubiquitin-related modifier). The resulting fusion protein SUMO-ORF4, -ORF5, and -ORF6 were highly expressed in Bm5 cells. The level of protein expression using the Bombyx mori larvae was higher than that using Bm5 cells. In addition, fusion to SUMOstar, which is not processed by native SUMO proteases, significantly enhanced protein expression levels compared to SUMO fusion. This study demonstrated that SUMO or SUMOstar, when fused with PRRSV structural proteins, was able to promote its soluble expression. This may be a better method to produce PRRSV structural proteins for vaccine development.

  • Fruit fly, Drosophila melanogaster has developed efficient immune mechanisms to prevent microbial infection, which are consisted of cellular and humoral responses. During the systemic or local infection, two distinct pathways (Toll and Imd) play major roles in antimicrobial peptide synthesis. The Toll pathway is required to defend Gram-positive bacterial and fungal infections, whereas the Imd pathway is important in Gram-negative bacterial infection. We have shown that the infection of the opportunistic Gram-negative bacterium, Pseudomonas aeruginosa strain PA14 (PA14) into fly dorsal thorax can kill the flies within 48 h ($100\%$ mortality) in our optimized infection condition, suggesting that the PA14 strain can cause disease progress in fly model system. We found that flies carrying a constitutively activated mutant form of the Toll receptor $(Tl^{10b})$ showed increased resistance to P. aeruginosa infection and that flies carrying mutations in the Toll signaling pathway as well as in the Imd signaling pathway was more susceptible to PA14 infection. All these results imply that the Toll pathway might be important in the resistance to this pathogenic Gram-negative bacterial infection.

  • In an attempt to investigate the current of clinical researches on spleen yang or vital energy deficiency syndrome, the results were as follows. 1. It is possible to occure spleen deficiency syndrome which come from genetic factor. 2. The absorption disturbance in spleen deficiency syndrome can be likely caused by gastrointestinal mucosa injury, disorder of vagus nerve funtion and impairment of excretion of exocrine gland in pancreas. 3. Owing to the failure of tansporting and converting funtion of spleen, minerals, hematogenic substance and nutritional substance are scanty and then imbalanced metabolism state which heat production is decreasing is appeared. 4. By the failure of vital energy and blood growth, decreasement of $O_2$ transportation ability of RBC, disoder of blood coagulation, immune system disturbance which humoral immunity is enhanced and cellular immunity is decreased, are noted. 5. While there is not still an attemt to study the spleen deficiency sydrome in muscle disease or disease of four extremities, but it is likely suggested that spleen-stomach supplyment thereapy is very excellent effect on muscle disease and disease of four extremities.

  • One of the practical limitations with the use of liposomes for delivery of the pharmaceutical substances such antigens is that liposomes are relatively unstable in storage. In order to extend the stability of liposome in storage without affecting their functional activity, solution-type liposomes were dehydrated to form a structurally intact dry liposomes. Comparative immunological evaluation was carried out for both dry and solution-type liposomes containing gag-V3 chimera, consequently it was found that dry liposomes elicited both humoral and cellular response as efficiently as solution-type liposemes did against the same gag-V3 antigen. Especially, long-term stability of the liposomes was remarkably enhanced by the dehydration made to loposomes without a significant change in its ability to elicit immune response in vivo. These results indicate that dry pH-sensitive liposome may become an effective delivery and adjuvant system for general vaccine development.

  • Oral administration of soluble antigen can induce peripheral tolerance to the antigen. This study was conducted to evaluate whether gamma-irradiated ovalbumin (OVA) can induce oral tolerance. To investigate this, we administrated intact or irradiated OVA to mice, induced allergic response using intact OVA and alum, then compared humoral and cellular immune responses. Mice treated with gammairradiated OVA had less OVA-specific IgE compared with those who were administered intact OVA. There was no difference in levels of OVA-specific IgG+A+M, IgG1, and IgG2a. Splenocytes of mice administered irradiated OVA showed similar OVA-specific T cell proliferation and secretion of IFN-γ and IL-4. However, there was an increase in IL-2 and a decrease of IL-6 secretion in mice treated with irradiated OVA. These results indicate that gamma-irradiated OVA have similar effects to intact OVA on antigen tolerance.

  • Scabies is a highly contagious skin infestation caused by the mite, Sarcoptes scabiei var. hominis. Complex responses to scabies mites in the innate, humoral, and cellular immune systems can cause skin inflammation and pruritus. Diagnosis can be challenging because scabies resembles other common skin conditions. We report the first Korean case of scabies in a hematopoietic cell transplant (HCT) recipient, initially suspected of skin graft versus host disease (GVHD). A T-cell acute lymphocytic leukemia patient underwent a sibling-matched allogeneic HCT and developed pruritus after cell engraftment. Treatment for GVHD did not improve the symptoms. He was diagnosed with scabies 30 days after the onset of symptoms.

  • Cowden's syndrome is a harmatomatous polyposis syndrome with characteristic mucocutaneous lesions and among the spectra of clinical disorders that has been attributed to germline mutations in the PTEN gene. Although Cowden's syndrome has rarely been reported, immunologic studies have revealed that patients with this syndrome have humoral and/or cellular immune abnormalities. We recently identified a 21-year-old woman with Cowden's syndrome who was diagnosed with candida esophagitis without a history of diabetes, carcinoma, or steroid therapy. We report the immunologic status of this patient and the relationship with candida esophagitis on the basis of a literature review.

  • This experiment shows cellular and humoral immune responses induced by soluble egg antigen of Schistosoma manscni, that is, change of the number of peripheral blood eosinophil, delayed hypersensitivity measured by the degree of ear swelling, granulomatous change of liver tissue and elevation of serum antibody titer by ELISA. SEA was given continuously by the insertion of a minipump into peritoneal cavity of mouse. In control group, same pump with HGG was inserted. New pump was eachansed once In two weeks and followed the result until 9 weeks after mini-pump insertion. 1. Highest peripheral blood eosinophil level was recorded at 2∼3 weeks after SEA punp insertion, 2. MaRirnum ear swelling was observed at 2 weeks arid then decreased gradually. 3. In liver tissue, several granulomas without egg were formed at 4 weeks. 4. Serum antibody titer was eleyated from 4 weeks after SEA pump insertion.
