To investigate the function of N-glycosylation of Cel5A (endoglucanase II) from Hypocrea jecorina, two N-glycosylation site deletion Cel5A mutants (rN124D and rN124H) were expressed in Saccharomyces cerevisiae. The weights of these recombinant mutants were 54 kDa, which were lower than that of rCel5A. This result was expected to be attributed to deglycosylation. The enzyme activity of rN124H was greatly reduced to 60.6% compared with rCel5A, whereas rN124D showed slightly lower activity (10%) than that of rCel5A. rN124D and rN124H showed different thermal stabilities compared with the glycosylated rCel5A, especially at lower pH value. Thermal stabilities were reduced and improved for rN124D and rN124H, respectively. Circular dichroism spectroscopy showed that the modification of secondary structure by mutation may be the reason for the change in enzymatic activity and thermal stability.
Objective: This study was conducted to evaluate the effects of tannins and cellulase on growth performance, nutrient digestibility, blood profiles, intestinal morphology, and carcass characteristics in Hu sheep. Methods: A total of 48 three-month-old meat Hu sheep ($25.05{\pm}0.9kg$) were blocked based on body weight, and randomly allotted to 4 treatments with 3 replicates of 4 sheep each. The experiment lasted for 80 d, and dietary treatments were as follows: i) CON, control diet; ii) TAN, CON+0.1% tannins; iii) CEL, CON+0.1% cellulase; iv) TAN+CEL, CON+0.1% tannins and 0.1% cellulase. Results: Compared with CON, CEL, and TAN+CEL had greater (p<0.05) final body weight (FBW) and average daily gain but lower (p<0.05) feed conversion ratio, while FBW of TAN+CEL was lower (p<0.05) than that of CEL. The apparent total tract digestibility (ATTD) of dry matter in TAN, CEL, and TAN+CEL groups were higher (p<0.05) than that in CON. CEL and TAN+CEL groups had greater (p<0.05) ATTD of crude fiber compared with TAN and CON, while TAN group had lower (p<0.05) ATTD of crude protein than other treatments. TAN, CEL, and TAN+CEL groups increased (p<0.05) serum globulin and alkaline phosphatase but decreased (p<0.05) albumin/globulin. Serum total protein was greatest for TAN+CEL, intermediate for TAN and CEL and least for CON (p<0.05). TAN+CEL group increased (p<0.05) dressing percentage compared with CON, while the backfat thickness of CEL was lower (p<0.05) than that of CON. The villus height of jejunum and ileum in CEL and TAN+CEL groups were greater (p<0.05) than that in CON, and the crypt depth and villus height: crypt depth of jejunum were increased (p<0.05) in TAN, CEL, and TAN+CEL groups. Conclusion: The addition of tannins and cellulase together promoted nutrient digestion, liver protein synthesis and intestinal development and thus improved growth performance and carcass characteristics.

The emergence of new technologies and their implementation by different manufacturers of electronic devices are experiencing an ascending trend. Most of the time, these protocols are expected to reach a certain degree of maturity, and electronic equipment manufacturers use simplified communication standards and interfaces that have already reached maturity in terms of their development such as ModBUS, KNX or CAN. This paper proposes an IoT solution of the Smart Home type based on an Analysis and Prediction System. A data acquisition component was implemented and there was defined an algorithm for the analysis and prediction of actions based on the values collected from the data update component and the data logger records.
대수심 부유 구조물의 하부기초 기술 중 하나인 dynamically penetration anchor (DPA 또는 흔히 torpedo anchor로 칭함)의 거동특성을 시험결과 및 수치 해석적 접근을 통해 분석하였다. 기존의 유한요소 해석기법으로는 이러한 대수심 anchor 구조물의 거동 특성을 적절히 모사하기 어렵기 때문에 본 연구에서는 이러한 부분을 해결하기 위해 Coupled Eulerian-Lagrangian (CEL) 법을 통해 지반-구조물 사이에서 발생하는 메쉬(mesh)의 distortion 현상 및 경계조건 등의 문제점을 대변형의 관점에서 해결하고자 하였다. 실측치와의 비교를 통해, CEL 기법의 타당성을 검증하였고, 그 결과 본 연구에서 적용한 CEL 기법이 기존 유한요소 기술로는 구현이 불가능한 대수심 anchoring system의 자유낙하에 의한 전반적인 거동 및 지반의 변형특성을 적절히 예측함을 알 수 있었다. 또한 검증된 기법을 바탕으로 dynamic anchor의 거동에 영향을 주는 여러 요소들에 대한 매개변수 연구를 추가로 수행하였다.
비 반추동물 특히 돼지의 경우 섬유소 소화율이 극히 낮은 것으로 알려져 있다. 따라서 본 연구는 섬유소분해효소유전자 (CelD) 를 췌장특이 프로모터인 rat Elastase Ⅰ 하류에 연결하여 구축된 벡터를 수정란내 미세주입한 후 생산된 형질전환 돼지를 이용하여 섬유소 소화능력을 측정하기 위하여 실시하였다. 먼저 35두의 수란돈으로 부터 65 두의 산자를 생산하였으며, PCR 분석 결과 4두가 형질전환돼지로 판명되었으며 이중 3두가 죽고, 생존한 l두를 다시 southern blot 분석결과 형질 전환된 것으로 판단하여 같은 복의 6두와 함께 섬유소 소화율을 측정하기 위하여 분리사육을 실시하였다. 최근 기존의 유한요소 해석기법으로는 항타 말뚝 관입과 같은 대변형 문제를 적절히 모사하기 어렵기 때문에 대변형 해석기법을 필요로 하고 있다. 본 연구에서는 대변형 수치해석 기법 중 하나인 Coupled Eulerian-Lagrangian(CEL) 기법을 이용하여 항타 관입의 3차원 대변형 해석을 수행하고자 한다. 현장 시험 결과와 비교를 통해, CEL 기법의 타당성을 검증하였고, 그 결과 본 연구에서 적용한 CEL 기법이 기존 유한요소 해석 기법으로는 구현이 불가능한 항타 말뚝 관입의 전반적인 거동을 합리적으로 모사할 수 있음을 알 수 있었다. 또한, 항타 개단말뚝의 특징인 선단부근에 응력이 집중되는 현상을 적절히 예측함을 알 수 있었다. 이를 통해 CEL 기법을 이용하여 항타 관입 해석이 가능한 것을 확인하였다.
The sequence coding for carboxymethylcellulase (CMCase, CelC) was isolated from the DNA of Salmonella typhimurium URl. Comparison between the deduced amino acid sequence of CelC (368 amino acid residues, Molecular mass 41 kDa) and that of the previously published CMCase revealed that this enzyme belongs to the cellulase family 8 and D. The protein was overproduced in Escherichia coli using T7 expression system, and its activity was confirmed by CMC-SDS-PAGE. When the overexpressed CelC protein was tested on cellulose-type substrates, the recombinant protein is able to degrade cellulose-type substrates, such as CM-cellulose, xylan, avicel, lichenan, and laminarin. Optimal temperature and pH for enzyme activity were found to be 50$^{\circ}C$ and pH 6.5, respectively.
본 연구에서는 토석류의 유하부 도달 속도 및 피해 영향범위 등의 흐름 특성을 파악하기 위하여 대변형 3차원 유한요소 해석을 수행하였다. 대변형 해석은 ABAQUS (Ver 6.13, 2013)의 Coupled Eulerian-Lagrangian (CEL) 기법을 이용하였으며, 실제 토석류 발생 지역의 관측 값과 해석 결과를 비교함으로써 CEL 대변형 해석기법의 타당성을 검증하였다. 그 결과, CEL 대변형 해석기법은 토석류 거동을 합리적으로 예측 할 수 있음을 확인하였다. 또한 토석류의 흐름에 사방댐이 미치는 영향을 확인하기 위하여 추가적인 해석을 수행하여 분석하였다. 그 결과, 사방댐에 의해 유하부로 흘러 내려오는 토석류의 속도와 체적의 감소효과를 확인하였고 토석류에 의해 발생하는 충격력을 산정하였다. 이로 인하여 대변형 토석류 해석기법은 기존 사방댐 안정성 평가 및 사방댐 설계에 활용이 가능할 것으로 판단된다.
This study was performed to test the cellulose digestibility using the transgenic pigs harboring cellulose degradation gene D (CelD). After delivered offsprings between normal pig and transgenic swine, DNA was isolated from piglets tail for PCR analysis. In first generation, five out of 65 piglets showed CelD positive. Unfortunately, four CelD-positive pigs were died during growing, but one survived pig was used as a transgenic founder to produce F$_1$ descendents. Among 3 F$_1$ transgenic pigs produced, one died and the remaining two pigs were used to test the fiber digest efficiency. An assorted feed was composite of 5% fiber with other ingredients. The feed of 3 kg per day was provided to the pigs including transgenic founders and littermate controls. The manure quantity was measured daily for a month, and all manures were dried for three days to analysis nitrogen, phosphate and fiber concentrations. The fiber digestion efficiencies of the transgenic F$_1$ pigs showed approximately 10% higher than those of control pigs. Fiber digestion was not greatly improved in transgenic pigs as it had been expected approximately 30%. Nitrogen concentration of transgenic pig's manure was slowly decreased compare to the control pigs. Because there were only two transgenic pigs tested, a large number of transgenic pigs may be necessary to obtain more reliable data. Breeding of animals to obtain sufficient transgenic pigs subjected for a further study is on progress. Taken together, this study demonstrated successful production of transgenic pigs with increase of cellulose digestibility in the porcine feed.
Clostridium thermocellum이 생성하는 섬유소분해 효소복합체인 cellulosome은 26개 의 서로 다른 단백질로 구성되어 있으며 그 구성물질로서 calcium을 포함하고 있다. 견고한 구조의 이 복합체에서 구성단백질을 분리하여 그 기능을 연구할 목적으로 이 복합체를 해체 (dissociation)하려 시도하였다. 이 복합체는 calcium을 제거하였을 대 해체되었다. 해체된 구성 단백질들은 MonoQ column chromatogrphy에 의해 구조단백질인 CipA를 포함한 분획, 91 kDa(CelK-tr), 60 kDa 과 57 kDa 단백질로 구성된 분획과 주로 46 kDa(CelA-tr), 또는 71 kDa(CelS-tr) 단백질을 포함하는 분획들로 크게 분리되었다. 대부분의 분획들은 crystalline cellulose 분해 활성을 보였다. 순수 분리된 71 kDa 단백질은 $60^{\circ}C$~$70^{\circ}C$에서 섬 유소 분해시 calcium에 의존적이었으나 46 kDa 단백질은 그렇지 않았다. 46 kDa 단백질은 cellodextrin을 celloviose 및 cellotriose 단위로 절단하며 cellotetraose 로부터 glucose가 생 성되는 것이 관찰되었다. Cellulosome의 섬유소분해 최성 산물이 cellobiose인 것을 고려할 때 개개 구성단백질의 활성이 이 효소 복합체내에서 조절되어 있음을 알 수 있다.
