Clinical and inflammatory response to antiviral treatments in dogs with parvoviral enteritis

  • Background: Canine parvoviral enteritis (CPE) is a fatal disease worldwide. The treatment of CPE is based mainly on supportive and symptomatic treatment. Antiviral addition to the treatment may result in a higher survival. Objectives: This study evaluated the effects of antiviral treatments with a standardized treatment (ST) on the clinical and inflammatory response of dogs with naturally occurring CPE. Methods: Twenty-eight dogs with CPE caused by canine parvovirus type 2 were divided randomly into treatment groups. The ST group received fluid, antibiotic, antiemetic, and deworming treatments. The antiviral treatment groups received the same ST with an additional antiviral drug, recombinant feline interferon omega (rFeIFN-ω), oseltamivir (OSEL) or famciclovir (FAM). Results: Compared to the healthy control, the tumor necrosis factor-α, interleukin-1β, interferon (IFN)-α, IFN-γ, haptoglobin, and C-reactive protein values were high (p < 0.05) on day zero. At presentation, mild lymphopenia, neutropenia, and a high neutrophil to lymphocyte (LYM) ratio (NLR) were also observed. Adding rFeIFN-ω to the ST produced the best improvement in the clinical score with a decreased NLR, while leucocytes remained low and inflammatory markers stayed high on day three. The survival rates of the groups were 85.7% in ST+IFN, 71.4% in ST+OSEL, 71.4% in ST+FAM, and 57.1% in ST groups on day seven. Conclusions: Antiviral drugs may be valuable in treating CPE to improve the clinical signs and survival. In addition, the decrease in NLR in favor of LYM may be an indicator of the early prognosis before the improvement of leukocytes, cytokines, and acute phase proteins in CPE.

Animals and diseases prevalence of the rescued dogs in an animal shelter in Incheon (인천지역 유기동물 현황 및 개 질병 조사)

  • Animals and disease frequency of the rescued dogs were investigated in Incheon Veterinary Medical Association Animal Shelter from January in 2012 to December in 2013. Three zoonoses (rabies, brucellosis, and dirofilariosis) and three infectious diseases (canine distemper, canine parvoviral enteritis, and canine influenza) were examined for stray dogs. Among 5,603 heads, 647 (11.5%) went back to their owner and 969 (17.3%) were adopted to new families. Prevalence of dirofilariosis, canine distemper and canine parvoviral enteritis were 2.2% (16/718), 6.0% (24/399) and 6.1% (24/396), respectively. Positive antibody rates against rabies, B. canis and canine influenza virus were 20.5% (41/200), 0.1% (1/718) and 2.0% (4/200), respectively. Protective antibody for canine distemper virus and canine parvovirus were shown in 47.0% (94/200). The data indicate that control measures including facility standards and disease control program are one of the important aspects of the shelter management because stray dogs are exposed to various infectious agents.

Histopathological and immunohistochemical studies on the intestinal lesions related to the infected age in spontaneous canine parvovirus enteritis (개 파보바이러스장염의 감염일령에 따른 병변의 병리조직학적 및 면역조직화학적 관찰)

  • Ninety seven cases of histopathologically diagnosed spontaneous canine parvovirus enteritis(CPE) were studied gross pathologically, histopathologically, immunohistochemically, to investigate histopathological types of small intestinal lesions, and antigen distributions in each pattern related to the infected age. And also, reliability of histopathological method in diagnosis of CPE was inspected with immunohistochemistry. The results were as follows : 1. Age-related occurring ratio in histopathologically diagnosed CPE was 53.6% in 4-8 weeks, 26.8% in 9-15 weeks, 8.25 in 16-19 weeks and 11.3% in 20-45 weeks of the clog age. 2. In histopathologic classification based on patterns of villi/crypts lesions of small intestine(jejunum), the ratio of A type (initial phase of necrosis of crypt epithelia, desquamated epithelial cells in the dilated lumen of the crypt) was 20.6%; the ratio of B type(middle phase of atrophy and fission of the villi, collapse of the mucosa, loss of normal crypt structure) was 62.9%, and C type(regenerative phase of the crypt architecture) was 16.5%. 3. The ratio of A, B, C type in 4-8 weeks old, respectively, was 23.5%, 61.5%, 15.4%; in 9-15 weeks old was 19.2%, 65.4%, 15.3% in 16-19 weeks old was 25.0%, 75.0%, 0.0%; and in 20-45 weeks old was 9.0%, 54.5%, 36.4%. 4. The antigen distribution in the nuclei of the crypt epithelial cells was higher than of the cytoplasm and numerous desquamated epithelial cells in dialated crypts in A type; The antigen cytoplasm and numerous desquamated epithelial cells in dialated crypts in A type; The antigen distribution in the nuclei of the collapsed crypt epithelial cells was not higher than that of the cytoplasm, crypts were lined by and filled with released viral antigens from the destructed epithelial cells in B type; and its distribution was also higher than in the epithelial cells adjacent to the tips of the villi, but it was not reacted in the regenerative crypt epithelial cells in C type. 5. Immunohistochemically detected antigen ratio in the small intestine of histopathologically diagnosed CPE was 94.6%, and this result indicates that histopathological diagnosis is very reliable method in diagnosis of CPE.

Detection of viral infections in wild Korean raccoon dogs (Nyctereutes procyonoides koreensis)

  • Wild raccoon dogs (Nyctereutes procyonoides koreensis) may play a role transmitting several pathogens to humans and pet animals. Information concerning the incidence of rabies, canine distemper virus (CDV), canine parvovirus (CPV), canine adenovirus type 2 (CAdV-2), canine parainfluenza virus type 5 (CPIV-5), and canine herpesvirus (CHV) is needed in wild raccoon dogs. In total, 62 brain samples of raccoon dogs were examined for rabies virus (RABV) and CDV, and 49 lung samples were screened for CDV, CAdV-2, CPIV-5, and CHV. No RABV, CAdV-2, CPIV-5, or CHV was identified, but nine CDV antigens (8.1%, 9/111) were detected. Moreover, 174 serum samples from wild raccoon dogs were screened for antibodies against the five major viral pathogens. The overall sero-surveillance against CDV, CPV, CAdV-2, CPIV-5, and CHV in wild raccoon dogs was 60.3%, 52.9%, 59.8%, 23.6%, and 10.3%, respectively. Comparisons of the sero-surveillance of the five pathogens showed that raccoon dogs of Gyeonggi province have slightly higher sero-positive rates against CDV, CPV, and CHV than those of Gangwon province. These results indicate high incidences of CDV, CPV, and CAdV-2 in wild raccoon dogs of two Korean provinces and a latent risk of pathogen transmission to companion and domestic animals.

Comparison of immune responses of dogs and guinea pigs inoculated with inactivated canine coronavirus vaccines (개 코로나바이러스 불활화 백신에 대한 개와 기니픽 간의 면역반응 비교)

  • Canine coronavirus (CCV) causes a mild gastroenteritis in dogs. The virus is highly contagious. Although the virus was isolated more than thirty years ago, canine coronavirus infection continues to be a widespread problem. Mixed infections with both CCV and canine parvovirus (CPV) are common. Four kinds of commercial killed CCV vaccines are available in Korea. All the commercial vaccines should pass the National Assay for Veterinary Biologicals prior to release. For the potency test of CCV vaccine, it is necessary to use CCV antibody free dogs. The test requires not only kennels but high cost. To develop easy, efficient and economic potency test method for killed CCV vaccine using laboratory animals, a series of experiments with rabbits and guinea pigs were carried out in this study. In the preliminary test, the guinea pigs showed better immune responses than rabbits. The guinea pig was also easy to manage. So guinea pig was selected for the potency test animals. When the guinea pigs were inoculated twice with one dose of vaccine intramuscuarly each, slower and a little lower SN antibody titers were induced in guinea pigs than in dogs (about 2 kg body weight Beagle strain) given the same posology as guinea pigs'. It was concluded that guinea pigs could be substituted for dogs in the potency test of killed CCV vaccine.

Papaya Ringspot Virus Coat Protein Gene for Antigen Presentation in Escherichia coli

  • The coat protein (CP) of Papaya ringspot virus (PRSV) was analyzed for presentation of the antigenic peptide of animal virus, Canine parvovirus (CPV), in Escherichia coli (E. coli). The 45 nucleotides fragment coding for the 15-aa peptide epitope of the CPV-VP2 protein was either inserted into the PRSV-cp gene at the 5', 3' ends, both 5' and 3' ends or substituted into the 3' end of the PRSV cp gene. Each of the chimeric PRSV cp genes was cloned into the pRSET B vector under the control of the T7 promoter and transformed into E. coli. The recombinant coat proteins expressed from different chimeric PRSV-cp genes were purified and intraperitoneally injected into mice. All of the recombinant coat proteins showed strong immunogenicity and stimulate mice immune response. The recombinant coat proteins containing the CPV epitope insertion at the C terminus and at both N and C termini elicited ten times higher specific antisera in immunized mice compared with the other two recombinant coat proteins which contain the CPV epitope insertion at the N terminus and substitution at the C terminus.

Outbreak of Canine Parvoviral Enteritis in Korea (개 파보바이러스성(性) 장염(腸炎)의 국내발생(國內發生))

  • The disease syndrome characterized by the acute vomiting and diarrhea with high mortality had been greatly epidemic in Korea since June, 1931 and it was followed serologically and electron microscopically for the clarification of the agent. The agent present in feces of dogs associated with this syndrome had characteristic feature in agglutinating pig red blood cells that was specifically inhibited by anti-CPV reference dog serum. This also showed the serological identity with the reference CPV antigen in immuno-diffusion. Electron micrograph of the material revealed parvovirus particles with size of 20nm and icosahedral structure. These results clearly indicated that CPV was the primary cause of canine epidemic prevailing in 1981 in Korea.

Effect of chicken egg yolk antibody on canine parvoviral enteritis in pups (개 파보바이러스성 장염에 대한 난황항체의 예방 및 치료 효과)

  • Preventive and therapeutic effects of egg yolk antibody, immunoglobulin Y (IgY), against canine parvovirus (CPV) was evaluated in 25 pups orally challenged with CPV-2a. Oral administration of IgY using powder, paste and coated paste delivery systems was compared. Each type of IgY was administered orally for 17 days from 3 days before challenge. The group of pups administered coated IgY showed mild symptoms such as a moderate decrease in total white blood cell count, no depression, vomiting and diarrhea when compared with other groups. The overall clinical score of the group of pups administered coated IgY was significantly lower than that of the challenge control group. However, mortality did not differ among groups because not all pups received symptomatic treatment. These results implied that oral treatment of coated IgY could improve therapeutic effects against CPV challenge if pups received symptomatic treatment.

Protective effect of chicken egg yolk antibody in colostrum-deprived neonatal puppies (초유결핍 신생자견에서 난황 항체의 방어효과)

  • 총 15두의 초유를 섭취하지 않은 신생자견을 대상으로 난황항체를 경구투여한 후 개 파보바이러스를 경구 접종하여 실험감염을 유발시켜 난황항체의 수동 면역에 의한 예방효과를 알아보고자 한다. 항체역가는 면역화된 산란계로부터 분리한 난황항체를 투여한 자견이 비면역 난황항체를 투여한 자견에 비해 높았다. 개 파보바이러스 접종 직전의 항체역가는 대조군의 경우 1:40에서 1:80, 실험군의 경우는 1:320에서 1:1280이었다. 모든 대조군의 자견들은 바이러스 접종후 4일에 임상증상을 나타내었고 총 7두중 6두가 폐사된 반면 실험군 자견은 2두만이 증상을 나타내었고 폐사 자견은 없었다(p<0.01). 개 파보바이러스를 경구 접종한 후 전체 자견의 혈구응집억제반응역가는 접종후 6일까지 감소하는 경향을 보였다. 접종후 5일의 분변내 혈구 응집반응역가는 실험군 자견의 경우 < 2에서 64였으며 대조자견은 216에서 2048로 높았다.

  • An acute enteric diseases accompanied with vomiting, diarrhea, anorexia, pyrexia, dehydration and high mortality has occured in dogs throughout the country since June, 1981 in Korea. According to epidemiological and clinico-pathological studies on the total of 44 dogs admitted to the animal clinic in the college, the cases were tentatively diagnosed as a viral enteritis and the possibility of canine parvovirus as the etiological agent for the cases was discussed.
