국내 소에서 Neospora caninum의 분리 (In vitro isolation of a bovine Neospora in Korea)

  • The Neospora sp. was isolated from the brain of 1 calf via continuous in vitro cultivation in Vero cell. Neospora tachyzoites were observed 45 days after inoculation of the homogenized brain suspension into the Vero cell. The isolated parasite (named tentatively as NCKB-1) was morphologically and ultrastructurally similar to the previously reported Neospora sp isolated in cattle (BPA-1, JPA-1). A comparison of the antigenic reactivity of cultivated tachyzoites with polyclonal antisera to Neospora caninum and Toxoplasma gondii confirmed that this protozoal isolate was similar to N caninum. This is the first report of successful isolation of Neospora sp from cattle in Korea.

출산 후 새끼와의 분리에 따른 유우의 발성음 특성 (Characteristics of Dairy Cow's Vocalization in Postpartum Related with Calf Isolation)

  • 본 연구는 출산 직후 송아지와의 분리에 따른 어미소의 발성음특징을 파악하고자 출산 후 이틀 이내의 홀스타인 암소 16두를 대상으로 실시하였다. 어미소와 송아지 사이의 후각적, 청각적 접촉이 가능한 경우(실험군 1)와 전혀 접촉을 하지 못하게 격리한 경우(실험군 2)로 나누어 오전 1시부터 4시까지와 오후 1시부터 4시하루 6시간 동안 3일에 걸쳐 디지털 녹음기와 지향성 마이크를 이용하여 발성음을 녹음하여 분석하였다. 어미소의 발성음은 특징에 따라 4가지 유형으로 구분되었다. 발성음의 빈도 및 스펙트로그램과 스펙트럼은 유형별로 차이가 있었으며 발성음의 주파수, 강도 및 길이 역시 차이를 보였다. 또한 날짜의 경과에 따라 어미소의 발성음은 급격하게 감소하였다. 본 연구를 통해 어미소와 송아지의 분리 여부에 따라 어미소의 발성음에는 차이가 있는 것으로 나타났으며, 발성음은 어미소와 송아지 사이 유대관계의 형성에 영향을 미칠 수 있는 매우 중요한 요인으로 판단된다.

소 Coronavirus 분리에 관한 연구 (Isolation of bovine Coronavirus from calves with diarrhea)

  • Fecal samples were collected from 47 calves with diarrhea and 12 clinically normal co-h-abitants, and tested for virus using MDBK cell cultures. Three cytopathic viruses were isolated from 8 fecal samples obtained from diarrheic calves. The isolated viruses were neutralized by bovine coronavirus hyperimmune serum In plaque reduction assay and were detected in the cytoplasm of MDBK cell by bovine coronavirus hyperimmune serum using immunofluorescence staining. The viruses agglutinated mouse erythrocytes only among the various animal erythrocytes tested and new isolates were identified as bovine coronavirus.

Babesia ovata의 순수분리와 형태학적 및 혈청학적 동정 (Pure Isolation and Identification of Babesia ovata by Morphological Characteristics and Complement Fixation Test in Korea)

  • This study was conducted to identify Babesia sp. strain isolated from the imported beef cattle, Aberdeen-angus heifers, in J farm of Jangsu Prefecture, Cheon-buk Province of Korea, morphologically, etiologically and serologically. Babesia sp. strain was purely isolated through the serial blood passages of three generations against splenectomized calves and one generation of blood passage against non-splenectomized calf(intact calf) by inhibiting the appearance of Theileria sergenti in the blood stream by means of injection of 20% oil pamaquine intramuscularly. In the splenectomized calves, the parasite multiplied markedly in blood stream soon after inoculation and parasitaemia was much severe. An elevated body temperature, anorexia, severe anaemia and icterus were observed clinically. Of three splenectomized calves, two were affected with haemoglobinuria and died. But in the non-splenectomized calf the clinical signs and parasitaemia were very mild. The means of the incubation period, the highest parasitaemia, the highest body temperature and the lowest PCV were 3.5 days, 41.1%, ${42^{\circ}C}$ and 9%, respectively, in the splenectomized calves. In calf erythrocytes Babesia sp. protozoa were polymorphic showing the round, oval, ameboid, piriform and paired piriform etc. The sizes of protozoa were $2.51{\sim}3.91{\times}1.32{\sim}2.19{{\mu}m}$ ($3.20{\times}1.60{\mu}m$). Serologically the isolated Babesia sp. were compared with other parasites, B. ovata, B. bigemina, B. bovis, T. sergenti, A. marginale and A. centrale by using the complement fixation(CF) test. As a result, the antibody titer between the homologous species were high. It was two tubes or more in the CF test. From the results obtained above the pure isolated Babesia species was identical with Babesia ovata Minami and Ishihara, 1980.

  • 고등한 진핵생물에서 nucleosome 구성에 중요한 역할을 하는 histone 단백질이 하등한 진핵생물에서도 존재하는지의 여부를 알아보기 위하여 실험을 하였다. 하등한 균류에 속하는 Rhizopus 속의 5종(R. acidus, R. arrhizus, R. formosaensis. R R. japoηicus, R. oryzae)을 재료로 하여 15% polyacrylamide acid-urea gel과 18% P이yacηlam ide SDS-gel을 이용하여 전기영동하였다. 분자량을 비교하기 위하여 marker protein으로 Cytochrome C와 함께 15% polyacrylamide SDS-gel을 이용하였으며 모든 gel에서 standard로써 calf thymus histone을 사용하여 electrophoreticmobility를 비교하였고 호 한 속내에서의 종간의 차이가 있는지를 설험하였다 실험결과는 한 속내에서 종에 따른 차이로 볼 수 없었으며 standard인 calf thymus histone HI. H2A, H2B, H3, H4와 유사한 electromobility흘 갖는 protein band를 확인할 수 있었으며 HI histone은 t성확하게 일치되지가않았다.따라서 하등한균류인Rhizopµs에서도고등 진핵생불과유사한단백질이 존재함을 주정할 수가 있었다.

좀개구리밥에서의 Histone 型 鹽基性 蛋白質의 單離 (Isolation of Histone-type Basic Proteins from Lemna paucicostata)

  • "좀개구리밥"에서 histone 型 鹽基性 蛋白質을 單離하는데 처음으로 成功했다. 鹽基性 單白質은 "좀개구리밥"의 細胞磨碎物에서 稀 錦酸으로 抽出 分離하였다. 그 中 Amberlite CG-50에 (pH 6.0에서) 吸着되는 蛋白質成分은 송아지 胸腺 histone과 비슷한 amino 酸組成을 보였고, 構成 amino 酸 殘基로서는 lysine 이 特히 많았다. Carboxymethyl cellulose에 (pH 4.2에서) 吸着되는 蛋白成分의 溶出 chromatogram은 송아지 胸腺 histone과 비슷했으나, 各 peak의 比率은 相遠했으며, 特히 非吸着 成分이 含有되어 있음도 相違했다. 또 吸着成分의 amino 酸組成은 histone 型 鹽基成을 나타냈으나, 非吸着 成分은 典型的인 histone과는 相異한 蛋白質임을 確認했다. 송아지 胸腺 histone과 "좀개구리밥"에서 單離한 蛋白 成分들의 黃酸 存在下에서 $HgSO_4$와의 沈澱反應을 histone아닌 蛋白質과 比較 檢討한 結果, "좀개구리밥"에서 單離한 것은 histone 型 蛋白成分이 分明하였다.

Isolation and In Vitro Culture of Vascular Endothelial Cells from Mice

  • In cardiovascular disorders, understanding of endothelial cell (EC) function is essential to elucidate the disease mechanism. Although the mouse model has many advantages for in vivo and in vitro research, efficient procedures for the isolation and propagation of primary mouse EC have been problematic. We describe a high yield process for isolation and in vitro culture of primary EC from mouse arteries (aorta, braches of superior mesenteric artery, and cerebral arteries from the circle of Willis). Mouse arteries were carefully dissected without damage under a light microscope, and small pieces of the vessels were transferred on/in a Matrigel matrix enriched with endothelial growth supplement. Primary cells that proliferated in Matrigel were propagated in advanced DMEM with fetal calf serum or platelet-derived serum, EC growth supplement, and heparin. To improve the purity of the cell culture, we applied shearing stress and anti-fibroblast antibody. EC were characterized by a monolayer cobble stone appearance, positive staining with acetylated low density lipoprotein labeled with 1,1'-dioctadecyl-3,3,3',3'-tetramethyl-indocarbocyanine perchlorate, RT-PCR using primers for von-Willebrand factor, and determination of the protein level endothelial nitric oxide synthase. Our simple, efficient method would facilitate in vitro functional investigations of EC from mouse vessels.

Progressively gangrenous limbs in Korean indigenous calves

  • This study investigated cases of progressively necrotizing limbs in Korean indigenous calves. The recent case (Case 1) involved a 3-month old, male calf in Jeonbuk province that presented a visibly dry form of gangrene affecting joints of the forelimbs and the hind limbs. Radiograph revealed osteoarthritis possibly resulting from pressure of disrupted joint skin, fasciae, deeper underlying musculatures and tendon. Histopathology of affected tissue showed necrotizing; severely thrombosed dilated blood vessels with rechanneling microvasculatures. The lack of substantial infectious inflammatory exudates in the vital organs and the inability to respond to antimicrobial treatment bolstered the notion that the observed thromboembolic and vascular lesion was attributed to possible vasoconstrictive effects of ergot alkaloids. Case 2: A previously encountered similar case in a 4-month old, male calf showing gangrene of hind limbs and posterior ataxia was likewise presented. These two cases were impressed as probable ergotism. Ergotism may be uncommon or underreported in Korea. Future isolation of ergot alkaloids in feeds or in pasture is highly suggested.

분변에 함유된 Cryptosporidium parvum 오오시스트의 순수분리 -Ether extraction과 discontinuous sucrose gradients의 병용 (Isolation of Cryptosporidium parvum oocysts from fecal samples - The combination of ether extraction and discontinuous sucrose gradients)

  • Cryptosporidium의 오오시스트를 분변에서 분리해 내는 방법은 많이 알려져 있으나 그러한 방법은 주로 설사변에 함유된 원충을 분리하는 데 적용되었다. 이 실험에서는 송아지와 마우스를 C. parvum으로 감염시키고, 그 분변으로부터 원충을 순수 분리하기 위해 ether extraction(EE)법과 discontinuous sucrose gradients(DSG)법을 병용하여 실시하였다. 먼저 EE법으로 분변 내의 지질이나 지방성분과 함께 큰 이물질을 제거한 후 돈망체를 사용하여 분변내의 이물질을 다시 한번 제거하였다. 동망체에 걸러진 분변 부유액을 2.5% potassium dichromate 현탄액으로 만들어 DSG법으로 원심한 결과 비중 1.103과 1.064의 sucrose gradient 경계부위에 횐 밴드가 관찰되었다. 흰 밴드 부위에서 이물질이 제거된 오오시스트를 회수할 수 있었는데 송아지와 마우스 분변을 트르롭으로 전처리한 부유액에 ml 당 오오시스트의 수가 많을수록 원충의 회수율은 높았다 송아지 분변에서 회수된 원충의 회수율은 EE법으로 처리된 부유액 내에 $3.8{\;}\times{\;}10^7/ml$개의 오오시스트가 함유되었을 때 81.6%였으며, 마우스 분변에서 회수된 원충의 회수율은 EE법으로 처리된 부유액내에 $3.2{\;} \times{\;}10^6/ml$개의 오오시스트가 함유되었을 때 51.6%이었다. 따라서 50% 정도의 원충을 회수하기 위해서는 EE법으로 전처리한 분변 부유액의 ml당 오오시스트의 수가 $2{\;}\times{\;}10^6$개 이상 되어야 할 것으로 나타났다. 또한 이 방법은 송아지의 설사분변은 물론 정상적인 분변이나 마우스 분변에 함유된, 오오시스트를 순수 분리하는데 효과적이었다.

경남 남부지방에서 사육 중인 송아지의 설사병에 관한 역학적 연구 (An epidemiological study on the calves with clinical diarrhea in southern Gyeongnam)

  • The present study was conducted to investigate the epidemiological characteristics such as isolation rate and antimicrobial drug susceptibility of etiological agents. The specimens (stool or intestine) were collected from 319 calves with clinical diarrhea from 195 farms in Gyeongnam south area (Gosung, Tongyung, Hadong) from June 2005 to August 2006. The isolation rate of Salmonella spp was higher in summer (8.4%) than in winter (4.8%) and the average was 7.2% (23/319 head). Some of Salmonella spp isolated were resistant to penicillin, oxytetracycline, tetracycline, and cephalexin (>90%), but some of them were susceptible to norfloxacin, ciprofloxacin, sulfamethoxazole/trimethoprim and amikacin(>30%). There was no statistical difference in the isolation rate of Eimeria spp between summer(48.9%) and winter(42.3%). For the evaluation of infection level of Eimeria spp oocyst per gram of feces (OPG) was examined, and severe, moderate and light infection level were 11.9%, 12.5% and 22.3%, respectively. In the isolation rates of Eimeria spp the calves under 19 days was lowerthan those over 60 days, but there was not different among herd size.
