Antioxidative Effect of Typha orientalis L. Extract on the Oxidative Stress Induced by Cytotoxicity of Cadmium Sulfate (황산카드뮴독성의 산화적 손상에 대한 부들 추출물의 항산화 효과)

  • Objectives: This study was carried out to analyze the cytotoxicity of cadmium sulfate ($CdSO_4$) and the antioxidative effect of Typha orientalis L. (TO) extract on the oxidative stress induced by cytotoxicity of $CdSO_4$ in the cultured NIH3T3 fibroblasts. Methods: For this study, the cell viability and the antioxidative effects such as the inhibitory activity of lipid peroxidation (LP) and superoxide dismutase (SOD)-like activity and xanthine oxidase (XO)-inhibitory activity were assessed. Results: The cadmium sulfate significantly decreased cell viability in dose-dependently, and $XTT_{50}$ value was measured at $47.4{\mu}M$ of $CdSO_4$. The cytotoxicity of $CdSO_4$ was determined as highly toxic by Borenfreund and Puerner's toxic criteria. The butylated hydroxytoluene (BHT) as antioxidant significantly increased cell viability injured by $CdSO_4$-induced cytotoxicity in these cultures. In the protective effect of TO extract on $CdSO_4$-induced cytotoxicity, TO extract remarkably increased the inhibitory ability of LP and XO as well as SOD-like ability. Conclusions: From the above results, it is suggested that the oxidative stress is involved in the cytotoxicity of $CdSO_4$, and TO extract effectively protected $CdSO_4$-induced cytotoxicity by antioxidative effects. The natural component like TO extract may be a putative therapeutic agent for treatment of the toxicity induced by heavy metallic compound like $CdSO_4$ correlated with the oxidative stress.

Relationship between Toxicity of Heavy Metals and Sludge Retention Time in Sequencing Batch Reactor Process (연속회분식반응조 공정에서 슬러지 체류시간과 중금속 독성의 관계)

  • In order to elucidate the relationship between the sludge retention time(SRT) and the toxicity of heavy metals, such as copper (Cu), cadmium(Cd), and zinc(Zn), in sequencing batch reactor(SBR) process, IC50 was estimated with measuring of INT-dehydrogenase activity in variable SRTs. When the concentrations of heavy metals were increased, the activity of INT-dehydrogenase was gradually decreased indicating the heavy metals inhibit bacterial activity. Cu showed higher toxicity than Zn and Cd. $IC_{50}$ of Cu, Cd, and Zn ranged from $0.37\sim1.96$ mg/L, $15.4\sim16.9$ mg/L, and $9.70\sim23.4$ mg/L, respectively. The toxicity of Cu and Zn was reversely proportional to the length of SRT. It is probably caused by the increased concentration of extracellular polymeric substances in longer SRT which absorb heavy metals. Therefore, the operation of SBR with increased SRT is desirable in treatment of industrial wastewater containing heavy metals.

Effects of Extract of Radix Achyranthis Bidentatae on Cadmium Inhalation Toxicity in Rats (우슬 추출물이 카드뮴 흡입폭로된 흰쥐의 독성해독에 미치는 영향)

  • To know the effects between Cd inhalation toxicity and extract of Radix Achyranthis Bidentatae, 4 rat groups were exposed to Cd aerosol in air using whole-body inhalation exposure for 6 hours/day, 5 days/week, and 4 weeks. Cd concentration in air was 1.03㎎/㎥ and mass median diameter(MMD) was 1.69㎛. 3 different dose intraperitoneal injections of extract of Radix Achyranthis Bidentatae to 3 inhalation exposure groups was done for 4 weeks and the results were as follows: The highest body weight gain for 4 weeks and food intake per day were from inhalation exposure group I and the highest lung and liver weight were also from inhalation exposure group I. The highest kidney weight was from inhalation exposure group III. The lowest Cd content in lung was 33.49㎍/g from inhalation exposure group I. The lowest Cd concentration in blood was 9.36㎍/㎗ from inhalation exposure control. Cd concentrations of 40.02㎍/g in liver and 69.18㎍/g in kidney were the lowest from inhalation exposure group I and III, respectively. The lowest Cd concentration in liver was 21.08㎍/g from inhalation exposure group III and The lowest Cd concentration in kidney was 15.78㎍/g from inhalation exposure group II. For weekly Cd concentration in urine, the value of the fourth week from inhalation exposure group III was the highest. For weekly Cd concentration in feces, the value of the first week from inhalation exposure group III was the highest. The highest metallothionein concentration in lung was 53.42 ㎍/g from inhalation exposure group III and the highest metallothionein concentration in liver was 188.18㎍/g from inhalation exposure group III. The highest metallothionein concentration in kidney was 143.92㎍/g from inhalation exposure group III. The highest Hct, Hb, and WBC values were from inhalation exposure group II and the highest RBC value was from inhalation exposure group III.

  • A herbivorous copepod, Tigriopus japonicus, was tested to study its acute and chronic responses to Cd tonicity. Under $20^{\circ}C,\;96hr-LC_{50}$ of T. japonicus in medium with $32\%_{\circ},\;26\%_{\circ},\;and\;18\%_{\circ}$ salinity were 11.1 ppm, 13.7 ppm, and 9.7 ppm, respectively. The $96hr-LC_{50}$ in medium with $32\%_{\circ},\;26\%_{\circ},\;and\;18\%_{\circ}$ salinity increased to 21.6 ppm, 15.0 ppm, and 11,0 ppm, respectively under $10^{\circ}C$ condition. Oxygen consumption rates during 96hrs exposure to Cd toxicity decreased more sharply at $20^{\circ}C$ than those at $10^{\circ}C$. Survival rate and $250dys-LC_{50}$ of T. japonicus in medium with $32\%_{\circ}$ salinity, in concentration of Cd 4 ppm were $5\%$ and 0.05 ppm, respectively under $10^{\circ}C$, and $0\%$ and 0.20 ppm under $20^{\circ}C$. It was that the copepod selectively feed on Skeletonema costatum, Nitzschia pungens, N. seriata, and Rhizosolenia hebetata f. semispina.

  • Amphibian population declines globally. Environmental contamination by heavy metals has been suspected to the one of the reason for distinction of amphibian which has obligate aquatic life style during larval period. Amphibians have been widely accepted as animal model for the study of endocrine disruption in aquatic ecosystem at molecular as well as individual levels. There are increasing need for toxicological data in amphibians at multiple endpoints for management of contamination and development of safety guidelines. Here, we reviewed toxicological information about toxicity of heavy metals (arsenic, cadmium, chromium, copper, lead, mercury, nickel, zinc) on various end-point of amphibian.

A Study on The Comparison of Leaching Methods and Stability of Cement Mortar Solidified Cadmium sludge (시멘트고화에 의한 카드뮴슬러지의 안정성 및 용출실험방법 비교 검토)

  • This study was Performed to evaluate the effective solidification of Cd sludge using cement and power plant fly ash as cement admixture, to identify the leaching characteristics of the heavy metal Cd sludge solidified, and to develope proper KLT(Korean Leaching Test) of hazardous waste. KLT was compared with EPT(Extraction Procedure Toxicity) and TCLP(Toxicity Characteristics Leaching Procedure). Fly ash contents ranged from 0% to 30% of cement weight. The experimental results showed that the optimum amount of fly ash replaced was 10% to 15% and KLT was less appropriate than EPA and TCLP. Also the purpose of the study was to suggest the modification factors on the leaching test currently used, based on the above mentioned aspects. The effects of pH, leaching time, leaching for agitating intensity, and leaching solvent were investigated. As the result of test, the leaching potential was relatively high when the pH and agitation intensity of leaching solution were 5 and 150rpm, respectively. Leaching time of six hours was found to be sufficient and the use of acetic acid as a leaching solvent is more useful in landfill site particularly.

  • The effects of environmental agents on health are great concern for all. It was recognized that each human has differential susceptibility to environmental effects. Susceptibility are changed by many factors includin gdevelopmpntal processes, genetic factors, nutritional stratus, preexisting disease conditions, life style and personal habits. Of all factors nutritional factors seem to be the area most modifiable. Consequently, It is an area that must be more thoroughly evaluated. In this paper, nutrient and environment interactions are reviewed briefly with published literatures. This paper deals with the influence of micronutrients(energy, protein and fat), Vitamins (vitamin 4, vitamin B-complex, vitamin C, vitamin D and vitamin I) and Minerals(calcium, iron, selenium, zinc and other minerls) on environmental effects. The role of arch nutrient was assessed in modifyine the expression of environmental pollutant toxicity with available litertures. In each nutrient section, the effect of environment was considered in following agents : heavy metals(lead, cadmium, mercury, silver and etc), inorganic agents(nitwits, sulfite, fluoride and etc), organic agents(benzene, carbon tatra-chloride, aflatoxin, auto dye, dialbrin etc), Irritant gas(ozone, carbon monooxide and etc), physical agents(X-irradiation, ultra violet, temperature and noise) and insectcides. The extent to which nutritional status modifies environmental effects 3nd its converse, how envirollments affects nutritional status is very complex. In deed, at the present time there are more than 50 chelnical/phycical agents that affect the nutrient metabolism and/or have their toxicity either directly diminished or enhanced by nutrients of those agents, small number of agents for each nutrients have sufficient evidence to warrant any reasonable degree of confidence in their hypothesized associtation. With these information at this present time it is hard to conclude that the recommended dietary allowance for each nutrient should be reconsidered.

Application of Weed Species as the Diagnostic Indicator Plants of Environmental Pollution (환경오염(環境汚染) 진단(診斷) 지표식물(指標植物)로서 잡초종(雜草種)의 활용(活用)에 관(關)한 연구(硏究))

  • The studies were conducted to obtain the basic information of the effects of pollutants on plant species and to select the plant species showing specific responses to the pollutants. For these purposes, paraquat, ammonium, and cadmium as a source of oxidative stress, nitrogen toxicity, and heavy metal toxicity respectively were treated to the plant species. Among the tested plants, Lamiaceae, Brassicaceae, and Caryophyllaceae were tolerant to paraquat, whereas Poaceae and Asteraceae were sensitive. Especially Mosla dianthera of Lamiaceae, Hemistepta lyrata and Aster pilosus of Asteraceae, and Paspalum thunbergii of Poaceae showed higher tolerance than others. Paraquat resistance was related with life style, overwintering capacity, so perennial and biennial species showed higher tolerance than annual species. In response to ammonium, Poaceae showed higher resistance while Fabaceae and Caryophyllaceae showed sensitiveness. Weed species having tolerance to ammonium were Echinochloa crus-galli var. praticola, Panicum dichotomiflorum, Setaria glauca, Chenopodium album, and Solanum nigrum, while Mosla dianthera, Arenaria serpyllifolia and Perilla frutescens var. japonica showed sensitiveness. In the response of plant species to cadmium, Digitaria sanguinalis, Amaranthus lividus showed higher resistance, whereas Galinsoga parviflora, Plantago asiatica, Ambrosia trifida, and Paspalum thunbergii showed sensitiveness. The injured degree on germination stage by pollutants did not related with injured degree on matured stage. During germination, the root elongation was more sensitive than shoot elongation by pollutants, paraquat, ammonium, and cadmium.

  • The effects of acute of marine toxicants on early life of coastal olive flounder were investigated. An increasing order of acute toxicity on embryo- and larva-stages of Paralichtys olivaceus was CdCl2$\mu\textrm{g}$/L and 29 $\mu\textrm{g}$/L, respectively, and those values at larva-stage were estimated 3.5 ng/L, 16.0 nL/L, 10.5 $\mu\textrm{g}$/L and 15.0 $\mu\textrm{g}$/L, respectively.

Comparison of Marine Luminescence Bacteria and Genetically Modified Luminescence E. coli, for Acute Toxicity of Heavy Metals (재조합 발광대장균과 해양 발광 미생물을 이용한 중금속 급성독성평가)

  • The responses of two luminescence-based biosensors were studied on various heavy metals in aqueous solutions. One was recombinant E. coli ($DH5{\alpha}$/pSB311), genetically modified luminescence-based bacteria, and the other was Vibrio fisheri used for the LumisTox system. The recombinant E. coli was marked with the lux CDABE gene from multicopy plasmid, pACYC184, originally isolated from Photorhabdus luminescens. The $DH5{\alpha}$/pSB311 had a characteristic of no organic substrate for its luminescence reaction. Among the tested heavy metals Zinc and cadmium were less toxic than copper and mercury. The recombinant E. coli was more sensitive to toxicity of heavy metals than the LumisTox. The order of toxicity of the heavy metals to the recombinant E. coli was $Hg^{2+}>Cu^{2+}>Zn^{2+}>Cd^{2+}$. In case of the LumisTox, the order of the toxicity of heavy metals was $Hg^{2+}>Cu^{2+}>Cd^{2+}>Zn^{2+}$. The genetically modified luminescence-based biosensor offers a range of sensitive, rapid, and easy to use methods for assessing the potential toxicity of heavy metals in aqueous samples.
