Phenotyping of Flavin-Containing Monooxygenase (FMO) Activity and Factors Affecting FMO Activity in Korean

  • Together with cytochrome P450 (CYP), flavin-containing monooxygenase (FMO) present in liver microsomes oxidizes various endogenous and exogenous chemicals. In an effort to determine the human FMO activity, we have developed two non-invasive urine analysis methods using caffeine (CA) and ranitidine (RA) as the probe compounds. As the production of theobromine (TB) and ranitidine N-oxide (RANO) from CA and RA is catalyzed primarily by the hepatic FMO, we have assigned the urinary molar ratios of TB/CA and RA/RANO as the in vivo FMO activity. In 200 age-matched Korean volunteers, the obtained TB/CA ratio ranged from 0.4 to 15.2 (38-fold difference) and the RA/RANO ratio from 5.7 to 27.2 (4.8-fold). The FMO activity of 20's, determined by caffeine metabolism, was the highest (2.5$\pm$l.9) and those of 30's, 40's, 50's, 60's and 70's were 40%, 50%, 24%, 39% and 36% of the 20's, respectively. Intake of grapefruit juice, known to contain flavonoids, inhibited the in vivo FMO (TB/CA) activity by 79%. Addition of the flavonoids like naringin, quercitrin and kaempferol, present in grapefruit juice, to the in vitro microso-mal FMO assay, thiobenzamide S-oxidation, produced 75%, 70% and 60% inhibition, respectively. Obtained Ki values of quercitrin, kaempferol and naringin on the in vitro FMO activity were 6.2, 12.0 and 13.9 $\mu\textrm{M}$, respectively. This suggested that the dose of drug should need to be adjusted to suit the individual FMO activities when the drugs metabolized by FMO are given to patients. As the intake of grapefruit juice has been identified to inhibit the FMO as well as CYP3A4 and lA2 activities, patients taking drugs metabolized by these enzymes should not drink grapefruit juice as the carrier.

Ion-Selectrodes based on Calcium Ferrite (칼슘 페라이트 이온-선택성 막전극)

  • The ion-selectrodes based on calcium ferrite were prepared by mixing calcium oxide with ferric oxide on molar ratio of 0.6:1.4, 0.8:1.2, 1.0:1.0, respectively. The specimens were sintered at $1400^{\circ}C$ for 2hrs in $O_2$ and for 20min in $H_2$ atmosphere. The selectrode sintered in hydrogen atmosphere showed better responded potential and wider range of responded concentration than selectrode sintered in oxygen atmosphere. The ion-selectrodes base on calcium ferrite(0.6:1.4 molar ratio.) showed the highest potential to bivalent cations such as $Ca^{2+}$, $Ba^{2+}$, $Mg^{2+}$, $Zn^{2+}$ and halogen anions such as $I^-$, $Br^-$ in the range of $10^{-1}{\sim}10^{-7}M$. It showed good agreement with theoretical nernstian values.

The Microstructure and Mechanical Properties of Fiber Reinforced Calcium Silicate Hydrates (섬유보강 규산칼슘수화물 경화체의 미세조직과 기계적 특성)

  • High flexible lightweight composites containing tobermorite as a main mineral is produced using various amorphous silicates, lime, cement and fibers. Here, Mechanical properties of the composites were studied by observing microstructures of hydrates and fibers. Amorphous silicates having better hydraulicity retarded the crystallization of tobermorite due to better formation of C-S-H gel in water bath curing, but, difficult conversion from C-S-H gel to tobermorite in hydrothermal reaction. In the low molar ratio of CaO/SiO2 (0.67), faster crystalization was observed dued to more impurities such as Al2O3 alkali, resulting in improving mechanical properties due to small crystal size and many contact points. It was identified that a lot of calcium silicate hydates formed at surface of pulps increase bonding strength and the crack-resistance of matrix in the composites, but decrease hardness and compressive strength. The choice of amorpous silicates having better hydraulicity, low CaO/SiO2 adding each fibers bellow about 5% in the raw mixs and lower molding pressure should be needed at improve mechanical properties of composites.

Removal Characteristics of $SO_2$ in the Coal Combustion Flue Gas Treatment Convergence System (석탄화력발전소 현장의 석탄연소 배가스 고도처리용 건식 분류층 반응 실증장치에서의 $SO_2$ 제거성능 특성)

  • The purpose of this study is to determine the feasibility of dry-type desulfurization process for actual application to coal-fired power plant. We used actual exhaust gas from Facility Y, Plant #2 to fabricate a demo-scale testing device to attempt to improve the efficiency of desulfurization. A spout-bed circulating dry scrubber convergence system connecting turbo reactor with bag filter was devised, then analyzed for performance characteristics of $SO_2$ removal for Ca/S mole ratio, superficial gas velocity, and ammonia injection, and for secondary reaction characteristics of the non-reactive sorbent at the bag filter. As a result, the installation of spout-bed circulating dry scrubber convergence system showed better economy and efficiency for removing sulfur than the existing wet/semidry-type desulfurization process. In addition, the best efficiency for desulfurization occurred when connected to the bag filter, with differential pressure maintained at 150 $mmH_2O$.

Properties of $CaO-P_2O_5-SiO_2$ Glasses ($CaO-P_2O_5-SiO_2$계 유리의 물성)

  • Properties in terms of the variation of the glass compositions, which were density (p), molar volume(Vm), atom/ion packing density (Dp), refractive index (nD), transformation temperature (Tg), dilatometric softening point (Td), thermal expansion coefficient (α), Young's modulus (E), and knoop hardness (KHN) were investigated in CaO-SiO2 glasses and CaO-P2O5-SiO2 glasses containing less than 10mole% of P2O5. Those properties were measured by density measurement kit, Abbe refractometer, dilatometer, ultrasonic pulse echo equipment, and micro hardness tester. When CaO content was increased in CaO-SiO2 glasses, p, Dp, nD, Tg, Td, α, E and KHN were increased, while Vm was decreased. When P2O5 was added to the CaO-SiO2 glasses with constant CaO/SiO2 ratio as 1.07, p, Dp, nD, Tg, Td, α, E and KHN were decreased, while Vm was increased. When the amount of P2O5 in glasses was kept constant, the changes of the properties with variation of CaO content in the CaO-P2O5-SiO2 glasses were very similar to those of CaO-SiO2 glasses. These phenomena could be explained by the structural role of P2O5 in the CaO-P2O5-SiO2 glasses, which was polymerization of siicate structures and resulted in [PO4] monomer structure in glasses. Due to this structural characteristics, the bond strength and packing density were changed with compositions. Proportional relationships between 1) np and Dp, 2) Tg, Td, α and CaO content, 3) E and Vm-1, and 4) KHN and P2O5 content were evaluated in this investigation.

Accumulation of Poly-$\beta$-Hydroxybutyrie Acid by Alcaligenes sp. (Alcaligenes sp.에 의한 Poly-$\beta$-Hydroxybutyric Acid의 축적)

  • Microorganisms capable of accumulating poly-p-hydroxybutyric acid(PHB) were isolated from soil by enrichment culture technique. Among them, the strain designated as FL-027 had high PHB productivity and was identified as Alcaligenes. The optimal medium composition for cell growth was 8.0 $g/\ell$ of fructose and 3.0 $g/\ell$ of $(NH_4)_2S0_4$, equivalent to C/N ratio 5.04 at pH 7.0 and $30^{\circ}C$. To investigate the optimal conditions for the PHB accumulation, we divided the process into two stages; the first stage for the growth of the cell in nutrient-rich medium and the second stage for the PHB accumulation in nutrient-deficiency medium. The optimal conditions for PHB accumulation were 8.0 $g/\ell$ of fructose and 0.25 $g/\ell$ of $(NH_4)_2S0_4$, equivalent to C/N ratio 60 at pH 6.5 and $30^{\circ}C$. PHB accumulation was stimulated by deficiency of nutrients such as $NH_4^+$, $Ca^{2+}$, $SO_4^{2+}$ in medium. Among them. $NH_4^+$ deficiency was chosen because of its effectiveness. We found the inhibition of cell growth by fructose in batch culture. In order to keep the fructose concentration at an optimal level, intermittent feeding fed-batch culture was employed, and the cell concentration as high as 10.83 $g/\ell$ whose PHB content was responsible for 43% of the dry cell weight. The purified PHB was identified as homopolymer of 3-hydroxybutyric acid by using IR and $^1H-NMR$.

A Study on the Experiments and Prediction of Desulfurization Efficiency in Fluidized Bed Combustor (유동층연소로에서 탈황효율 실험 및 예측에 관한 연구)

  • We have studied that the prediction of desulfurization efficiency by limestone in fluidized-bed coal combustor. The results were presented as follows : Firstly, the bed temperature had a great deal of effect on the desulfurization and the optimum temperature of limestone was 85$0^{\circ}C$~90$0^{\circ}C$. Secondly, as the velocity and temperature increased, $K_{s}$, K and the desulfurization efficiency increased. So, $k_{s}$, $k_{d}$ highly depended on the air velocity and bed temperature, and $k_{s}$, $k_{d}$ were 82.53 mm/sec, 0.0041/sec at 0.2 m/sec, 85$0^{\circ}C$, $k_{s}$, $k_{d}$ were 125.62 mm/sec. 0.00532/sec at 0.3 m/sec, 80$0^{\circ}C$ respectively. And $k_{s}$, $k_{d}$ were 143.78 mm/sec, 0.00568/sec at 0.3 m/sec, 85$0^{\circ}C$. Thirdly, as a result of desulfurization modeling, there was good agreement between theory and experiments as anthracite fraction increased. At 3.0 of optimum Ca/S molar ratio, there was very good agreement between theory and experiments.riments.riments.s.

Effects of Caprylic Acid and Cyclodextrin Complex on In vitro Fermentation Characteristics and Methane Production (Caprylic Acid와 Cyclodextrin 복합물이 In vitro 반추위 발효성상 및 메탄 생성에 미치는 영향)

  • This study was conducted to evaluate the effects of dietary addition of caprylic acid(CA)-cyclodextrin (CD) complex on in vitro fermentation characteristics, total gas and methane production. Experiment was done with six treatment groups; 1) no CA-CD complex(control), 2) CA 20 mg(T1), 3) CD 830 mg(T2), 4) CA-CD complex 425 mg(T3), CA-CD complex 850mg(T4), CA-CD complex 1,700 mg(T5). Ruminal pH, ammonia and total VFA concentrations of T2, T3, T4 and T5 were lower(P<0.05) than those of control and T1 for the 12h incubation. The increase in molar percentage of propionate was observed in T4 and T5 compared with control and T2 for the 8h incubation(P<0.05), however, the ratio of acetate to propionate was unchanged in all treatments. Total gas of T1 was lower than that of control, but T2, T3, T4 and T5 were higher compared with control for 12h incubation(P<0.05). If the methane ratio (as %) to total gas for all treatments was compared, T3, T4 and T5(CA-CD supplemented groups) averaged 2.7% whereas control, T1 and T2 showed 3.4, 2.8 and 5.1%, respectively. Therefore, according to these results, it might be concluded that supplementation of CA-CD complex could reduce methane production without disrupting ruminal fermentation.

Spectrophotometric Determination of Nizatidine and Ranitidine Through Charge Transfer Complex Formation

  • Two Spectrophotometric procedures are presented for the determination of two commonly used H2-receptor antagonists, nizatidine (I) and ranitidine hydrochloride (II). The methods are based mainly on charge transfer complexation reaction of these drugs with either ${\rho}-chloranilic$ acid (${\rho}-CA$) or 2, 3 dichloro-5, 6-dicyanoquinone (DDQ). The produced colored products are quantified spectrophotometrically at 515 and 467 nm in chloranilic acid and 000 methods, respectively. The molar ratios for the reaction products and the optimum assay conditions were studied. The methods determine the cited drugs in concentration ranges of 20-200 and $20-160\;\mu\textrm{g}/mL$ for nizatidine and ranges of 20-240 and $20-140\;\mu\textrm{g}/mL$ for ranitidine with chloranilic acid and DDQ methods, respectively. A more detailed investigation of the complexes formed was made with respect to their composition, association constant, molar absorptivity and free energy change. The proposed procedures were successfully utilized in the determination of the drugs in pharmaceutical preparations. The standard addition method was applied by adding nizatidine and ranitidine to the previously analyzed tablets or capsules. The recovery of each drug was calculated by comparing the concentration obtained from the spiked mixtures with those of the pure drug. The results of analysis of commercial tablets and the recovery study (standard addition method) of the cited drugs suggested that there is no interference from any excipients, which are present in tablets or capsules. Statistical comparison of the results was performed with regard to accuracy and precision using student's t-test and F-ratio at 95% confidence level. There is no significant difference between the reported and proposed methods with regard to accuracy and precision.

Size Distribution of Ambient Aerosol Measured at a Coastal Site in Jeju Island (제주도 해안가에서 측정된 에어로졸의 성분별 입경분포 특성)

  • During the period from April to September 2002, the size distributions of ambient aerosol were measured at the coastal site at Hamduk in Jeju Island. Na$\^$+/, K$\^$+/, Mg$\^$2+/, Ca$\^$2+/and Cl$\^$-/ exhibited mostly a bimodal coarse mode size distribution, while ammonium and sulfate were mainly in the fine size range, with maximum at around 0.54$\mu\textrm{m}$. The average molar concentration ratio of ammonium to sulfate for fine particles was equal to 2.0${\pm}$0.9. Nitrate was evenly found in both the coarse and fine modes. Elements like Al, Fe, Cu, Mg, Na, Ti, Sr and Mn were dominant in coarse particles, with the maximum at around 5.25$\mu\textrm{m}$. S and Pb were mainly in the submicrometer size range. Other elements with a fine and coarse modes were V, Ni, Cu, Ba and Mo. The patterns of the size distributions of trace elements measured at the downtown in Jeju City were very similar to those at the coastal site in Hamduk. However, the amplitude of size fractional concentrations at Jeju City was narrower than that at Hamduk. While the mass median diameters for the chemical species originated from the natural origin such as marine and crust were relatively large, those for ammonium, sulfate, S and Pb were very small.
