Caffeoyl Shikimate Esterase has a Role in Endocarp Lignification in Peach (Prunus persica L.) Fruit

  • Caffeoyl shikimate esterase (CSE) is a key enzyme in lignin synthesis in Arabidopsis thaliana. To determine the role of CSE in lignification of the endocarp in peach (Prunus persica L.) fruit, we cloned and characterized the P. persica CSE homolog, which we designated PpCSE. The 954 - bp PpCSE gene encoded a 317 - amino acid polypeptide. PpCSE expression patterns in the mesocarp and endocarp changed during peach fruit development. There was no significant difference between the expression levels of PpCSE in the mesocarp and endocarp at 39 and 44 days after full bloom (DAFB), but the expression level of PpCSE in the endocarp at 50 and 55 DAFB was 80.73 and 72.75 times higher, respectively, than that in the mesocarp. During peach fruit development, PpCSE expression in the endocarp increased rapidly; the relative PpCSE expression level at 50 DAFB was 122.70 times higher than that at 39 DAFB. At the protein level, CSE was detected in the peach fruit endocarp at 50 and 55 DAFB. Our study suggests that PpCSE expression during peach fruit development is closely related to the degree of endocarp lignification.

Cigarette Smoke Extract-induced Reduction in Migration and Contraction in Normal Human Bronchial Smooth Muscle Cells

  • The proliferation, migration, cytokine release, and contraction of airway smooth muscle cells are key events in the airway remodeling process that occur in lung disease such as asthma, chronic obstruction pulmonary disease, and cancer. These events can be modulated by a number of factors, including cigarette smoke extract (CSE). CSE-induced alterations in the viability, migration, and contractile abilities of normal human airway cells remain unclear. This study investigated the effect of CSE on cell viability, migration, tumor necrosis factor (TNF)-${\alpha}$ secretion, and contraction in normal human bronchial smooth muscle cells (HBSMCs). Treatment of HBSMCs with 10% CSE induced cell death, and the death was accompanied by the generation of reactive oxygen species (ROS). CSE-induced cell death was reduced by N-acetyl-l-cysteine (NAC), an ROS scavenger. In addition, CSE reduced the migration ability of HBSMCs by 75%. The combination of NAC with CSE blocked the CSE-induced reduction of cell migration. However, CSE had no effect on TNF-${\alpha}$ secretion and NF-${\kappa}B$ activation. CSE induced an increase in intracellular $Ca^{2+}$ concentration in 64% of HBSMCs. CSE reduced the contractile ability of HBSMCs, and the ability was enhanced by NAC treatment. These results demonstrate that CSE treatment induces cell death and reduces migration and contraction by increasing ROS generation in normal HBSMCs. These results suggest that CSE may induce airway change through cell death and reduction in migration and contraction of normal HBSMCs.

A Hydraulic and Feasibility Study of New Tower Internal in Gas Processing Plants

  • A new tower internal, which is called CSE, is presented. The CSE is composed of a nozzle perforated in its bottom along the entire periphery and equipped with a multi vane axial swirler at the inlet and hollow cylindrical separator at the outlet of the nozzle. According to the experimental work for obtaining the necessary hydraulic information of the CSE, which is used for preliminary design of a separation column, the CSE showed a stable operation over the wide rage of gas/liquid ratio. However, it caused large pressure drop due to the high gas velocity which should carry liquid droplets through the element. The high pressure drop may cause problems in energy recovery and the application of the CSE can be limited to the high pressure columns. Assuming that the tray efficiency of the CSE is the same with the existing separation columns, the results of the column design showed the size reduction of the column diameters by 30 to $40\%$ and investment cost saving, depending on operating conditions. The application of the CSE to separation column may also contribute to the de-bottlenecking the existing column.

  • According to adaptive structuration theory, faithfulness of appropriation(FOA), and consensus on appropriation(COA) are variables for measuring appropriation in use of IT. FOA is the level of interpreting and using the intentions of the IT designers objectively and COA is the level of agreement on ways of using IT among or with user group independently of designers' intention. In traditional theories of IT adoption, computer self-efficacy(CSE), self evaluation on abilities of using computer, has been considered as one of the important individual differences. which would influence on beliefs of using IT. Therefore, considering the relationship among CSE, FOA, COA and attitude, we can hypothesize that CSE would play a moderation varaible among appropriation related variables relationships, FOA-attitude and COA-attitude. The objective of this study is an analysis of CSE's moderation effects in causality relationship among appropriation measuring variables. For this purpose, we propose a research model where there are four hypotheses for moderation effects of CSE on the relationships between appropriation variables. We test these hypotheses by sampling ERP and groupware users. In result, our proposed hypotheses except moderation effect of COA-usefulness are accepted. Main contribution of this study is to identify the relationship between CSE and AST. This study implies that very careful consideration of users' CSE is necessary in order to perform managerial activities based on FOA and COA in IT management department.

Suppression of Cellular Apoptosis Susceptibility (CSE1L) Inhibits Proliferation and Induces Apoptosis in Colorectal Cancer Cells

  • The cellular apoptosis susceptibility (CSE1L) gene has been demonstrated to regulate multiple cellular mechanisms including the mitotic spindle check point as well as proliferation and apoptosis. However, the importance of CSE1L in human colon cancer is largely unknown. In the present study, we examined expression levels of CSE1L mRNA by semiquantitative RT-PCR. A lentivirus-mediated small interfering RNA (siRNA) was used to knock down CSE1L expression in the human colon cancer cell line RKO. Changes in CSE1L target gene expression were determined by RT-PCR. Cell proliferation was examined by a high content screening assay. In vitro tumorigenesis was measured by colony-formation assay. Cell cycle distribution and apoptosis were detected by flow cytometric analysis. We found CSE1L mRNA to be expressed in human colon cancer cells. Using a lentivirus based RNAi approach, CSE1L expression was significantly inhibited in RKO cells, causing cell cycle arrest in the G2/M and S phases and a delay in cell proliferation, as well as induction of apoptosis and an inhibition of colony growth capacity. Collectively, the results suggest that silencing of CSE1L may be a potential therapeutic approach for colon cancer.

  • The part-surface of mold or stamping-dies consists of a compound surface which consists of lots of composite surfaces, and may have various types of feature shapes including convex sharp edge (CSE). Those CSE features should be considered with care in machining the surface, which necessitates extraction of CSE curves on a compound surface. This work can be done rather easily for a solid model which has a complete topology information. In case of the compound surface without topology information, however, such CSE curves must be gathered through some geometrical calculations paying much computation time. In the paper, extracting CSE curves by the construction of a CSE region-map which can reduce time, and detecting various common edge types are presented.

  • Objectives Cigarette smoking had been recorded as the main cause of impaired endothelium-dependent vasodilation in smokers by reducing nitric oxide (NO), a production of endothelial nitric oxide synthase (eNOS). However, the mechanism of NO impairment via eNOS activity is unclear until now. In this study, cell passage is suggested to be a relevant factor to eNOS expression under cigarette smoking stress. Methods Bovine aortic endothelial cells (BAECs) were chosen as the research subject with passages ranking from 6 to 9 (6P to 9P). After exposure of cigarette smoking extract (CSE) solution, MTT assay and Western blot method were performed to check the cell viability as well as eNOS protein concentration. In these experiments, four concentrations of CSE at 0.5, 1, 2, and 4% were selected for treatment. Results Our results showed that cells almost died at 4% of CSE. Besides, eNOS protein mass had a linear decrease under the increase of CSE concentration. In addition, the effect of CSE on eNOS expression was dissimilar between different passages. Conclusions This study indicated that CSE had effect on both cell viability and eNOS expression. Besides, a reduction in protein mass was matched with the decrease of cell viability due to CSE tress. Last but not least, the response of eNOS protein to different concentration of CSE at different passages was disparate, making the hypothesis about cell passage related inhibition of eNOS caused by CSE solution.

Effects of Antioxidant on Oxidative Stress and Autophagy in Bronchial Epithelial Cells Exposed to Particulate Matter and Cigarette Smoke Extract

  • Background: We evaluated the effect of particulate matter (PM) and cigarette smoke extract (CSE) on bronchial epithelial cell survival, as well as oxidative stress and autophagy levels. Moreover, we aimed to assess the effect of the antioxidant N-acetylcysteine (NAC) on the adverse effects of PM and CSE exposure. Methods: Normal human bronchial epithelial cells (BEAS-2B cells) were exposed to urban PM with or without CSE, after which cytotoxic effects, including oxidative stress and autophagy levels, were measured. After identifying the toxic effects of urban PM and CSE exposure, the effects of NAC treatment on cell damage were evaluated. Results: Urban PM significantly decreased cell viability in a concentration-dependent manner, which was further aggravated by simultaneous treatment with CSE. Notably, pretreatment with NAC at 10 mM for 1 hour reversed the cytotoxic effects of PM and CSE co-exposure. Treatment with 1, 5, and 10 mM NAC was shown to decrease reactive oxygen species levels induced by exposure to both PM and CSE. Additionally, the autophagy response assessed via LC3B expression was increased by PM and CSE exposure, and this also attenuated by NAC treatment. Conclusion: The toxic effects of PM and CSE co-exposure on human bronchial epithelial cells, including decreased cell viability and increased oxidative stress and autophagy levels, could be partly prevented by NAC treatment.

Effect of Coicis Semen Extract on Streptozotocin-Induced Diabetic Nephrophthy Rats (의이인(薏苡仁) 추출물의 경구투여가 Streptozotocin에 의한 흰쥐의 당뇨병성 신증에 미치는 영향)

  • Objectives : The present study was carried out to investigate the beneficial effect of Coicis Semen extract(CSe) on streptozotocin(STZ)-induced diabetic nephropathy rats. CSe was given to rats with oral administration. Methods : The experimental animals were divided into 3 groups : normal group of rats, control group of STZ-induced diabetic rats, sample group with CSe treatment. Experimental diabetes was induced by the injection of STZ(60 mg/kg) to the rat via the peritoneum. The effects of CSe on STZ-induced diabetic nephropathy were observed by measuring the serum level of creatinine, BUN and uric level of glucose. Kidney level of lipid peroxidation and the activities of reduced glutathione(GSH) were also examined Results : STZ-induced increase of serum creatinine was lowered by CSe treatment, but BUN and uric level of glucose did not show significant changes. CSe oral administration showed statistical decrease of lipid peroxidation in renal cortical tissues, but it has no effect on the activities of GSH. Conclusion : CSe treatment showed protective effect on rat diabetic nephropathy model, but action mechanism of the effect was still not dear. We thought to be concerned with anti-oxidative stress.
