Ganoderma lucidum, an oriental polypore fungus and medicinal mushroom, has a long history of use for promoting health and longevity in Korea, China, and other Asian countries. This study was aimed at determining the anti-inflammatory activity and mechanism of action of Ganoderma lucidum in murine macrophage RAW 264.7 cells. Ganoderma lucidum was extracted with ethanol and freeze-dried. The anti-inflammatory effect (nitrite production) of Ganoderma lucidum extracts was tested using a nitric oxide (NO) colorimetric assay. Semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) was performed to quantify the mRNA expression of cytokines including tumor necrosis factor-${\alpha}$ ($TNF-{\alpha}$), interleukin $(IL)-1{\beta}$, and IL-6. Western blotting was performed to measure the expression levels of inflammation-related proteins, such as inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), nuclear factor kappa B ($NF-{\kappa}B$) p65, and phosphorylated $NF-{\kappa}B$ p65. The NO colorimetric assay showed that NO production increased with the treatment of lipopolysaccharide in (LPS)-activated RAW 264.7 macrophages and decreased with the cotreatment of Ganoderma lucidum extracts and LPS. Ganoderma lucidum extracts repressed the mRNA expressions of cytokines, which were increased after the LPS treatment. In addition, Ganoderma lucidum extracts inhibited the LPS-induced expression of iNOS and COX-2 and the LPS-induced phosphorylation of $NF-{\kappa}B$ p65. These results suggest that the Ganoderma lucidum extracts exert an anti-inflammatory activity by inhibiting $NF-{\kappa}B$ related proteins and cytokines.
Cnidium monnieri (L). Cussion is used as a tonic agent in traditional oriental medicine. However, the molecular mechanism of mast cell-mediated anti-inflammatory modulation has not been fully understood. The aim of the present study was to demonstrate the effects of Cnidium monnieri (L). Cussion eathyl acetate fraction on the expression of pro-inflammatory cytokines, as well as to elucidate its mechanism of action in the human mast cell line (HMC-1). Cells were stimulated with phorbol 12-myristate 13-acetate (PMA) plus A23187 in the presence or absence of Cnidium monnieri (L). Cussion eathyl acetate fraction. Cnidium monnieri (L). Cussion eathyl acetate fraction significantly inhibited the PMA plus A23187-induction of inflammatory cytokines such as tumor necrosis factor (TNF)-$\alpha$, interleukin (IL)-6 and IL-8. Moreover, EtOAc fraction attenuated cyclooxygenase (COX)-2 expression. In activated HMC-1 cells, phosphorylation of extra-signal response kinase (ERK) 1/2 decreased after treatment with EtOAc fraction. Moreover EtOAc fraction inhibited PMA plus A23187-induced nuclear factor (NF)-${\kappa}B$ activation, $I{\kappa}B$ degradation. EtOAc fraction suppressed the expression of TNF-$\alpha$, IL-6, IL-8 through a decrease in the ERK 1/2, as well as activation of NF-${\kappa}B$. These results indicated that Cnidium monnieri (L). Cussion EtOAc fraction exerted a regulatory effect on inflammatory reactions mediated by mast cells.
We previously reported that anti-inflammatory properties of poncirin, isolated from fruit of Poncirus trifoliata, might be the result from the inhibition of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), tumor necrosis $factor-{\acute{a}}$ ($TNF-{\alpha}$) and interlukin-6 (IL-6) expression via the down-regulation of $NF{-\kappa}B$ binding activity. In this study, we investigated whether poncirin has an inhibitory effect on the production of pro-inflammatory mediators ex vivo and whether poncirin could relieve the symptoms of dextran sulfate sodium (DSS)-induced colitis in mice model of inflammatory bowel disease. Poncirin significantly inhibited the productions of NO, IL-6 and $TNF-{\alpha}$ in lipopolysaccharide (LPS)-induced mouse peritoneal macrophage. In addition, poncirin-treated mice when compared to control mice not receiving treatment recovered better from the weight loss caused by DSS-induced colitis. Changes in disease activity index (DAI) of poncirin-treated mice were also more favorable than for control mice and were comparable with mice treated with a typical anti-inflammatory-drug, 5-aminosalichylic acid (5-ASA). In addition, suppression of plasma NO and IL-6 productions of poncirin-treated mice was also observed in DSS-induced colitis. These results suggest that poncirin has potentially useful anti-inflammatory effects mediated by suppression of inflammatory mediator productions.
The aim of this study was to explore the role and effect of ginsenosides Rg1 on osteoblasts cultured with Ti particles. Osteoblasts from neonatal rats were cultured with particles and different doses of Rg1, the main active ingredient in ginsenosides Rg1. We found that the COX-2, $PGE_2$, TNF-${\alpha}$, IL-1, and IL -6 concentrations in the medium of cells cultured with Ti particles significantly increased as compared with that of the control cells (p<0.05 or p<0.01). In addition, cells cultured with Ti particles alone exhibited the highest concentrations of these molecules. The $PGE_2$, TNF-${\alpha}$, IL-1, and IL-6 levels in the medium of cells cultured with Rg1 were in between those of the control cells and the cells cultured with Ti particles alone. The IL-1ra level in the group cultured with Ti and medium-dose Rg1 was the highest followed by the cells cultured with Ti and high-dose Rg1 and those cultured with Ti and low-dose Rg1 (p<0.05). In conclusion, ginsenosides can reduce the levels of infl ammatory cytokines produced by osteoblasts on induction with Ti particles and can prevent prosthesis loosening.
본 연구는 제주에서 자생하는 산박하(Isodon inflexus (Thunb.) Kudo, I. inflexus (Thunb.) Kudo )의 80% 메탄올 추출물과 분획물, 그리고 분리 물질인 henryin의 항산화능 및 항염증에 관하여 조사한 것이다. 항산화 효과는 DPPH 라디칼 소거, xanthine oxidase 억제 및 superoxide radical 소거 활성 측정을 통하여 수행할 수 있는데, 산박하 추출물의 효능을 측정한 결과 superoxide radical 소거 활성에서 에틸아세테이트 분획물과 부탄올 분획물의 $IC_{50}$값이 각각 $0.9{\mu}g/mL$, $0.2{\mu}g/mL$로 대조군인 allopurinol ($2.2{\mu}g/mL$)에 비해 우수한 억제 효능을 나타내었다. RAW 264.7 세포주를 사용한 항염 효능 평가에서, 에틸아세테이트 분획물이 강한 NO 억제 효과를 나타내었고, 이 분획으로부터 순수 분리하여 구조 동정된 물질인 henryin 역시 농도 의존적으로 NO 억제시킴을 확인하였다. 특히, 에틸아세테이트 분획물과 henryin은 iNOS, COX-2와 염증관련 cytokine인 TNF-${\alpha}$, IL-6, IL-$1{\beta}$의 mRNA 발현을 농도 의존적으로 억제하였다. 이상의 결과들로부터 산박하 에틸아세테이트 추출물이 항산화 및 항염증 효능을 갖는 화장품 원료로서 개발 가능성이 있고 henryin은 기능성 지표물질로 활용될 수 있음이 시사되었다.
Objectives : This study aimed to investigate the unknown mechanisms behind the anti- inflammatory activity of Hyeonto-dan(HT) 70% ethanol extract on LPS-stimulated RAW 264.7 cells. Methods : Cells were treated with Hyeonto-dan 1 h prior to addition of 200 ng/mL of LPS. Cell viability was measured by the MTS assay. Nitric oxide levels were determined by the Griess assay. $PGE_2$ were measured using EIA kit. Pro-inflammatory cytokine production was measured by the enzyme-linked immunosorbent assay (ELISA). The expression of COX-2, iNOS, and MAPKs was investigated by Western blot, qRT-PCR. $NF-{\kappa}B$/p65 localization and interaction of the TLR-4 receptor with LPS was examined by immunofluorescence assays. Results : Hyeonto-dan had no cytotoxicity at the measured concentration. Hyeonto-dan inhibited NO production and pro-inflammatory cytokines such as IL-6, $TNF-{\alpha}$, and PGE2 as well as the protein and mRNA expression of iNOS and COX-2. Moreover, Hyeonto-dan inhibited the interaction between LPS and TLR-4 in murine macrophages. It suppressed phosphorylation of extracellular signal-regulated kinase (ERK 1/2), c-jun N-terminal kinase (JNK 1/2) and p38. Finally, it inhibited translocation of $NF-{\kappa}B$ in response to competitive LPS. Conclusions : Based on the results of this study, Hyeonto-dan inhibited the binding of TLR-4 receptor to LPS and inhibited the phosphorylation of extracellular signaling pathway MAPKs. These inhibitory effects are thought that the amount of $NF-{\kappa}B$ delivered to the nucleus was decreased and the inflammatory reaction was prevented by decreasing the production of LPS-induced $PGE_2$, NO, IL-6 and $TNF-{\alpha}$.
회향(Foeniculum vulgare) 열매의 메탄올 추출물과 분획물(hexane, methylene chloride, ethylacetate, n-butanol)을 cell culture model system을 이용하여 항염증 활성을 평가하였다. 이를 위해, LPS처리로 염증이 유도된 RAW 264.7 macrophage에서 시료가 세포독성을 나타나지 않는 $100{\mu}g/ml$의 농도를 처리한 후 염증매개물질인 NO, PGE2, 염증성 사이토카인($IL-1{\beta}$, IL-6 및 TNF-${\alpha}$) 생성 및 mRNA 발현을 측정하였다. 그 결과 hexane, methylene chloride 및 ethylacetate 분획물은 대조군에 비해 NO, PGE2 생성을 억제하였으며, 이는 iNOS, COX-2 mRNA 발현 저해에서 기인함을 확인하였다. Methylene chloride와 ethylacetate 분획물은 $100{\mu}g/ml$의 농도에서 $IL-1{\beta}$, IL-6 생성 및 mRNA 발현을 효과적으로 억제하였으며, 특히 ethyl acetate 분획물은 TNF-${\alpha}$ 의 생성과 mRNA발현을 유의적으로 저해하였다. 본 연구 결과를 통해 회향 및 회향 분획물의 항염증 활성을 확인하였으며, 이는 염증 매개물질인 NO, PGE2 그리고 염증성 사이토카인 mRNA 활성 및 분비 억제에 의한 것으로 나타났다.
본연구는 콜라비 추출물의 항산화 및 항염 효능을 조사하기 위하여 수행하였다. 콜라비는 70% 에탄올을 이용하여 조추출한 후 헥산, 에틸아세테이트, 부탄올을 이용하여 용매 극성에 따라 순차적으로 분획하였다. 항산화 활성은 총폴리페놀 함량 측정과 ABTS 라디칼 소거활성을 측정하여 평가하였다. 에틸아세테이트 분획물이 총폴리페놀 함량($27.33{\pm}0.26mg\;GAE/g$)과 ABTS 라디칼 소거활성($IC_{50}\;172.9{\pm}1.6{\mu}g/mL$)이 가장 높게 측정되었다. 항염 활성은 RAW 264.7 세포를 이용하였으며, 에틸아세테이트 분획물이 가장 높은 항염 활성을 보였다. 에틸아세테이트 분획물의 LPS로 유도된 RAW 264.7 세포에서 전염증성 매개인자들에 대한 저해효과를 농도별로 측정하여 확인하였다. 콜라비 에틸아세테이트 분획물은 NO, $PGE_2$ 생성과 iNOS와 COX-2 및 TNF-${\alpha}$, IL-6, IL-$1{\beta}$와 같은 전염증성 사이토카인들의 단백질 발현을 농도의존적으로 저해하였다. 이러한 결과들은 콜라비가 항산화 및 항염 효능을 가지는 소재로서의 개발 가능성이 있음을 시사한다.
This study investigated the anti-inflammatory effects of processed (Beopje) curly dock (Rumex crispus L.) in LPS (lipopolysaccharide)-stimulated murine RAW 264.7 cells. The experimental group was classified into five groups : LPS no treatment, CD (curly dock), CD-B (CD processed through Beopje), LPS, LPS+CD-B (LPS+CD processed through Beopje) and LPS+CD (LPS+CD). Treatment of the Raw 264.7 cell lines using LPS led to a significant increase in NO production, pro-inflammatory cytokines ($TNF-{\alpha}$, IL-6 and $IL-1{\beta}$), and inflammation related genes (COX-2 and iNOS). Investigation of the inhibitory effects of CD and processed CD on NO production and expression of iNOS and COX-2 was done in LPS-induced RAW 264.7 cells. There was significant inhibition of NO production by LPS+CD and LPS+CD-B in a dose-dependent manner (p<0.05). Particularly, LPS+CD-B exhibited reduced mRNA expression of iNOS and COX-2 and NO production as compared to LPS+CD in Raw 264.7 cell lines (p<0.05). These results may explain some known biological activities of curly dock including the anti-inflammatory effects. CD-B in particular exhibited the highest anti-inflammatory effects of inhibiting production of NO, through the regulation of inflammatory related genes and pro-inflammatory cytokines. These results of Beopje processing might help decrease the anti-biological effects and increase several active substances of curly dock.
Objectives: This study was performed to investigate the effects of Gagamsosokmyeong-tang(Jiajianxiaoxuming-tang) on the monosodium iodoacetate(MIA) induced early state osteoarthritis in rats. Methods: Osteoarthritis was induced by injection of MIA(0.25 mg) into knee joints of rats. Osteoarthritis rats were divided into control(n=8) and treated=8 group respectively, Control group was taken distilled water and treated group was taken extracts of Gagamsosokmyeong-tang(Jiajianxiaoxuming-tang) by orally for 20 days. Body weight was measured at 0, 5, 10, 15, 20 days after MIA injection. At the end of experiment, gross and histopathological examination on the articular cartilages of the knee joints were performed. Proteoglycan(PG) content of articular cartilages were analysed by safranine O staining method. The content of tumor necrosis $factor-{\alpha}(TNF-{\alpha})$, $interleukin-1{\beta}(IL-1{\beta})$ in synovial fluids were analysed by enzyme-inked inmunosorbent assay(ELISA) method. And also cycloxygenase-2(COX-2), matrix metalloproteinase 3(MMP-3), inducible nitric oxide synthase(iNOS), calpain immunochistochemical examination on the knee joints were performed. Results: PG content in articular cartilages of the treated group was significantly increased compared with control group. Histopathological osteoarthritic score of the treated group was significantly decreased compared with control group. $TNF-{\alpha}$ content in synovial fluids, expression of iNOS and calpain in synovial membrane of the treated group were significantly decreased compared with control group. But body weight, $1L-1{\beta}$ content in synovial fluids, expression of iNOS and MMP-3 of the treated group were not significantly changed compared with control group. Conclusions: On the basis of these results, we conclude that Gagamsosokmyeong-tang(Jiajianxiaoxuming-tang) has anti-arthritic effects on the MIA induced early stage osteoarthritis in rats.
