Stem length, main root length and main root diameter showed rapid increasement in two-years-old and three-years-old peony. And there after, the increasement was almost finished. Occurrence of disease was increasing every cultivated year, especially the development of leaf spot was most serious. In propagation by root dividing method, the radix yield (kg/10a) was increasing every cultivated year. However, the difference of radix yield at four and five-years-old peony were not significant. The radix yield of four-years-old peony was higher 26% than three-years-old one. As cultivated year goes by, content of paeoniflorin was increasing then the content was highest at four-years-old peony (4.06%). In 1995, the content, had no certain tendency, was highest at three-years-old peony (3.14%). At hot air drying, browing of peony radix was increasing every cultivated year. If we consider radix yield and color, three-years-old peony was proper object of harvesting.
Purpose : Empirical antimicrobial treatment is indicated before bacteriological results are available for young children with febrile UTI to minimize renal scarring. To ensure appropriate therapy, knowledge of the prevalence of causative organisms and their susceptibility patterns to antimicrobials is mandatory. We performed a retrospective analysis investigating the local prevalence and resistance patterns of uropathogens, primarily E. coli, isolated from community-acquired UTIs. Methods : A total of 103 positive urine cultures from children with febrile UTI collected at Bundang CHA General Hospital from February 2004 to February 2005 were analyzed. Inclusion criteria were fever higher than $37.5^{\circ}C$, significant bacteriuria with single strain growth of at least 10s colony forming units/mL urine, and leukocyturia >5/HPF. Results : E. coli(89.3%) was the leading uropathogen followed by Enterococcus spp.(3.9%) Klebsiella spp.(2.9%), Citrobctcter spp.(1.9%) and Enterobacter spp.(1.9%). E. coli strains revealed a low proportion of antimicrobial susceptibility to ampicillin(AMP; 27.2%) ampicillinsulbactam(AMS; 34.8%) and trimethoprim-sulfamethoxazole(SXT; 65.2%). Susceptibility patterns to cephalosporins were as follows; cefazolin(1st generation; 91.3%), cefoxitin(2nd; 100%), ceftriaxone(3rd; 97.8%) and cefepime(4th; 97.8%). Three E. coli isolates produced ex tended - spectrum beta-lactamase(ESBL). Conclusion : Empirical treatment with AMP, AMS and SXT, which are commonly used in pediatric clinics, is not recommended for childhood UTI due to high incidence of resistance. The high level of susceptibility to cephalosporins makes these drugs reasonable alternatives. However the emergence of ESBL-producers, even though they are quite few, may have an impact on cephalosporin treatment in the future. (J Korean Soc Pediatr Nephrol 2006;10:18-26)
BACKGROUND: The buds of persimmon trees are susceptible to cold damage, often with the late frost, at the time of budburst. This study was conducted to determine effect of the cold damage on shoot and fruit growth the current season. METHODS AND RESULTS: 'Fuyu' trees, grown in 50-L pots, were placed for 1 h at $-2.2{\pm}0.5$, $-2.6{\pm}0.5$, or $-3.0{\pm}0.5^{\circ}C$ within a cold storage, at their budburst on April 5. Some trees under ambient temperature at $10-17^{\circ}C$ served as the control. Cold damage of the buds containing flower buds was 54% at $-2.2^{\circ}C$, and significantly increased to 95% at $-3.0^{\circ}C$. The bud damage included the complete death of all, complete death of main buds only, or the late and deformed shoot growth in the spring. Number of flower buds in early May dramatically decreased as the damage ratio increased. Since the thinning of flower buds in mid-May and fruitlets in early July was done in no or slightly damaged trees, the final number of fruits and yield did not decrease compared with the control when the damage increased by 60% and 70%, respectively. Average fruit weight and skin coloration tended to be better with increasing bud damage. Shoot growth was more vigorous in those trees whose buds were severely damaged by low temperature. CONCLUSION(S): Shoot growth and the yield may depend on the number of flower buds and percent fruit set after the cold damage.
This study was conducted to assess the usefulness of the temperature characteristics of the tidal flat sediments during low tide as a environmental factor, and burrowing behaviour, fatness, total hemocyte counts(THCs) and differential hemocyte counts(DHCs) of hard clam Meretrix lusoria as biological activity indices for the management of hard clam farms located in Taean(Chungnam province) and Gimje(Jeonbuk province) tidal flat in Korea. Temperature ranges of the sediment at 1cm depth during spring(March to May) and summer(June to August) in Taean(where the exposure time was about $5.5{\sim}6$ hours during low tide) were $8.7{\sim}26.8^{\circ}C\;and\;27.6{\sim}32.8^{\circ}C$, respectively. Even though there was no significant difference(P>0.05), temperatures of the surface sediment where submerged with remained seawater were generally higher than that of uncovered with seawater. Burrowing depths of normally digged hard clams were 0.9{\sim}3.6cm from March to October, 2002. In the field experiment performed at Taean farming ground covered with seawater, burrowing times of the clams under natural water temperatures were $41.6{\pm}10minutes$ in February and $5.4{\pm}1.3minutes$ in August, respectively, and these were influenced by water temperatures. Fatness of hard clams began to decrease from May(at Taean tidal flat) and June(at Gimje tidal flat), showed the lowest level in August and increased again from September. Total hemocytes counts in the hemolymph of the hard clams were decreased to the lowest level in July($24.7{\times}10^4cells/mL$, at Taean tidal flat) and August($28.2{\times}10^4cells/mL$, at Gimje tidal flat), and significantly increased again from September(at Taean tidal flat, P<0.01) and October(at Gimje tidal flat, P<0.001), respectively. We observed three types of hemocytes from the hemolymph of hard clams according to whether hemocytes retain the granules or not and the size of the granules. As a results, we could found that periodical monitoring of the sediment temperatures, clam burrowing behaviour and hemocyte parameters were very helpful for the management of hard clam farming.
Purpose: Mucopolysaccharidosis type II (MPS II or Hunter syndrome) is a rare lysosomal storage disorder caused by iduronate-2-sulfatase (IDS) deficiency. MPS II causes a wide phenotypic spectrum of symptoms ranging from mild to severe. IDS activity, which is measured in leukocyte pellets or fibroblasts, was reported to be related to clinical phenotype by Sukegawa-Hayasaka et al. Measurement of residual plasma IDS activity using a fluorometric assay is simpler than conventional measurements using skin fibroblasts or peripheral blood mononuclear cells. This is the first study to describe the relationship between plasma IDS activity and clinical phenotype of MPS II. Methods: We hypothesized that residual plasma IDS activity is related to clinical phenotype. We classified 43 Hunter syndrome patients as having attenuated or severe disease types based on clinical characteristics, especially intellectual and cognitive status. There were 27 patients with the severe type and 16 with the attenuated type. Plasma IDS activity was measured by a fluorometric enzyme assay using 4-methylumbelliferyl- ${\alpha}$-iduronate 2-sulphate. Results: Plasma IDS activity in patients with the severe type was significantly lower than that in patients with the attenuated type ($p$=0.006). The optimal cut-off value of plasma IDS activity for distinguishing the severe type from the attenuated type was 0.63 $nmol{\cdot}4hr^{-1}{\cdot}mL^{-1}$. This value had 88.2% sensitivity, 65.4% specificity, and an area under receiver-operator characteristics (ROC) curve of 0.768 (ROC curve analysis; $p$=0.003). Conclusion: These results show that the mild phenotype may be related to residual lysosomal enzyme activity.

The objective of this study was to determine the processing conditions for salt dried rockfish Sebastes schlegeli by sun drying and cold-air drying, as measured by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activity. We processed salt dried rockfish samples. The salinity of rockfish samples was within 1% following salting with 25% salt brine for 3 h. The moisture content of salt dried rockfish was found to reduce linearly from 70.12 to 39.5 g/100 g over the same time interval. The water activities of salt dried rockfish by sun and cold-air drying were 0.94 and 0.87, respectively, after three days of drying. Acid values (AV) were 10.71 and 5.96 mg KOH/g, respectively, after the three day drying period. The ADH activity in a water extract from salt dried rockfish following sun and cold-air drying for 24 h was 228.5% and 226.1% at 13.3 mg/mL, respectively, and was higher than that when drying lasted for 48 and 72 h. The ALDH activity was not affected but both ADH and ALDH activity tended to decrease as the drying time increased from 24 to 72 h. The conditions of processing for the best quality of salt dried rockfish were determined to be drying with a cold-air system for 24 h. These results indicated that water extracts from salt dried rockfish have valuable biological attributes owing to the metabolizing of alcohol and can provide useful information for the design of drying systems for salt dried rockfish.
Purpose : We attempted to compare the independent factors such as age, sex, C-reactive protein(CRP), and white blood cell count(WBC) in children with radiologic studies and assess the necessity of performing voiding cystourethrography(VCUG). Method : 98 children who have been diagnosed their first time febrile urinary tract infection from Janurary 2002 to Januray 2005 were enrolled. In all patient, the duration of fever which occurred before and after treatment was recorded, and CRP, WBC, $^{99m}Tc$-2,3-dimercaptosuccinic acid($^{99m}Tc$-DMSA) renal scans, renal ultrasound and VCUG were analyzed. Results : Of the 98 children diagnosed with urinary tract infection(UTI), 52 were male and 46 were female. 18 had abnormalities in VCUG, 17 had abnormalities in kidney ultrasound, and 20 had partial defects or diffuse uptake decrease in $^{99m}Tc$-DMSA renal scans. There were no significant relationship between incidence of radiologic abnormalities and age. The risk of renal scar was significantly higher in children who had a longer febrile period before treatment than in those with shorter period. Both CRP and WBC were significantly elevated in children with the radiological abnormalities. A positive of $^{99m}Tc$-DMSA renal scans and renal ultrasound were highly associated with vesicoureteral reflux(VUR). Conclusion : If there are abnormalities in the kidney ultrasound and $^{99m}Tc$-DMSA renal scan of a child with initial UTI, a VCUG is recommended. Even in cases without abnormal findings in $^{99m}Tc$-DMSA renal scan and renal ultrasound, clinical data such as CRP and WBC should be assessed, and VCUG should be Performed for the undetected VUR.

Purpose: The general test method of the Korean Pharmacopeia specifies the test method on the clauses of quality control after manufacturing. According to KFDA Guidance for Medicines, standards of residual solvents regulates the maximum permissible dose of acetonitrile as 400 ppm, ethanol as 5,000 ppm, and acetic acid as 5,000 ppm. This study aims at identifying the type of resiual solvents in the final [F-18]FDG vial of an automatic synthesizer and measure its residual quantity. Materials and Methods: The center carried out residual solvents test of [F-18]FDG injection using Agilent Technologies 7890A with a Flame Ionization Detector. The column of Agilent Technologies 7890A used in measuring of residual solvents was CP WAX column ($30m{\times}0.53mm{\times}1.0{\mu}m$) and analysis condition was split mode 1:1 at the initial temperature $70^{\circ}C$ which was increased $20^{\circ}C/minute$ after two minutes and maintained at the final $140^{\circ}C$ for two minutes. The analysis method was as following: Firstly, ethanol-acetonitrile-acetic acid mixture was classified into four types of concentration (250-25-250 ppm, 1,000-100-1,000 ppm, 3,000-300-3,000 ppm, and 6,000-600-6,000 ppm), and $1.0{\mu}L$ of each type of concentration was injected into gas chromatography followed by an analysis of its peak domain. Then, a calibration-curve by the external standard method was drawn based on the analysis result. Results: While ethanol and acetonitrile were detected in TRACERlab MX, FASTlab had additional acetic acid. The residual quantity of the ethanol-acetonitrile-acetic acid mixture evaluated using the calibration-curve was average 72 ppm ethanol, 54 ppm acetonitrile, and 1030 ppm acetic acid for FASTlab, whereas average 439 ppm ethanol and 79 ppm acetonitrile for TRACERlab MX. This indicated that both of them were within the maximum permissible dose. Conclusion: Solvent residues in the [F-18]FDG injection were all within maximum permissible doses and proper to be used to examine a patient. The result indicated that types and quantities of solvent resides of radioactive pharmaceuticals vary depending on the automatic synthesizer.
For callus culture of leaves and internodal tissue of Quercus acutissirna Carr. and Quercus variabitisBL, MS and GD media were supplemented with combinations of different levels of growth substances such as NAA and BAP, NAA and kinetin, 2,4-D and BAP, and 2,4-D and kinetin. The results were obtained as follows; 1. The rates of formation and growth of callus were good on the GD medium supplemented with NAA and BAP, but the values were quite variable on the MS medium. Root formation occurred at the ratio of 1 : 10-100 in the concentrations of BAP and NAA. 2. The GD medium supplemented with the combination of NAA and kinetin showed 100% callus formation. The callus grew well on the other medium except for that with low concentration of NAA and kinetin. Most of roots were developed from leaf tissue cultured on GD media. 3. The GD medium supplemented with the combination of 2,4-D and BAP also showed 100% callus formation, and growth of callus was good on both of MS and GD media. In particular, the callus derived from internodal tissue showed. the most vigorous growth and reached above 22mm diameter, when they were cultured on MS media supplemented wi th the combinations of $10{\mu}M$ 2,4-D and $1.0{\mu}M$ BAP, and $10{\mu}M$ 2,4-D and $0.1{\mu}M$ BAP. 4. The GD medium supplemented with the combination of 2,4-D and kinetin showed high rate of callus formation, as compared with MS medium Callus grew well on both of MS and GD media.
In this study, we investigated the properties of incubation and growing of Pleurotus eryngii in addition to the mycological properties to use them as basic data for breeding. The speed of mycelial growth on the MCM was faster than on the PDA. The biomass in the PDB broth culture was higher than in the MCM and YMG broth culture. KNR2515 and KNR2516 required 19 days for growth of mycelia on commercial sawdust media. KNR2503 required 6.5 days and 15.3 days for pin-heading and harvesting, respectively. In morphological properties by the mushroom, the heights of KNR2312 and KNR2322 were 122.7 and 121.0 mm, respectively. The thickness of KNR2322 and KNR2513 were 39.8 mm and 31.3 mm, respectively. The weight of KNR2524's fruiting body was 36.3 g, which is good as wild strain. The quality of fruiting body of KNR2503 was 4.0 in comparison to the score 7 of commercially cultivated strains. KNR2512 had the darkest color of pileus with L value 43.6. The slow growing strains, KNR2511, KNR2513, and KNR2512 had the bright pileus with L value 80. In morphological characteristics, KNR2511, KNR2513, and KNR2515 had white lamellar and plane pileus. The three strains are supposed to be the same group and KNR2516 and KNR2518 appeared to be related to the group. The commercially cultivated strains had convex pileus, KNR2502, KNR2503, KNR2504, KNR2521, and KNR2525 had infundibuliform, and the other strains had plane pileus. Several strains were valuable for breeding, JNR2503 for growth rate, KNR2512 for pileus color, and KNR2312, KNR2322, KNR2503, and KNR2513 for the quality.
