Leukemia Stem Cells in Blood Cells; Focused on Acute Myeloid Leukemia

  • It is known that acute myeloid leukemia (AML) is a heterogeneous blood cancer, which is enormously propagated by self-renewing leukemia stem cells (LSCs). The persistence of LSCs after chemotherapy can contribute to minimal residual disease and relapse by LSCs can be evoked promptly. Elucidating special molecules and cellular activity of LSCs is an extremely important to eliminate AML. Despite an increasing understanding of the origin of LSCs by incessant study, AML still remains a notorious disease with high mortality. An exact identification of the LSCs that sustain the proliferation of neoplastic clone is a fundamental issue in AML treatment. CD34+CD38- conventional phenotype is overall regarded as LSCs, but it has a limitation that is still hard to demarcate exactly due to similarity with normal hematopoietic stem cells (HSCs). Not all primary blasts and progenitors have equal function, thus a bona fide marker for identifying LSCs from HSCs is needed in hematologic malignancy, especially in AML. These findings have direct important implications in both in mechanistic study of LSCs as well as in the strategies of more effective therapies. In this review, I briefly summarized current advances in LSCs biology, focusing on membrane markers and a functional behavior of LSCs in AML treatment with monoclonal antibodies. Ultimately, it may be helpful in overviewing the status of LSC research, while expecting the clinic benefits of target therapy by specific inhibition.

동시진공증발법을 이용한 고효율 CIGS 박막 태양전지 개발 (Development of High Efficiency CIGS Thin Film Solar Cells by co-evaporation process)

  • CIGS 박막 태양전지는 제조단가가 낮고 박막 태양전지 중에서 변환효율이 가장 높아 발전 가능성이 큰 태양전지로 인식되고 있다. 이미 일본, 독일, 미국을 비롯한 선진국에서는 30-50 MW 급의 양산 라인이 구축되고 있어 2010년 이후에는 본격적인 상용화가 진행될 것으로 보인다. CIGS 광흡수층은 진공증발, 셀렌화, 나노입자, 전기도금등 다양한 방식으로 제조가 가능한데 이 중에서도 동시진공증발공정은 고효율 CIGS 박막 태양전지 제조에 적합하다. 본 연구에서는 동시진공증발법을 이용하여 CIGS 박막을 증착하였으며 소다회유리/Mo/CIGS/CdS/i-ZnO/n-ZnO/Al/AR 구조의 태양전지를 제조하였다. 기판온도 모니터링을 통한 Cu 이차상 조절 기술을 이용하여 결정립이 매우 큰 CIGS 박막을 증착하였으며 Ga/(In+Ga) 조성비의 조절을 통하여 밴드갭 에너지를 최적화하였다. 또한 QCM 장치를 활용하여 용액 속에서 성장되는 CdS 박막의 두께와 특성을 조절하였다. 이러한 공정최적화를 통하여 개방전압 0.65 V, 단락전류밀도 38.8 $mA/cm^2$, 충실도 0.74 그리고 변환효율 18.8% 의 CIGS 박막 태양전지를 얻었다.

한국연안 진주담치와 굴의 중금속농도에 관한 연구 (Study on the Heavy Metal Concentration in Mussels and Oysters from the Korean Coastal Waters)

  • 1985-1989년간 한국연안의 서로 다른 서식지로부터 채집된 진주담치 Mytilus edulis와 굴 Crassostrea gigas중의 중금속 농도를 알아보았다. 두이매패류에서 가장 높은 중금속농도는 조사점 1, 2, 5, 23 그리고 25에서 발견되었던 반면 가장 낮은 중금속농도는 진해만 주위의 조사점에서 발견되었다. 5개년간 굴과 진주담치의 평균중금속농도비는 Cu 16:1, Cd 1.63:1, Pb 1.36:1, Zn 7.61:1 그리고 Hg 2.08:1로서 굴이 진주담치보다 Cu, Zn, Hg에서 보다 효과적인 생물농축자로 나타났다. 진주담치의 크기별 분포는 크기가 클수록 중금속농도도 증가하였다.

Hydroquinone, a Reactive Metabolite of Benzene, Reduces Macrophage-mediated Immune Responses

  • Hydroquinone is a toxic compound and a major benzene metabolite. We report that it strongly inhibits the activation of macrophages and associated cells. Thus, it suppressed the production of proinflammatory cytokines [tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-$1{\beta}$, IL-3, IL-6, IL-10, IL-12p40, IL-23], secretion of toxic molecules [nitric oxide (NO) and reactive oxygen species (ROS)] and the activation and expression of CD29 as judged by cell-cell adhesion and surface staining experiments. The inhibition was due to the induction of heme oxygenase (HO)-1 in LPS-activated macrophages, since blocking HO-1 activity with ZnPP, an HO-1 specific inhibitor, abolished hydroquinone's NO inhibitory activity. In addition, hydroquinone and inhibitors (wortmannin and LY294002) of the phosphatidylinositol-3 kinase (PI3K)/Akt pathway had very similar inhibitory effects on LPS-induced and CD29-mediated macrophage responses, including the phoshorylation of Akt. Therefore, our data suggest that hydroquinone inhibits macrophage-mediated immune responses by modulating intracellular signaling and protective mechanisms.

온산공단(溫山工團) 주변 농작물(農作物)의 중금속(重金屬) 농도조사 (A Survey on the Heavy Metal Concentrations of Crop Materials Grown near Onsan Industrial Complex)

  • 온산(溫山)공업단지 조성(造成)에 따른 농작물(農作物)의 오염(汚染)가능성을 조사하기 위하여 1978년 3월부터 12월에 걸쳐 주변 농경지에서 재배된 농작물(農作物)(수도, 보리, 대두, 채소, 과일)중의 중금속 농도를 분석한 결과는 다음과 같다. 중금속 함량은 농작물의 종류, 부위, 재배지역에 따라 약간의 차이가 있었으나 일정한 경향은 찾아 볼 수 없었다. 분석한 농작물 시료의 평균치(平均値)를 보면(풍건물(風乾物)기준) As 0.23 ppm, Cd 0.46 ppm, Cu 4.88 ppm, Hg 0.09 ppm, Pb 3.86 ppm, Zn 41.3 ppm으로서 이 지역에 대한 자연함유량(自然含有量) 기준(基準)으로 이용할 수 있을 것이다.

피라졸 유도체를 함유한 폴리알킬플루오렌 공중합체의 향상된 EL 특성 (Highly Enhanced EL Properties of PF Copolymers with Pyrazole Derivatives)

  • We have synthesized new blue electroluminescent polyalkylfluorene-based copolymers [poly(F-co-Py)x:y, where x:y = 99:1 or 95:5 mole ratios] containing the hole-injecting pyrazole derivative [3,3'-(4,6-bis(octyloxy)-1,3-phenylene)bis(1,5-diphenyl-4,5-dihydro-1H-pyrazole] through Ni(0) mediated polymerization, and their electroluminescent properties were investigated. Electroluminescent (EL) devices were fabricated with ITO / PEDOT:PSS (110 nm) / copolymers or PF homopolymer (80 nm) / Ca (50 nm) / Al (200 nm) configuration. Each EL device constructed from the copolymer exhibited significantly enhanced brightness and efficiency compared with a device constructed from the PF homopolymer. The EL device constructed with poly(F-co-Py)99:1 exhibited the highest luminous efficiency and brightness (0.95 cd/A and $2,907\;cd/m^2$, respectively). The achieved luminous efficiency was an excellent result, providing almost a 4-fold improvement on the efficiency obtainable with the a PF homopolymer device. This enhanced efficiency of the copolymer devices results from their improved hole injection and more efficient charge carrier balance, which arises from the HOMO level (~5.83 eV) of the poly(F-co-Py)99:1 copolymer, which is higher than that of the PF homopolyme (~5.90 eV).

T Lymphocyte Development and Activation in Humanized Mouse Model

  • Humanized mice, containing engrafted human cells and tissues, are emerging as an important in vivo platform for studying human diseases. Since the development of Nod scid gamma (NSG) mice bearing mutations in the IL-2 receptor gamma chain, many investigators have used NSG mice engrafted with human hematopoietic stem cells (HSCs) to generate functional human immune systems in vivo, results in high efficacy of human cell engraftment. The development of NSG mice has allowed significant advances to be made in studies on several human diseases, including cancer and graft-versus-host-disease (GVHD), and in regenerative medicine. Based on the human HSC transplantation, organ transplantation including thymus and liver in the renal capsule has been performed. Also, immune reconstruction of cells, of the lymphoid as well as myeloid lineages, has been partly accomplished. However, crosstalk between pluripotent stem cell derived therapeutic cells with human leukocyte antigen (HLA) mis/matched types and immune CD3 T cells have not been fully addressed. To overcome this hurdle, human major histocompatibility complex (MHC) molecules, not mouse MHC molecules, are required to generate functional T cells in a humanized mouse model. Here, we briefly summarize characteristics of the humanized mouse model, focusing on development of CD3 T cells with MHC molecules. We also highlight the necessity of the humanized mouse model for the treatment of various human diseases.

Effects of rosmarinic acid on immunoregulatory activity and hepatocellular carcinoma cell apoptosis in H22 tumor-bearing mice

  • Rosmarinic acid (RA) is a natural polyphenolic compound that exists in many medicinal species of Boraginaceae and Lamiaceae. The previous studies have revealed that RA had therapeutic effects on hepatocellular carcinoma (HCC) in the H22-xenograft models by inhibiting the inflammatory cytokines and $NF-{\kappa}B$ p65 pathway in the tumor microenvironment. However, its molecular mechanisms of immunoregulation and pro-apoptotic effect in HCC have not been fully explored. In the present study, RA at 75, 150, and 300 mg/kg was given to H22 tumor-bearing mice via gavage once a day for 10 days. The results showed that RA can effectively inhibit the tumor growth through regulating the ratio of $CD4^+/CD8^+$ and the secretion of interleukin (IL)-2 and interferon-${\gamma}$, inhibiting the expressions of IL-6, IL-10 and signal transducer and activator of transcription 3, thereby up-regulating Bax and Caspase-3 and down-regulating Bcl-2. The underlying mechanisms involved regulation of immune response and induction of HCC cell apoptosis. These results may provide a more comprehensive perspective to clarify the anti-tumor mechanism of RA in HCC.

Transcriptional and Epigenetic Regulation of Context-Dependent Plasticity in T-Helper Lineages

  • Th cell lineage determination and functional specialization are tightly linked to the activation of lineage-determining transcription factors (TFs) that bind cis-regulatory elements. These lineage-determining TFs act in concert with multiple layers of transcriptional regulators to alter the epigenetic landscape, including DNA methylation, histone modification and threedimensional chromosome architecture, in order to facilitate the specific Th gene expression programs that allow for phenotypic diversification. Accumulating evidence indicates that Th cell differentiation is not as rigid as classically held; rather, extensive phenotypic plasticity is an inherent feature of T cell lineages. Recent studies have begun to uncover the epigenetic programs that mechanistically govern T cell subset specification and immunological memory. Advances in next generation sequencing technologies have allowed global transcriptomic and epigenomic interrogation of CD4+ Th cells that extends previous findings focusing on individual loci. In this review, we provide an overview of recent genome-wide insights into the transcriptional and epigenetic regulation of CD4+ T cell-mediated adaptive immunity and discuss the implications for disease as well as immunotherapies.

과자금(瓜子金) 추출물과 Cyclosporin A가 OVA로 천식이 유발된 생쥐의 염증성 면역세포에 미치는 영향 (Effects of Extract of Herba Polygalae Japonica on Inflammation Cells of Lung in Asthma-induced Mice by OVA Exposure)

  • Objectives : The present study was carried out to investigate the effect of Herba Polygalae Japonica on the proliferation and activation of eosinophils which were prepared from lung cells of asthma-induced mice by ovalbumin (OVA) treatment. Methods : C57BL/6 mouse was exposed to OVA three times a week for 6 weeks. The mouse lung tissues were dissected out, chopped and dessiciated with collagenase (1 ${\mu}g$/ml). Eosinophils were activated by rIL-3/rmIL-5 co-treatments. The lung cells were treated with extract of Herba Polygalae Japonica (EPJ), incubated for 48 hr at $37^{\circ}C$, and analyzed by flow cytometer, ELISA, RT-PCR and immunocytochemistry stain. Results : A significant cytotoxicity by drug treatment was not observed. The cell number ratio of granulocyte, CD3e-/CCR3+, CD3e+CD69+, CD4+, CD23+/B220+ cells was increased in rmIL-5/rIL-3 treated control group compared to the normal group. Cells numbers in the experimental animal group treated with EPJ was all decreased. In ELISA analysis, IL-4, IL-5, IL-13 levels and histamine release level were increased in the control group compared to the normal animal group, then significantly decreased in the experimental group with 100 ${\mu}g$/ml of EPJ treatment. In RT-PCR analysis, mRNA expressions of IL-4, IL-5, IL-13, CCR3 and eotaxin were increased in the control group compared to the normal animal group, then decreased in the experimental group with 100 ${\mu}g$/ml of EPJ treatment. And eosinophil proliferation levels were 18847${\pm}$,1527 (cpm) in the control group, 4676${\pm}$972 (cpm) in the positive control group, and 7709 ${\pm}$ 549 (cpm), 16839 ${\pm}$ 1403 (cpm), 16385 ${\pm}$ 1723 (cpm) in the experimental group with 100 ${\mu}g$/ml, 10 ${\mu}g$/ml, 1 ${\mu}g$/ml of EPJ treatment. Conclusions : The present data suggested that Herba polygalae japonica may have an effects on the inhibition of parameters associated with asthma responses in eosinpophils, and thus implicate the possibility for the clinical application of EPJ.
