In the present study, baicalin, baicalein, and wogonin, a major flavone isolated from Scutellaria Radix were examined for their effects on PMA-induced Interlukin-6 (IL-6), $interferon-\gamma(IFN-\gamma)$, tumor necrosis factor $(TNF)-\alpha$, IL-4, IL-10, and IL-13 productions in the PMA-stimulated $CD4^+$ Jurkat T cells. These three compounds inhibited PMA-induced Th1 cytokine $(IL-6,\;IFN-\gamma,\;TNF-\alpha)$ and Th2 cytokine (IL-4 and IL-13) productions in a concentration-dependent manner. But wogonin, but not baicalin baicalein, increased PMA-induced IL-10 production. These results suggest that baicalin, baicalein, and wogonin, a major flavone modulate Th1 and Th2 cytokine productions in $CD4^+$ Jurkat T cells and these properties may contribute to the anti-atopic dermatitis activity of Scutellaria Radix.

The cyclodextrin glycosyltransferase (CGTase, EC 3.2.1.19) from Bacillus brevis CD162 was purified by precipitating with ammonium sulfate, DEAE-Sepharose CL-6B column chromatography and Sephadex G-150 column chromatography. The molecular mass and pI of the purified enzyme were estimated to be 74,000 and 6.3 by SDS-PAGE and isoelectric focusing, respectively. The purified enzyme was clearly identified as the CGTase by zymogram after SDS-PAGE. The optimum pH and temperature for the enzyme activity were 8.0 and $55^{\circ}C$, respectively. The enzyme was stable at the range of pH $5.5{\sim}9.0$, and up to $50^{\circ}C$. The amino acid sequence from the $NH_2-terminal$ of the purified CGTase was Ser-Val-Thr-Asn-Lys-Val-Asn-Tyr-Ser-Lys-Asp-Val-Ile-Tyr-Gln. The yields of the products from starch as the substrate were 1.3% for ${\alpha}-$, 33.9% for ${\beta}-$, and 9.7% for ${\gamma}-cyclodextrin$.

The immunogenetic analysis was performed to characterize the Korean native chickens (KNC) determined by monoclonal antibodies specific to chicken leukocyte differentiation antigens and flow cytometry. A total of 174 chickens including 58 KNC (black, brown and darkbrown colored), 77 foreign breed (Nagoya, White Reghorn, Rhode Island and Cornish) and 39 mixed breed (19 KNC with Nagoya and 20 KNC with Rhode Island) separately growing at Animal Science and Technology Institute were examined. The proportion of cells expressing MHC class II molecule (B-L in chicken) was significantly high in KNC. Proportion of CD4+ T helper cells was also higher in KNC and two mixed breed than that in foreign breed. However, proportion of CD8+ cells and TCR1 + (${\gamma}^{\delta}$ T cell receptor) cells was the lowest among the breed examined. Otherwise, those proportions were significantly high in White leghorn and two mixed breeds with two exclusive subpopulations. The two subpopulations were also typically shown in MHC class $II^+$ cells in KNC and one mixed breed, black-colored KNC with Nagoya. Although genotypic analysis was not pursued to characterize the immunogenetic properties of KNC, difference of phenotypic expression based on leukocyte differentiation molecules could be elucidated in KNC in this study.
Purpose : The Purpose of this research was to investigate the effects of Haingkyunghonghwatang (HKHHT) on anti-inflammatory effects. Methods : As for the parameters of inflammation, levels of several inflammatory cytokines and chemical mediators were determined in mouse lung fibroblast cells(mLFC) and RAW264.7 cells. Also, changes in pathological features by drug treatment were investigated in the in vivo edema-induced rats by carrageenin/arachidonic acid or in the colitis-induced mice by DSS treatment. Results : The cytotoxicity of HKHHT on mLFC and RAW264.7 cells wasn't observed at 100, 50, 10, and $1{\mu}g/ml$ of The treatments. $IL-1{\beta}$, IL-6 and NOS-II mRNA expression of RAW264.7 cells was inhibited by The treatments in a dose-dependent manner. HKHHT treatment of RAW264.7cells(HtRc) inhibited $TNF-{\alpha}$ and COX-2 mRNA expression. HtRc significantly inhibited IL-6 and NO production. HtRc inhibited ROS production. HKHHT inhibited rat's paw edema induced by carrageenin or arachidonate treatment in all concentrations examined. The body weight and colon length of colitis-induced mice were recovered to a normal level by DSS treatment. Clinical disease levels were significantly improved compared to the control animals. HKHHT treatment of colitis-induced mice(HtCm) significantly increased hematological values such as WBC and RBC counts, Hgb and HCT levels, but decreased PLT values. HtCm decreased IL-6 and $TNF-{\alpha}$ production significantly HtCm significantly increased CD3+(T) cell counts. In contrast, HKHHT treatment decreased CD19+ B cell counts and CD3+/CD69+ significantly, and also decreased B/T ratio (%) though not significant. Conclusion : These results indicated that HKHHT could be used for treating diverse female diseases caused by the inflammation.
To investigate the degree of contamination from Korean coastal region, the concentrations of Cu, Pb, Zn and Cd in sediments, seawater and oyster (Crassostrea gigas) at Masan Bay, Onsan Bay, Daesan industrial complex and their control areas were analysed. Values for sediments, seawater and oyster in the industrial complex coastal region were higher than those in the control area except for seawater in Daesan. The values for dissolved phasc of Cu, Pb, Zn and Cd in seawater showed 0.3~1.75 ${\mu}g/l$, 0.013~0.12 ${\mu}g/l$, 0.20~6.14 ${\mu}g/l$ and 0.007~0.021 ${\mu}g/l$, respectively. The concentrations of Cu, Pb, Zn and Cd in sediments werd 12.0~47.8 ${\mu}g/g$, 6.16~59.5 ${\mu}g/g$, 43.0~230 ${\mu}g/g$ and 0.52~11.2 ${\mu}g/g$, respectively. The concentrations of Cu, Pb, Zn and Cd in oyster showed 12.1~85.6 ${\mu}g/g$, 0.267~1.48 ${\mu}g/g$, 1, 070~3, 250 ${\mu}g/g$ and 3.23~7.71 ${\mu}g/g$, respectively. The contents of heavy metals in oysters at industrial complex coastal region showed that they were not seriously contaminated compared with those of Mussel Watch (1992).
In Iri industrial area, Heavy metals in ait of the factories, 10 metal and 8 non-metal factories, were examined for ten months(from Feb. to Nov, 1991). The methals in blood, urin and hair of 232 employees who have worked in the factories were also examined at the same time The results are summarized as follows 1. Heavy metals Pb, Cd and Mn in the metal factories were 0.031mg/m$^{3}$, 0.0065mg/m$^{3}$, and 0. 035mg/m$^{3}$ respectively, but 0.017mg/m$^{3}$, 0.021mg/m$^{3}$ and 0.014mg/m$^{3}$ in non-metal factories. 2. Heavy metals such as Pb, Cd and Mn in blood from employees in the metal factories were measured : 22.36$\mu$g/dl, 0.27$\mu$g/dl and 1.26$\mu$g/dl respectively, The values in the non-metal factories were 19.84$\mu$g/dl, 0.21$\mu$g/dl and 1.24$\mu$g/dl. 3. Heavy metals such as Pb, Cd and Mn in urine from employees in the metal factories were measured 32.94$\mu$g/l, 0.16$\mu$g/dl, and 1.60$\mu$g/dl respectively, whereas the values in the non-metal factories were 28.79$\mu$g/l, 0.13$\mu$g/dl and 1.35$\mu$g/l. 4. Heavy metals such as Pb, Cd and Mn in hair from employees in the metal factories were measured 8.92mg/kg, 0.33mg/kg and 3.71mg/kg respectively, but 8.14mg/kg, 0.31mg/kg and 3.26 rng/kg in the non-metal factories.
Purpose : The Purpose of this research was to investigate the effects of Cheongyeolhawlhyeoltanggagyehyeoldeung (CYHHT) on anti-inflammatory effects. Methods : As for the parameters of inflammation, levels of several inflammatory cytokines and chemical mediators were determined in mouse lung fibroblast cells(mLFC) and RAW264.7 cells. Also, changes in pathological features by drug treatment were investigated in the in vivo edema-induced rats by carrageenin /arachidonic acid or in the colitis-induced mice by DSS treatment. Results : The cytotoxicity of CYHHT on mLFC and RAW264.7 cells was not observed at 100, 50, 10, and $1{\mu}g/ml$ of CYHHT treatments. $IL-1{\beta}$, IL-6 and NOS-IImRNA expression of RAW264.7 cells was inhibited by CYHHT treatments in a dose-dependent manner. CYHHT treatment of RAW264.7 cells inhibited $TNF-{\alpha}$ and COX-2 mRNA expression. CYHHT treatment of RAW264.7 cells significantly inhibited IL-6 and NO production. CYHHT treatment of RAW264.7 cells inhibited ROS production. CYHHT inhibited rat's paw edema induced by carrageenin or arachidonate treatment in all concentrations examined. The body weight and colon length of colitis-induced mice were recovered to a normal level by DSS treatment. Clinical disease levels were significantly improved compared to the control animals. CYHHT treatment of colitis-induced mice significantly increased hematological values such as WBC and RBC counts, Hgb and HCT levels, but decreased PLT values. CYHHT treatment of colitis-induced mice decreased IL-6 and $TNF-{\alpha}$ production significantly CYHHT treatment of colitis-induced mice significantly increased CD3+(T) cell counts. In contrast, CYHHT treatment decreased CD19+ B cell counts and CD3+/CD69+ significantly, and also decreased B/T ratio (%) though not significant. Conclusion : These results indicated that CYHHT could be used for treating diverse female diseases caused by the inflammation.
The interaction between UN and CdCl2 in the LiCl-KCl molten eutectic was studied at 773 K. The reaction was controlled by sampling the melt, as well as by analysis of the resulting precipitate. The process was shown to proceed according to several parallel reactions. The summary reaction was determined to have two stages: a fast one and a slow one. The 19-53% UN → UCl3 conversion was obtained for the molar ratio of CdCl2/UN = 1.22-14.9. The rest of UN converts into the precipitate of complex composition (UNCl + U2N3 + U4N7 + UN2). The increase in the CdCl2/UN molar ratio from 1.22 to 14.9 resulted in the decrease in duration of the first "fast" stage of the process from 18 h to 1 h.
This study was performed to investigate the dose-response relationship between average daily cadmium dose (ADCD) from rice and the occurrence of urinary cadmium (U-Cd) in individuals eating that rice. This was a retrospective cohort designed to compare populations from two areas with different levels of cadmium contamination. Five-hundred and sixty-seven participants aged 18 years or older were interviewed to estimate their rice intake, and were assessed for U-Cd. The sources of consumed rice were sampled for cadmium measurement, from which the ADCD was estimated. Binary logistic regression was used to examine the association between ADCD and U-Cd (cut-off point at $2{\mu}g/g$ creatinine), and a correlation between them was established. The lowest estimate was $ADCD=0.5{\mu}g/kg\;bw/day$ [odds ratio (OR) = 1.71; with a 95% confidence interval (CI) 1.02-2.87]. For comparison, the relationship in the contaminated area is expressed by $ADCD=0.7{\mu}g/kg\;bw/day$, OR = 1.84; [95 % CI, 1.06-3.19], while no relationship was found in the non-contaminated area, meaning that the highest level at which this relationship does not exist is $ADCD=0.6{\mu}g/kg\;bw/day$ [95% CI, 0.99-2.95]. Rice, as a main staple food, is the most likely source of dietary cadmium. Abstaining from or limiting rice consumption, therefore, will increase the likelihood of maintaining U-Cd within the normal range. As the recommended maximum ADCD is not to exceed $0.6{\mu}g/kg\;bw/day$, the consumption of rice grown in cadmium-contaminated areas should not be more than 246.8 g/day. However, the exclusion of many edible plants grown in the contaminated area from the analysis might result in an estimated ADCD that does not reflect the true level of cadmium exposure among local people.
Objective: To testify whether the increased peripheral blood natural killer (pbNK) cells fraction and their cytolytic activity could coincide with patient's history of recurrent spontaneous abortion (RSA) and to evaluate these factors are can be valuable diagnostic markers in RSA. Methods: Women with a history of RSA comprised the patient group (n=35). Normal fertile women, who were experienced at least one healthy term birth without history of infertility or recurrent miscarriage, were included as the healthy control group (n=15). The pbNK cells of $CD3^-/CD56^+/CD16^+$ and their cytolytic activities against K562 cells were measured by flow cytometry and the values were compared between study and control groups. Results: Proportions of pbNK cells among peripheral blood monocytes (PBMC) ($14.2{\pm}5.2$ vs. $9.4{\pm}3.7%$, p=0.002, 95% confidence interval [CI], 1.8 to 7.8) was significantly higher in the patient group. The odds ratio of having RSA history was increased as 8.4 folds (59% of sensitivity, 80% of specificity, and 95% CI: 2.0 to 35.8) in patients who showed pbNK cells fraction above 12.1% which was determined as cut-off value by using ROC curve analysis. The cytolytic activities of pbNK cells which measured by three different ratio of effecter pbNK cells to target K562 cells and calculated by the percent of cytolytic K562 cells, were significantly higher in study group than that of control group (in 50:1 ratio, $48.3{\pm}19.0$ vs. $31.3{\pm}11.9%$, p=0.002; in 25:1 ratio, $37.0{\pm}18.1$ vs. $20.2{\pm}9.2%$, p<0.001; in 12.5:1 ratio, $23.5{\pm}12.7$ vs. $12.4{\pm}7.3%$, p=0.001). With the cut-off values of cytolytic activity of pbNK cells as 43.1% (50:1), 26.9% (25:1), and 17.4% (12.5:1) each, the risk of having RSA history was increased by 10.0, 11.4, and 15.0 folds in patients who had increased in each effector of pbNK to target of K562 cells ratio. Conclusion: The analysis of pbNK cells fraction and their cytolytic activity can be valuable diagnostic markers for RSA. We are going to planning the large scaled studies which include the data of obstetric outcomes in subsequent pregnancies to clarify our results of this study.
