Background: The molecular basis of follicular dendritic cells (FDC)-germinal center (GC) B cell interaction is largely unknown, although this cellular interaction is thought to be important for the whole process of GC B cell differentiation. Methods: Using FDC-like cells, HK, and highly purified GC B cells, we attempted to identify the molecules that play critical roles in the interactions between FDC and B cells. GC B cells were co-cultured with HK cells and soluble CD154 in the presence or absence of various function-blocking monoclonal antibodies to examine their effect on GC B cell binding to HK cells and B cell proliferation. Results: Anti-CD11a and anti-CD54 antibodies inhibited GC B cell binding to HK cells while anti-CD49d and anti-CD106 antibodies did not. GC B cell proliferation was not impaired by the disruption of GC B cell-HK cell adherence. Conclusion: Our results suggest that CD11a/CD18-CD54 interactions play an important roles in the initial binding of GC B cells to FDC and diffusible growth factors from FDC may be responsible the massive proliferation of GC B cells.

Macrophages are important for the first line of defense against microbial pathogens. Integrin CD11b, which is encoded by Itgam, is expressed on the surface of macrophages and has been implicated in adhesion, migration, and cell-mediated cytotoxicity. However, the functional impact of CD11b on the inflammatory responses of macrophages upon microbial infection remains unclear. Here, we show that CD11b deficiency resulted in increased susceptibility to sepsis induced by methicillin-resistant Staphylococcus aureus (MRSA) infection by enhancing the pro-inflammatory activities of macrophages. Upon infection with MRSA, the mortality of Itgam knockout mice was significantly higher than that of control mice, which is associated with increased production of TNF-α and IL-6. In response to MRSA, both bone marrow-derived macrophages and peritoneal macrophages lacking CD11b produced elevated amounts of pro-inflammatory cytokines and nitric oxide. Moreover, CD11b deficiency upregulated IL-4-induced expression of anti-inflammatory mediators such as IL-10 and arginase-1, and an immunomodulatory function of macrophages to restrain T cell activation. Biochemical and confocal microscopy data revealed that CD11b deficiency augmented the activation of NF-κB signaling and phosphorylation of Akt, which promotes the functional activation of macrophages with pro-inflammatory and immunoregulatory phenotypes, respectively. Overall, our experimental evidence suggests that CD11b is a critical modulator of macrophages in response to microbial infection.
연구배경 : 패혈증에서 백혈구의 활성화는 미생물 및 숙주 독성 물질의 청소에 중요한 역활을 하며 한편으로 활성화된 호중구와 그의 특성 물질은 조직 손상파 장기의 기증 부전을 초래한다. 백혈구의 CD11/18 유착 분자는 감염 및 염증 부위로 호중구 이동의 첫 단계인 호중구-내피 세포 유착을 조절한다. 패혈증 치료 전락으로 호중구 억제 효과의 가능성을 알아보기 위해 rat 에서 Escherichia coli 폐렴을 유발하여 CD11b에 대한 단클론 항체(MAb 1B6)의 효과를 평가하였다. 방법 : 1 mg/kg CD11b에 대한 단클론 항체와 1 mg/kg BSA출 폐렴 유발 6 시간전, 유발시 및 유발 6 시간후 무작위적으로 피하 투여하였다. 폐렴 유발후 무작위적으로 24, 60 및 90%의 산소를 투여하였으며 폐렴 유발후 4시간 및 3일간 하루에 한번씩 100 mg/kg의 ceftriaxone을 투여하였다. 말초 및 폐포 호중구와 폐 손상의 지표인 D(A-a)O2, W/D LW ratio 및 폐포 세척액 단백 농도를 12시간과 96시간까지 생존한 동물에서 측정하였다. 결과 : CD11b에 대한 단클론 항체는 말초 및 폐포 호중구를 감소시켰으며 96시간 보다 12시간에 더욱 감소시켰다. 폐손상 지표는 단클론 항체 투여군과 대조군을 비교시 차이를 보이지 앉았으나 생존 동물 수의 부족으로 통계학적 평가의 의미가 없었다. 조기(6 시간) 사망률은 항체 투여군 51%로 대조군 31%보다 의미(P=0.02) 있게 증가하였으나 후기 사망률(12 시간에서 72 시간)은 항체 투여군 44% 대조군 36%로 의미(P=0.089) 있는 차이가 없었다. 결론 : CD11b/18 유착 분자는 감염 및 염증 부위로 호중구의 이동을 조절하는 것으로 알려져 있다. CD11b에 대한 단클론 항체는 rat의 폐인성 패혈증에서 폐포 호중구를 감소시키고 조기 사망률을 증가시켰다. 따라서 CD11b에 대한 단클론 항체가 rat의 폐인성 패혈증의 초기에 호중구의 폐포내 이동을 억제시켜 숙주의 방어 기전에 장애를 초래하여 조기 사망률을 증가시키는 것으로 사료된다.
CD137 (4-1BB/tnfrsf9) has been shown to co-stimulate T cells. However, agonistic anti-CD137 monoclonal antibody (mAb) treatment can suppress $CD4^+$ T cells, ameliorating autoimmune diseases, whereas it induces activation of $CD8^+$ T cells, resulting in diverse therapeutic activity in cancer, viral infection. To investigate the CD137-mediated T cell suppression mechanism, we examined whether anti-CD137 mAb treatment could affect $CD11b^+Gr-1^+$ myeloid-derived suppressor cells (MDSCs). Intriguingly, anti-CD137 mAb injection significantly increased $CD11b^+Gr-1^+$ cells, peaking at days 5 to 10 and continuing for at least 25 days. Furthermore, this cell population could suppress both $CD8^+$ T cells and $CD4^+$ T cells. Thus, this study demonstrated that, for the first time, anti-CD137 mAb treatment could induce $CD11b^+Gr-1^+$ MDSCs under normal conditions, suggesting a possible relationship between myeloid cell induction and CD137-mediated immune suppression.
본 연구는 중환자의 개에서 전신성염증반응증후군 및 패혈증에 의하여 유발되는 백혈구 활성화 지표를 분석하기 위하여 실행되었다. 분석된 지표로는 백혈구 수치, 혈소판 수치, 혈액 도말 표본의 백혈구 변화상 및 백혈구 표면의 CD11b발현 정도 등을 검사하였다. 그 결과, 패혈증군에서 백혈구의 CD11b 발현 값 (유세포 분석)이 현저하게 높게 관찰되었으며(P<0.05), 퇴행성 변화 정도도 같은 군에서 현저하게 높게 관찰 되었다(P<0.05). 또한 비 생존군의 CD11b 발현 값도 생존군에서의 값보다 현저하게 높았다. 이러한 연구 결과로 볼 때, 백혈구의 CD11b 발현은 중환자의 개에서 그 예후를 평가하는데 유용하다고 여겨진다.
It is well reported that tumor cells can regulate host immune systems. To identify the detailed changes of immune cells between tumor bearing mice and normal mice, we evaluated the systemic immune cell phenotype of B16F10 tumor bearing mice in a time dependent manner. The lymphocytic population (CD4+ and CD8+ T cells) of tumor bearing mice significantly decreased compared to that of normal mice. We found that the Foxp3+CD25+ CD4 T cell decreased, but the Foxp3+$CD25^{high}$ CD4 T cell significantly increased. All subpopulations of CD8 T cells decreased, except the CD62L-CD44+ CD8 T cell subpopulation. The myeloid cell population (CD11b+ and Gr-1+ cells) of tumor bearing mice significantly increased. Specifically, Foxp3+$CD25^{high}$ CD4 T cell and CD11b+Gr-1+ cells significantly increased in early phase of tumor progression. These results are helpful to understand the change of the systemic immune cell subpopulation of tumor bearing mice in a time-dependent manner.
CD40L를 발현하는 KU812세포와 CD40를 발현하는 항원 특이적 B 세포의 배양을 통해 IgE 클라스 스위치가 유도된다. 본 연구자는 체외면역법에 의해 건강인의 말초혈액을 이용하여 쌀 알레르겐 특이적 IgM 항체를 생성하는 B 세포주를 수립하였다. 본 연구에서는 KU812세포에 의한 쌀 알레르겐 특이적 IgE 항체의 유도를 시도하였다. 배양 전, 쌀 알레르겐 특이적 B 세포주, RA9G11과 PMA와 ionomycin을 6시간 처리하여 활성화된 KU812세포에서 CD40와 CD40L 발현량을 flow cytomerty를 통화여 각각 확인하였다. RA9G11세포와 활성화된 KU812세포는 높은 수준의 CD40와 CD40L를 각각 발현하였다. 쌀 알레르겐 특이적 IgE항체 합성 유도를 위해, 여러 가지 농도의 IL-4 존재하에, RA9G11세포와 활성화된 KU812세포를 12일 동안 배양하였다. IgE 정상영역의 mRNA 발현량과, 쌀 알레르겐 특이적 IgE 항체 생성 세포가 모든 조건에서 확인되었으며, 특히 50U의 L-4 농도 하에서 같은 비율의 RA9G11과 활성화된 KU812세포를 배양했을 때, IgE 합성이 가장 높았다. 따라서 활성화된 KU812세포에 의한 쌀 알레르겐 특이적 IgE 합성은 알레르기 질환의 치료 및 예방에 관한 연구에 기여할 것으로 사료된다.
본 연구는 소의 유선 내 감염(유방염)과 체세포 면역 표지자의 상관 관계 분석을 위하여 자연적으로 감염된 젖소와 건강한 젖소의 우유를 비교한 단면조사 연구이다. 유방염에 이환된 31마리의 국내 젖소의 우유에서 세 가지의 체세포 면역 표지자(CD11b, CD4, CD8)의 발현과 체세포 수(SCC), 그리고 세균학적인 분석(배양 및 PCR검사)을 통한 감염 여부 및 병원체의 종류에 대하여 분석하였다. 그 결과 감염된 젖소의 우유는 건강한 젖소의 우유보다 체세포 내의 CD11b와 CD4발현이 유의적으로 증가하였으며, CD4/CD8비율도 높았다. 그러나, 병원체의 종류에 따른 증가된 체세포 면역 표지자와는 커다란 연관성을 보이지 않았으나, 감염된 병원체의 수와 관련해서는, 체세포 면역 표지 인자 CD11b, CD4의 발현 그리고 CD4/8 비율의 증가와 현저한 관련이 있었다. 이러한 연구 결과로 볼 때, 면역 표지자를 이용한 우유의 체세포 분석을 통하여 특발성 소 유방염의 진단 및 관리에 유용하게 활용될 수 있을 것이다.

Background: Epstein Barr virus (EBV) infected B cells are transformed into lymphoblastoid cell lines. Some researchers suggested some a few similarities between this process and carcinogenesis. We observed the expression of CD80 and CD86, co-stimulatory molecules on EBV-transformed B cells and changes of CD54 expression after stimulation of CD80 and CD86. Methods: CD80 and CD86 were stimulated using anti-CD80 and anti-CD86 monoclonal antibodies. To assess apoptosis and surface protein expression, flow cytometric analysis was performed. Intracellular signal molecules were evaluated by RT-PCR and immunoblot. Morphology and localization of proteins were examined using inverted or confocal microscope. Results: Cross-linking of CD80 and CD86 induced apoptosis and interfered with proliferation of EBV-transformed B cells, and dispersion of clumped cells. We also examined that their stimulation induced ROS accumulation and reduced CD54 expression. Interestingly, we observed that CD80 and CD86 diminished the expression of CD54 in different methods. Both CD80 and CD86 downregulated activation of focal adhesion kinase. CD80 stimulus inhibited CD54 expression through mainly RhoA inactivation, while CD86 down-regulated Ras and JNK phosphorylation. Conclusion: These results suggest that co-stimulatory CD80 and CD86 molecules, expressed EBV-transformed B cells, may play a role in apoptosis and cell adhesion.
Background: CD27 is recently known as a memory B cell marker and is mainly expressed in activated T cells, some B cell population and NK cells. CD27 is a member of tumor necrosis factor receptor family. Like CD40 molecule, CD27 has (P/S/T/A) X(Q/E)E motif for interacting with TNF receptor-associated factors (TRAFs), and TRAF2 and TRAF5 bindings to CD27 in 293T cells were reported. Methods: To investigate the CD27 signaling effect in B cells, human CD40 extracellular domain containing mouse CD27 cytoplamic domain construct (hCD40-mCD27) was transfected into mouse B cell line CH12.LX and M12.4.1. Results: Through the stimulation of hCD40-mCD27 molecule via anti-human CD40 antibody or CD154 ligation, expression of CD11a, CD23, CD54, CD70 and CD80 were increased and secretion of IgM was induced, which were comparable to the effect of CD40 stimulation. TRAF2 and TRAF3 were recruited into lipid-enriched membrane raft and were bound to CD27 in M12.4.1 cells. CD27 stimulation, however, did not increase TRAF2 or TRAF3 degradation. Conclusion: In contrast to CD40 signaling pathway, TRAF2 and TRAF3 degradation was not observed after CD27 stimulation and it might contribute to prolonged B cell activation through CD27 signaling.
