Acanthamoeba in Southeast Asia - Overview and Challenges

  • Acanthamoeba, one of free-living amoebae (FLA), remains a high risk of direct contact with this protozoan parasite which is ubiquitous in nature and man-made environment. This pathogenic FLA can cause sight-threatening amoebic keratitis (AK) and fatal granulomatous amoebic encephalitis (GAE) though these cases may not commonly be reported in our clinical settings. Acanthamoeba has been detected from different environmental sources namely; soil, water, hotspring, swimming pool, air-conditioner, or contact lens storage cases. The identification of Acanthamoeba is based on morphological appearance and molecular techniques using PCR and DNA sequencing for clinico-epidemiological purposes. Recent treatments have long been ineffective against Acanthamoeba cyst, novel anti-Acanthamoeba agents have therefore been extensively investigated. There are efforts to utilize synthetic chemicals, lead compounds from medicinal plant extracts, and animal products to combat Acanthamoeba infection. Applied nanotechnology, an advanced technology, has shown to enhance the anti-Acanthamoeba activity in the encapsulated nanoparticles leading to new therapeutic options. This review attempts to provide an overview of the available data and studies on the occurrence of pathogenic Acanthamoeba among the Association of Southeast Asian Nations (ASEAN) members with the aim of identifying some potential contributing factors such as distribution, demographic profile of the patients, possible source of the parasite, mode of transmission and treatment. Further, this review attempts to provide future direction for prevention and control of the Acanthamoeba infection.

Synthesis of Various Biomass-derived Carbons and Their Applications as Anode Materials for Lithium Ion Batteries (다양한 바이오매스 기반의 탄소 제조 및 리튬이온전지 음극활물질로의 응용)

  • In this study, various plant-based biomass are recycled into carbon materials to employ as anode materials for lithium-ion batteries. Firstly, various biomass of rice husk, chestnut, tea bag, and coffee ground are collected, washed, and ground. The carbonization process is followed under a nitrogen atmosphere at 850℃. The morphological and chemical properties of materials are investigated using FE-SEM, EDS, and FT-IR to compare the characteristic differences between various biomass. It is noticeable that biomass-derived carbon materials vary in shape and degree of carbonization depending on their precursor materials. These materials are applied as anode materials to measure the electrochemical performance. The specific capacities of rice husk-, chetnut-, tea bag-, and coffee ground-derived carbon materials are evaluated as 65.8, 80.2, 90.6, and 104.7 mAh g-1 at 0.2C. Notably, coffee ground-based carbon exhibited the highest specific capacity owing to the difference in elemental composition and the degree of carbonization. Conclusively, this study suggests the possibility of utilizing as energy storage devices by employing various plant-based biomass into active materials for anodes.

Effects of Gibberellin Treatment on the Dormancy-breaking and Germination Promotion of Carpinus laxiflora Seeds Stored for 16 Years (지베렐린 처리에 따른 서어나무 16년 저장 종자의 휴면타파 및 발아촉진 효과)

  • This study assessed the vitality of Carpinus laxiflora seeds stored for more than 15 years in order to discover optimal pre-treatment conditions for removingthe causes of seed dormancy and improving revitalization rate. Seeds were collected in October 2000 and stored at -18℃ for 16 years. Experiments to assess the revitalization ability of the seeds were performed under the following conditions: a controlled environment; in warm stratification (30 days at 23℃); in cold stratification (30 days at 4℃ andfor 30, 45, 60, and 120 days); gibberellin (GA3) treatment (24 hours per day in GA3 solutions of 100, 500, 1000, 2000, and 3000 mg·L-1); and both cold stratification and GA3 treatment (30 days at 4℃, then in a GA3 solution of 100, 500, and 1000 mg·L-1 for 2 hours). The average germination percentage(GP) of untreated seeds was 2%, and the average GP of warm-stratification seeds was 10%. Cold-stratification seeds had the highest GP at 81% for the 45-day process, while the 120-day cold-stratification seeds had the lowest GP at 67.3%. The average GP of seeds treated with GA3 ranged from 77% (100 mg·L-1) to 99% (1000 mg·L-1), indicatingsignificant differences between the treatment concentrations. The treatment effect of GA3 was highest at 500 mg·L-1 and 1000 mg·L-1, and lowest at 100 mg·L-1. The average GP of seeds treated with GA3 following cold stratification was 68%, which was lower than the cold stratification-only (73.2%) and GA3-only (88.4%) treatments. A comprehensive comparison of the seed germination characteristics according to the four treatments determined that a GA3 500 mg·L-1 pre-treatment, with the highest average GP, was ideally suited to the revitalization of long-term stored C. laxiflora seeds. Consequently, C. laxiflora stored at -18℃ for 16 years indicated strong vitality and could be regenerated by proper pre-treatment.

Effects of 1-methylcyclopropene (1-MCP) on Fruit Quality and Occurrence of Physiological Disorders of Asian Pear (Pyrus pyrifolia), 'Wonhwang' and 'Whasan', during Shelf-life (동양배 '원황' 및 '화산'의 상온유통 중 품질 및 생리장해 발생에 미치는 1-methylcyclopropene (1-MCP) 처리의 영향)

  • This study was conducted to investigate the effect of 1-methylcyclopropene (1-MCP) on fruit quality and incidence of physiological disorders for keeping freshness during marketing period in Asian pear (Pyrus pyrifolia Nakai) 'Wonhwang' and 'Whasan'. Fruits were treated with $1{\mu}L{\cdot}L^{-1}$ 1-MCP for 12 hours at $25^{\circ}C$, at two or three stages of ripeness as determined by days after full bloom (DAFB). Fruits were harvested at 130 and 140 DAFB in early season cultivar 'Wonhwang' and 135, 145, and 150 DAFB in mid-season cultivar 'Whasan', respectively. Fruits were stored at $25^{\circ}C$ for 21 days and measured the flesh firmness, weight loss, soluble solids, acidity, ethylene, respiration and severity of physiological disorders at week interval. 1-MCP treatment to 'Wonhwang' pears harvested at 130 and 140 DAFB effectively delayed firmness loss during storage at $25^{\circ}C$. Untreated fruits of 'Wonhwang' pears harvested at 130 DAFB showed 32.3 and 10.1N of firmness after 14 and 21 days of shelf-life at $25^{\circ}C$, respectively, while those of the 1-MCP treated fruits showed 39.4 and 33.1N during same period. In the fruits harvested at 140 DAFB, the firmness of untreated fruit was lowered to 14.8 and 6.6N after 14 and 21 days, respectively, but those of 1-MCP treated fruit were 35.0 and 33.3N, respectively. Whereas, 1-MCP treatment delayed firmness loss only in the fruit harvested late (150 DAFB) in 'Whasan' pears. Higher soluble solids content and acidity during extended shelf-life were apparent in 1-MCP treated 'Wonhwang' pears, while those of 'Whasan' pears were little changed. 'Wonhwang' pears showed a relatively high ethylene production (maximum $0.58{\mu}l{\cdot}L^{-1}$) in the fruits harvested late than early harvested one. 'Whasan' pears showed little amount of ethylene production regardless of extended shelf-life. 1-MCP treatment to 'Wonhwang' pears decreased respiration rate following shelf-life, 42 and 50% reduction were observed at 14 days of shelf-life when compared with those of untreated ones harvested at 130 and 140 DAFB, respectively. No reduction of respiration rate by the treatment of 1-MCP was detected in 'Whasan' pears which showed considerably low respiration rate compared with 'Wonhwang' pears. Harvest time influenced the level of physiological disorders together with extension of shelf-life in both the cultivars. 1-MCP treatment completely blocked the incidence of internal browning of 'Wonhwang' pears harvested at 130 DAFB, and reduced the incidences of pithiness and core browning, while it promoted the flesh spot decay disorder regardless of harvest time. 1-MCP treatment was of little benefit for the prevention of physiological disorders in 'Whasan' pears compared with those of 'Wonhwang'.

The Screening of Aflatoxin Producing Fungi from Commercial Meju and Soy Bean Paste in Western Gyeongnam by Immunoassay (면역분석기법을 이용한 서부경남 시판 메주 및 된장에서의 Aflatoxin 생성균 검색)

  • Generally, non-aflatoxigenic fungi, such as Aspergillus oryzae, and Aspergillus are main microflora in Korean traditional fermented foods including Meju and soybean paste, but sometimes, Aspergillus flavus and Aspergillus parasiticus can be contaminated and accumulated aflatoxins during fermentation and storage. So the screening of aflatoxigenic strains in fermented traditional food is very important to improve the sanitary quality of those foods. In this work, we screened aflatoxin producing fungi from commercial Meju and soybean paste in Western Gyeongnam by immunoassay. Samples were randomly purchased from market of the commercial Meju(10 EA) and soybean paste(20 EA) in nine areas of Western Gyeongnam. Of the samples collected,24 strains and 22 strains of Aspergillus sp. were isolated from Meju and soybean paste, respectively. The isolated strains were cultured on SLS media at $25^{\circ}C$ for 15 days. The cultured broth were extracted with ethyl acetate and were analysed to determine aflatoxin B$_1$(AFB$_1$) by direct competitive ELISA(DC-ELISA). Six strains(25%) isolated from Meju, and 2 strains(9%) isolated from saybean paste, were confined as aflatoxin producing strains. The average range of aflatoxin productivity of isolates from Meju was 54.6 $\pm$ 38.7 ng/ml and that from soybean paste was 11.1 $\pm$ 8.6 ng/ml, respectively. Among them, isolated strain No. M-5-4 produced a high level of AFBl and showed 98.26 ng/ml of AFB$_1$. Every isolates were also re-confined their AFB$_1$productivity by thin layer chromatography(TLC). The TLC results also showed same trend as DC-ELISA results. As the above results, the screening of hazard mycotoxigenic fungi from traditional fermented foods should be necessary for the safety and the application of HACCP system in the food manufactory in Korea.

STUDIES ON THE PROCESSING AND UTILIZATION OF SARDINE PROTEIN CONCENTRATE (정어리 분말단백질 가공 및 이용에 관한 연구)

  • Since 1976 the catches of sardine increased rapidly in Korea. However due to the poor facilities of preservation, most sardine landed was used only for fish meal as feeds. The aims of this study are to investigate the processing of sardine as a protein. concentrate and to solve related problems under our particular circumstances. Using the ethyl alcohol and isopropyl alcohol, the storage effect for further processing, the optimum processing conditions of sardine protein concentrate and amino acid composition of the product were determined. The utilization of sardine protein ,concentrate as a supplement of bread and noodles was also studied. Chopped sardine meat could be stored in isopropyl and ethyl alcohol without significant deterioration as a raw material for tile further processing. High qualify sardine Protein concentrate could be produced by the method, that is five times five minutes extraction with isopropyl or ethyl alcohol at $80^{\circ}C$ under adequate mixing. In the first step of the extraction, the solvent was added as much as 10 times tile sample amount and the equal volume of additional solvent was also used for the second to fifth step extraction. In the products extracted using isopropyl alcohol and ethyl alcohol, the yields of sardine protein concentrate were $21.2\%$ and $20.3\%$ respectively, and the dry basis contents of protein in the two products were $80.5\%$ and $75.8\%$, the lipid being $0.22\%$ and $0.27\%$ respectively. Isoproyl alcohol was superior to superior alcohol for the extraction of fresh sardine. In amino acid composition of sardine protein concentrate, no difference was found between the products of isopropyl and ethyl alcohol extraction except a little difference in the amount of amino acid between them. In the supplementation of bread and noodles, taste panel showed that supplemented bread and noodles were well accepted when $3\%$ of wheat flour was replaced by sardine protein concentrate.

Alternative Concept to Enhance the Disposal Efficiency for CANDU Spent Fuel Disposal System (CANDU 사용후핵연료 처분시스템 효율향상 개념 도출)

  • There are two types of nuclear reactors in Korea and they are PWR type and CANDU type. The safe management of the spent fuels from these reactors is very important factor to maintain the sustainable energy supply with nuclear power plant. In Korea, a reference disposal system for the spent fuels has been developed through a study on the direct disposal of the PWR and CANDU spent fuel. Recently, the research on the demonstration and the efficiency analyses of the disposal system has been performed to make the disposal system safer and more economic. PWR spent fuels which include a lot of reusable material can be considered being recycled and a study on the disposal of HLW from this recycling process is being performed. CANDU spent fuels are considered being disposed of directly in deep geological formation, since they have little reusable material. In this study, based on the Korean Reference spent fuel disposal System (KRS) which was to dispose of both PWR type and CANDU type, the more effective CANDU spent fuel disposal systems were developed. To do this, the disposal canister for CANDU spent fuels was modified to hold the storage basket for 60 bundles which is used in nuclear power plant. With these modified disposal canister concepts, the disposal concepts to meet the thermal requirement that the temperature of the buffer materials should not be over $100^{\circ}C$ were developed. These disposal concepts were reviewed and analyzed in terms of disposal effective factors which were thermal effectiveness, U-density, disposal area, excavation volume, material volume etc. and the most effective concept was proposed. The results of this study will be used in the development of various wastes disposal system together with the HLW wastes from the PWR spent fuel recycling process.

Effects of Gamma Irradiation and Ethylene Oxide Fumigation for the Quality Preservation of Spices and Dry Vegetables (건조향신 조미식품의 품질보존을 위한 효과적인 살균방법에 관한 비교연구)

  • Gamma irradiation as a new physical treatment was applied to comparative investigates with a conventional ethylene oxide fumigant on the microbiological and physicochemical qualities of selected spices and dry vegetables such as powdered red pepper, black pepper, welsh onion, onion, garlic, carrot, korean cabbage and instant ramyon soup. The microorganisms contaminated in the sample, including total viable count, thermophilic bacteria, aerobic spore and fungi counts between the $10^4\;to\;10^6/g$ range. Coliforms were found only in black pepper and welsh onion powder as the $10^2\;to\;10^3/g$ level. A radiation dose of 7 to 10 KGy were sterilized completely to the contaminated microorganisms, while ehthylene oxide (E.O.) fumigation reduced of them to the $10^3/g$ level. An optimum dose of irradiation was less detrimental than E.O. fumigation to the physicochemical properties of the sample. Sensory evaluation after three months of storage at room temperatures showed that the overall acceptability of irradiated sample was higher than that of the non treated control as well as E.O. fumigated samples. Comparison gamma irradiation with E.O. gas treatment showed that E.O. treatment was less effective than radiation in cotrolling microbial contamination of spices and vegetables.

Extending the Shelf-life of Yukwa Using Secondary Packaging (이차포장을 통한 유과의 저장성 연장)

  • This study aimed at improving the packaging technology of Yukwa to improve the quality and extend the shelflife using secondary packaging. After packaging the Yukwa using an OPP film, P2, P3, and P4 packaging materials were applied secondarily. Various films including (1) P1: OPP (oriented polypropylene), (2) P2: P1+OPP/LLDPE (linear low density polyethylene), (3) P3: P1+PET (polyethylene terephthalate)/NY (nylon)/CPP (cast polypropylene) and (4) P4: P1+PET/AL (aluminum)/NY/CPP (P4) were used for packaging Yukwa. The experiment was conducted at $25^{\circ}C$ for 12 weeks. P1 showed the highest acid value score (1.26 mg KOH/g), and P3 had the highest peroxide value score (32.91 meq/kg) among all packaging groups. Nevertheless, these values did not exceed the guideline values of 2.0 g KOH/g and 40 meq/kg specified in the Korean food code. The overall color difference showed a tendency for decreasing Hunter 'L' values and increasing 'a' and 'b' values; however, no noticeable difference in the outer appearance was observed in any of the packaging treatments except in the P1 for greater than 10 weeks of storage. Some texture defects were observed in the Yukwa when the moisture contents dropped below 5%. The P4 packaging treatment had the lowest moisture permeability and showed the least rheological deterioration change, followed by P3 and P2. In conclusion, the use of a secondary packaging with less gas and moisture permeability was more effective for maintaining the quality and extending the shelf-life of Yukwa than other types of packaging material.

Studies on Stability for the Quality of Ginseng Products -3. Determination of Sorption Properties and Optimum Moisture Contents in Extract, Extract Powder and Extract Tea of Red Ginseng- (인삼제품(人蔘製品)의 품질안정성(品質安定性)에 관한 연구(硏究) -3. 홍삼정(紅蔘精), 정분(精粉) 및 정차(精茶)의 흡습특성(吸濕特性)과 적정수분함량(適正水分含量) 기준설정(基準設定)-)

  • To investigate stability for the quality of selected ginseng products, their sorption properties were clarified in red ginseng extract(RGEP), and red ginseng extract powder(RGEP), and red ginseng extract tea(RGET). Simultaneously, the BET monolayer value of each product was determined in order to inquire out the possibility of establishment as a criterion for the optimum moisture content of the ginseng products. Based on the BET monolayer moisture level of spray dried RGEP which ranged from 4.08 to 4.65%, it would be desirable to establish the optimum moisture content of the products at 4.4${\pm}$0.3%. This is 1.3 to 1.9% lower than the criter on, "less than 6.0%". The optimum moisture level for RGET of which monolayer value ranged 0.93 to 1.37% would be 1.2${\pm}$0.17%. In this case, the maximum permissible limit of moisture content could presumably be raised up to 1.37% in place of current criterion, "less than l.2%". From the results of a study on the growth of molds, the optimum moisture content for RGE assumed to be extended up to 40.0${\pm}$1.07 despite 36.0${\pm}$1.0% of the present criterion. On the other hand, a storage study under the maltreated condition, $48{\pm}2^{\circ}C$ 75%RH, was also carried out in order to make it clear whether the BET monolayer values were able to be used as indices for optimum moisture level of the products. In all samples tested adsorption occurred at even higher levels of moisture than the monolayer values. However, since there are many other possible factors affecting the quality of products the optimum moisture content is preferable to be reduced to the monolayer value. As a result, it was proved that the optimum level of moisture for both RGEP and RGET could be established by the monolayer values.
