Antioxidant Effect of HotWaterExtract of Muraenesox cinereus's Skin (갯장어껍질 열수 추출물의 항산화 효과)

  • For the development of functional food and cosmetics using hot water extract of Muraenesox cinereus's skin, contents of vitamin, amino acid and element, and antioxidant activity were investigated. The results are shown as follows: among vitamins, A(0.21mg/100 g), C (78.12mg/100 g), $D_3$ (0.03 mg/100 g), E (1.97 mg/100 g) and Niacin (2.53mg/100 g) were detected, respectively.Mineral contents were an order of K > P > Na > Mg > Ca > Fe and Zn. Contents of total amino acids were an order of Pro > Gly > Arg > Glu > Phe > Ala. Especially, the sum of total amino acids was 27.17 mg/100 mL, which was about 4.0 fold higher than that of free amino acid. DPPH radical scavenging activity of hot water extract of M. cinereus's skin at 25 mg/mL was 63.5% and did not increase at above 50 mg/mL. Activities of antioxidant enzymes in the liver of ethanol-treated rats using hot water extract of M. cinereus's skin were investigated. Compared to control group, activities of ADH and GSH-px were decreased. In the case of CAT and SOD activity, they were increased. These results showed that the hot water extract of M. cinereus's skin can be applied to raw macterial for functional food and cosmetics.

  • The trp2 gene encoding trifunctional enzyme in Coprinus cinereus was investigated the expression in the heterothallic mushroom species. To identify the homology of trp2 gene to Schizophyllum commune and Pleurotus ostreatus, southern hybridization was performed with plasmid pHIONA8 containing C. cinereus trp2 gene as a probe, which resulted in a strong signal indicating an homologous sequence to the chromosomal DNA of S. commune. About 50 transformants per dish was appeared in the complementation test by pHIONA8 using S. commune tryptophan auxotroph as host. In the mating test between transformants and other mating type alleles, the fruiting body of S. commune was formed at $30^{\circ}C$ in $2{\sim}3$ weeks.

Phylogenetic Relationships of Coprinoid Taxa and an Agaric-like Gastroid Taxon Based on the Sequences of Internal Transcribed Spacer (ITS) Regions (ITS 영역의 염기서열을 이용한 먹물버섯류 및 주름버섯 유사 복균류와의 계통학적 유연관계)

  • The phylogenetic relationships of black-spored basidioid taxa (mainly coprinoid taxa) and an agaric-like gastroid taxon were studied. The sequences of internal transcribed spacer regions (ITS) partially including 17S, 25S and 5.8S from 14 species (Coprinus comatus, C. atramentarius, C. micaceus, C. cinereus, C. disseminatus, C. rhizophorus, C. radians, C. echinosporus, Psathyrella candolleana, Podaxis pistillaris, Conocybe lactea, Bolbitius demangei, Agaricus balzei, and Stropharia rugosoannulata) were compared. The reciprocal homologies of ITS sequences among these species were in the range of $38.7{\sim}77.2%$. Black-spored taxa were classified into four clusters. Cluster I comprised C. micaceus, C. radians, and C. disseminata. Cluster II is consisted of C. cinereus, C. echinosporus, C. rhizophorus, and C. atramentarius. On the other hand, C. comatus is in cluster III with Agaricus balzei and Podaxis pistillaris even though this species is belonging to the section Coprinus in morphological aspect. Psathyrelloid taxon is included in cluster II. The question of the origin of secotioid (agaric-like) fungi has been taken, though largely on theoritical suggestions as to whether gastroid taxa give rise to agarics with secotioid taxa as intermediates or whether secotioid taxa are evolutionary novelities arising from many distinct groups of agarics. In this relationship, it was shown that secotioid taxon evolved from within agaric species.

  • The internal transcribed spacer II regions (ITS II) of the ribosomal DNA gene repeat from Coprinus spp. were amplified using polymerase chain reaction (PCR) and sequenced. Sequences from 11 species including Coprinus comatus, C. atramentarius, C. micaceus, C. lagopus, C. cinereus, C. rhizophorus, C. flocculosus, C. radians, and C. echinosporus were compared. The spacer regions of them were $253{\sim}275$ nucleotide in length and partially contained 5.8S and 25S. The reciprocal homologies of each ITS II sequence among these strains were in the range of $50.6{\sim}100%$. According to the analysis of ITS II sequences, Coprinus spp. were classified into three clusters. Cluster I consisted of Coprinus lagopus, C. cinereus, C. echinosporus, C. rhizophorus, C. niveus, and C. atramentarius. Cluster II comprised C. micaceus, C. flocculosus, C. radians, and C. disseminatus. On the other hand C. comatus is in Cluster III even though this species is belonging to the section Coprinus in morphological aspect. These results suggest that Coprinus comatus, which was considered as a type species of the genus Coprinus in morphological classification, gives a doubt of monophyletic evolution and is assumed to be paraphyletic or polyphyletic.

Seasonal Variation of Lipids and Fatty Acids of Sharp Toothed Eel(Muraenesox cinereus) (갯장어의 일반성분과 지방산의 계절적 변화)

  • Seasonal variation of uraenesox cinereus) muscle was investigated. Crude lipid content varied from 3.85 to 12.59 g/100g, comprising the highest content in November. The major fatty acids of total lipid, neutral lipid, and phospholipid were C16:0, C23:0, C16:1, C18:1, C20:5, and C22:6, but in phospholipid, Cl8:3n-6 was also the major fatty acid. The C22:6 content of the neutral lipid was much lower compared to that of the total lipid and phospholipid. The content of polyunsaturated fatty acids (ranged from 73.93 to 66.23%) in phospholipid was higher than that of any other lipid fraction. In glycolipid, C20:1 and C14:1 were higher compared to those of any other lipid fraction, but C20:5 and C22:6 were lower. The annual average ratio of n-3 to n-6 of total lipid, neutral lipid, phospholipid, and glycolipid was 10.82, 12.27, 6.63, and 6.50, respectively. The particular trend of seasonal variation of fatty acid composition was not showed in total and neutral lipid. However, the samples caught in September and November had a high crude lipid content with a significantly lower content of polyunsaturated fatty acids in phospholipid. Also, the content of monounsaturated fatty acids in glycolipid was lower in samples of September, November, and January.

Cluster II comprised C. micaceus, C. radians, C. disseminatus, Psathyrella candolleana, and P. spadiceogrisea. On the other hand C. comatus is in Cluster III with Stropharia rugosoannulata even though this species is belonging to the section Coprinus in morphological aspect. These results suggest that taxonomic position of Psathyrella would better be inculded in genus Coprinus. Coprinus comatus, the type species of Coprinus, gives a doubt on monophyletic evolution and is assumed to be paraphyletic or polyphyletic.

  • Coprinus cinereus 2249 that is a kind of basidiomycetes constitutively produced alkaline carboxymethyl cellulase (CMCase), filter paper cellulase (FPase) and xylanase. Crude enzymes prepared with optimal conditions showed higher FPase activity than CMCase activity. The FPase was most active at pH 9 at 50$^{\circ}C$. When applied on deinking of the old newsprint (ONP), it increases the freeness and brightness due to effect of hydrolysis at 0.1% enzyme concentration. Also, The physical properties of deinked pulp were improved.

Seasonal Variation in Abundance and Species Composition of Fishes Collected by a Beam Trawl around Naro-do, Korea (남해 나로도 주변해역에서 조망에 채집된 어류의 종조성과 계절변동)

  • Seasonal variations in abundance and species composition of fish assemblage around Narodo were examined. Fish samples were monthly collected by a beam trawl from December 2000 to November 2001. A total of 62 fish species representing 37 families were collected. Of these, Thryssa kammalensis, T. hamiltoni, Chaeturichthys hexanema, Johnius grypotus, Leiognathus nuchalis, Cynoglossus robustus and Setipinna taty were dominant species comprising $82\%$ of the total number of individuals. Species diversity indices ranged from 1.2 to 2.2 showing the highest values from September to November 2001. The fish assemblage was divided into three different groups by cluster analysis of the fourteen dominant species. The main residence species were J. grypotus, L. nuchalis, C. hexanema, C. robustus, Setipinna tenuifilis, T. kammalensis and Ctenotrypauchen microcephalus. Of the temporal species T. hamiltoni, Liparis tanakai and Erisphex pottii were collected In spring, and Muraenesox cinereus and Trichiurus lepturus were in summer and autumn. Principal component analysis revealed that seasonal variation in the fish assemblage was attributed to the abundance of temporal species due to seasonal variation of environmental factors such as water temperature and ecological pattern (polagic/demersal) of fish species.

Changes in Available Lysine and Lipid Oxidized Products during the Storage of Dried Sea Eel, Muraesox cinereus (건조(乾燥) 갯장어의 저장중(貯藏中) 지방(脂肪)의 산화(酸化)와 유효성(有效性) Lysine의 변화(變化))

  • Lipid oxidation is one of the major factors affecting on deterioration of nutritional quality in dried fish products. In this paper, the relationship between oxidized products of lipid and brown pigments, free amino acids and available lysine during the storage of dried sea eel, Muraesox cinereus, was investigated. And the inhibiting effect of antioxidant to lipid oxidation and its role to the protein quality were also discussed. From the results, TBA and carbonyl value rapidly increased while amino-N and available lysine diminished during hot air drying. This suggests that drying conditions greatly affected to the oxidation of lipid and making amino acids 'unavailable'. TBA value increased up to 20 days, and hereafter gradually diminished. Increase in TBA and carbonyl value and formation of fat oxidative brown pigment were closely related to the loss of free amino-N and available lysine. The loss of available lysine seemed to be affected by the formation of unsaturated carbonyl compounds rather than saturated carbonyl compounds. By the treatment of antioxidant, the loss of amino acids and available lysine was somewhat retarded. This may suggests that the oxidation of lipid or oxidative browning reactions are functioning to the loss of available lysine. In antioxidant treated sample, 23% of amino-N to the total amino-N in the fresh sample was lost after 20 days storage at $30^{\circ}C$ while the loss of amino-N to 39% in case of the control, and afterward the value treated to be slightly reduced or remained steady.

Production and Characteristics of Protein Hydrolysate from Sharp Toothed Eel (Muraenesox cinereus) (갯장어 효소 가수분해물의 제조 및 특성)

  • Protein hydrolysate was prepared as a natural flavor stock from the sharp toothed eel (Muraenesox cinereus) mince using com-mercially available proteolytic enzymes, Alcalase, Neutrase, Protamex, and Flavourzyme. A 6 hr hydrolysis of mince, to which water of the equal weight of the mince was added, with $2\%$ (w/w, protein weight) Elavourzyme at $50^{\circ}$ yielded a hydrolysate of the highest acceptability. Removing the access lipid in liquified hydrolysate (not dehydrated) after enzyme hydrolysis, five times repetitive extraction using n-hexane (liquified hydrolysate : n-hexane=4 : 1, v/v) was effective, resulting in less than $1\%$ lipid content of the dehydrated-hydrolysate. The amino acid composition of the hydrolysate (prepared with Flavourzyme) was similar to that of the starting material. Hydrolysis led to an increase in concentration of not only total free amino acid but also free amino acid such as serine, glutamic acid, alanine, and methionine responsible for umami taste, especially up to about 40 times for methionine. Major free amino acids in amount were leucine, phenylalanine, valine, alanine, and isoleucine and they comprised about half of the total free amino acids, Moisture adsorption, fat adsorption, emulsifying capacity, and foaming capacity of the hydrolysate were 870.1 $\pm$ $7.9\%$, 352.0$\pm$ $5.3\%$, 50.3 $\pm$ $1.2\%$, and $87.5\pm$ $2.5\%$, respectively, and solubility was 83$\~$$84\%$ at acid pH range of 2$\~$4.
